Abstract

Ekstrak Teh Hijau mengandung polifenol utama dalam daun teh, yaitu katekin yang terdiri dari Epigallocathecin 3-gallate (EGCG), epigallocathecin (EGC), epicathecingallate (ECG) , epicatechin (EC), gallocathecin (GC). Diantara keempat komponen tersebut EGCG (Epigallocathecin Gallate) merupakan komponen yang paling potensial. Polifenol teh hijau memiliki efek peredaman terhadap ROS (Radical Oxidative Superoxide), sehingga dapat mencegah kerusakan kulit akibat dari sinar UV-B. Penelitian ini bertujuan untuk membuktikan pemberian krim ekstrak teh hijau 70% dapat mencegah penurunan jumlah kolagen dermis dan peningkatan ekspresi matriks metalloproteinase-1 pada mencit BALB-C. yang dipapar sinar ultraviolet-B.Penelitian ini adalah penelitian eksperimental laboratorium, menggunakan rancangan the randomized post-test only control group. Sebanyak 30 ekor mencit balb/c yang digunakan pada penelitian ini dibagi menjadi 3 kelompok yang masing-masing terdiri dari 10 ekor mencit. Kelompok Kontrol (O) tidak diolesi apapun, Kelompok 1 diolesi bahan dasar krim (Kelompok Perlakuan 1), dan Kelompok 2 diolesi krim ekstrak teh hijau 70% (Kelompok Perlakuan 2). Semua Kelompok Perlakuan diberikan paparan sinar UVB dengan dosis total 840 mJ/cm2 selama 4 minggu, kemudian dilakukan biopsi untuk permeriksaan jumlah kolagen dan ekspresi MMP-1. Data dianalisis menggunakan one way Anova untuk mengetahui adanya perbedaan signifikan pada ketiga kelompok perlakuan kemudian dilanjutkan uji post hoc dengan menggunakan tes LSD (Least Significance Difference) untuk mengetahui beda nyata terkecil pada taraf kemaknaan <0,05. Hasil penelitian menunjukkan terjadinya penurunan yang lebih rerata kolagen yang bermakna pada Kelompok Kontrol (47,83%) dan Kelompok Perlakuan 1 (50,73%) setelah diberikan paparan sinar UV-B dibandingkan dengan rerata kolagen pada Kelompok Perlakuan 2 (67,59%). Hasil rerata ekspresi MMP-1 terjadinya peningkatan pada rerata kontrol (66,76%) dan rerata Kelompok Perlakuan 1 (74,46%) dibandingkan rerata ekspresi MMP-1 Kelompok Perlakuan 2 (39,31%). Melalui uji post hoc tidak ada perbedaan hasil dari Kontrol dan Kelompok Perlakuan 1 baik pada jumlah kolagen dan ekspresi MMP-1 (P>0,05).Simpulan penelitian ini adalah krim ekstrak teh hijau 70% dapat mencegah penurunan jumlah kolagen dermis dan peningkatan ekspresi MMP-1 pada mencit Balb/C yang diberi paparan UV-B.

Abstract

Background: Our laboratory assessment indicated that the Creatine Monohydrate® supplement contains phytoestrogen and phytoprogesteron. This study was aimed to prove that the administration of Creatine Monohydrate® supplement could increase the estrogen levels and decrease testosterone levels in male albino rats (Rattus norvegicus) with moderate physical activity given. Methods: A true experimental with pre-test and post-test randomized control study was conducted using 14 albino male rats. The samples were divided into two groups; the control groups (P0) that received aquadest and moderate physical activity, and treatment group (P1) that received 0.4 mg Creatine Monohydrate® supplement twice a day and moderate physical activity given. Moderate physical activity (swimming) was given five times a week for 14 days. Estrogen and testosterone level were assessed before and after treatment was given. Results: Before the treatment, the estrogen and testosterone levels between P0 group and P1 group were comparable. Post-test results showed a significant different in estrogen levels between the P0 and P1 group (23.17±2.86 and 27.82±3.06 ng/ml respectively; p<0.05) as well as testosterone levels (18.15±2.95 and 21.69±2.14 ng/ml respectively; p<0.05) after 14 days of treatment. A paired analysis showed an elevating of estrogen levels but no effect on testosterone levels in P1 group that treated with Creatine Monohydrate® Conclusion: This study indicated that the administration of Creatine Monohydrate® increased the estrogen levels but have no effect on testosterone levels in male albino rats (Rattus norvegicus) with moderate physical activity.

