Abstract

Rice washing water is household waste that is often wasted and is no longer used, even though rice washing water still contains nutrients in the form of vitamins A, C, B1, carbohydrates, phosphorus, potassium, magnesium, nitrogen and iron. Vitamin B1 (thiamin) dissolves in water when washing rice. Vitamin B1 contained in the water used to wash rice has a role in metabolism. The nutritional content in rice washing water can be used to support the growth and development of chili plants. In this study, rice washing water was used as liquid fertilizer for chili plants. The research design was a completely randomized design with the holding variable being the concentration of rice washing water (0%, 25%, 50% 75% and 100%) with 3 repetitions each. The data obtained were analyzed using SPSS Anava followed by Duncan's test. The results showed that the concentration of rice washing water at 100% was the treatment that had the best effect on the growth of chili plants (Capsicum frutescens).

Abstract

Lactic Acid Bacteria (LAB) are bacteria that are widely found in traditional fermented products from Indonesia. Processed Indonesian products containing LAB are made by involving the process of fermenting foodstuffs with starch-based ingredients such as glutinous tape. Processed tape made from glutinous rice fermented by yeast. This study aims to determine the number of isolates contained in fermented sticky rice and find out how the characteristics of LAB isolates in fermented sticky rice are circulating in Citra Niaga Market, Jombang. This research is expected to be the basis for further research to determine the potential of LAB and diversity in glutinous tape. In this study, BAL was isolated from 3 brands of sticky tape circulating in the Legi Jombang Market using the isolation method with a multilevel dilution technique and showed a different number of microbes in each sample. In sample 1 there were 1.7 × 106 CFU/ml, in sample 2 there were 3.0 × 107 CFU/ml while in sample 3 there were microbes with a total of 1.14 × 105 CFU/ml. In this study it was suspected that the isolates obtained belonged to the genus Streptococcus.

Abstract

Market waste is a component of organic waste that has not been widely used. One of the efforts to utilize market waste is to use market waste as a raw material for making ecoenzymes. This study aims to describe the characteristics of the color, pH, aroma, and total dissolved solids (TDS) of ecoenzymes produced from market waste including mustard greens, kale, banana peels, pineapple peels, and various combinations of these market wastes. Ecoenzyme fermentation was carried out for 90 days. The results of the observations showed that there were differences in color, pH, aroma, and TDS in the resulting ecoenzymes. The color of the ecoenzyme produced is generally brown with different color degradation. The aroma of the ecoenzymes produced also varies, but ecoenzymes containing banana peels, as well as a combination of fruit and vegetable peels, apart from the treatment of banana peels + kale, produce an unpleasant odor. The pH of the ecoenzyme produced was entirely acidic with a pH range of 2.9-5.6. The lowest ecoenzyme pH was made from kale + mustard greens, while the highest pH was made from banana skin treatment. Ecoenzyme TDS values ​​also varied, with the lowest value being 955 in the banana peel treatment, and the highest value in the mustard greens treatment with a value of 2920. The results showed that market waste has the potential to be used as a raw material for making ecoenzymes.

Abstract

Tanah merupakan salah satu media tumbuh yang biasanya digunakan dalam budidaya pertanian. merupakan salah satu faktor lingkungan yang tanah memiliki berbagai sifat fisik, kimia, dan biologi. Sifat fisik tanah terdiri dari tekstur dan struktur. Tekstur tanah terdiri dari ukuran fraksi pasir, debu, dan liat. Tekstur tanah mempengaruhi kemampuan tanah dalam menahan air dan juga pertumbuhan akar tanaman. Tanah yang baik harus mengandung unsur hara makro dan mikro esensial, bahan organik, kapasitas pertukaran kation, dan pH tanah yang sesuai. Tujuan dari penelitian ini adalah untuk mengidentifikasi karakteristik tanah pertanian di Dusun Banjarsari, Kecamatan Bandarkedungmulyo, Kabupaten Jombang. Penelitian ini dilakukan dengan pengambilan sampel tanah dilakukan pada Desember 2021. Tempat pelaksanaan penelitian ini adalah di Desa Banjarsari, Bandarkedungmulyo, Jombang, Jawa Timur dan laboratoriun Fakultas Pertanian Universitas KH. A. Wahab Hasbullah Jombang. Berdasarkan penelitian yang dilakukan dapat disimpulkan bahwa tanah di lokasi penelitian menunjukkan bahwa memiliki porositas rendah, sedang, hingga tinggi. Tanah dengan porositas tinggi terdapat pada horizon III. Kapasitas pertukaran kation tertinggi juga terdapat pada horizon III. Aerasi tanah yang baik akan dipengaruhi oleh tingginya porositas tanah. Pada horizon I tanah memiliki porositas rendah karena banyak mengandung fraksi lempung, sehingga lebih sesuai untuk pertumbuhan akar tanaman.

Abstract

relatively shorter adult phase than the larval stage. The larval stage of this organism lives in piles of organic matter, including in piles of household waste. The ability of larvae to utilize organic matter in their environment is inseparable from the role of potential microbes that produce enzymes associated with their gut. This study aims to isolate, screen, and identify the potential bacteria in producing hydrolytic enzymes from gut of O. rhinoceros larvae. The third instar larvae were dissected aseptically to obtain gut bacterial isolates. The isolates were tested for their ability to produce amylase, cellulose, lipase, and protease using agar plate containing specific substrates. There were eight different isolates, and three of them had the ability to produce all four enzymes. The three isolates were identified as Bacillus. These results can be used as a reference for handling household organic waste.

