Abstract

The impact of free radicals on testicular Leydig cells can result in the disruption of male reproductive health. The present study aimed to evaluate the effects of a high-fat diet on Leydig cells and the potential of kepok banana peel extract as a medication to mitigate these effects. In total of 20 adult male Wistar rats were assigned to five groups and treated with standard feed (P1), high-fat feed (P2), high-fat feed with simvastatin (P3), and high-fat feed with banana peel extract at doses of 100 and 200 mg/kg BW (P4 and P5). After 60 days of treatment, the number of Leydig cells was determined using HE staining methods. Data were analyzed using an One Way ANOVA test. The results showed that the administration of 100 mg/kg BW of kepok banana peel extract (P4) was able to maintain the number of Leydig cells and counteract the negative effects of a high-fat diet. These findings suggest that kepok banana peel extract may have the potential as a herbal medicine for supporting male reproductive health.

Abstract

The purpose of this study was to analyze the potential of tomato extract (Lycopersicum esculentum L.) in increasing the Sertoli and spermatogenic cells count of the contralateral testis in white rats (Rattus novergicus) with unilateral torsion. A total of 24 male Wistar white rats, aged 3-4 months old with a weight of 180-200 g were used in this study. The rat samples were randomly divided into 4 treatment groups: Group 1 as a negative control (NC) which consisted of rats without testicular torsion and given tomato extract; Group 2 as a positive control (PC) which consisted of rats with unilateral testicular torsion 720 for 6 hours without given tomato extract; Group 3 as treatment 1 (T1) consisted of rats that were subjected to unilateral testicular torsion 720o for 6 hours and were given tomato extract at a dose of 100 mg/kg BW/day; and Group 4 as treatment 2 (T2) consisted of rats that were subjected to unilateral testicular torsion 720 for 6 hours and were given tomato extract at a dose of 200 mg/kg BW/day. Each group consisted of six rats. Tomato extract was administered orally for 30 days. The observations of the number of Sertoli cells and spermatogenic cells (spermatogonia cells, spermatocytes, and spermatids) were carried out histologically with Hematoxylin-Eosin (HE) staining and the slides were examined with a light microscope. The data obtained was analyzed by ANOVA and continued with Duncan's test. The results showed that the administration of tomato extract significantly increased (P0.05) the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubule. Giving tomato extract 100 mg/kg BW/day resulted in an increase in the number of Sertoli cells and better spermatogenic cells than tomato extract 200 mg/kg BW/day. In conclusion the administration of tomato extract after detorsion can increase the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubules in white rats subjected to unilateral torsion of 720 for 6 hours

Abstract

This study was conducted to validate a commercial testosterone enzyme-linked immunosorbent assay (ELISA) kits (DRG EIA-1559) inanalytic and biological manner for measuring serum testosterone concentrations in kacang goats. This study used 18 healthy kacang goats, six bucks (2 years), six kids (6 months), and six does (2 years). Blood samples were collected from jugular vein and prepared as serum. Two validation tests were performed, an analytical validation comprises a parallelism, accuracy, precision and sensitivity and a biological validation by comparing testosterone concentration from bucks, kids, and does. Testosterone concentrations were measured using ELISA technique. Data of analytical validation were analyzed descriptively and test of equality of slope was performed to see the parallelism between samples and standard curves. Analysis of variance (ANOVA) was used for biological validation data. Results of parallelism showed that sample curve was parallel to the standard curve. Accuracy, precision (% CV of intra-and inter-assay) and sensitivity of the assay were 99.654.27%, 10%, 15% and 0.083 ng/ml, respectively. Results of biological validation showed that the assay used were accurately measured testosterone which testosterone concentrations in bucks were significantly higher compared to kids and does (P0.05). In conclusion, a commercial testosterone ELISA kits (DRG EIA-1559) is a reliable assay for measuring serum testosterone concentration in kacang goats. Abstract

Herbal antioxidants have gained attention recently as possible treatments for disorders of male reproductive function linked to oxidative stress because of their ability to neutralize free radicals. The investigation into herbal flavonoids for preserving oxidative stress-impaired testicular physiology has been effectively conducted through the visualization of bibliometric analysis. Bibliometric data of published papers collected from the PubMed and Scopus databases in 2018-2023. Network visualization was performed using VOSviewer software on the set keywords that occur (n=15). The literature review was conducted according to the research trends identified by the clusters that appeared in the network visualization. The result revealed that a total of 266 records have been published based on the specified search set keywords since 2018. Through the analysis of keywords, inclusion and exclusion criteria, and the relevant content of papers, 11 articles were identified as suitable for conducting a literature review. Network visualization revealed that the research falls into two general categories: (1) toxicity to the testicles caused by oxidative stress and (2) prevention of oxidative stress impact on tissue through herbal medicine. Examination of the paper's content revealed that certain antioxidant compounds from plants can stabilize free radicals in the testicles. These antioxidants include vitexin, diosmin, hesperidin, phenolic acids, naphthoquinones, fisetin, pachypodol, sciadopitysin, carvacrol, and morin. However, further research is needed to compare active antioxidant content in different herbal plants and identify optimal concentrations for therapeutic effects.