Abstract

Dislipidemia adalah kelainan metabolisme lipid yang ditandai terjadinya peningkatan kolesterol total, trigliserida, kolesterol LDL dan penurunan kolesterol HDL. Pada saat ini banyak penelitian untuk mencegah dan mengobati dislipidemia dengan bahan alami. Teh putih merupakan teh tanpa proses fermentasi yang berasal dari daun teh (camellia sinensis) yang sangat muda dan masih menggulung serta terlindung dari sinar matahari sehingga mencegah degradasi polifenol. Ekstrak teh putih mengandung derivat katekin tertinggi dibanding teh lainnya , ECGC (Epigalocathecin 3-Gallate) dan kafein ini dapat memperbaiki profil lipid darah dan memiliki efek vasoprotektif, juga memiliki kemampuan untuk menginhibisi (Cholesteryl ester transfer protein) CETP, yang bisa meningkatkan kadar kolesterol HDL dan menurunkan kadar kolesterol total ,trigliserida dan kolesterol LDL. Penelitian ini bertujuan untuk mengetahui kegunaan Ekstrak Teh Putih (camellia sinensis) sebagai alternatif untuk mencegah dislipidemia dan mengetahui dosis pemberian ekstrak teh putih untuk mencegah dislipidemia pada tikus jantan galur wistar. Penelitian ini adalah penelitian eksperimental murni dengan randomized posttest only control group design. Tikus putih jantan dipilih secara random dan dibagi menjadi 3 kelompok, masing-masing berjumlah 10 ekor tikus, yaitu kelompok kontrol diberikan diet tinggi lemak dan plasebo yang berupa akuades , kelompok perlakuan I diberi diet tinggi lemak dan ekstrak teh putih 14,4 mg, dan kelompok perlakuan II diberi diet tinggi lemak dan ekstrak teh putih 28,8 mg masing-masing 1 kali sehari. Setelah perlakuan selama 28 hari sampel darah diambil dari medial kantus sinus orbitalis, untuk pemeriksaan kadar kolesterol total, trigliserida, kolesterol LDL dan kolesterol HDL. Hasil penelitian menunjukkan bahwa pada kelompok pemberian ekstrak teh putih 14,4 mg dan pemberian ekstrak teh putih 28,8 mg terdapat penurunan kolesterol total secara bermakna masing-masing 137,31% dan 156,65% (p < 0,05), penurunan trigliserida secara bermakna masing-masing 77,29% dan 101,01%(p < 0.05) dan penurunan kolesterol LDL masing-masing 53,58 % dan 75,12%(p < 0,05), serta peningkatan kolesterol HDL secara bermakna masing-masing 44,31% dan 66,39% (p < 0,05). Penelitian ini menyimpulkan ekstrak teh putih mencegah peningkatan kolesterol total, trigliserida, kolesterol LDL dan mencegah penurunan kolesterol HDL, jadi ekstrak teh putih mencegah dislipidemia.

Abstract

Introduction: Overweight and obesity are abnormal conditions characterized by excessive fat accumulation in adipose tissue, to the extent that health is impaired. It is associated with a greater risk of various metabolic diseases. Binahong (Cordifolia (Ten) Steenis Anredera) leaves extract is known to contain flavonoids, saponins, and tannins that have an effect on weight loss and a decrease abdominal fat mass. This study aims to prove that the administration of binahong leaves extracts reduces body weight and visceral abdominal fat mass in obese male rats (rattus norvegicus) with moderate physical exercise. Methods: This research was a true experimental study with the post-test only control group design. The research subjects were 36 male rats divided equally into 2 groups, (18 rats each). The positive control group (P1) were treated with moderate physical exercise (20 min of swimming/day for 2 weeks) and oral aquadest (placebo) for 2 weeks, while the treatment group (P2) were treated with moderate physical exercise (20 min of swimming/day for 2 weeks) and oral binahong leaves extract of 400 mg/kg bW once a day for 2 weeks. Body weight and abdominal visceral fat mass were measured after 2 weeks of treatment. The daily remaining feed was observed to observe the food intake. Results: The results showed that the average body weight of P1 group was 288.6±6.11 g, whereas the P2 group was 272±6.48 g (p<0.001). The abdominal visceral fat mass in the P1 group was 9.24±0.48 g, while in the P2 group was 7.43±0.58 g (p<0.001). The median remaining feed in group P1 was 3 g, while in the P2 group was 9.54 g (p<0.001). Conclusion: Together, the results of this study suggested that binahong leaves extracts reduced body weight and visceral abdominal fat mass in obese male rats (rattus norvegicus) with moderate physical exercise.

