Abstract

The aims of the research was to determine the growth response and to identify the potential embriogenic callus of the hypoxia condition tolerant of soybean varieties in vitro. The research was conducted at the Plant Tissue Culture Laboratory and the Laboratory of Food Technology, Faculty of Agriculture, North Sumatera University, Medan, Indonesia, from March 2016 to September 2016. The result were analyzed with t-test with the combination between soybean varieties (Baluran, Gepak kuning, Willis) and the plant growth regulators (2,4-D, NAA, IAA) 10 mg/l. The parameters observed were: the percentage of plants forming callus, the callus colour, the callus texture, the callus histological, the fresh weight of callus, the total chlorophyll content, the protein concentration, the SOD activity and the POD activity. The result showed that the combination between varieties Gepak Kuning and Willlis with 10 mg/l 2,4-D significantly affected the growth of embriogenic callus, the fresh weight of callus, the total chlorophyll content, the protein concentration, the SOD and POD activities.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh terhadap induksi akar (rhizogenesis) pada P. amboinicus secara in vitro. Penelitian ini dilaksanakan di Laboratorium Kultur Jaringan UPT. Benih Induk Hortikultura Gedung Johor, Dinas Pertanian, Sumatera Utara, Indonesia. Penelitian ini dimulai pada bulan April 2016 sampai dengan Juli 2016. Rancangan yang digunakan adalah rancangan acak lengkap non faktorial dengan 7 perlakuan yaitu: MS + 1 mg/l NAA + 0,1 mg/l kinetin; MS + 2 mg/l NAA + 0,1 mg/l   kinetin;  MS + 3 mg/l NAA  + 0,1 mg/l kinetin; MS + 1 mg/l 2,4 D + 0,1 mg/l kinetin; MS + 2 mg/l 2,4 D + 0,1 mg/l kinetin; MS + 3 mg/l 2,4 D + 0,1 mg/l kinetin; MS + 0,1 mg/l kinetin. Penelitian ini menggunakan 15 ulangan. Hasil penelitian menunjukkan bahwa perlakuan zat pengatur tumbuh memberikan pengaruh nyata terhadap berat segar akar dan jumlah akar tetapi belum memberikan pengaruh nyata terhadap persentase terbentuknya akar dan umur muncul akar. Medium MS + 3 mg/l NAA + 0,1 mg/l kinetin memberikan hasil terbaik pada persentase terbentuknya akar, berat segar akar, dan jumlah akar.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi karakter morfologis Durian (Durio zibethinus Murr) di Kecamatan Tigalingga dan Pegagan Hilir Kabupaten Dairi Sumatera Utara.Penelitian ini dilakukan di Kecamatan Tigalingga dan Kecamatan Pegagan Hilir Kabupaten Dairi pada bulan Desember 2016 sampai dengan Januari 2017, data diidentifikasidengan metode survei deskriptif menggunakan panduan descriptors for durian(Bioversity Internasional)dengan teknik pengambilan sampel secara accidental sampling.Hasil penelitian menunjukkan bahwa di dua Kecamatan di Kabupaten Dairi terdapat 21 genotipe durian yang teridentifikasi. Hasil karakterisasi morfologis untuk bentuk tajuk (piramida, jorong, bulat, stengah membulat, lonjong  dan tidak beraturan), permukaan batang (kasar), daun (empat persegi, bulat panjang dan bulat telur), buah (bulat ujung datar, bulat, bulat telur, lonjong, bulat panjang, dan ovoid), biji (lonjong jorong dan bulat).   Kata kunci : karakter morfologi, durian, kabupaten Dairi

Abstract

This study aims to get individuals selected of some soybean Glycine max L. (Merrill) M5 generation with character resistant stem rot disease Athelia rolfsii (Curzi)and high production. This research was conducted in Plant Disease Laboratory and experimental field Faculty of Agriculture Universitas Sumatera Utara Medan, Indonesia. This research was conducted from December 2016 until July 2017. In this study using augmented design. The treatments were 15 mutant lines, the check variety were Anjasmoro, Agromulyo, and Kipas Putih varieties. The results showed that the appearance of agronomic characters observed in the inoculated medium of the fungus causes stem rot disease is lower than the optimum field without inoculation of fungus. Differences in the means values of the mutant line are in the larger stem diameter character of plant than with the inoculation of fungus.  Broad sense heritability values were found in the number of pods per plant, number of seeds per plant, and seed weight per plant on the inoculated medium of the fungus and the optimum field. Selection performed on population M5resulted 7 individuals with high production character and 62 individuals with tolerance of stem rot disease character.

Abstract

This reserach aimed to identify the morphological chracteristics and phylogenetic relationship of Dragon Fruit (Hylocereus sp.) plants. The research was started from April 2017 until May 2017 at Sitinjo district of  Dairi Regency North Sumatera with survey methode using dragon fruit descriptor of Hylocereus by International Union For The Protection Of New Varieties (UPOV). Sampling technique using purposive sampling. Exploration results was obtained 24 genotypes of Dragon Fruit plants based on identified fruit morphological characteristic. Based on clustering analyze the closest genetic relationship is on genotype G17 and G23 which has 2 differences of 7 characteristic with dissimilarity coeficient is 1,000 and the furthest genetic relationship is on genotype G18 and G26 with dissimilarity coeficien is 4,796 which has 5 differences of 7 characteristic.

