Abstract

Background: Hibiscus sabdariffa known as roselle, which belongs to the family Malvaceae, grows in sub-tropical and tropical region including Indonesia. Several studies have reported the benefits of roselle for health. Roselle extract has been shown to have antibacterial effect as a support for antibacterial therapy, especially for case of antibiotic resistance, and antioxidant effect which can neutralize free radicals. Purpose: The aim of this study was obtaining the minimum inhibitory concentration (MIC) and inhibition concentration (IC50) of roselle extract. Methods: Antibacterial effect of roselle extract against Methicillin-Resistant Staphylococcus aureus (MRSA) was tested in nutrient agar media using diffusion method. Antioxidant activity of roselle extract was performed by DPPH. The violet color of DPPH solutions that was reduced by roselle extract were measured using visible spectrophotometer at the wavelength of 516 nm. Result: The result of this study obtained the (MIC) and IC50 of roselle extract were 2,5% and 1251±202,32 ppm, respectively. Conclusion: It can be concluded that roselle extract have antibacterial effect against MRSA and antioxidant effect.

Abstract

The main ingredients of roselle (Hibiscus sabdariffa L.) which have antioxidant and antibacterial effect were anthocyanins, and flavonoids. Flavonoid compounds were unstable at high temperatures. Meanwhile, the processing of roselle extract powder into ready-to drink or food preparations generally requires the thermal processes. The aims of this study were to determine the effect of temperature and heating time of roselle extract powder solution on the inhibition of microbial growth of Staphylococcus aureus ATCC 25923. The method of this study uses a ready-to- process roselle powder extract sample. The roselle extract powder solution in a screw cap tube was heated at a temperature of 40°C, 50°C, 60°C, 70°C, 80°C, and 90°C on a waterbath, with heating times at each temperature were 15 minutes and 30 minutes. Anti bacterial activity of the sample solution was measured based on the ability to inhibit the growth of Staphylococcus aureus ATCC 25923 by the agar-well diffusion method. The inhibition zone of the sample was measured by caliper. The result of this study showed that inhibition zones caused by the sample that had been heated at those temperature for 15 minutes were 9.55±0.70 mm, 9.53 ±0.04 mm, 9.70±0.14 mm, 9.68±0.24 mm, 10.10±0.14 mm, and 10.25±0.21 mm. Meanwhile, after heating for 30 minutes the inhibition zones were 10.08±0.25 mm, 10.20±0.28 mm, 10.43±0.18 mm, 10.08±0.18 mm, 10.78±0.04 mm, 9.70±0.14 mm. Two ways statistical test ANOVA Randomized Control Block Design with 95% confidence level was used to prove that there was a significant difference berween the mean inhibition zones of each sample. The conclution of this research was, the temperature in the range of 40 -90 oC has no effect on the anti-bacterial activity of the sample. However, the heating time of the sample affected the antibacterial activity of the roselle extract powder solution against Staphylococcus aureus ATCC 25923.

Abstract

Fifteen Streptomyces isolates were isolated from soil in some different location on vegetable plantation at agriculture standard condition. The isolates were assessed for their antibacterial activity against Mycobacterium tuberculosis (MTB) ATCC H37RV and mycobacterial which isolated from Dr. Soetomo Hospital patients in Surabaya. The Interna-tional Streptomyces Project 4 (ISP4) and Middlebrook 7H9 (MB7H9) wwere used as growth or fermentation medium. The screening of inhibition activity was performed using turbidimetry and spot-test on agar medium. Results shown that 33.3% of the isolates (5 isolates) have anti-mycobacterial activities. The first line anti tuberculosis drug rifampicin, (RIF), ethambutol (EMB), isoniazid (INH), and pyrazinamide (PZA) were used as standards or positive controls with concentration 20 ppm. Optical density of crude fermentation broth concentrated from five isolates relatively lower than five anti-tuberculosis drug activity standard, although their activities against some microbial were similar to the stand-ard at spot-test. The most efficient isolate shown anti-mycobacterial activity was Streptomyces B10 which identified as Streptomyces violaceousniger. In addition, fatty acid methyl ester (FAME) profile of gas chromatography-mass spec-trometry chromatogram of each isolates were studied and compared to Streptomyces spp.

