Abstract

Blood cholesterol level which were higher than normal is a trigger of various diseases. The objective of the research was to know the anti-hypercholesterol effect of bark African plant. The research about utilization of ethanol extract of bark African plant (Vernonia amygdalina Delile) has been done, it proved could decrease total cholesterol levels in male mice (Mus musculus). In this research sample macerated with ethanol 96% and then concentrated by rotary evaporator, and gained 9,7% of yield. Based on the results of phytochemical test, secondary metabolites compound that contained in bark African plant are alkaloid, flavonoid, triterpenoid, steroid, phenolic and saponin. On antihiperkolesterolemia activity test by using male mice, there are several treatments that negative control (-), positive control (+) and three treatment with different doses i.e. a low doses (7.5 mg/kg BB), middle doses (15 mg/kg BB) and high doses (30 mg/kg BB). The parameter measured was blood serum of male mice by using a Cholesterol Test Strips. From this test it showed that the effectiveness of decreased total cholesterol level at middle doses of 15 mg/kg BB. Abstract

The research about the biosorpsion of hexavalent chromium heavy metal [Cr(VI)] with Streptococcus mutans bacteria has been carried out. Biosorption has been done by exposure of chromium to broth culture, the consentration and optimum time happened on the 4 day with the biosorption percentage of Cr(VI) by Streptococcus mutans bacteria was obtained at a concentration of Cr(VI) 5 ppm, with a percentage of 90.80%. It shows Streptococcus mutans has potential as biosorbent of Cr(VI). Keywords: Biosorption, Cr(VI), Streptococcus mutans

Abstract

Dyes are one source of heavy metal waste originating from the textile industry, this is caused by the manufacture of dyes that use heavy metals as catalysts. Heavy metals are very dangerous because they cannot be broken down or degraded. Wastewater produced from the textile industry can contain dangerous metals, namely chromium (Cr), copper (Cu) and lead (Pb), where if these metals enter water bodies they can damage the food chain. Therefore, an alternative method is used that can reduce this waste, namely the biosorption method using microorganisms in the form of bacteria. This research uses gram positive bacteria in the form of Streptococcus mutans bacteria which will be exposed to chromium (VI) metal and gram negative bacteria in the form of Flavobacterium sp bacteria which will be exposed to lead (Pb) metal and Pseudomonas sp bacteria which will be exposed to chromium (VI) metal, lead. (Pb), and copper (Cu). The aim of this research was to determine the absorption ability of negative and gram-positive salt bacteria on these heavy metals. Literature results show that Streptococcus mutans bacteria are able to absorb chromium (VI) metal with a maximum of 90.80% with a concentration of 5 ppm on the 5th day. Flavobacterium sp bacteria are able to absorb lead metal (Pb) with a maximum of 96% with a concentration of 2 ppm on the 18th day, while Pseudomonas sp bacteria are able to absorb chromium (VI) metal with a maximum of up to 96.192% with a concentration of 5 ppm on the second day. -14, lead(Pb) metal can be absorbed to a maximum of 36.874% with a concentration of 4 ppm on the 16th day and Cu(II) metal can be absorbed to a maximum of 84.447% with a concentration of 3 ppm on the 16th day. Abstract

Mahakam Ulu District is geographically located in the western part of East Kalimantan Province, where the Mahakam River flows. This upstream segment of the Mahakam River is the main river that functions as a transportation route, as raw water processed by Municipal Waterworks, used as a place for bathing, washing as well as serving as a lavatory by the community and a source of water for local community agriculture. Surface water sampling points consist of 14 stations, including ST I: Mahakam River (Long Pahangai Village), ST II: Melaseh River (Long Pahangai Village), ST III: Pahangai River (Long Pahangai Village), ST IV: Mahakam River (Tiong Ohang Village), ST V: Sebunut River (Long Bagun Village), ST VI: Alan River (Long Bagun Village), ST VII: Mahakam River (Ujoh Bilang Village), ST VIII: Melahan River (Long Melahan Village), ST IX: Mahakam River (Laham Village), ST X: Sungai Ratih (Damum Paroy village), ST XI: Mahakam River (Datah Bilang village), ST XII: Pari River (Datah Bilang village), ST XIII: Pataq River (Long Bagun sub-district) and ST XIV: Medang River (Long Bagun sub-district). Determination of water quality status using the STORET Method or Pollution Index Method. The STORET method is one of the commonly used methods for determining water quality status. With this STORET method can be known parameters that have met or exceeded water quality standards. From the results of the study, the water quality parameters that partially did not meet the class I quality standards were 6 parameters, among others: TSS, BOD, COD, DO and Faecal coliform and Total coliform, while for classes II, III and IV all parameters meet the quality standards according to regulations. Abstract

