Abstract

Capability of Rhodobacterium bacteraceae in producing biosurfactant were investigated. Biosurfactant has been produced using medium of Mineral Salt Medium with two sources of carbon, which is crude oil (RB-CO) and waste coconut oil (RB-WCO) as carbon sources. Biosurfactant extent was determined by the yield of biosurfactant and the surface tension reduction. Biosurfactant were investigated for their efectiveness on PAHs biodegradation. The results showed that R. bacteraceae can produce biosurfactant, and showed the highest biosurfactant yield (2,791 g/l) when grown on RB-WCO, while we found 2,122 g/l yield on MSM-CO after 72 hours. The best biosurfactant activity was obtained when using waste coconut oil as carbon source, resulted in surface tension reduction up to 29,17 mN/m. Addition of 50 mg of biosurfactant per 200 ml of the mineral salt medium containing 50 ppm PAHs significantly enhanced the PAHs degradation indicating that the organism utilized crude oil as carbon source. Our concluding remarks suggest that biosurfactant from R. bacteraceae when grown on waste coconut oil substrate can be used as an effective agent to be applied on site contaminated with PAHs by enhancing biodegradation rate.

Abstract

A study was carried out to investigate the concentrations and contamination levels of heavy metals in the sediments of Segara Anakan, Cilacap  for three heavy metals (Cd, Pb, and Cr). sediments samples were collected from six sites during February 2013. The samples were analyzed to determine their heavy metals, pH and total organic carbon (TOC) content. Environmental assessment  of  sediments  pollution  by  heavy  metal  was  carried  out  using Contamination Factor (CF), Enrichment Factor (EF) by comparing to CCME,   ANZECC and NOAA targets for sediment quality Guidelines. The results of this simple comparison revealed that, the Segara Anakan  sediment concentrations of heavy metals are within the permissible limits of standards for Pb. CF value of Pb, Cd, and Cr (0.86, 10.8, dan 3.3)   indicated no contamination for Pb (0.86), very strong contamination for Cd (10.8), and moderately to strong contamination for Cr. Calculated EF of the Pb, Cd, Cr  in sediment were 1.50, 18.8, dan 5.7 indicating  deficiency to low enrichment  for Pb anrophogenically, and significant enrichment for Cd and  Cr. According to the CF and EF value, Segara Anakan may face a severe heavy metal pollution contamination problem in the future.Keywords: sediments, contamination, anthropogenic

Abstract

The excessive consumption of artificial dyes can lead to negative effects on human body. Thus, the invention of natural dyes, such as carotenoids, is needed in order to reduce the negative impacts. Carotenoids are yellow, orange, and reddish yellow pigments produced by plants, animals, algae, and microorganisms. This study was aimed to analyze the carotenoid pigments of mangrove sediment bacteria in Segara Anakan, Cilacap, and to identify species of bacteria that can produce carotenoids. Carotenoids were analysed by using Thin Layer Chromatography (TLC) and UV-Vis spectrophotometer. Meanwhile molecular identification of bacteria was carried by 16S rDNA PCR and DNA sequence was analysed through a program called Basic Local Alignment Search Tool (BLAST). The kinship of bacteria was shown in Phylogenetic tree by using Bioedit and MEGA 5 software. Qualitative analysis by using TLC produced several pigments like: β-carotene, β-isorenieraten, lycopene, flavonoids, chlorophyll a, chlorophyll b and a feofitin with Rf value of 0.36-0.95. Quantitative analysis showed that bacteria KH (greenish yellow), KT (dark yellow) and KM (light yellow) produced carotenoids at 95.30 mg.g-1, 110.34 mg.g-1 and 25.349 mg.g-1. KH, KT and KM were suspected to be bacteria known as Streptomyces chartreusis, Bacillus megaterium and Streptomyces chromofuscus with similarity of 99%. Mangrove sediment bacteria had the potential of producing carotenoids as an alternative of eco-friendly natural dyes.

Abstract

Batik Industry has been developed in Banyumas regency, Central JavaIndonesia. This industry produced liquid waste as the side of production process.The liquid waste decreased the water quality surrounds it. One of the pollutantwhich brought trough the liquid waste was heavy metal chromium (Cr) whichknown toxic to the organism in the water. Therefore, there should be preventionaction to minimize the bad impact of liquid waste. One of method which could beuse to prevent the water quality was by phytoremediation. There were three kindsof aquatic plant in this research; Eceng gondok (Eichornia crassipes), kayu apu(Pistia stratiotes) and ganggang (Hydrilla verticillata). The aim of using aquaticplant was to observe the ability of aquatic plant to decrease chromium level in thewater and decide which was the most effective. It was use completely randomizeddesign with 4 treatments and 6 repetitions, which will analyze by ANAVA andcontinued with LSD. The result showed that aquatic plant could be used asremediator agent to heavy metal Cr. Eceng gondok had 49.56%, kayu apu had33.61% and ganggang had 10.84% on decreasing Cr level in liquid waste of batikindustry. It can conclude that Eceng gondok was the most effectivephytoremediator agent on decreasing Cr level in liquid waste of batik industry.

