Abstract

The objective of the research was to determine the effect of fermentation time and the optimal fermentation time producing the highest alcohol content of the product. This study used a randomized block design with fermentation time as a treatment which was consists of 11 levels namely 0, 1, 2, 3, 4, 5, 6, 7, 8, 9, and 10 days. The results showed that the naturally anaerobic fermentation time did not affect the alcohol content, pH, total dissolved solids of the alkohol product. The treatment of natural fermentation for 5 days was an appropriate treatment to produce alcohol with the characteristics of the alcohol content of 2.05%(w/w), acidity (pH) 3.41, total dissolved solids 5,30 brix, 0.216% total acid. Abstract

The purpose of this study was to determine the concentration and size of Na-alginate beads that have the highest degradation activity of dibenzothiophene. Biodesulfurization (BDS) of dibenzothiophene (DBT) was performed using 3 Na-alginate concentrations and 3 different beads sizes in the oil model system. Biodesulfurization was performed with incubation for 24 hours. The previous research showed that sodium alginate (Na-alginate) was an appropriate immobilizing agent compared to other immobilized materials. Na-alginate 4% show the activity of the degradation of the most good that is 66.33% (bead size 2 mm), 62.99% (bead size 3 mm), 59.93% (bead size 4 mm), for concentration of 3% Na-alginate showed 65.58% (bead size 2 mm), 61.68% (bead size 3 mm) and 60.43% (bead size 4 mm), while concentration 5% showed the most low that is 64.86% (bead size 2 mm), 61.01% (bead size 3 mm), and 58.89% (bead size 4 mm). The stability test showed Na-alginate 4% have the stability and durability of the bead stronger, the test showed Na-alginate can be used up to five repeat and still have degradation activity. Abstract

After being heated at 100o Celcius, the bacteriocins from the pickled LAB of tabah bamboo shoots hypothetically predicted to be able to inhibit the growth of pathogenic bacteria, especially S. aureus ATCC 25923 and E. coli ATCC 8739. The LAB isolates thus were heated at a temperature of 100o C for various of 5, 10, 15 minutes heating time and subsequently measured by regression and correlation analysis of its influence towards the inhibition of the growth of S. aureus ATCC 25923 and E. coli ATCC 8739 bacterias. This study shows the variations of heating time at a temperature of 100°C resulted with of inhibition zone’s diameter formation (mm), activity values (mm2/ml), and the effects (r dan R2) of these two bacteriocins towards its inhibition on the bacterias. Abstract

The aim of this study was to find out initial infusion temperature and infusion time effects on of antibacterial and antioxidant activities the Robusta Coffee leaves herbal drink extract. This study also determined the best initial infusion temperature and infusion time that can produce Robusta Coffee leaves herbal drink extract with the best antibacterial and antioxidant activities. The study used factorial completely randomized design. The first factor is infusion temperature, consisting of 70±2°C, 80±2°C, and 90±2°C. The second factor is infusion time which consists of 5, 10, and 15 minutes. The data was analyzed by analysis of variance (ANOVA) and continued with Tukey test. The results showed that the treatment of initial infusion temperature, infusion time, and interaction between the two treatments had a very significant effect on antibacterial activity Abritrary Unit (AU) against Salmonella typhimurium and antioxidant activity Inhibitory Concentration 50 (IC50). The treatment of infusion time had a significant effect on antibacterial activity AU against Escherichia coli, but the initial infusion temperature and interaction between the two treatments had a very significant effect on it. The best treatment to infuse herbal drink of Robusta Coffee leaves as a source of antibacterial and antioxidant is using initial infusion temperature of 90±2°C and infusion time of 5 minutes with antibacterial activity AU against Escherichia coli of 1.248,34 ± 132,58 mm2/mL, antibacterial activity AU against Salmonella typhimurium of 1.473,37 ± 79,57 mm2/mL, and antioxidant activity IC50 of 185,01 ± 1,66 ppm. Abstract