Abstract

The amylase is an enzyme that widely used in industry. In this research, amylase was isolated from endosymbiont bacteria (EKA 4) of cockroach Pycnoscelus surinamensis from household garbage dump Surabaya. This research aims to identify bacteria EKA 4, produce and characterize amylase from it at various temperature, pH and effect of metal ions. Amylase screening assay was done by imoculating the isolate into medium contain 2% of starch then incubated for 24 hours. Detection of amylase was done qualitatively by measured presence of halozone and quantitatively using DNS method. 16S rRNA analysis found that bacteria EKA 4 has similarity to Bacillus subtilis with 100% of percent identity. Result showed that optimum activity of amylase from B. subtilis were at 40°C, pH 6 and could use ion CaCl2 as a cofactor in a reaction. Thus, utilization of amylase from endosymbiont P. surinamensis could be as an alternative source in big industries.

Biofertilizer is an alternative to chemical fertilizer that can enhance plant growth but is specific to certain plants and soils. Various biofertilizers have been produced, but the specific biofertilizers to enhance durian growth have not been widely available. This study aims to determine the potential of hydrolytic enzyme-producing bacteria, one of the microbial components of biofertilizers, in the De Durian Park (DDP) area, Wonosalam, Jombang District, Indonesia. The bacteria were isolated using the selective medium plating technique. The obtained isolates were screened for their ability to produce cellulase, amylase, protease, and lipase. The clear zone around the colony indicated the ability of the isolates to produce hydrolytic enzymes, and then the clear zone index was obtained. The best three isolates for each enzyme production were characterized macroscopically, microscopically, and biochemically, then identified using 16S rRNA gene sequence analysis. Fifty-two isolates were successfully obtained from the soil of DDP. The best three cellulolytic bacteria had indices ranging from 1.83 to 2.14, categorized as moderate-strong, identified as Bacillus anthracis, B. pacificus, and B. cereus. The amylolytic bacteria ranged from 1.25 to 1.35, categorized as moderate, and identified as Bacillus anthracis, B. paranthracis, and B. paramycoides. The proteolytic bacteria ranged from 1.33 to 1.50, categorized as moderate, identified as B. pumilus, B. paramycoides, and B. cereus. The lipolytic bacteria ranged from 1.94 to 3.87, categorized as moderate-strong, and were identified as Enterobacter sichuanensis, B. pacificus, and E. asburiae. These cellulolytic, amylolytic, proteolytic, and lipolytic bacteria have the potency as candidates for a biofertilizer formula.

Diversity of Vachellia nilotica rhizosphere bacteria in the savanna of Baluran National Park, Indonesia and their potential for hydrolytic enzyme production. Biodiversitas 26: 1754-1767. Vachellia nilotica, a leguminous plant introduced to Baluran National Park as a firebreak, is now causing environmental problems due to its invasiveness, which hinders the growth of native plants. Despite significant ecological changes, there is no data regarding the impact of this plant invasion on soil microorganisms. This study aimed to assess the bacterial diversity in the rhizosphere of V. nilotica and investigate the bacteria's ability to produce hydrolytic enzymes. The bacterial community was identified using Next-Generation Sequencing (NGS), and a conventional isolation technique. A total of 99,520 Operational Taxonomic Units (OTUs) were obtained, encompassing a rich diversity of 4,631 species. These species belonged to four phyla (i) Pseudomonadota; (ii) Actinomycetota; (iii) Bacillota; and (iv) Acidobacteriota. Additionally, 26 isolates were obtained through conventional isolation, and all of them can produce hydrolytic enzymes, including protease, lipase, amylase and cellulase. A total of 9 isolates showed highest proficiency in producing hydrolytic enzymes, of which 8 belonged to the Bacillota phylum and 1 isolate showed similar characteristic with Acidobacteriota phylum. The isolates SAV 2.NA.1.5.6 and SAV 2.NA.2.4.3 exhibited multiple enzyme activities and were classified as Bacillota. The findings indicate that the rhizosphere of V. nilotica contains a high diversity of bacteria, including those capable of producing hydrolytic enzymes. This information can serve as a foundation for managing V. nilotica and promoting sustainable development by leveraging the potential of rhizosphere bacteria in this plant. To our knowledge, no prior research has been conducted on this topic in the savanna area of BNP.

Abstract

Food security remains a critical challenge in rural communities with limited land resources. This community service program aimed to enhance household food independence in Plosokerep Village, Jombang, through the integration of mini gardens, organic horticulture education, and environmentally friendly technology. The program applied a participatory action research approach involving 60 households and three educational institutions. Activities included the distribution of vegetable seedlings, training on organic fertilizer and botanical pesticide production, and the establishment of demonstration gardens. Pre- and post-tests were conducted to measure knowledge improvement, while field observations assessed adoption rates and sustainability potential. Results showed a 71.9% increase in participants’ knowledge, a 92% plant survival rate, and the formation of informal farmer groups. The program also reduced household organic waste by 25% and pesticide use by 70%. These findings indicate that simple, community-based agricultural interventions can significantly improve food security, environmental awareness, and social cohesion, and can be replicated in other areas with similar conditions.