Abstract

The study aimes to examine the accumulation level of selected heavy metal Cd in various of common mud crab (Scylla spp.) samples from Segara Anakan Lagoon-Cilacap. Mangrove crab and sediment samples were taken from Segara Anakan (108046’–109005’E; 7034’–7048’S) and were analyzed using Atomic Absorption Spectrophotometer (AAS). The Cd extent in Scylla spp. varied from 0.084 - 0.273 ppm which still below the European Commission (2006) threshold value (0.5 ppm). On the other hand Cd concentration in sediment for most stations value (0.625 - 1.635 ppm) have exceeded the Interim marine sediment quality guidelines (ISQGs value; 0.7 ppm) except for station 2 and 4. Concerning the sediment, to interpret and assess the contamination status four indices were used, namely Contamination Factor (CF), Enrichment Factor (EF), Pollution Load Index (PLI), and Geoaccumulation Index (Igeo). The results show the Segara Anakan sediment value for CF varied 3.125 - 8.175, EF was 50.481, PLI was 5.381 and Igeo varied 0.319 - 0.736. The finding result shows the Segara Anakan sediment could be considered as moderately polluted.Keywords: cadmium, contamination, scylla, sediment.

Abstract

Polycyclic Aromatic Hydrocarbons (PAHs) have the potential to contaminate the coastal area of Plawangan Timur, Cilacap, Indonesia. Therefore, this research aims to determine the composition, distribution, source, and contamination level of PAHs in this region. Sediment samples were taken at seven stations. The research findings indicate that among the 16 types of PAHs, the proportion of each PAH varied between 3-12%. Notably, Benzo(a) Pyrene (BaP) had the highest proportion at 12%, while Naphthalene had the lowest proportion at 3%. According to the distribution pattern, PAHs with a high molecular weight (HMW) are more prevalent than those with a low molecular weight (LMW). The total amount of PAHs ranged from 185 to 1289.9 mg.kg-1 (dw), with an average of 1016.97 mg.kg-1 (dw). Using a diagnostic ratio, it was determined that the origin of the PAHs was either petrogenic, pyrogenic, mixed petrogenic and pyrogenic, or pyrolytic. As a result, Plawangan Timur's degree of pollution was classified as a very high polluted. The average concentration value of each PAHs is higher than the values for the Effect Range Low (ERL) and Effect Range Median (ERM) ranges, indicated there is a significant ecological risk that could have negative consequences on marine life and the ecosystem. It is urgently to handle PAHs pollution due to their relatively high content in sediments. This can be achieved by improving the management of wastewater treatment plants (WWTPs) for industry, optimizing the role of mangrove forests in reducing pollution in the waters, as well as building and increasing collaboration between related agencies in the prevention of marine oil spills.

Abstract

Sungai Pelus merupakan salah satu sungai yang terletak di Kabupaten Banyumas, sungai yang memiliki panjang ±28 km dan melalui 8 wilayah ini mengalir dari lereng Gunung Slamet sampai Sungai Serayu. Penelitian ini mengkaji konsentrasi dan tingkat pencemaran logam berat Cd dan Cr pada Sungai Pelus Kabupaten Banyumas, Jawa Tengah. Logam berat yang dihaslikan oleh industry ataupun limbah rumah tangga memiliki dampak yang berbahaya bagi Kesehatan manusia dan lingkungan. Kandungan Cr dan Cr secara alami terdapat dalam batuan, tanah, dan air kemudian terdistribusi pada media air dan terakumulasi dalam sedimen dan biota. Metode yang digunakan pada penelitian ini adalah metode survei dengan teknik pengambilan data yaitu Purposive Random Sampling, penelitian ini terdiri dari 7 stasiun mulai dari hulu hingga hilir sungai serayu. Data dibandingkan dengan baku mutu kemudian dianalisis dan dihitung menggunakan Nemerrow Index. Kandungan Logam Berat Cd pada matriks air tertinggi bernilai 25,59 mg/l pada stasiun 4 yang berada di tengah sungai pelus dengan aktivitas warga yang cukup padat, sedangkan kandungan Cr pada matriks air tertinggi bernilai 11,05 mg/l pada stasiun 6. Tingkat pencemaran pada sungai pelus melebihi status baku mutu logam berat pada matriks air dengan nilai rerata 16,119 mg/l dan sungai ini dapat dikategorikan sebagai sungai yang sangat tercemar.