This research aimed to determine the ability of each bacterial isolate to produce ethanol, isolate and characterize bacteria that have the ability to produce potential ethanol from lau and air in the Arak Industry of environment in the Tri Eka Buana Village, Sidemen District, Karangasem Bali. Samples were obtained from 3 Hamlets in Tri Eka Buana Village. Bacteria were taken from lau before fermentation, during fermentation, and after fermentation. Bacteria from air were taken from the fermentation, distillation, and under the coconut tree and the variation of time is 15, 30, and 60 minutes of exposure. Bacteria from lau were isolated by growing on growth media, periodically (after 30, 60, 90, 120, and 150 seconds) the isolates were taken from suspension to be inoculated in growth media for gas production. Isolation was carried out using YEPD-fluconazole media to obtain pure isolates. The fluconazole concentration used was 0,2 g/L. Pure isolates were then carried out qualitative tests for gas production and quantitative tests with alcohol oxidation reactions. The results of UV-Visible spectrophotometry, there were 6 best isolates and then fermented in YEPD media on a scale of 900 mL for 10 days. The fermentation results were then distilled using a reflux distilator. BU3.111E1 isolates are determind the best isolates with 8,50% brix ethanol. The BU3.111E1 were obtained from air bacteria in Pungutan Hamlet, place of sampling during the fermentation process for 15 minutes. From the characterization results, isolate BU3.111E1 was Gram positive bacteria, basil bacteria, motile bacteria, and catalase negative. Abstract

Dregs of solid waste brem industry contained 12 percent of sugar and 10,8 percent of starch. Component contained in the dregs are expected to produce citric acid. The aims of this study were to determine the effect of static cultivation on the production of citric acid from liquid hydrolyzate brem waste by using Aspergillus niger. The experimental design used in this study was a simple randomized block design (RAK), which the static cultivations (0, 1, 2, 3 days) as a treatment. The results showed that the static cultivation treatment significantly affected the level of acidity (pH), total soluble solids, biomass, reducing sugar, calcium citrate, citric acid and yield. The static cultivation for 1 day was the best treatment to produce Ca-citrate which the production level was 3,84 ± 0,05 g/L. By the stoichiometry method the citric acid production could be calculated, which equal to 2,12 ± 0,03 g/L. Keywords: Aspergillus niger, dregs of brem, static cultivation, Ca-citrate, citric acid

Abstract

This study was conducted to observe the potential aphrodisiac activity of Gigantochloa nigrociliata in male mouse Mus musculus. The twenty four male mice, aged 12 weeks were divided randomly into four groups, each group consisted of 6 mice. One group was used as control (P0) where the mice were treated with aquadest. Three other groups were given treatment. The extracts contained of water (P1), etanol (P2), and n-hexane (P3). The animals in each treatment were treatments given 200 mg/kg bw Gigantochloa nigrociliata of extract orally (gavage) once daily as much as 0,2 mL for 33 days. The observations of sexual behaviour performed every three days during the treatment. Variables observed for the sexual behaviour are both the number and latency of mount and intromission. Mice were anaesthetized after 33 days. Cauda epidydimis were analyzed for motility, number and viability of sperms. The result showed that testosterone serum level increased significantly on etanol extract group (p<0.05). There were significantly increased the mounting number, intromission number with reduction in mounting latency, and intromission latency of male mice (P<0.05). There was significant an increase in the number of (a) sperm motility and number of sperm on etanol extract group (p<0.05). The results of the present study demonstrate that Gigantochloa nigrociliata extract improve sexual behaviour in male mouse. ABSTRAK Telah dilakukan penelitian mengenai potensi rebung bambu tabah sebagai bahan afrodisiak yang diujikan pada mencit jantan (Mus musculus). Sebanyak 24 mencit dikelompokkan menjadi empat perlakuan dan enam ulangan. Satu kelompok digunakan sebagai kontrol (P0). Tikus pada kelompok kontrol diberikan aquades. Tiga kelompok lainnya diberikan perlakuan ekstrak air bambu tabah (P1), ekstrak etanol bambu tabah (P2), dan ekstrak n-hexane bambu tabah (P3). Setiap hewan pada masing masing perlakuan diberikan ekstrak 200 mg/kg bb (bobot badan) sebanyak 0,2 mL ) secara oral selama 33 hari. Pengamatan perilakukawin berupa mount dan intromission dilakukan setiap 3 hari sekali. Setelah 33 hari hewan dikorbankan untuk dikoleksi epididimis bagian cauda yang digunakan dalam analisis spermatozoa dan pengambilan darah yang digunakan dalam analisis hormon testosteron serum. Hasil analisis dengan menggunakan uji one way Anova menunjukkan adanya perbedaan kadar hormon testosteron serum secara signifikan (p<0,05). Kelompok hewan dengan perlakuan etanol memiliki kadar hormon yang lebih tinggi dibandingkan kelompok lain. Kelompok hewan dengan perlakuan heksan memiliki kadar hormon terendah dengan perbedaan yang signifikan dibandingkan dengan kelompok perlakuan lain. Terjadi pula perbedaan motivasi seksual yang diamati pada perilaku kawin berupa mount dan intromission. Perbedaan jumlah dan waktu terjadinya mount dan intromission terjadi secara signifikan di antara kelompok perlakuan dan kontrol (p<0,05). Peningkatan jumlah mount dan intromission, serta semakin singkatnya waktu untuk mencapai mount dan intromission terjadi secara signifikan dibandingkan dengan kontrol dan diantara kelompok perlakuan (p<0,05). Hasil analisis data juga menunjukkan peningkatan motilitas spermatozoa tipe (a) yakni spermatozoa dengan gerakan yang progresif maju ke depan dan jumlah spermatozoa, terutama pada esktrak etanol rebung secara signifikan. Disimpulkan bahwa rebung bambu berpotensi sebagai bahan afrodisiak karena dapat meningkatkan hormon testosteron yang merupakan salah satu faktor penting dalam pengaturan sistem reproduksi serta dapat meningkatkan motivasi seksual yang diuji pada mencit jantan.

