Abstract

Umbi wortel (Daucus carota L.) mengandung senyawa golongan fenolik dan flavonoid yang memiliki berbagai aktivitas farmakologis. Senyawa golongan fenolik dan flavonoid dapat diekstraksi dengan berbagai metode ekstraksi. Perbedaan metode ekstraksi dapat menghasilkan kadar fenolik dan flavonoid yang berbeda. Penelitian ini bertujuan untuk membandingkan kadar fenolik dan flavonoid total ekstrak metanol umbi wortel dari berbagai macam metode ekstraksi. Umbi wortel diekstraksi dengan metode perkolasi, maserasi, soklet, dan refluks menggunakan pelarut metanol sehingga diperoleh ekstrak metanol umbi wortel. Penetapan kadar fenolik dan flavonoid total dilakukan menggunakan spektrofotometer UV-VIS dengan pembanding asam galat dan kuersetin. Kadar fenolik total dari empat metode ekstraksi dianalisis statistic menggunakan uji One Way Anova sedangkan kadar flavonoid total menggunakan uji Kruskal Wallis kemudian dilanjutkan Mann-whitney pada taraf kepercayaan 95%. Hasil penelitian menunjukkan kadar fenolik total ekstrak metanol umbi wortel dengan metode perkolasi, maserasi, soklet, dan refluks berturut-turut 40,423; 33,311; 45,164; dan 41,663 mgGAE/gram ekstrak sedangkan kadar flavonoid total secara berturut-turut 14,348; 6,065; 11,861; dan 4,704 mgQE/gram ekstrak. Analisis statistik menunjukkan ada perbedaan kadar fenolik dan flavonoid total ekstrak metanol umbi wortel hasil ekstraksi menggunakan metode perkolasi, maserasi, soklet, dan refluks. Kadar fenolik total tertinggi diperoleh dengan metode soklet sedangkan flavonoid total tertinggi diperoleh dengan metode perkolasi.

Abstract

ABSTRAKKelurahan Kalisegoro merupakan salah satu kelurahan yang ada di Kecamatan Gunung Pati Kota Semarang. Rata-rata penduduk Kalisegoro bekerja di sektor swasta, wiraswasta dan buruh serta sebagian yang lain beragam. Adanya pandemi covid-19 yang terjadi beberapa waktu yang lalu, menyebabkan masyarakat banyak kehilangan pekerjaan dan sumber mata pencaharian. Hilangnya pendapatan suatu kepala keluarga akibat pandemi Covid-19 ini mengharuskan setiap keluarga mencari alternatif lain untuk mencukupi kebutuhan sehari-hari. Tujuan pengabdian ini untuk memberikan pengetahuan kepada ibu-ibu warga kelurahan Kalisegoro tentang pembuatan sabun eco-friendly serta produk yang dihasilkan dapat dijual sehingga dapat meningkatkan perekonomian keluarga dan masyarakat setempat. Metode pelaksanakaan dengan memberikan pelatihan pembuatan sabun eco-friendly kepada ibu-ibu di Kelurahan Kalisegoro. Peserta pelatihan terdiri dari ibu-ibu PKK di RT 06 RW 01 yang berjumlah 22 orang. Hasil dari kegiatan ini yaitu dihasilkan sabun eco-friendly dengan bahan alami yang aman bagi lingkungan dan pemakai serta memberikan pelatihan kepada masyarakat Kelurahan Kalisegoro mengenai langkah pembuatan sabun eco-friendly yang nantinya mitra dapat mengembangkan keterampilan dalam berwirausaha. Kata kunci: pelatihan; sabun eco-friendly; kewirausahaan ABSTRACTKalisegoro Village is one of the villages in Gunung Pati District, Semarang City. The average Kalisegoro resident works in the private sector, is self-employed and is a laborer, as well as a variety of others. The Covid-19 pandemic that occurred some time ago caused many people to lose their jobs and sources of livelihood. The loss of income for the head of the family due to the Covid-19 pandemic requires each family to look for other alternatives to meet their daily needs. The aim of this service is to provide knowledge to mothers of Kalisegoro sub-district residents about making eco-friendly soap and the resulting products can be sold so that they can improve the economy of their families and local communities. The implementation method is to provide training in making eco-friendly soap to mothers in Kalisegoro Village. The training participants consisted of 22 PKK women in RT 06 RW 01. The results of this activity are producing eco-friendly soap with natural ingredients that are safe for the environment and users as well as providing training to the Kalisegoro Village community regarding steps for making eco-friendly soap so that partners can later develop entrepreneurial skills. Keywords: training; eco-friendly soap; entrepreneurship.

