Abstract

Abstract Aeromonas hydrophila is a motile Aeromonas bacteria that cause Septicemia (MAS) or the disease kwon as red spots (Plumb, 1994). These bacteria attack the various types of freshwater fish such as African catfish, carp and prawns. Treatment of MAS is still a lot of use antibiotics, whereas antibiotic a lot side effects. This study tries for find an alternative use  antibacterial substances using natural chemicals derived from medicinal plants of leaves extracs red betel (P. crocatum) which is antibacterial is a volatile oil, flavonoids, alkaloids and tannins.  The purpose of this study was to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of leaves  extracts red betel (P. crocatum) against of bacteria A. hydrophila. The research was conducted on July-August 2011, at the faculty of veterinary medicine, Airlangga University and Organic Chemistry Laboratory Faculty Of science and Technology Airlangga University. The method used in this study is an experimental method, using completely randomized design (RAL) with 11 treatments and three replications. Variables observed in this study is the minimum inhibitory concentration (MIC) and Minimum Bactericidal Concentration (MBC) of leaves extracts red betel (P. crocatum). The observations were analyzed using analysis of variance (ANAVA). The results showed that extracts leaves red betel (P. crocatum) has efficacy against the bacteria A. hydrophila in vitro. Based on the results of MIC testing, leaves exracts of red betel (P. crocatum) inhibits growth of bacteria A. hydrophila at a concentration of 12,5% and based on test results of MBC, leaves extracs of red betel (P. crocatum) effectively kills the bacteria A. hydrophila at a concentration of 25%. It can be concluded that the concentration of antibacterial substances contained in red betel able to inhibit and kill bacteria A. hydrophila. This study recommends further to determine the effects of leaves extracts  red betel (P. crocatum) A. hydrophila against bacteria in vitro, so that can know the effectiveness of leaves effects of leaves extracts  red betel (P. crocatum) in suppressing populations of bacteria A. hydrophila

Abstract

Abstract Giving antibiotics continuously can cause A. salmonicida smithia become resistant and residues of antibiotics may pollute the water environment. One alternative to using traditional medicinal plants that are antibacterial. Advantages of using traditional medicinal plants are relatively more secure, easily available, inexpensive, does not cause resistance, and relatively harmless to the surrounding environment. The content of natural chemicals from cucumber fruit is known to have antibacterial effects namely, flavonoids and phenols. The purpose of this study to determine ability the cucumber fruit juice inhibit bacterial growth A. salmonicida smithia and to determine the best concentration to inhibit bacterial growth A. salmonicida smithia with ampicillin drugs for comparison by in vitro. This research was conducted in the Bacteriology Laboratory of Fish Quarantine Center of Sidoarjo and Human Genetic Laboratory the Institute of Tropical Disease Centre, Airlangga University, Surabaya. This research use done eksperimental method by in vitro, with a large measure of cucumber fruit juice inhibition zone around the existing paper disc and comparing it to a large zone of inhibition using ampicillin. Concentration dilution series cucumber fruit juice used during the research, began 2 gr/ 2 ml to the concentration of 0,0625 gr/ml. Result of research show at concentration 0,125 gr/ml still has ability to pursue bacterium A. smithia salmonicida, so that can be anticipated that effective cucumber fruit juice if used as by drug, because at small concentration can to pursue bacteria A. smithia salmonicida by in vitro.

Abstract

Abstract Bacteria are single-celled organisms, reproduce by dividing themselves (asexual) that reproduce by way of extending the cell nucleus followed by cell division called binary fission (Prajitno, 2005) and can only be viewed with a microscope because of their small size (Dwijoseputro, 1989 .) One of the bacteria that cause the greatest failures in the cultivation of freshwater fish and sea water is furunculosis (presence of a number of furuncles at the same time) caused by the bacterium Aeromonas salmonicida (Cipriano and Bullock, 2001). A. salmonicida is divided into several subspecies, among others, A. salmonicida subspecies salmonicida, subspecies masoucida, subspecies achromogenes and subspecies smithia (Hiney and Olivier, 1999). Fish infected with these bacteria will show symptoms such as fish body color becomes darker, the ability to swim down, resulting haemorrhagic septicemia, the abdomen looks bloated, the eyes and slightly protruding damaged fish (exopthalmia), all damaged fins, and gills become pale and damaged (Buller, 2004). The method used in the research is experimental method. This study aims to determine whether fruit majapahit juice (C. cujete L.) has antibacterial power against bacteria A. salmonicida and find out whether or not sensitive majapahit juice (C. cujete L.) in inhibiting the growth of bacteria A. salmonicida with standard comparator antibiotic kanamycin. Research conducted at the Laboratory of Bacteriology Hall Fish Quarantine Airport Highway Ir. H. Juanda Street No.23, Semambung Village Sedati- Sidoarjo District, East Java Province in February 2011. The results showed that the juice majapahit (C. cujete L.) could inhibit bacterial growth of A. salmonicida against in vitro, but when compared with standard antibiotic kanamycin 30 µg still relatively insensitive to inhibit bacterial growth of A. salmonicida.

