Abstract

The aim of this research was study of Insulin Like Growth -I Complex to level of malondialdehide (MDA) and percentage of intact membrane plasm. Report of several species suggest that seminal plasma contains factors that may influence sperm viability. Seminal plasma was reported to be important for maintaining spermatozoa motility in bull and ram. For improving ram sperm viability and for increasing the resistance of boar spermatozoa to cold shock damage. IGF-I has been identified in the testis, where secreted by Leydig and Sertoli cells. Collection of semen by using artificial vagina from male goats and then characterized of motility and viability of spermatozoa. Identification of Insulin Like Growth Factor –I Complex were done by Native-PAGE 12% techniques, after several bands were identified and was done isolate of first band protein Insulin Like Growth Factor-I Complex. Insulin Like Growth Factor – I (IGF-I) Complex seminal plasma have molecular weight of 150,288 kDa, respectively. Sperm preparation for this research by centrifugation of semen with Bracket and Oliphan’s medium. These sperm preparation were characterized of motility, viability. Then after sperm preparation divided into three groups. The group I, added with Bracket and Oliphan’t medium, group II added with Bracket Oliphan’t and IGF – I Complex medium, group III added with IGF – I Complex medium and then were exposed 45 minutes for incubation and then percentage of MPU and level of MDA was evaluated. The result of this research showed that percentage of level MDA has significant different between group I,group II and group III, (p<0,05). The percentage of MPU sperm has significant different between groups I, group II and group III (p<0,05). The highest of percentage MPU was found in group III and the lower of level MDA was found in group III. Regresion test showed that negative correlation between level of MDA and percentage of MPU sperm.

Abstract

Carbon black is one of the components of particulate matter (PM), known as cytotoxic, causing inflammationin the respiratory and cardiovascular system, and able to pass through the placental barrier, which may affect the fetus. The effects caused by pollutants depend on the intensity and time length of the exposure.The objective of this study is to identify Vascular Endothelial Growth Factor (VEGF) and Mitogen Activating Protein Kinase (MAPK) as an indicator of maternal placental inflammation in pregnant rats (Rattus norvegicus) exposed to carbon black with immunohistochemical technique. This study was a laboratory experiment with three treatment groups and seven replications using pregnant female white rats (Rattus norvegicus). Pregnant female rats were exposed to carbon black with a dosage of 523 mg/m3 and 1064 mg/m3 during pregnancy of day 6 up to day 11. The next phase was the observation and examination of the expressions of VEGF and MAPK by immunohistochemical staining toward placenta of the white rats exposed to carbon black. The result of the study showed an increase in the expression of VEGF in the placenta of the rats exposed to carbon black with a dosage of 523 mg/m3 and 1064 mg/m3 per inhale during the mid-period (6-11) of pregnancy. In addition, there was also an increasein the expression of MAPK on the placenta of the rats exposed to carbon black with a dosage of 523 mg/m3and 1064 mg/m3 per inhale during the mid-period (day 6-11) of pregnancy. In conclusion, the expression of VEGF and MAPK are increased along with the increased dosage of carbon black and age of gestation. ABSTRAK Carbon black merupakan salah satu komponen partikulat matter (PM) yang diketahui bersifat sitotoksik serta menyebabkan terjadinya inflamasi pada sistem pernapasan dan kardiovaskular serta dapat melewati barier plasenta fetus sehingga dapat memengaruhi fetus. Efek yang ditimbulkan oleh polutan tergantung dari besarnya paparan dan lama waktu paparan. Penelitian ini bertujuan untuk mengidentifikasi Vascular Endothel Growth Factor (VEGF) dan Mitogen Activating Protein Kinase (MAPK) sebagai indikator imflamasi pada plasenta maternal tikus (Rattus norvegicus) yang dipapar carbon black dengan teknik imunohistokimia. Penelitian ini dilakukan dengan tiga kelompok perlakuan dan tujuh kali ulangan yang menggunakan tikus betina bunting. Tikus betina bunting dipapar dengan carbon black dosis 523 mg/m3 dan 1064 mg/m3 selama kebuntingan hari ke-6 sampai 11. Tahap berikutnya adalah pengamatan dan pemeriksaan ekspresi VEGF dan MAPK dengan pewarnaan imunohistokimia terhadap plasenta tikus putih yang telah dipapar dengan carbon black. Hasil penelitian adalah terjadi peningkatan ekspresi VEGF pada plasenta tikus yang dipapar carbon black dosis 523 mg/m3 dan 1064 mg/m3 per inhalasi selama masa pertengahan (6-11) kebuntingan. Selain itu terjadi peningkatan ekspresi MAPK pada plasenta tikus yang dipapar carbon black dosis 523 mg/m3 dan 1064 mg/m3 per inhalasi selama masa pertengahan(6-11) kebuntingan. Simpulan penelitian ini adalah ekspresi VEGF dan MAPK meningkat seiring dengan peningkatan dosis carbon black dan umur kebuntingan.

