Abstract

Penelitian ini dilakukan untuk mengetahui ragam alel masyarakat Dayak Kaharingan di Kota Palangka Raya.  DNA diekstraksi dari sel epitel mukosa mulut, dari 26 individu dengan metode fenol kloroform. DNA mikrosatelitautosom lokus D2S1338, D13S317 dan D16S539 diamplifikasi pada mesin PCR. Pengamatan hasil PCR dilakukan dengan Polyacrylamide Gel Electrophoresis (PAGE) dan visualisasi DNA hasil PCR dengan pewarnaan perak nitrat.  Hasil penelitian ini menunjukkan terdapat 29 alel dari ketiga lokus yaitu lokus D2S1338 sebanyak 11 alel, serta masing-masing sembilan alel pada lokus D13S317 dan lokus D16S539. Nilai heterozigositas tertinggi terdapat pada lokus D2S1338 yaitu 0,8971 dengan kekuatan pembeda (PD) 0,9682, diikuti lokus D13S317 dengan kekuatan pembeda 0,9339 dan lokus D16S539 dengan kekuatan pembeda 0,9226.

Abstract

Penelitian ini bertujuan untuk mengetahui keragaman genetik ikan cakalang (Katsuwonus pelamis) yang ada di Kabupaten Jembrana dan Karangasem, Bali. Sampel DNA diambil dari 30 ekor di kabupaten Jembrana dan 30 ekor di kabupaten Karangasem. Ekstraksi DNA dilakukan dengan menggunakan chelex 10% dan amplifikasi menggunakan metode PCR dengan konfigurasi HotStart pada control region DNA mitokondria, menggunakan primer forward CRK dan primer reverse CRE. Analisis filogenetika dilakukan dengan menggunakan metode Neighbor Joining dengan model Kimura 2-parameter. Variasi dari setiap sekuen diperoleh dengan menghitung jumlah, persentase dari tiap haplotipe, keragaman haplotipe (hd) dan keragaman nukleotida (?) menggunakan program DnaSP 5.10. Analisis Fst (Fixation Index) berdasarkan frekuensi haplotipe menggunakan Arlequin 3.1. Haplotipe pada sampel Jembrana berjumlah 28 haplotipe dan sampel Karangasem berjumlah 15 haplotipe dengannilai hd secara keseluruhan yaitu 0,997±0,006 dan untuk keragaman nukleotida (?) sebesar 0,04694. Nilai Fst yang diperoleh yaitu -0,00334 dan Fst P sebesar 0,90918±0,0100, menunjukkan bahwa populasi ikan cakalang di Kabupaten Jembrana dan Karangasem berasal dari populasi induk yang sama.

Abstract

The Balinese clans embraced patrilineal system.They divided into groups of soroh or clans which means respect for ancestors. Pandeclan is a unique soroh which comes from ancestors who have expertise in memande. The genetic identityof this clan in Bali is unknown.Therefore the aim of this study is to find out the genetic identity from the varieties of allelic diversity which found on Pande community inside and outside Seririt using microsatellite DNA analysis of Y chromosome, namely DYS19, DYS390, DYS393, and DYS395. Resultsof this study have foundthat in Pande clan there are 21 varieties of allelic diversity and 25 haplotypes with the average heterozygosity of four loci is 0.72 ± 0.03. The dendogram indicates that Pande clanfrom district of Seririt did not cluster based on it’s dadia.

Abstract

Smoking habits have been around since ancient times, but nowadays this habit is considered to be detrimental, especially to health. The impact that is often felt by smokers is difficulty in breathing because the lungs are exposed to cigarette smoke. Cigarette smoke contains about 1015-1017 oxidants or free radicals, as well as 4700 harmful chemicals, including aldehydes / carbonyls, NO2, and SO2. Herbal cigarettes are tobacco cigarettes with added ingredients from plants. Gurah terapi sin cigarettes are herbal cigarettes that are sold commercially. The aim of this study was to determine the effect of gurah cigarette smoking on the leukocytes and lung histology of mice. This study used a comparative method consisting of 3 groups, namely the control was not exposed to cigarette smoke, treatment 1 was exposed to commercial cigarette smoke and treatment 2 was exposed to cigarette smoke with herbal ingredients and each group consisted of 10 replications. The results showed that there were significant differences (p <0.05) regarding the number of cell necrosis, type II pneumocytes, inflammatory cell infiltration, hemorrhage, and alveolar dilation. While the results of the analysis of the number of leukocytes showed no significant difference where p > 0.05. The results showed that there was no significant difference in the number of leukocytes in the control group, treatment 1 and treatment 2 (p > 0.05). herbs containing various kinds of antioxidants cause a tendency for differences in the number of leukocytes where there is a decrease in the number of lymphocytes and neutrophils and an improvement in the histopathological structure of the lung against type I pneumocyte cell necrosis, hemorrhage, alveolar dilation, type II pneumocyte cell proliferation, and inflammatory cell infiltration in exposed mice. commercial cigarette smoke without herbal ingredients.