Abstract

Shiitake mushroom (Lentinus edodes) is one of the wood mushroom types that can be consumed as a food as well as for a medical purpose. Lentinan, a polysaccharide contained in shiitake, is well known for its use on cancer medication. Mycelium of Shiitake mushroom contains lentinan the same as other part of the mushroom like fruity body. Toxicity of the lentinan in mycelium compare to the fruity body has been first conducted by using Brine Shrimp Lethality Test (BST). Using Potato Dextrose Broth media with the growth rate of 3.88% did mycelium multiplications. Probit analysis showed that the toxicity of the mushroom’s cap, stem, and mycelium of Shiitake mushrooms is LC50 = 648.76507 mg/ml LC50 = 489.39444 mg/ml, and LC50 = 481.16941 mg/ml respectively.

Abstract

This study aims to determine the minimum inhibitory concentration (MIC) and bactericidal concentration (MBC) of the fermentation filtrate (FF) on red passion fruit pulp (Passiflora edulis Sims.) against Escherichia coli Extended Spectrum Beta Lactamase (ESBL) and Methicillin Resistant Staphylococcus aureus (MRSA). The method used was the fermentation of red passion fruit pulp for 24 h using De Man Rogosa Sharpe Broth (MRS-broth) media, and the result showed that the FF of red passion fruit pulp made a dilution series concentration of 50%, 25%, 12.5%, 6.25%. Considering this result, the MIC FF of red passion fruit cells against ESBL and MRSA has a 25% dilution concentration, while MBC has a 50% dilution concentration. This shows that red passion fruit has the potential to be developed as antibacterial material, especially to fight bacteria that are already resistant.

Abstract

Community Service Activities (PENMAS) were held in August 2018 in Kapas, Bojonegoro with a target audience of SMKN 4 communities, totaling 107 people, consisting of students majoring in Cooking, members of Dharma Wanita, Chefs from 2hotels, several UMKM Food and Beverages around the school, teachers and partisipants from other schools. The activity was also attended by the Principal, the Head of the Education Office who witnessed the signing of the MOU between the PENMAS Team and the school. One of the Departments managed by SMK 4 is Tata Boga with thelearning outcome to prepare entrepreneurs or businessmen of food products. As a vocational school that has held the ISO 9001 2008 certificate, the school is obliged to prepare graduates according to the demands of their competence. Based on preliminary studies, information was obtained that there were several problems related to food consumption and production and government regulationin the case of traded food businesses. The training materials provided in this training were: Food business production regulation (1), How to make good food (2),Ingredients additional allowable food (3), Prohibited food additives and substitute alternatives (4), Good packaging methods and techniques for food and beverage (5), Halal Assurance System (SJH) and Halal food product Certification Process (6) and Simple marketing for food business (7). Based on the evaluation of the pre-test and post-test resulted that after the training participants' understanding showed an increasing trend. Three topics that showed a significant increase in understanding were: Requirements for clean and healthy food (1), Introduction of food coloring logos (2), and Examples of halal food ingredients (3). PENMAS activities will be followed up with intensive coaching activities for students of SMKN4 which are included in the group of beginner beverage food businesses.