Abstract

AbstractCurrently catfish is experiencing a fairly rapid growth, this is because the catfish is one of the important commodities in the fishery. Efforts to increase catfish production is to improve the quality of feed. The aim of this study is to observe effect of snail meat meal for growth rate, food conversion ratio and survival rate of catfish seeds. This research method is experimental with a Completely Randomized Design (CRD) with five treatments and four replications. The treatments were used: (A) 0% snail meat meal, (B) 10% snail meat meal and 30% fish meal, (C) 20% snail meat meal and 20% fish meal, (D) 30% snail meat meal and 10% fish meal and (E) 40% snail meat meal. The main parameters measured were growth, feed conversion ratio and survival rate. Supporting parameters measured were water quality. Analysis of the data used is Analysis of variance (ANOVA) and to know the difference among treatment using Duncan's Multiple Range Test. The results of this study showed that use of snail meat meal in feed formulation stastistically difference (p<0.05) for growth and survival rate of catfish seeds but not statistically difference (p>0.05) for feed conversion ratio. Water quality in maintenance media seed catfish is the temperature 0range between 26-31C, pH 7-8 and dissolved oxygen was 7.2-8.0 mg/L.

Abstract

Abstract The purpose of this research was to know the effect of kinds of feeding on growth and survival rate of spiny eel. This research used experimental method and Completely Random Design method with six treatment and each treatment was replicated four times. The treatments are : artificial feed (A), nila fish (B), shrimp (C), combination of artificial feed with nila fish (D), combination of artificial feed with shrimp (E) and combination of nila fish with shrimp (F). The main parameter observed are specific growth rate, growth of absolute length and survival rate. The support parameter is water quality. The data analiysis of this research used a Analysis of Varian (ANAVA) and to determine the best performed used Duncan's Multiple Range test. The result of this research showed that given kinds of feeding was not significantly difference (p>0,05) to specific growth rate, growth of absolute length and survival rate of spiny eel. The average value of growth of absolute length, specific growth rate and survival rate of each treatment are treatment C (2,5062 mm), treatment C (0,9375 %BT/hari) and treatment C (71,6495%).

Abstract

Abstract The development of fish culture is strongly influenced by hatchery technology, especially in the stock of fish seed. Often, the problem arises in the stock of seeds, due to the maturation of gametes, broadstock fish sometimes do not happen simultaneously, one of alternative solution to the problem through the application of reproductive biotechnology, is preservation of sperm (Dirjennak, 2007). Preservation sperm is to optimize the male broadstock (Dirjennak, 2007). This storage process requires diluent and cryoprotectant material that could sustain sperm motility (Zaenab, 2007). Diluent material is used to reduce the activity of spermatozoa that inhibit energy usage and prolong the life of sperm (Rustidja, 2000). The use of honey as a diluent material is expected to support the vitality of spermatozoa. Material of honey there are various combinations of materials a simple sugar and salt ions. With the combination of basic ingredients and simple sugar salt ions spermatozoa can produce an energy source so it can defend itself and can fertilize an egg cell after Cryoperservation process. The purpose of this research is to know the effect of adding extender honey in enhancing sperm motility and viability of fish comet after the freezing process and to determine the best dose of honey in the process of freezing sperm comet fish (Carassius auratus auratus). This research was conducted at the Laboratory Center for Artificial Insemination Singosari. The design use Completely Randomized Design (CRD) followed by Duncan's multiple range test. Test materials in this research is sperm comet fish are packed in mini straw and stored in liquid nitrogen container with 9 treatments and 3 replications. Media diluent used was physiological NaCl is added to honey, glucose, fructose, and Tris Aminomethane. Honey dosages of the experiments were 0.3% (A); 0.4% (B), 0.5% (C), 0.6% (D) and 0.7% (E) and 0.05% glucose (KG); fructose 0.05% (KF), Tris Aminomethane (KT) and without the addition of physiological NaCl (KN). The main parameters of observed percentage of live sperm and duration of motion. The supporting parameters measured were fresh sperm concentration, percentage of live sperm fresh, long fresh sperm, pH, volume and color of sperm. The results of the research shows that the addition of honey with different dosage at diluents material physiological NaCl gived significant effect (P <0.05) against the percentage of sperm motion and long life. Average percentage of long life and highest sperm obtained by treatment of the addition of honey 0.6% (D), ie 63.89% and 101.33 seconds. Average lowest percentage of life gained by treatment of honey 0.3% (C) that were 39.67 second while the lowest was in motion treatment control without the addition of (KN) that were 28.33 seconds.

