Abstract

The aim of this study was to optimize the use of Ca-Alginates beads as a drug reservoir in the patch system with and without freeze-drying process. Meloxicam, an anti-inflammatory non-steroidal drug, was used as a drug model. Ca-alginate beads were prepared by ionic gelation method using polymer alginate and cross-linker CaCl2. The beads were evaluated for its melting temperature, FTIR spectra, and entrapment efficiency. The FTIR study showed the loss of guluronic peaks of alginate caused by a cross-linking process with Ca2+, characteristics peaks of meloxicam still appeared indicating the compatibility of the drug with the polymers used. The using Ca-alginate beads without freeze-drying process as a drug reservoir resulted in a very wet patch that was difficult to dry, cracked surface, and pharmaceutically not acceptable. Meanwhile, patch using Ca-alginate beads with the freeze-drying process as a drug reservoir demonstrated satisfactory characteristics with minimum weight variation among the patches that lead to giving uniformity in the drug content.

Abstract

This research formulate erythropoietin-alginate microspheres and to evaluate characteristics of erythropoietin-alginate microspheres at different amount of drug using aerosolization. Amount of erythropoietin are 10.000 IU (F1); 20.000 IU (F2); 60.000 IU (F3). The mixture of erythropoietin-alginate was sprayed into CaCl2 and was stirred at 1000 rpm for 30 minutes. Formulas resulted spherical shape of microspheres. The size of microspheres was 2.77 µm for F1; 3.89 µm for F2; and 4.42 µm for F3. The results of swelling index showed that swelling index of microspheres increased by increasing the concentration of erythropoietin. The results were in accordance with the size of the microspheres that increased with increasing concentration of drug. The yields of microspheres obtained were respectively 91.92%; 87.53%; 86.50% for F1, F2 and F3. It can be concluded that the particle size of microspheres, swelling index increased by increasing concentration of erythropoietin. In contrast, yield of microspheres decreased by increasing drug concentration. In conclusion, formula of microspheres were potential in terms of characteristics and may recommend for further in vivo study.

Abstract

The study was designed to investigate the optimation of preparation of transdermal patch sodium diclofenac matrix type. In this study, matrix-type base containing diclofenac sodium were prepared using polymeric combination of EC N-20  and PVP K-30 in different ratios ( 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) and 6:4 (Formula IV). The moisture content was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using Scanning Electron Microscope (SEM) Released test using dissolution tester were carried out in 500 mL of  phosphate buffer salin pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. The concentration of sodium diclofenac released was determined using spectrophotometric method at l 276 nm. Data was analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0.05).  It is concluded that the polymeric combination EC N-20 and PVP K-30 in composition 7:3 (Formula III) was the best formulation. ABSTRAK Telah dilakukan penelitian tentang optimasi sediaan transdermal patch natrium diklofenak tipe matriks. Pada penelitian ini, basis matriks mengandung natrium  diklofenak dibuat dengan kombinasi polimer  etilselulose (EC) N-20  dan polivinilpirolidon (PVP) K-30 dengan berbagai perbandingan yaitu 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) dan 6:4 (Formula IV). Moisture content (MC) ditentukan dengan cara berat awal dikurangi berat akhir dan dibagi dengan berat akhir. Homogenitas permukaan patch ditentukan dengan menggunakan Scanning Electron Microscope (SEM). Uji pelepasan natrium diklofenak dilakukan dengan alat disolusi dan menggunakan dapar fosfat salin  pH 7,4 ± 0.05 sebanyak 500,0 mL, kecepatan pengadukan 50 rpm dan percobaan dilakukan pada suhu 37 ± 0,5°C. Kadar natrium diklofenak yang terlepas ditentukan dengan metoda spektrofotometer pada panjang gelombang (l) 276 nm. Data yang diperoleh dianalisa secara statistik menggunakan program SPSS dengan derajat kepercayaan 95% (α = 0,05).  Kesimpulan yang didapatkan adalah komposisi polimer EC N-20 dan PVP K-30 dengan perbandingan  7:3 (Formula III) adalah formula yang terbaik.

Abstract

The use of microspheres nowadays is important in the pharmaceuticals formulation. The aim of the present research was to evaluate physical characteristics of ovalbumin-loaded alginate microspheres using aerosolisation technique at various concentrations of alginate and BaCl2. Alginate microspheres were characterized in terms of encapsulation efficiency, yield, particle size, surface morphology and protein integrity. The comparative study between concentrations of BaCl2 and alginate polymer was investigated. Increasing concentration of alginate polymer from 2.5 to 3.5% reduced their particle size and formed smoother and spherical microspheres. Increasing Ba2+ concentration, simultaneous increased the encapsulation efficiency and yield. Scanning electron microscope (SEM) photomicrographs revealed that with the increase in the electrolyte concentration the density of the cross link is also increased. Smoother surface was demonstrated when the electrolyte concentration is increased from 0.5M to 0.75M. Formula prepared using 0.75M BaCl2 and 3.5% alginate polymer resulted the highest encapsulation efficiency (92%), the highest microspheres yield (73%) and the smallest microspheres size (3.73µm). Most of the formulations maintain the integrity of ovalbumin after 2 hours incubation in pH 1.2 followed by 8 hours incubation in pH 7.4. The different formulations produced different physical characteristics of ovalbumin-loaded alginate microspheres.

Abstract

ABSTRACT: The study was conducted to investigate the effect of composition of surfactant (Span 80 and Tween 80) and cosurfactant (butanol) on characteristics and ovalbumin entrapment in the microemulsion water in oil (w/o).The composition of surfactant (Span 80 and Tween 80) and cosurfactant (butanol) used were 6:3, 7:3 and 8:3. Evaluation was conducted on the surface tension, viscosity, droplet size, pH and the efficiency of ovalbumin trapping in microemulsion system. Results were analyzed by statistic using one way analysis of variance (ANOVA) with 95% confidence level (α = 0,05). From the results it was concluded that the composition of surfactant and cosurfactant 7:3 resulted in the best microemulsion. Abstract

Abstract

Hand sanitizer gel is one of the alternative methods for hand hygiene when soap and water are not available. Binahong leaf (Anredera cordifolia) is a natural herb that has phenolic, flavonoids, saponins, and the steroid-terpenoids compound, which is potentially used as the active ingredient in the hand sanitizer gel. The primary purpose of this research was to determine the antibacterial potential and physicochemical properties of a hand-sanitizer gel containing binahong leaves’ infusion against Staphylococcus aureus. The efficacy of this formulation was tested against a bacterial strain using the disc diffusion method and determined by measuring the inhibition zone of each preparation. The testing of physicochemical properties includes the organoleptic test, pH, viscosity, and gel spreadability. The physicochemical characteristics of the gel showed excellent properties for all formulas. The antibacterial test showed that hand sanitizer gel containing an infusion of binahong’s leaves has an inhibition zone (6.02 mm; 6.24 mm; 6.55 mm), almost close to a positive control (9.15 mm). In conclusion, the formulated hand sanitizer gel possesses the antibacterial potential to inhibit S. aureus.