Abstract

Repeated use of antibiotic ampicillin sulbactam after reconstitution, therefore in storage various many factor can affect it stability such as type of solvent used, storage temperature and storage time. Objective of study is to analyze the chemical stability product of ampicillin sulbactam in repeated use with the parameters of the solvent and storage temperature Sample products were weighed and reconstituted using WFI and NS solvents, diluted ad 20 ppm and put into sterile vials. Then stored at room temperature (25-30 ° C) and refrigerator temperature (4-8 ° C). Storage is carried out for 24 hours which is divided into 4 -time series, namely 0, 1, 4 and 24 hours. Each time series is microbiological tested and after 18 hours incubation the inhibition zone is observed and its diameter measured using calipers. Result of study is at 0th hour showed a significant difference in different solvents. Samples dissolved with NS solvents have a greater inhibition zone than those dissolved using WFI. This happens because there is a primary salt effect that occurs with the influence of electrolytes (salt) or variations in the strength of ions can affect the coefficient of activity that affects the reaction rate. From the statistical analysis at the 1st and 4th hours, the storage temperature that provides significant result, storage in the refrigerator produces better results compared to storage at room temperature. At 24 hours the temperature and solvent did not give the significant result on the stability of the sulbactam ampicillin sample. Conclusion is NS solvents give better results than WFI solvents. Samples are more stable if stored in a refrigerator. After 4 hours based on the above study the sample no longer meets the requirements of Indonesian Pharmacopoeia 5th edition

Abstract

The sodium diclofenac realeased from transdermal patch of combination of EC N–20 and PVP K–30 with ratio 6:4 with plasticizer PEG 400 5% (formula 1), PEG 400 10% (formula 2) and PEG 400 20% (formula 3) was investigated. The percentage of moisture contents was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using fluorescence microscope and scanning electron microscope (SEM). Released test using dissolution apparatus were carried out in 500 mL of phosphate buffer saline pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. Results were analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0,05), showed that that the combination of EC N–20 and PVP K–30 at ratio 6:4 with plasticizer PEG 400 5% was the best choice for manufacturing transdermal patch based on physicochemical and the release profile. ABSTRAK Telah dilakukan penelitian pelepasan sodium diklofenak dari transdemal Patch kombinasi matriks EC N–20 dan PVP K–30 dengan rasio 6:4 dan plasticizier PEG 400 5 % (formula 1), PEG 400 10% (formula 2), dan PEG 400 20 %. Persentase kadar air dihitung sebagai perbedaan antara berat awal dan akhir. Homogenitas dari permukaan Patch di determinasi menggunakan Mikroskop flourescence dan SEM. Uji pelepasan dilakukan menggunakan alat uji dissolusi dalam 500 mL larutan salin fosfat buffer pH 7.4 ± 0.05 pada suhu 37 ± 0.5°C dengan kecepatan pengadukan 50 rpm. Hasil di analisis menggunakan program statistik SPSS analisis one way variance (ANAVA) dengan tingkat kepercayaan 95% (α = 0,05), menunjukkan bahwa kombinasi dari EC N–20 and PVP K–30 pada rasio 6:4 dengan plasticizer PEG 400 5% merupakan pilihan terbaik untuk industri Patch transdemal berdasarkan fisikokimia dan profil pelepasan.

Abstract

ABSTRAK Pelepasan natrium diklofenak dari Sediaan Transdermal Patch Tipe Matriks dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 telah diteliti. Pada penelitian ini sediaan transdermal patch tipe matriks yang mengandung natrium diklofenak dibuat dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 pada perbandingan yang berbeda ( 9:1 (Formula I); 8:2 (Formula II); dan 7:3 (Formula III)). Persentase (%) moisture content dihitung dari perbedaan berat relatif pada penimbangan akhir. Uji homogenitas permukaan patch dilakukan dengan menggunakan metode Scanning Electron Microscopy (SEM). Uji disolusi dengan media larutan dapar fosfat salin pH 7,4 ± 0.05 sebanyak 500,0 mL yang terlebih dahulu dipanaskan sampai mencapai temperatur percobaan 37 ± 0.5°C diaduk pada kecepatan 50 rpm. Hasil dianalisis menggunakan SPSS dengan metode ANOVA one way dengan tingkat kepercayaan 95% (a = 0,05). Berdasarkan penelitian ini, penggunaan polimer kombinasi antara etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 dapat meningkatkan fluks pelepasan natrium diklofenak dari sediaan patch natrium diklofenak. Perbandingan 7:3 dari etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 adalah kombinasi yang terbaik untuk sediaan patch natrium diklofenak karena konsistensi bentuk fisik serta nilai fluks pelepasan dan penetrasi yang dihasilkan lebih besar dari perbandingan lainnya.