Abstract

Carrot (Daucus carota L.) contains phenolic and flavonoid compounds that contribute to various pharmacological activities. Phenolic and flavonoids can be extracted using various solvents. Solvent polarity difference will produce amounts and types of secondary metabolites differently. This research aims to compare the total phenolic and flavonoid content of carrot extract from various solvents. Carrot was extracted using the maceration method with n-hexane, ethyl acetate, and 70% ethanol as solvent to obtain n-hexane, ethyl acetate, and 70% ethanol extract of carrot. Spectrophotometry carried out the determination of total phenolic content by comparing gallic acid and total flavonoids with a comparison of quercetin. The maximum wavelength of gallic acid is 739.60 nm and the operating time of 120 minutes. The maximum wavelength of quercetin is 427.50 nm and an operating time of 30 minutes. Total phenolic and flavonoid levels of carrot extract from the three solvents were analyzed statistically through one-way ANOVA followed by the Bonferroni test at a 95% confidence level. The results showed that the total phenolic contents of the n-hexane extract, ethyl acetate, and 70% ethanol extract of carrot 11.881±0.31; 24.308±1.33 and 39.398±1.65 mg GAE/g extract. The flavonoid content was 2.650±0.13; 20.675±2.58 and 9.063±0.99 mg QE/g extract respectively. The highest total phenolic content was obtained from 70% ethanol solvent and the highest total flavonoid was obtained from ethyl acetate solvent.

Abstract

Pagertoyo Village, Limbangan District, Kendal Regency is one of the villages that has the potential for growth and development of herbal plants. One of them is the binahong plant (Anredera cordifolia). This plant has the property of curing various diseases, including lowering blood pressure and gout. This community service aims to provide education about the benefits of the binahong plant and training in making binahong pudding to PKK women in Pagertoyo Village, Limbangan District, Kendal Regency. Implementation of activities was carried out in 3 stages. The first stage is a health check (check blood pressure and uric acid). The second stage is education about the benefits of the binahong plant. The participants were given a lecture about the benefits of the binahong plant followed by a discussion and question and answer session. The third stage is training in making binahong pudding. The implementation of this community service activity ran smoothly and successfully. The participants actively participated in question-and-answer sessions and discussions as well as training.

Hasil penelitian menunjukkan rendemen ekstrak metode maserasi selama 3 dan 5 hari sebesar 24,66% dan 24,88%, sedangkan metode digesti selama 3 dan 6 jam sebesar 10,06% dan 16,59%. Nilai Rf kuersetin sebesar 0,80, dengan nilai Rf ekstrak berturut-turut 0,41; 0,65; 0,68; dan 0,80. Kadar flavonoid total pada metode maserasi 3 dan 5 hari sebesar 61,40 mgQE/g dan 67,62 mgQE/g, sedangkan metode digesti 3 dan 6 jam sebesar 64,45 mgQE/g dan 73,32 mgQE/g. Uji statistik menunjukkan perbedaan signifikan (p<0,05), dengan kadar flavonoid tertinggi diperoleh dari metode digesti selama 6 jam. Dengan demikian, metode dan lama ekstraksi berpengaruh terhadap kadar flavonoid total dalam ekstrak buah bit.

Abstract

A comprehensive QSAR analysis, in conjunction with molecular docking, molecular dynamics simulations, MM-PBSA binding energy estimations, and ADMET profiling, was conducted to facilitate the development of novel anticancer agents based on hydroxyxanthone derivatives. Molecular and electronic descriptors were calculated using the DFT method with the 3-21G basis set. The best QSAR model identified several descriptors that significantly influence anticancer activity, including the atomic charges at positions C1, C3, C4a, and C7, as well as the highest occupied molecular orbital (HOMO), surface area (SA), molecular volume (VOL), and molecular weight (MW). This model was used to design novel hydroxyxanthone derivatives (X27 to X47). The docking result showed that compounds 7-bromo-3-hydroxy-1-(methylamino)-9H-xanthen-9-one (X43), 6-hydroxy-8-(methylamino)-9-oxo-9H-xanthene-2-carbonitrile (X44), and 3-hydroxy-7-mercapto-1-(methylamino)-9H-xanthen-9-one (X45) had stronger binding energy values than gefitinib as a native ligand. Gefitinib had a binding energy of -6.84 kcal/mol, while those compounds had values of -6.92, -7.12, and -6.92 kcal/mol, respectively. In a molecular dynamics simulation of 100 ns, compounds X43, X44, and X45 exhibited stability comparable to that of gefitinib against the EGFR protein. Additionally, the binding energy MM-PBSA of compound X43 was the lowest (-29.18 kcal/mol), followed by X44 (-27.11 kcal/mol), gefitinib (-26.06 kcal/mol), and X45 (-25.21 kcal/mol). Furthermore, these compounds met Lipinski's rule parameters and the minimal standard parameters in terms of ADMET characteristics, as predicted by physicochemical properties. In conclusion, compounds X43, X44, and X45 are potential anticancer agents for MDA-MB-231 breast cancer cells.