Abstract

This research aimed to identify the morphological characteristics and phylogenetic relationship of several genotypes of coconut in Silau Laut district Asahan regency Sumatera Utara. The research was conducted from April to Mei 2017 by survey method based on coconut descriptor of International Plant Genetic Resources Institute (IPGRI) with sampling technique using accidental sampling. This result of research showed that in Silau Laut district there were 27 genotypes of coconut been identified. The result of morphologycal characteristics for crown shape (spherical, hemispherical, X-shaped ‘silhouette’), flower shape (normal and additional spathes or bracts), fruit shape (oblong, ovoid, angled, round), nut shape (pointed, ovoid, almost round, oblate). The closest phylogenetic relationship is G41 and G42 with coefficient dissimilarity 3,00 and farthest phylogenetic relationship is G40 and G45 with coefficient dissimilarity 12,53. The result of dendogam showed at scale 18 was formed 5 goups. Phylogenetic relationship of between genotypes of coconut in Silau Laut district Asahan regency still have a close level relationship.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi fenotipe tanaman pisang barangan (Musa acuminata Linn.) yang ada di Kabupaten Deli Serdang Sumatera Utara. Penelitian ini dilaksanakan di Kecamatan Sinembah Tanjung Muda Hilir Kabupaten Deli Serdang mulai bulan Maret sampai dengan Agustus 2017 menggunakan metode survei deskriptif dengan panduan descriptors for banana (International Plant Genetic Resources Institute) dengan teknik pengambilan sampel secara purposive sampling. Hasil penelitian ini menunjukkan bahwa hubungan kekerabatan terdekat ada pada sampel D1P1S1 dan D1P1S3 dengan nilai jarak terdekat senilai 0,000 dan hubungan kekerabatan terjauh pada sampel D1P3S3 dan D3P3S2 dengan nilai jarak terjauh senilai 5,000.

Abstract

To fulfill the demand of soybeans, the expansion of the planting area can be one of the choices through the conversion of marginal land by utilizing suboptimal land, such as saline soil. One of the plant breeding techniques to obtain tolerant plants to salinity is the use of chemical mutations, such as colchicine. The study aimed to reveal the effect of salinity on the characters of soybean plants (mutated with colchicine) and the effect of colchicine on the number of chromosomes and ploidy level. This study was conducted from January to March 2020 and carried out in the plastic house of Faculty of Agriculture, Universitas Sumatera Utara. The experiment was arranged in a Randomized Block Design with salinity levels (0 dS.m-1, 2 dS.m-1, 4 dS.m-1, 6 dS.m-1) as treatments and consisted of ten replications in each treatment. The result showed that the soybean plants were not successfully induced by colchicine at a concentration of 0.04 % with soaking duration of 10 hours, as indicated by the number of chromosomes and analysis of ploidy level. The soybeans that had been induced were planted in saline soil. There were significant effects observed on the productive branch, number of filled pods, and the weight of seeds per plant. Colchicine treatment did not successfully affect the number of chromosomes of soybeans but had an impact on the chromosome pattern. In addition, the salinity treatment of 4 dS.m-1 significantly affected the number of filled pods compared to the treatment of 6 dS.m-1.

Abstract

Amarilis is one of the flower of the park that is attractive and has high economic value. This study aims to determine the effect of vernalization on the appearance of local amaryllis plants in Tanah Karo and West Sumatra. This research was conducted at the Plant Breeding Basic Laboratory of the Faculty of Agriculture, University of North Sumatra, Medan, from February 2018 to July 2018. This research was carried out by storing tubers at room temperature (control) and storage of tubers at low temperatures (vernalization) at 10 oC for 6 weeks, and with differences in accession of plants namely G1 (Accession of West Sumatra Payakumbuh), G2 (Bukit Tinggi West Sumatra Accession), G3 (Padang Panjang West Sumatra Accession), G4 (Local Kabanjahe Accession Tanah Karo), G5 (Berastagi Local Accession Tanah Karo ), G6 (Tanah Karo Local Accession Accession). The results showed that vernalization gave a different appearance compared to the control treatment.

Abstract

The aims of the research were to know The Root Growth Response of Some crosses F1 Corn Populations (Zea mays L.) at Iron Stress (Fe) in Medium of Nutrient Culture. This research was conducted at the greenhouse Faculty of the Agriculture University of Sumatera Utara, Medan from March to April 2017. The research used Randomized Block Design (RBD) with two using two factors i.e : F1 population (NEI 9008 x CLA 46, CLA 84 x NEI 9008, CLA 106 x NEI 9008, CLA 16 x CLA 84, and CLA 84 x P1027) and iron stress treatment (0, 50 ppm, and 100 ppm). The results showed that the F1 population had a significant effect on root-canopy weight, root volume, and root canopy ratio. Iron stress treatment with different concentrations had a significant effect on root distribution parameters, root canopy ratio, root length, and the extension of root length. The interaction between F1 population and iron stress has a significant effect on root volume character. Iron treatment with a concentration of 100 ppm can still be tolerated by the F1 population of maize crops on nutrient culture. The CLA 16 x CLA 84 populations had a better adaptation to the iron stress treatment in nutrient culture media. The CLA 16 x CLA 84 had lower stress sensitivity index value and a smaller productivity gap on root dry weight.
