Abstract

AbstrakTelah dilakukan penelitian untuk mengetahui uji aktivitas anti-osteoporosis secara in vivo dari tanaman Spilanthes acmella terhadap sel osteoblas tulang trabekula proksimal femur mencit jantan model osteoporosis induksi deksametason. Penelitian ini dilakukan dengan rancangan penelitian posttest only control group, 60 ekor mencit jantan sehat (usia >5 bulan) dikelompokkan secara random menjadi 10 kelompok yaitu kelompok sehat tanpa induksi deksametason dan kelompok osteoporosis induksi deksametason. Kontrol positif mendapat suspensi alendronat dan kelompok latihan fiik menggunakantreadmill mencit berjalan 10m/menit bertahap selama 5 -12 menit, 3 kali dalam satu minggu, kelompok kombinasi ekstrak etanol 70% dan latihan fiik serta kelompok uji fraksi butanol, heksan, etil asetat dan air. Penelitian ini dilakukan selama 4 minggu. Osteoporosis akibat pemakaian glukokortikoid menjadi penyebab osteoporosis sekunder yang meningkatkan risiko fraktur. Telah banyak bukti klinik tentang peran fioestrogen dalam pengobatan osteoporosis pada pascamenopause. Spilanthes acmella, atau yang dikenal masyarakat dengan sebutan Legetan, termasuk famili Asteracea merupakan satu tanaman obat di Indonesia yang mempunyai kandungan senyawa polifenol dan flvonoid. Dari hasil penelitian sebelumnya diketahui bahwa baik fraksi butanol dan air dari tanaman tersebut mampu meningkatkan aktivitas enzim alkalin fosfatase yang merupakan marker pembentukan tulang. Hasil menunjukkan bahwa ekstrak etanol 70% Spilanthes acmella dikombinasikan dengan latihan fiik meningkatkan jumlah sel osteoblas secara bermakna bila dibandingkan dengan hanya ekstrak etanol atau latihan fiik. Ini membuktikan bahwa ekstrak etanol 70% Spilanthes acmella mempunyai efek additive terhadap efek latihan pembebanan aksial pada kondisi osteoporosis. Fraksi heksana, etil asetat dan air juga meningkatkan secara bermakna jumlah sel osteoblas. Fraksi non polar merupakan fraksi yang lebih potensial untuk pembentukan tulang karena fraksi tersebut banyak mengandung senyawa turunan amida dan sesquiterpena. Kesimpulan penilitian ini menunjukan adanya efek additive ekstrak etanol 70% dari Spilanthes Acmella pada latihan fiik dengan meningkatkan jumlah sel osteoblas.Kata kunci: Spilanthes acmella, osteoporosis sekunder, osteoblasAbstractStudy was conducted to analyse the anti-osteoporosis effect of Spilanthes acmella plant in vivo on osteoblast cell trabecular bone of femur male mice with dexamethasone- induced osteoporosis. This study using a post test control group design, 60 male healthy mice (>5 months old) were randomizely devided into 10 groups, there are healthy control group (without induction dexamethaxone), and osteoporosis groups (induction with dexamethaxone). The positive control receive suspension alendronat suspension and the exercise group walking using mice treadmill 10m/minute, 5-12 minutes 3 times a week,the combination group of 70% extract ethanol and exercise, the butanol, hexane, etil asetat and water fraction group. The study conducted for 4 weeks. Glucocorticoid-induced osteoporosis is leading cause of secondary osteoporosis and an increase in fracture risk. There were clinical evidence suggest a role for phytoestrogen in the treatment of post-menopausal osteoporosis. Spilanthes acmella, or already known by public society as Legetan, include in Asteracea family, is one of Indonesia medicinal plants that contain of polyphenol and flvonoids. Previously in vitro study showed that buthanol and water fraction from this plant have increased alkaline phosphatase that known as marker of bone formation.The results showed that 70% ethanol extract of Spilanthes acmella that combined by physical exercise increasing osteoblast cells signifiantly than only 70% ethanol extract or physical exercise. Additionally hexane, ethyl acetate and water fractions from Spilanthes acmella also increasing osteoblast cells signifiantly. Non-polar fraction is a potential fraction of bone formation because it contains many nonpolar compounds such as amides derivatives and sesquiterpene. The study concluded that 70% ethanol extract of Spilanthes acmella have an additive effect to weight bearing exercise in osteoporotic condition.Keywords : Spilanthes acmella, secondary osteoporosis, osteoblast