Abstract

Green mussel (Perna viridis) is one of the spesies in Bivalve class which filter the food particles from their surroundings. As filter feeder, it captured anything including plankton, detritus, and even heavy metals. Heavy metals content in green mussels could become dangerous due to the bioaccumulation process. This research aimed to determine the levels of heavy metals Cadmium (Cd) and Chromium (Cr) not only in the tissues of green mussels but also in the sediments of Sawojajar waters, Brebes; current study also determined the feasibility of green mussels for consumption in the perspective of food safety. The method used in this research was a field survey method which conducted in June 2022. The analysis method used AAS refers to the American Public Health Association (APHA) with laboratory tests conducted at the Productivity and Aquatic Environment Laboratory of IPB. The results showed that the content of heavy metal Cd in green mussel samples was at 0.19 mg.kg-1 which was meet the threshold concentration. Whereas, the heavy metal Cr in green mussels’ tissue exceeded the threshold of chromium concentration at 0.21 mg.kg-1. In addition, the heavy metal content of Cd and Cr in sediment were still meet the threshold concentration at 3.2 mg.kg-1 and 0.13 mg.kg-1 respectively. In terms of food safety, the EDI (Estiate Daily Intake) value based on the results of heavy metals Cd and Cr concentrations was 0.11 and 0.13 µg.kg-1.day-1 respectively. The result indicated that these heavy metals concentration had low risk in causing health issue. In the same way, the target hazard value (THQ) of Cd and Cr were not exceeded the RfD (Reference Dose) at 0.109 and 0.036 respectively. These value of THQ for both Cd and Cr indicated low risk in causing cancer and the green mussels are meet the food safety standard

Abstract

Sungai Pelus merupakan sungai yang terdapat di Kabupaten Banyumas, Jawa Tengah. Tingkat aktivitas masyarakat yang ada di sempadan Sungai Pelus cukup tinggi seperti permukiman, perkebunan, sawah, pabrik kayu, dan tambang pasir yang berpotensi menghasilkan limbah padat maupun cair yang akan bermuara ke badan sungai dan sampai ke sedimen sehingga membuat sungai tersebut tercemar, parameter pencemaran tersebut adalah logam berat. Penelitian ini bertujuan untuk mengetahui perbedaan kandungan Cd dan Cr pada sedimen antar stasiun, distribusi kandungan Cd dan Cr pada sedimen, serta apakah kawasan Sungai Pelus tercemar oleh Cd dan Cr berdasarkan Contaminant Factor (CF), Enrichment Factor (EF), Indeks Geoakumulatif (Igeo) dan Pollution Load Index (PLI). Metode yang digunakan yaitu metode survey dengan teknik pengambilan sampel purposive random sampling pada tujuh stasiun. Kandungan logam berat Cd dan Cr dianalisis menggunakan Spektrofotometri Serapan Atom (SSA). Hasil yang didapat menunjukan adanya perbedaan kandungan Cd dan Cr pada sedimen antar stasiun. Sungai Pelus terdeteksi memiliki nilai CF dengan kategori kontaminasi tinggi untuk Cd dan rendah untuk Cr, EF dengan kategori sedang untuk Cd dan tidak ada faktor pengayaan untuk Cr, Igeo dengan kategori tercemar sedang untuk Cd dan tidak tercemar untuk Cr serta PLI dengan kategori tercemar untuk Cd dan tidak tercemar untuk Cr.

Abstract

Muara Kali Ijo is the place where rivers flow in Kebumen Regency, which carry a lot of domestic wastes resulting from sanitation activities, namely detergent. The study aims to determine the quality of Muara Kali Ijo Waters based on MBAS concentration (Methylen Blue Active Substances). MBAS is a standard method for determining the concentration of anionic surfactants in detergents. This study uses descriptive methods with purposive random sampling techniques. Sampling was carried out on March 11, 2023 with a total of five stations. The results of the analysis of the MBAS concentration in the BBSPJPPI laboratory ranged from 0.031 – 0.064 mg/L and the measurement of water quality parameters was still relatively good, below the quality standard threshold. PCA results show that there is a negative correlation between MBAS concentration and water quality parameters. Productive anthropogenic activities around the area have a potential to contribute detergent waste from washing activities and increase MBAS concentrations.