Abstract

Introduction: The aging process is caused by several factors, one of which is the accumulation of oxidative damage caused by high free radicals in the body. Ashitaba (Angelica keiskei) leaves extract contains flavonoids, polyphenols, tannins, and antioxidant capacity. This study aimed to prove that administration of Ashitaba leaves extracts prevent superoxide dismutase (SOD) reduction and malondialdehyde (MDA) elevation in overtraining male Wistar rats (Rattus norvegicus). Methods: This study was an experimental study with randomized pretest-posttest control group design. The subjects used in this study were 16 male Wistar rats, healthy, aged 6 months, and weighing 180- 200 grams. The control group was given overtraining two hours after aquabidest 2 cc treatment (P0), and the treatment group was given overtraining two hours after administration of Ashitaba leaves extract of 20 mg/kg BW/day (P1). Before and after treatment for 14 days, blood sample were drawn through canthus medial orbital sinus to measure the level of SOD and MDA using the ELISA method. Results: The results showed in control group there was a decrease in SOD levels (from 8.58±0.78 ng/mL to 5.93±1.27 ng/mL; p<0.001) and increased MDA levels (from 2.75±0.19 nmol/mL to 3.28±0.23 nmol/mL; p= 0.001). In treatment group there was no change in SOD levels (from 8.90±0.61 ng/mL to 8.57±0.49 ng/mL; p= 0.217) and there was no change in MDA levels (from 2.67±0.33 nmol/mL to 2.62±0.24 nmol/mL; p= 0.693). Conclusion: It can be concluded that administration of Ashitaba leaves extracts prevented SOD reduction and MDA elevation in overtraining male Wistar rats.

Abstract

Introduction: Moderate exercise is one of the pillars of weight management. Supplementation of ingredients containing bioactive compounds such as Sauropus androgynus (L.) Merr leaves extract which contains flavonoids, tannins, and saponins may help obesity management. The purpose of this study was to prove that supplementation of Sauropus androgynus (L.) Merr leaves extract reduced more body weight and triglyceride in obese male Wistar rats with moderate exercise. Methods: This research was an experimental study with a randomized pretest-posttest control group design. The subjects were 20 rats (Rattus norvegicus). Obesity and hypertriglyceridemia were induced by a high-fat and high-carbohydrate diet. Rats were divided into two groups, namely the control group and the treatment group. Results: The control group there was a decrease in body weight (from 284.5±3.17 to 278.3±4.27 grams; p <0.001) and a decrease in triglyceride levels (from 186.7±14.8 to 181.1±13.6; p = 0.047). In the treatment group there was also a decrease in body weight (from 282.1±5.72 to 272.2±3.55 grams; p <0.001) and a decrease in triglyceride levels (from 185.4±9.24 to 160.2±22.5; p = 0.003). The decrease in body weight in the treatment group was more than in control group (-9.90±3.37 vs -6.20±2.74; p = 0.018). Triglyceride levels in treatment group also decreased more than in control group (-25.2±19.3 vs -5.60±8.13; p = 0.008). Conclusion: Based on the results of this study, it can be concluded that supplementation of Sauropus androgynus (L.) Merr leaves extract reduced more body weight and triglyceride in obese male Wistar rats with moderate exercise.

Abstract

Introduction: Management of obesity are including consumption of supplements. Nutrilite GreenTrim® Supplement contains green tea extract and EGCG and squalene. This study aimed to prove the effects of Nutrilite GreenTrim® on body weight, waist circumference, and body fats percentage in obese men. Methods: This clinical trial used pretest-posttest control group design in 22 obese men, aged 25-40 years with a BMI 25-35 kg/m2. Subjects were divided into the control group (K) and the treatment group (P). Subjects were given moderate physical exercise and a low-calorie diet. Results: The results showed K group lost weight (from 85.1±8.99 to 82.3±9.21 kg; p<0.001), while the P group lost weight (from 84.1±10.7 to 80.1±10.9 kg; p <0.001) and the difference showed that the P group lost weight greater than the K group (p = 0.007). K group experienced a decrease in waist circumference (from 96.9±7.69 to 93.8±8.84 cm; p <0.001), while the P group (from 96.5±7.78 to 92.2±7.59 cm; p <0.001) the difference showed that the decrease in waist circumference in the P group was better than the K group (p = 0.041). In the K group, body fat percentage decreased (from 27.5±3.31 to 25.8±3.77%; p <0.001), the P group also decreased (from 26.5±3.78 to 23.3±3.67%; p<0.001) the analysis showed that the decrease in body fat percentage in the P group was better than the K group (p <0.001). Conclusion: Supplementation of Nutrilite Green Trim® reduced body weight, waist circumference, and body fats percentage in obese men with moderate physical exercise and low-calorie diet.