Abstract

This research employs an artificial intelligence (AI)-driven molecular docking approach to identify potential antidiabetic compounds from Syzygium polyanthum phytochemicals targeting the α-glucosidase enzyme. The docking simulations were conducted using the PLANTS software, which utilizes an ant colony optimization (ACO) algorithm, a nature-inspired AI technique that mimics the foraging behavior of ants to explore ligand binding conformations efficiently. PLANTS integrates multiple empirical scoring functions, including ChemPLP, to evaluate protein-ligand interactions by modeling steric complementarity, hydrogen bonding, and torsional potentials, enabling accurate prediction of binding affinities. The protein structure with PDB code 2JKE was validated with a root-mean-square deviation (RMSD) of 0.2912 Å, confirming the reliability of the docking protocol. Screening results revealed seven phytochemical compounds Hexadecanoic acid 2-hydroxy-1-(hydroxymethyl), Methyl oleate, Methyl palmitate, Phytol, 9,12,15-Octadecatrien-1-ol, Nerolidol, and Eicosane exhibited lower docking scores (-96.2919 to -80.5188) than both the reference drug miglitol (-80.2642) and the native ligand (-77.2910), indicating stronger and more stable binding to the α-glucosidase active site. These findings suggest that the identified compounds have superior theoretical inhibitory potential compared to miglitol, a clinically used α-glucosidase inhibitor. The AI-based in silico screening using PLANTS thus provides a powerful, cost-effective strategy for accelerating antidiabetic drug discovery by prioritizing promising natural compounds for further experimental validation.

Abstract

Chlorochalcone derivatives (chalcones 1–3) were synthesized using ultrasound-assisted Claisen-Schmidt condensation, yielding > 80%. Antioxidant activity was evaluated through DPPH and ABTS assays, demonstrating strong activity with IC50 values ranging from 61.52 ± 0.97 to 98.27 ± 1.42 ppm. Chalcones 1 and 2 show SPF potential at 40 ppm and chalcone 3 at 20 ppm (SPF 19.47 ± 0.46). ADMET analysis using the pkCSM tool confirmed favorable pharmacokinetic profiles and low toxicity, supporting their safety for potential applications. Additionally, density functional theory calculations provided more profound insights into molecular stability and reactivity, including electronic properties such as HOMO-LUMO gaps, further corroborating their pharmacological efficacy. These results collectively indicate that chalcones 1–3 exhibit potent antioxidant activity, adequate UV protection, and promising pharmacokinetic properties. Integrating in vitro, in silico, and DFT analyses underscores their potential as multifunctional compounds for antioxidant and sunscreen applications.

Abstract

Post-harvest management is essential because it will affect the final results. Post-harvest management products can be classified as simplicia because the stages include wet sorting, washing, draining, changing shape, drying, packaging, and storage. These steps must be paid attention to to ensure the content of active compounds in simplicia. The Bejo Bintang Toedjoe herbal garden at the Plalangan Gunungpati sub-district is managed by women from the Plalangan sub-district, both housewives and workers. Community service was implemented by preparing and making leaflets/brochures, conducting site surveys, conducting pretests and posttests, and monitoring and evaluating the practical results. This activity was attended by 25 participants, not only participants from the managers of the Bejo Bintang Toedjoe herbal garden but also residents from representatives of several community associations. The discussion session was opened after the counseling had been delivered. After the discussion session is finished, it continues with practice. The results of the pretest analysis showed that most participants still need to learn the effect of direct heating with sunlight for drying medicinal plant materials. Some participants also needed to learn that the tools used to cut medicinal plant materials were stainless steel. The posttest results showed that there was an increase in knowledge after conducting counseling and post-harvest management practices of medicinal plants.