Abstract

Penelitian ini bertujuan mengetahui efek sentrifugasi semen domba terhadap persentase kapasitasi dan reaksi akrosom spermatozoa. Penelitian ini terdiri atas 3 kelompok semen domba yaitu kelompok kontrol (P0) semen tanpa sentrifugasi, P1 semen disentrifugasi selama 5 menit dengan kecepatan 1800 rpm, dan P2 semen disentrifugasi selama 10 menit dengan kecepatan 1800 rpm. Hasil penelitian menunjukkan bahwa pada kelompok P2 kapasitasi tidak berbeda nyata dengan kelompok P1, tetapi pada kelompok P0 efek kapasitasi dan reaksi akrosom paling rendah. Reaksi akrosom spermatozoa pada kelompok P2 menunjukkan hasil yang paling tinggi. Lama sentrifugasi semen domba ekor gemuk dengan kecepatan sentrifugasi 1800 rpm selama 10 menit dapat meningkatkan persentase kapasitasi dan reaksi akrosom spermatozoa.

Abstract

This study aims to determine the best dose of basil leaf extract to add to AndroMed® semen extender to improve spermatozoa quality after freezing and thawing. This study used healthy, normal four-year-old Simmental bulls with good libido. The sample used was fresh semen from Simmental bull collected using an artificial vagina. This study used a completely randomized design (CRD) with three treatments and six replicates. Ejaculate that met the motility qualification of more than 70%, was divided into three groups, each extended in AndroMed® without the addition of basil leaf extract (T0), with the addition of 0.1% basil leaf extract (T1) and with the addition of 0.3% basil leaf extract (T2). Straw containing semen with 30 million spermatozoa/straw was processed using an IceCube automatic freezer (Minitube, Germany). The results showed that spermatozoa motility, viability, and spermatozoa plasma membrane integrity in the T2 group were higher (p <0.05) than those in the T0 and T1 groups, while spermatozoa morphological abnormalities in the T2 group were lower (p <0.05) than those in the T0 and T1 groups. There were no significant differences (p >0.05) in these parameters between groups T0 and T1. It could be concluded that the addition of 0.3% basil leaf extract to the AndroMed® extender could improve the quality of the semen of Simmental bulls after thawing.

Abstract

This study aimed to evaluate the protective effects of α-tocopherol and zinc sulfate on oxidative stress parameters in ovarian granulosa cells of female Wistar rats (Rattus norvegicus) exposed to lead acetate (Pb(C₂H₃O₂)₂). A total of 25 rats were randomly divided into five groups: Control (C), lead exposure only (T0), α-tocopherol + lead (T1), zinc sulfate + lead (T2), and α-tocopherol + zinc sulfate + lead (T3). Treatments were administered orally for 21 days. Superoxide dismutase (SOD) levels were measured spectrophotometrically, and p53 expression was analyzed using immunohistochemistry. The results showed that SOD levels significantly decreased in group T0 compared to the control (p <0.05), while treatment with either α-tocopherol (T1), zinc sulfate (T2), or their combination (T3) significantly improved SOD levels compared to T0. Moreover, p53 expression was markedly elevated in T0, indicating enhanced oxidative stress and potential apoptosis, whereas all antioxidant-treated groups showed reduced p53 expression, with T3 demonstrating values comparable to the control group. In conclusion, the combination of α-tocopherol and zinc sulfate provided a synergistic antioxidant effect, effectively enhancing SOD levels and suppressing p53 expression in granulosa cells of lead-exposed rats. This suggested a promising therapeutic potential of these compounds in mitigating lead-induced ovarian toxicity.

Abstract

This study aimed to determine the effect of the addition of cherry tomato, california papaya, and honey melon juice into skim milk extender in maintaining the quality of Sapudi ram semen at chilled temperature (5oC) storage. Five replication of ejaculates were divided equally into four groups. In control group (T0) semen was diluted in skim milk extender, while in T1, T2, and T3 groups semen were diluted in skim milk extender contained 20% of cherry tomato, california papaya, and honey melon juice, respectively. The extended semen was stored at a chilled temperature, and semen quality (based on sperm motility, viability, and plasma membrane integrity) was observed daily for five days. The result showed that semen quality declined day by day during the five days of storage. Based on the minimum standard of post-thawed semen motility (40%), the spermatozoa of the control group only lasted by the second day. Meanwhile, in the groups with the addition of fruit juice motility could last up to the third day, with the highest motility (p <0.05) on the addition of california papaya juice. This study concluded that the addition of 20% (v/v) california papaya juice in skim milk extender could maintain the percentage of spermatozoa motility up to three days at a chilled temperature.