Abstract

Background: Glucocorticoid-induced osteoporosis is leading cause of secondary osteoporosis by decreasing formation activity and increasing resorption activity. Spilanthes acmella, is one of Indonesia medicinal plants that contain of polyphenol and flavonoids. Previously in vitro study showed that buthanol and water fraction from this plant have increased alkaline phosphatase that known as marker of bone formation. The objective of this study to analyze the effect of Spilanthes acmella and physical exercise in increasing testosterone and osteoblast cells of femoral’s trabecular glucocorticoid-induced osteoporosis male mice. Method: This study using a posttest control group design, 36 male healthy mice (5 months old) were randomizely devided into 6 groups, there are : 1.Healthy control group (without induction dexamethaxone), 2.Osteoporosis groups (induction with dexamethaxone without treatment), 3.Positive control receive suspension alendronat, 4.70% Ethanol extract of Spilanthes acmella group, 5.Combination group of 70% extract ethanol of Spilanthes acmella and exercise, and 6.Exercise group (walking using mice treadmill 10m/minute, 5-12 minutes 3 times a week). All of the intervention were given for 4 weeks. Abstract

Isolation, identification, and structure elucidation of terpenoid compounds from an n-hexane extract of Marsilea crenata Presl., had been done. The leaves of M. crenata was extracted using n-hexane solvent. The extract then separated with vacuum column chromatography and open column chromatography to obtain the isolate. Furthermore, the isolate was identified and elucidated using UV-Vis, FT-IR, 1H-NMR, 13C- NMR, and 2D-NMR (COSY, HMQC, and HMBC). The identification and elucidation of the isolated structure from an n-hexane extract of M. crenata leaves indicate the isolate was a pentacyclic triterpenoid.

Abstract

Dusun Kiringan-Jetis, Bantul, merupakan desa wisata jamu gendong dengan penduduk mayoritas berprofesi sebagai pengrajin jamu. Jamu yang mereka jual secara turun-temurun terbatas dalam bentuk jadi dan setengah jadi. Perlu adanya peningkatan diversifikasi produk jamu untuk meningkatkan nilai jualnya. Kegiatan pengabdian pada masyarakat ini bertujuan untuk meningkatkan ketrampilan usaha dalam meningkatkan kualitas dan mengembangkan produk jamu yang akan dijual. Untuk dapat mencapai tujuan yang diinginkan dari kegiatan ini maka dilakukan upaya: (1) Edukasi dan pelatihan ketrampilan membuat es krim herbal dan (2) edukasi dan pelatihan ketrampilan membuat inovasi kemasan produk jamu yang aman dan menarik.Tim pelaksana kegiatan ini terdiri dari dosen dan mahasiswa S1 Fakultas Farmasi Universitas Airlangga dan Universitas Ahmad Dahlan. Dari kegiatan tersebut terlihat antusias para peserta pelatihan, peningkatan pengetahuan dan ketrampilan dalam pembuatan es krim herbal dan inovasi kemasan produk. Hal ini terlihat dari peningkatan skor post-testsetelah dilakukan pelatihan.Besar harapan para peserta kegiatan untuk dapat mengaplikasikan pengetahuan yang didapat sehingga dapat meningkatan perekonomian Dusun Kiringan-Jetis, Yogyakarta

Abstract

Kegiatan asuhan mandiri Toga atau yang juga dikenal dengan asman TOGA diintegrasikan dengan program PKK yang dalam pelaksanaannya dikoordinir oleh kader Asman dan didampingi fasilitator dari Puskesmas. Program ini dibawah pembinaan oleh Dinas Kesehatan Kabupaten/Kota. Desa Canditunggal yang terletak di wilayah Kecamatan Kalitengah Kabupaten Lamongan mempunyai kebun TOGA yang asri terbentang di tengah jalan desa. Kebun TOGA ini merupakan hasil kegiatan ibu-ibu PKK yang juga merupakan kader asman TOGA. Pada awalnya kebun TOGA ini hanya untuk keindahan saja. Untuk itu pada kegiatan pengabdian masyarakat yang dilakukan kali ini bertujuan untuk memberdayakan ibu-ibu PKK tersebut agar dapat memanfaatkan TOGA khususnya dalam membuat produk dari daun katuk menjadi produk bernilai ekonomi untuk memperbanyak ASI. Kegiatan dilakukan dengan pemberian materi dan pelatihan pembuatan produk. Antusiasme, kreatifitas dan inovasi dari ibu-ibu PKK kader asman TOGA Desa Canditunggal ini merupakan potensi yang sangat besar untuk keberhasilan pemanfaatn TOGA untuk menciptakan produk-produk yang bernilai ekonomi.  Kata Kunci: Asman, daun katuk, TOGA