Abstract

Introduction: Obesity increases the risk of many degenerative disease. Abdominal fat is a sign that someone has great risk for metabolic syndrome. Moderate physical exercise is suggested to reduce weight and body fat percentage. Cinnamon (Cinnamomum burmannii) extract can reduce weight by the lipolysis activity in visceral adipose tissue and supress the appetite. This research aimed to prove that administration of cinnamon extract reduced weight and abdominal visceral fat in obese male wistar strain ratswith moderate intensity physical exercise. Methods: This research was a true experimental with post-test only control group design. The subjects were 36 male albino rats (Rattus norvegicus), Wistar strain, healthy, 4-5 months age, and obese (body weight of > 250 g), which divided into two groups (n= 18). The positive control group (P1) was treated with placebo (2 ml aquadest), standard food, and moderate physical exercise. The experimental group (P2) was treated with 1000 mg/kg BW/day cinnamon extract standard food, and moderate physical exercise. After 2 weeks of treatment, body weight and abdominal visceral fat mass were measured. Results: The average body weight of the P2 group was significantly lower than the P1 group (271.9±5.48 and 284.7±5.05 g respectively, p<0.001). The median abdominal visceral fat mass in the P1 group was 9.0 g, whereas the P2 group was 7.9 g (p<0.001). The median remaining food in the P1 group was 3.43 g, whereas the P2 group was 10.07 g (p<0.001). Conclusion: This study indicated that administration of cinnamon extract reduced weight and abdominal visceral fat in obese male wistar strain ratswith moderate intensity physical exercise.

Abstract

Introduction: A moderate-intensity diet and exercise regimen is highly recommended as a non-pharmacological therapy, which can be used in type 2 diabetes mellitus. It can increase the GLUT-4 protein serum activity in the receptor of the muscle cell membrane. This research aimed to prove that moderate physical exercise was able to reduce post-prandial blood glucose and increase GLUT-4 protein serum on diabetic male Wistar rat (Rattus norvegicus). Methods: This research employed an experimental pre-test post-test control group design by using 26 diabetic male Wistar rats that were randomly selected. The first group without moderate exercise, the second group with moderate exercise. The measurement of post-prandial blood glucose and GLUT-4 protein serum was performed after 14 days of treatment. Results: The median of post-prandial blood glucose levels for the control group (P0) in pre-test was 177.00 mg/dl and in post-test was 168.00 mg/dl, moreover for the treatment group (P1) in pre-test was 158.00 mg/dl and in post-test was 157.00 mg/dl. Moreover, the median of GLUT-4 for the control group (P0) in pre-test was 8.97 ng/dl and in post-test was 9.34 ng/dl, and for the treatment group (P1) in pre-test was 8.44 ng/dl and in post-test was 9.38 ng/dl. The results of the analysis in both groups showed that there was no significant difference between the control group and the treatment group. Conclusion: It can be concluded that moderate physical training did not significantly reduce post-prandial blood glucose, and did not significantly increase GLUT-4 serum on diabetic male Wistar rat.

Abstract

Background: Excessive physical activity triggers hormonal imbalances, one of which is beta endorphin and lead to increasing leukocytes as a response to the inflammation. The purpose of this study was to prove that excessive physical activity increased leucocyte and decreased endorphin levels in male Wistar rats (Rattus norvegicus). Methods: This study was an experimental study with randomized pretest-posttest group design. The research subjects were male rats (Rattus norvegicus), Wistar strain, aged 8-10 weeks with a body weight of 150-200 grams and healthy. A total of 14 rats were randomly divided into two groups. The first group was treated with balanced physical activity (P1), the second group was treated with excessive physical activity (P2). On day 8 and day 28, leukocytes were examined using whole blood method and beta endorphin were examined using ELISA. Results: The results showed that in the group P1, both leukocyte levels (from 14.0±2.73x103/µL to 17.1±5.07x103/µL, p= 0.170) and beta endorphin levels (from 176±30.2 to 161±11.1 ng/mL, p= 0.110) remained stable. Whereas, the P2 group experienced leukocyte reduction (from 12.3±2.19x103/µL to 23.7±2.92x103/µL, p <0.001) and decreased beta endorphin levels (from 212±46.0 ng/mL to 55.3±8.05 ng/mL, p <0.001). Conclusion: This study suggesting that excessive physical activity increased leucocyte and decreased endorphin levels in male Wistar rats (Rattus norvegicus).
