Abstract

Effects of Casein Hydrolisate and Light Intensity on Production of Biomass and Canthinone Alkaloid in Cell Suspension Cultures of Pasak Bumi (Eurycoma longifolia Jack). A study on the effect of several concentrations of casein hydrolysate and light intensity to produce biomass and canthinone alkaloid in cell suspension culture of Pasak Bumi (Eurycoma longifolia Jack) was conducted. The research was an experimental study with block random design non factorial. For study of casein hydrolysate was used 8 (eight) of concentration treatments, i.e. 0.00, 0.10, 0.25, 0.50, 0.75, 1.00, 2.00, dan 5.00% (w/v). Beside for study of different light intensity effect was used five of light intensity, i.e. 0, 190, 290, 585, and 1525 lux. The results showed that addition 0.1–2.0% (w/v) of casein hydrolysate into culture medium did not show the effect on production of cell biomass compare with without casein hydrolysate. Total of alkaloid produce increased two fold in the culture medium containing 0.1% casein hydrolysate. The medium with addition 5% casein hydrolysate significantly decrease of increased fresh weight, dry weight and total of alkaloid compare with medium without or additions of casein hydrolysate at 0.1–2.0% concentration. Besides, the modifications on light intensity (0–1525 lux) did not effect the production of cell biomass in culture medium, but the place of E. longifolia Jack cell suspension culture at light of 1525 lux was found have to stimulate increased the total of 9- methoxycanthin-6-one in cells.

Abstract

Penyakit busuk pangkal batang yang disebabkan oleh Ganoderma boninense menyebabkan kerusakan signifikan pada perkebunan kelapa sawit di Asia Tenggara, termasuk di Indonesia. Penggunaan fungisida kimia yang intensif terhadap Ganoderma dapat berdampak negatif terhadap lingkungan, kesehatan, dan terganggunya keseimbangan ekologi. Aplikasi mikrob sebagai agens hayati menggunakan cendawan endofit antagonis, salah satunya dark septate endophyte (DSE) berpotensi sebagai pengendalian Ganoderma. Penelitian ini bertujuan mendapatkan isolat DSE lokal Sumatera Utara dari areal endemik Ganoderma yang berpotensi untuk mengendalikan G. boninense. Eksplorasi cendawan DSE dilakukan pada areal dengan insidensi Ganoderma moderat (5%-30%) di Kecamatan Galang, Kabupaten Deli Serdang dan pengujian dilakukan secara in vitro. Hasil penelitian didapat lima isolat cendawan DSE, yaitu isolat II 3 A CM, Gelap A, II 4 CM, II 3 A CC, dan abu-abu. Kemampuan daya hambat terhadap perkembangan G. boninense secara in vitro dari yang paling tinggi berturut-turut adalah isolat Gelap A (100%), abu-abu (90.87%), II 3 A CC (82.83%), II 3 A CM (82.23%), dan II 4 CM (74.88%) pada 7 hari setelah inokulasi. Temuan ini merupakan data awal akan potensi cendawan DSE sebagai agens hayati G. boninense dan merupakan laporan pertama untuk cendawan DSE asal tanaman kelapa sawit di Sumatera Utara.

Abstract

The aim of the research was to determine the best paclobutrazol treatment to obtain superior quality seed potato cultivars. The research was conducted at the Tissue Culture Laboratory of Horticulture Technical Centre Service Unit, Gedung Johor, Medan, North Sumatra, Indonesia from January 2020 to March 2020. The complete randomized design with a single factor, 4 treatments, and 6 replications was used in this research. The results showed that the concentration of paclobutrazol affected the number of leaves, the shoots length, the number of shots, the number of nodes, the number of internodes, the stem diameter, and the percentage of roots formed but not significantly affected the number of roots. The M2 treatment, MS + 9 ppm paclobutrazol, showed the best treatment with an average plantlet performance value of 7.33 and the plantlets showed the compact morphology with larger stem diameter and smaller thick dark green leaves.

Abstract

Phosphate (P) nutrient plays a significant role in plant growth and yield. P is an essential element that plays an important role in photosynthesis and root development. Phosphate nutrient availability is deficient in some soil types due to retention, such as in Andisol soil types. High phosphate retention in Andisol soil types causes P nutrients to be unavailable to plants and can reduce crop yields. The availability of P in Andisol soils can be done, among others, by applying phosphate solubilizing microbes. Phosphate solubilizing microorganisms are soil microorganisms consisting of bacteria and fungi that can mineralize organic P, dissolve inorganic P minerals, and store large amounts of P to make it available to plants. This literature review aims to determine the mechanism of phosphate-solubilizing microbes in P dissolution in Andisol soil. The methods used in this systematic review are collecting data through the internet and utilizing recognized sources such as Science Direct, Research Gate, Google Scholar, and Web of Science. Content analysis was performed on the collected data, and the results were organized into thematic categories. Furthermore, the findings are presented descriptively with the help of tables to facilitate understanding. Since phosphate-solubilizing microorganisms can dissolve P in the soil through chemical and biological mechanisms, it can be concluded that phosphate-solubilizing microorganisms also have an important role in the soil P cycle. The implications of this literature review are to understand the retention of P nutrients in Andisols and how the dissolution mechanism works, as well as the use of microbes as a solution to increase phosphate dissolution so that it is available to plants.