Abstract

Abstract

Background: Justicia gendarussa Burm. f., has been used traditionally in Indonesia for antifertility. Nowadays, a capsule containing 70% ethanol extract of J. gendarussa leaves has been studied for safety. Objective: This study aimed to determine the acute and sub-chronic toxicity of 70% ethanol extract of J. gendarussa leaves in vivo. Methods: In the acute toxicity study, a single dose of 2,000"‰mg/Kg BW was orally administered to mice (n = 10), which were monitored for 24"‰days. For the subchronic toxicity study, rats were randomly divided into four groups (n = 10). The control group received distilled water, while the treatment groups received a repeated dose of 40, 200, and 1,000 mg/Kg BW orally for 90 days. Blood samples were collected for hematological and biochemical evaluations. Gross pathology and histopathology of liver and kidneys were assessed. Results: No mortality and non-observed adverse effect level (NOAEL) were observed in the acute toxicity study. The hematological analysis did not show significant differences in the subchronic toxicity study. The SPGT, SGOT, and creatinine values showed no change in groups 2 and 4, but the level of SGPT increased in groups 3. The increasing level of BUN was observed in all treated groups. Abnormalities or histopathological changes were observed in the liver and kidney in groups 3 and 4. Conclusions: Using 70% ethanol extract of J. gendarussa leaves at a therapeutic dose is safer, but it needs attention at a higher dose.

Abstract

Background: Justicia gendarussa from Papua has traditionally been used for the treatment of several diseases, and phytochemical studies have been carried out since 1987. Objective: This study aimed to determine the use of this plant as a male contraceptive that the Nimboran Ethnic and their perspectives have long used. Method: A qualitative method with an ethnographic approach was used, while data were collected through interviews, observations, and documentation. The informants were selected using purposive and snowball sampling. Result: The results showed that 44% of people use it to delay pregnancy, 24% due to young marriage with poor economic conditions, 14% after moving to another place 12% because of tribal wars over fertile land to multiply offspring. The preparation and usage of this plant through the collection, mixing, and manufacturing method and in terms of dosage, time, and duration of use were explored more deeply by conducting interviews with 50 informants. The effectiveness and success as a method of contraception are presented in the way the community has known the plant over different generations, and this method is still used as an alternative option considering the very wide area profile from one place to another. Conclusion: Justicia gendarussa is used by the Nimboran Ethnic group as an ingredient in traditional medicines, especially for male contraception.

Abstract

AbstractJusticia gendarussa Burm.f has been traditionally used as male contraceptionin in Indonesia. Gandarusa leaves contain gendarusin A which has potential as an antifertility agent. To prove that gandarusa leaves are safe to use as a male contraceptive, a subchronic toxicity test was carried out on this plant. Acute and subchronic oral toxicity studies of Justicia gendarussa Burm.f in rats were performed in the previous study in order to evaluate its safety. The aim of this study is to identify the metabolite profile of rat organ results of their subchronic toxicity test. Seven organs of rat such as kidney, hepar, testis, limph, instestine, lung, and heart were extracted using methanol and their profile metabolites were identified using UHPLC Vanquish Tandem Q Exactive Plus Orbitrap HRMS by positive and negatif mode. The results showed that all sampels contain 22 compounds from the positive ionization mode and 35 compounds from the negatif ionization mode. Compounds identified from the positive mode were dominated by amino acid groups, then compounds identified from the negatif mode were dominated by fatty acids and their derivatives. There were 35 metabolites that identified from the organ samples of the rat resulting subchronic toxicity test using the UHPLC Vanquish Tandem Q Exactive Plus Orbitrap HRMS method.Keywords: Justicia gendarussa Burm.f, rats, organ, metabolite profile