Abstract

Preservation of ram semen has an impact on the exposure of artificial conditions to the spermatozoa which was followed by overproduction of reactive oxygen species (ROS) and resulted in functional damages of spermatozoa. This study aimed to determine the effect of the combination of chicken, quail, and duck egg yolks with green tea extract (GTE) on the quality of Sapudi ram spermatozoa at 5°C storage. Ejaculates of Sapudi ram with more than 70% spermatozoa motility were divided into four groups. Semen were extended 1 : 10 in citrated-chicken egg yolk extender (control group, T0), and citrated yolk of chicken (T1), quail (T2), and duck (T3) egg added with 0.05 mg GTE per 100 mL of extender. Extended semen was kept in a tube, which was put in a beaker containing distilled water and stored in a refrigerator (5°C). Spermatozoa motility, viability, and plasma membrane integrity (PMI) were evaluated daily for five days. The results showed that GTE in duck egg yolk citrate extender could maintain the highest spermatozoa motility, viability, and PMI for five days (p <0.05). It could be concluded that the duck egg yolk citrate extender with the addition of GTE (Camellia sinensis) was beneficial for maintaining the semen of Sapudi ram spermatozoa at chilled temperatures. Further study is needed to use the extender for freezing Sapudi ram semen.

Abstract

Livestock production requires Sapudi rams, a breed native to Indonesia, to meet meat demand and food security. In artificial high-quality frozen semen is needed to spread Sapudi rams. To maximize the survival of spermatozoa during cryopreservation, semen should be stored in an extender. Green tea leaf extract (GTLE) and skim milk-goat egg yolk (SM-GEY) may be a good cryoprotectants due to their antioxidant properties. This study aimed to determine the effect of adding GTLE to the SM-GEY extender on the quality of Sapudi ram spermatozoa stored at 5°C. The fresh semen sample was divided into four different GTLE treatment groups, which each contained a 0.1 mL semen sample and a 25-mL extender of SM-GEY. Group T0: no GTLE added to SM-GEY; Groups T1, T2, and T3: 0.1 mL semen diluted in 25 mL SM-GEY with 0.05, 0.10, and 0.15 mg GTLE. Extended semen was then stored at 5°C, and its quality was evaluated daily for five days. The variables observed included spermatozoa motility, viability, and membrane integrity. Data were analyzed using one-way analysis of variance followed by Duncan's test using Statistical Program and Service Solution version 23. The result of this study was that adding 0.05 mg GTLE to 25 mL of SM-GEY extender significantly maintained the spermatozoa motility, viability, and plasma membrane integrity of Sapudi ram spermatozoa for three days at 5°C (p <0.05). Therefore, it could be concluded that adding 0.05 mg of GTLE to the SM-GEY extender preserved Sapudi ram spermatozoa's motility, viability, and membrane integrity for three days at 5°C.

Abstract

This study aims to determine the effect of adding melon (Cucumis melo L.) juice to Lactated Ringer's-egg yolk extender on the intact plasma membrane (IPM) and morphological abnormalities of native rooster spermatozoa. Five ejaculate each from two native roosters (Gallus gallus domesticus) were divided into four equal volumes. Ejaculate was extended 1:10 (v/v) in Lactated Ringer's-egg yolk without or with the addition of of 2, 4, and 8% (v/v) melon flesh juice, respectively for groups T0, T1, T2, and T3. Extended semen was stored in a refrigerator (5°C) and evaluated every two hours. The results showed that IPM of all groups decreased (p <0.05) consistently, lasting more than 40% for six hours in the T0 and T1 groups and four hours in the T2 and T3 groups. The percentage of spermatozoa morphological abnormalities in all groups stored for six hours was higher (p <0.05) than before storage. However, it was not significantly different (p >0.05) from that which had been stored for two and four hours. It was concluded that native rooster semen extended in Lactated Ringer's-egg yolk without (group T0) or with 2% melon flesh juice (group T1) and stored at 5°C retained spermatozoa plasma membrane integrity of more than 40% and morphological abnormalities of less than 20% for six hours. While the addition of 4 % (T2) and 8% (T3) melon flesh juice maintained the percentage of IPM and spermatozoa morphological abnormalities for only four hours.

Abstract

The purpose of this study was to determine the motility and viability of kampung rooster (Gallus gallus domesticus) spermatozoa in 0.9% Sodium chloride (NaCl) solution as an extender with the addition of egg yolk at different concentrations at cool temperature (5°C). This study was performed using two 1.5 years old healthy local roosters. Semen was collected through massage in the morning. The pooled semen sample was divided into four treatment groups. In the T0 group the semen was diluted in 0.9% NaCl, while in the T1, T2 and T3 groups the semen was diluted in 0.9% NaCl added with 5, 10 and 15% egg yolk. The results showed that the motility and viability of spermatozoa decreased when stored at cool temperatures for ten hours (p <0.05). Semen of roosters stored at 5°C in saline solution without the addition of egg yolks showed the lowest motility and viability of spermatozoa (p <0.05). The addition of egg yolk into the saline extender increased the motility and viability of spermatozoa. Concentration of 15% egg yolk in saline solution resulted in the highest spermatozoa motility and viability when stored for up to 8 hours (p <0.05). However, motility and viability of spermatozoa at 10 hours of storage were not significantly different (p >0.05) with the addition of 10% and 15% egg yolks. Therefore, it could be concluded that the addition of 15% egg yolk into a saline solution as an extender could maintain the motility and viability of kampung rooster spermatozoa when stored at 5°C for 10 hours.