Abstract

This research aims to determine the effect red calliandra leaf extract on weight, anatomy andhistology of male mice liver. Treatment was administered orally with varying doses (2, 4, 6mg/ kg). Control grups was given 0,9% NaCl. This study used a completely randomizeddesign (CRD), consisting of 12 groups with three replications. Histological observationsshowed that red calliandra leaf extract significantly affected the damage of male mice liver.Damage level of nucleus pyknotic and fatty degeneration increased in line with the increasingdoses in treatment for 7 and 21 days, as well as the dilation of central venous in treatment for7 days. The damage increase in line with the leght of treatment in the fatty degeneration (dose2 mg/kg bw), nuclues pyknotic (doses 2 and 6 mg/kg bw), infiltration of inflammatory cells(dose 4 mg/kg bw), sinusoid congestion (doses 2, 4 and 6 mg/kg bw), and central venousdilatation (dose 2 and 4 mg/ kg bw).Keywords : Red calliandra, male mice, liver histology

Abstract

The presence of spermatozoa DNA is reliably counted as a solid evidence when dealing with rape cases. However, police department often finds difficulties to obtain it, mainly because it has been destroyed by the perpetrator. Such act can be done by washing semen stains on the clothing with detergent. The purpose of this research is to analyze the impact of detergent as a washing property towards spermatozoa DNA extract on a cotton fabric. Observed variables in this study are quantity and quality of spermatozoa DNA extract from semen stain on cotton fabric. Samples are divided based on two separate treatments; detergent-washed and unwashed. Furthermore, samples are stored with different storage time. This research applied two methods; DNA extraction using kit (Roche) and DNA quantification using spectrophotometer Nanodrop. The research was held at Biomedic Laboratory, Faculty of Medicine, Udayana University, Denpasar with ethical clearance. Seminal plasma sampel was obtained from a donor with informed consent. This research showed that seminal stain washing on cotton fabric doesn't remove the DNA until two-week-storage time.

Abstract

Infection is a major problem mortality and morbidity in the world, a lot of an infection in the digestive tract is causes bacteria. The Escherichia coli O157 and Escherichia coli O157: H7 strains are bacteria that cause bloody diarrhea to death. Ultraviolet light has the potential to eliminate bacteria. The purpose is to know decontamination of E. coli O157 and E. coli O157: H7 on meat after ultraviolet exposure. The method is a factorial RBD with two factors (distance and time) with variations in distances (5, 10, 15, and 20 cm) and times (5, 10, 15, 20 dan 25 minutes). All treatments were repeated twice and obtained 160 samples. The quantitative data obtained were using univariant analysis followed by the Least Significant Difference with a significant level of 5%. The irradiation for E. coli O157 on meat with 5 cm for 25 minutes showed a significant reduction (88,62%), while the farthest distance 20 cm for 25 minutes decreased (73,42%). The nearest irradiation E. coli O157: H7 on meat 5 cm for 25 minutes show decrease (86,78%), while the farthest distance was 20 cm for 25 minutes (75,16%), there was an increase colonies in several treatment variations but overall decreased. In conclusion there was significant decrease number of bacterial colonies in meat given E. coli O157 culture, while there was a fluctuation decrease number of colonies in meats given E. coli O157: H7 culture.

Abstract

Abstract Objective: P-glycoprotein (P-gp) overexpression on neoplastic cells can deteriorate the therapeutic outcome on cancer patients. P-gp plays important role on drug efficacy and toxicity. This research aimed to measure P-gp expression on children with Acute Lymphoblastic Leukemia (ALL) on Sanglah Hospital, Denpasar. Method: Flowcytometry method was used to measure P-gp expression level on Bone Marrow samples from pediatric patients (0-12 years old) who were newly diagnosed with ALL in Sanglah Hospital. P-gp overexpression were based on the percentage of cell stained. Ten percent of P-gp expression were considered as the cut-off value of P-gp overexpression. Result: On this study, 11 samples were obtained with the range value of 56-97% on P-gp expression. Conclusion: All 11 patients had P-gp overexpression.

Abstract

Advances in molecular biology has brought a new branch of multidiscipline study named biotechnology. To comprehend the complex nature of concepts in biology, practical and hands-on approaches are often required. This activity was a collaboration between Biology Teachers Working Group (MGMP Biologi) in Klungkung Regency with university as the facilitator, and aimed to train senior high school biology teachers in compiling learning modules as well as supplying them with the appropriate materials required in simple practical sessions, such as those in animal physiology and reproduction, as well as biotechnology. The event was held successfully, attended by total of 20 biology teachers from six senior high schools (SMA) in Klungkung District, Klungkung Regency, started by introductory lectures and followed by practical sessions on how to perform mice vaginal and sperm smear, human blood smear, and DNA extraction of plant tissues. Abstract

Sodium nitrite (NaNO2) is a chemical compound that is often used in food industry. NaNO2 widely used for food preservatives, mainly in processed meat products. However, NaNO2 could cause anemia by interfering with the process of transporting oxygen in the blood. The effects caused by NaNO2 could be repaired with the use of vitamin C. This study was aimed to determine the effect of vitamin C on the blood profile of rats induced by NaNO2, and the effective dose to improve the blood profile of rats. The study used a completely randomized design (CRD) with 4 groups, namely K- (Aquades); K+ (NaNO2 37,5mg/kgBW); C1 (NaNO2 37,5 mg/kgBW + vitamin C 1,8 mg/animal/day); C2 (NaNO2 37,5 mg/kgBW + vitamin C 3,6 mg/animal/day). The samples used in each treatment group were 6 rats, therefore the total sample used was 24 male white rats (Rattus norvegicus) strain Wistar. NaNO2 and vitamin C solutions were administered orally by the gavage method. NaNO2 solution was given until 35th day, then vitamin C solution was given on 22nd to 35th day. Blood samples were taken three times, the day before the administration of NaNO2, on the 21st day, and on the 36th day, further analysis of blood used a haematology analyzer and blood smears preparations Giemsa staining. Data were statistical analyzed by ANOVA and continued with Duncan's test. The results showed that Vitamin C can improve the blood profile, with the most effective dose of vitamin C is 1,8 mg/animal/day.
