Abstract

Processing fish with the fermentation method can increase the nutritional content, bring out a distinctive taste, and make the fish last longer. Rusip is a type of complementary food from Bangka Belitung in the form of fish sauce that has been rotten or fermented. Rusip is made from fermented anchovy (Clupeoides borneensis). The concept of fermentation in Rusip is the breakdown of complex protein molecules by proteolytic bacteria into simple amino acid components. The purpose of this study was to determine the presence of proteolytic bacteria that play a role in rusip fermentation. The media used in screening proteolytic bacteria is skim milk agar (SMA) media. Then it was incubated at 37oCfor 24 hours. Bacterial colonies that form a clear zone on the media indicate that these bacteria can degrade protein. After the isolation process and isolates were obtained, the bacteria were characterized by Gram staining method and molecular identification. The results showed that there were proteolytic bacteria in Rusip food, and the 3 best isolates that produced protease enzymes were identified as Bacillus cereus.

Abstract

Lactic acid bacteria (LAB) play an important role in robusta coffee fermentation by soaking coffee beans. Spontaneous fermentation of coffee beans can improve coffee quality by producing organic acids and volatile compounds. The ability of LAB to produce organic acids also has the potential as an antifungal compound. Penicillium is one of the predominant contaminants of coffee beans products in Indonesia. Coffee beans can become contaminated by Penicillium during various stages such as harvesting, processing, transport, and storage. To prevent contamination of coffee beans, new preservation methods involving biological agents, such as lactic acid bacteria, may be used instead of chemicals. The secondary metabolites produced by LAB can be utilized for bio preservation. Lactic acid bacteria in robusta coffee fermentation should be further explored and analyzed for antifungal activity against Penicillium sp., and molecularly identified to determine the spesies of lactic acid bacteria that have antifungal potential. The isolation method was carried out using multilevel dilution and spread plate on MRSA+CaCO3 media. Identification was conducted through macroscopic and microscopic observation of cells and their characteristics, as well as molecular identification. Antifungal testing was performed using the agar well diffusion method. The results of LAB isolation yielded 12 LAB isolates with characteristics of round colony shape, gram-positive, rod and round cell shape, with negative catalase. The test results showed that isolates BKR 4, BKR 11, BKR 12 exhibited antifungal activity. BKR11 isolate was identified as Leuconostoc mesenteroides.

Abstract

Aquatic mammals in Indonesia are officially protected. However, there is a lack of research on these mammals, particularly in relation to potential disease-causing bacteria. A study was conducted in a conservation pond to address this gap, where swab samples were taken from the blowholes of aquatic mammals. The focus was on identifying bacteria that could potentially cause infectious diseases in these animals. The results revealed V. parahaemolyticus bacterial isolates, which showed a 98% similarity to the registered V. parahaemolyticus in NCBI. These bacterial isolates exhibited hemolysin properties and demonstrated resistance to trimethoprim, streptomycin, cephalothin, and penicillin antibiotics.

Abstract

Burns are skin tissue damage that occurs as a result of direct contact with a heat source. One of the natural treatments for burns is watermelon (Citrullus lanatus) rind. Watermelon rind contains citrulline, an alkaloid that plays a role in wound healing. This study aims to explore the potential of watermelon rind to become extract gel as a burn remover in male Balb/C strain mice. This research method is a true experiment that begins with the preparation of watermelon rind extract by maceration with ethanol. Furthermore, the polyphenol, flavonoid, saponin, and alkaloid content of the extract was identified through the phytochemical screening. The next stage was the preparation of a gel with three variations of extract concentrations, namely 25%, 50%, and 75% w/v. The gels were characterized by organoleptic, pH, and spreadability. The final stage tested the gel’s activity as a burn wound healing in male Balb/C strain mice. The results showed that the extract contained saponins, polyphenols, flavonoids, and alkaloids. The spreadability test results of the gel showed that all gels met the requirements. All gels had activity against burns in male Balb/C strain mice. Thus, watermelon rind extract can be used as a medicine for burns.