Abstract

AbstractShrimp are susceptible to a wide variety of pathogens, one of each were viruses. Whitespot Baculovirus (WSBV) is one of virus attack in vannamei. Shrimp had been infected by WSBV will showed high mortality. One of Strategies for prophylaxis and control of WSBV is enhancement of disease resistant by using vaccines. Recently, quasi-immune response have been reported by which Penaeus japonicus surviving from WSBV infections possess a resistance against challenge WSBV (Venegas et al., 2000). This study was used Lithopenaeus vannamei that been vaccination with inactived-formaline WSBV virus cell or it called Whole Cell Killed Virus (WCKV) to know its responses to WSBV. The aim of this study was to know the effect Whole Cell Killed Virus vaccine to survival rate vannamei shrimp (Lithopenaeus vannamei) infected by Whitespot Baculovirus (WSBV). This study used descriptive study. The efficacy of vaccines made of inactivated WSSV with different dose. The dose are P1 (dose 0,01µg/ml each shrimp), P2 (dose 0,02 µg/ml each shrimp), P3 (dose 0,03 µg/ml each shrimp) dan P4 (non-vaccine (kontrol) injected by PBS). Primary parameter was survival rate (%). Secondary parameter was water quality which of temperature, pH, salinity and dissolved oxygen. The result showed that survival rate of L. Vannamei with different dose of WCKV vaccines was increases.

Abstract

Abstract Daphnia magna has good nutrition to carry on egg produce and fry of fish cupang, maskoki, oscar, tetra, and also can used for food source of fry and seed. Daphnia magna not only supply from natural environment, cause their growth deppend on fertilizer as food and environment condition. The other side: high population density, limited of food source low temperature and shorter irradiation can produce dorman egg (ephippia). The dorman eeg can use for Daphnia magna stock which way by hatching. The factor that can be influence for ephippia hatch are temperature and density. Based on this fact, a research about influence of different temperature and ephippia density for ephippia hatch of Daphnia magna. The purpose of this research was to know the influence of different temperature and ephippia density for ephippia hatch of Daphnia magna. This research has experimental characteristic and use Factorial Completely Randomized Design with three experiments of temperature (A factor), three experiments of density (B factor) and three times replicated. There are 9 kinds of combination between density and temperature. The parameter which checked in this research is degree of hatch of ephippia Daphnia magna and water quality that is dissolve oxygen rate, NH3 and of pH hatch media. The result of the analysis showed that the temperature and density influence of hatching Daphnia magna ephippia. The best result can be used for hatching Daphnia magna ephippia is temperature at 25oC with density 50 ephippia /100 ml giving highest result for the hatch of ephippia Daphnia magna. There are interaction betwen temperature and density for hatching.

Abstract

Abstract African catfish (Clarias gariepinus) is one of fish consumption use to fulfill animal protein. The fulfillment need of animal protein with aquaculture effort, where as in the aquaculture effort will larvae quality increased. Increasingly of larvae quality with natural food had been adding feed supplement. Natural food to used is Daphnia spp. and then food supplement to used is viterna. The viterna will increase Daphnia nutritions, because of the viterna contains many nutritions to survival and growth of African catfish. The aim of this study was to know the influence of Daphnia spp. enrichment with viterna to survival and growth of African catfish (Clarias gariepinus) larvae. The rearing of larvae during 40 days. This study used Complete Random Design. The treatment were A (present Daphnia spp. without viterna), B (Daphnia spp. enrichment with viterna 10 ml/L dose), C (Daphnia spp. enrichment with viterna 10 ml/L dose), D (Daphnia spp. enrichment with viterna 10 ml/L dose), E (Daphnia spp. enrichment with viterna 10 ml/L dose) dan F (Daphnia spp. enrichment with viterna 10 ml/L dose), each treatment with 4 replications. The result show that viterna applications to Daphnia was significantly influenced (p<0,05) to survival African catfish larvae. The highest survival was E treatment (7,25 %). The viterna leaf to Daphnia spp. Was significantly influenced (p<0,05) to specific growth rate African catfish larvae. The highest specific growth rate was B treatment (1,885 %BW/day).