Abstract

ABSTRACT In Vitro Study of the effectiveness of Essential Oil Extracted from Lemongrass (Cymbopogon citratus) to Aspergillus Sp. Study on determining of the effectivenes of essential oil extracted from lemongrass (Cymbopogon citratus) in order to suppressing the growth of fruit rot fungi (Asoergillus sp.) and also to determining the minimum concentration that able to inhibit colony growth and spore formation of the Aspergillus sp. in vitro, was conducted in Laboratory of Plant Diseases from June into November 2012. The result showed that the fungi were isolated from rot fruit mango has similarities with the fungi Aspergillus sp. The characteristic of Aspergillus has cepted hyphae, growth appear colonies, conidiophores uncepted and stand up, the tip of conidiophore is swollen and produce vesicle form, the surface of vesicle is covered by fialid that usually simple and colored or colorless, fialid produced conidia which were form green, brown or black chain. Essential oil of lemongrass have an antifungal activity and it was found effective to inhibit growth of the fruit rot fungi Aspergillus sp. on PDA. The inhibition zone of the lemongrass essential oil against Aspergillus sp. were 85 mm which is include a very strong category. Effective concentration of essential oil of lemongrass to inhibit the growth of Aspergillus sp. are 0,6% - 1%. The persentase of inhibition in this concentration reached to 100%. On the other hand, the control treatment, 0,2%, and 0,4% concentration were 0%, 0%, and 28,53%, respectively. Minimum concentration which is capable to inhibit the formation of spores and growth of fungi colonies were 0,2% and 0,3% concentration. The compounds suspected as antifungal against Aspergillus sp. were ?-citral (geraniol) and ?-citral (neral). Keywords: Lemongrass (Cymbopogon citratus), essential oil, antifungi, Aspergillus sp.

Abstract

ABSTRAK Pemanfaatan Minyak Atsiri Sereh Dapur (Cymbopogon citratus) sebagai Biopestisida untuk Mengendalikan Plutella xylostella (Lepidoptera: Plutellidae) Informasi tentang efikasi dan pitotoksisitas dari minyak atsiri sereh dapur sangat diperlukan dalam memanfaatkan minyaknya untuk mengendalikan Plutella xylostella. Penelitian efikasi minyak sereh di laboratorium sudah dilakukan sebagai penelitian pendahuluan sebelum melakukan penelitian di lapangan. Penelitian lapangan yang bertujuan untuk mengetahui efikasi dan pitotoksisitas minyak sereh dapur dilakukan di Desa Pancasari, Kecamatan Sukasada, Kabupaten Buleleng (1200 m dpl). Hasil penelitian menunjukan, minyak atsiri sereh dapur pada konsentrasi 10 % menimbulkan gejala terbakar pada daun kubis. Sedangkan pada konsentrasi 2.5 % dan 5 % menunjukan gejala perubahan warna dari hijau menjadi ungu. Tetapi pada konsentrasi rendah 1 %, 0.5 %, dan 0.1 % minyak sereh tidak memiliki pengaruh pitotoksik terhadap tanaman kubis. Pengaruh minyak sereh terhadap populasi P. xylostella menunjukan tidak beda nyata pada konsentrasi rendah. Tetapi pada umumnya dapat menekan populasi P. xylostella dibandingkan dengan kontrol. Berdasarkan penelitian tersebut maka minyak sereh dapur dapat dimanfaatkan sebagai pengendali P. xylostella pada konsentrasi rendah yaitu dibawah konsentrasi 1 %, karena pada konsentrasi tersebut minyak sereh tidak bersifat pitotoksik dan mampu menekan pupulasi P. xylostella di laboratorium maupun di lapangan.