Amplification of pncA genes from water bacterial post mining in Samarinda has been done. The aim of this research is to isolate chromosomal DNA and amplificated the pncA gene from water bacterial post mining in Samarinda. The research methode consist of chromosomal DNA isolation and amplification of pncA gene with polymerase chain reaction (PCR) technique. The result showed the size of chromosomal DNA is ± 10 kb. By using primer pncA gene, the result of amplification of the pncA gene showed that there were two sizes of amplicon, namely 0.7 kb and 0.2 kb. The conclusion that can be drawn was there is two sizes of amplicon pncA gene. Keywords: chromosomal DNA isolation, amplification of pncA gen, PCR

Abstract

Kumis kucing plant (Orthosiphon spicatus B. B. S.) are one of the plants that have been used as diuretic, antidiabetic and antihypertensive drugs. Identification of chemical compounds, and antidiabetic activity assay of bioproduction of endophytic fungi with code B.Os.1F that isolated from the stems of kumis kucing have been carried out. Two fermentation methods (dynamic and static fermentation) to determine the pH. The tlc profile analysis and pH showed that B.Os.1F isolate by dynamic fermentation for 12 days had the potential to produce chemical compounds as antidiabetics at pH 4 ~ 6. The product of fermentation are extracted with ethyl acetate, then fractionated by column chromatography (SiO2): (i) n-hexane-ethyl acetate [10:1 ~ 1:1]; (ii) chloroform-methanol (20:1 ~ 1:1) based on “bioassay guided fractionation” with a-glucosidase enzyme inhibition assay. The isolate of B.Os.1F.2-3 had an activity of 76 %, and identify based on UV-Vis, Fourier-Transform Infra Red (FT-IR) spectroscopies and gas chromatography-mass spectrometer (GC-MS) are estimated to be Ergosta-5,7,9(11),22-tetraen-3-ol compounds. Abstract

This literature review aims to determine the potential of various carbohydrate sources as raw materials for bioethanol production through fermentation to achieve maximum ethanol concentration. The hydrolysis process is conducted enzymatically using microbes during the liquefaction stage and gluco-amylase during the saccharification stage, followed by fermentation by microbes. The literature results show that the carbohydrate source, Ganyong tuber (Canna edulis Kerr.), produces a maximum ethanol concentration of 84.451% with a fermentation period of 7 days. Abstract

Fermentation has been used by traditional societies as far back as 6000 AD involving microorganisms such as yeast and bacteria that can convert carbohydrates into alcohol and carbon dioxide. This process is essential for the production of bioethanol produced from agricultural residues. Fermentation testing can be done using raw materials such as tubers, sticky rice, sago, water hyacinth and cassava to produce bioethanol as an alternative renewable energy. Methods This research utilizes 8 articles from national and international literature studies in the last 10 years that focus on the fermentation process to produce bioethanol. Research and development of efficient fermentation techniques is urgently needed to increase the yield and sustainability of bioethanol production, thus becoming an environmentally friendly solution in meeting global energy needs. Abstract

Antibacterials are compounds used to eradicate microbes, especially microbes that harm humans. These antibacterial compounds can be extracted directly from plants, where these extracts contain secondary metabolite compounds that have antibacterial activity. Antibacterial testing is carried out on gram-positive bacteria such as Staphylococcus, Streptococcus and others. The method used in this research was a literature study of 10 national and international articles published in the last 10 years. The articles cited focus on research articles on plants that have the potential as antibacterials of several gram-positive bacteria which include extract solvents, metabolite compound content in extracts and their activity capabilities based on the diameter value of the inhibition zone and/or minimum inhibitory concentration using the disc diffusion method. Abstract

Cancer is one of the leading causes of death worldwide, including in Indonesia. This review explores the potential of various plants as sources of anticancer compounds through secondary metabolite analysis. Plants such as Rosa damascena, Wild Onion (Scorodocarpus borneensis Becc), and Acalypha Indica have been studied for anticancer activity using in vitro methods. The results showed that extracts from Rosa damascena had varying IC50 values ​​against various types of cancer cells, including 16.28 μg/mL in liver cancer cells (HepG-2) and 18.09 μg/mL in breast cancer cells (MCF-7). In addition, Wild Onion showed very strong anticancer activity with an IC50 value of 1.1061 μg/mL in L1210 leukemia cells. Secondary metabolites such as flavonoids, alkaloids, and tannins were identified as key components contributing to the anticancer effects. These findings highlight the importance of further research to develop more effective and safe plant-based cancer therapies. Keyword: Anticancer, Cancer, Secondary Metabolites, Plants
