Abstract

This study aimed to evaluate the effects of ciplukan (Physalis angulata L.) leaf ethanol extract on L-NG-nitroarginine methyl ester (L-NAME)-induced hypertensive rats. We randomly divided twenty-five Wistar rats into five groups. The sham group was given a PBS injection. The hypertensive group was injected with L-NAME on days 1 to 28. Three groups of hypertensive rats were given the extract on days 4 to 28. Blood pressure was measured using the tail-cuff method on days 0, 4, 10, and 27. The endothelial progenitor cells (EPCs) in the blood were measured by flow cytometry as a percentage of circulating angiogenic cells (CACs, CD34+/CD309+/CD45+) and endothelial colony-forming cells (ECFCs, CD34+/CD309+/CD45-). Serum NO and MDA levels, as well as serum SOD activity, were measured colorimetrically. Serum TNF-α levels were measured by the ELISA method. The ciplukan leaf extract reduced systolic and diastolic blood pressure, reduced the percentage of EPCs in the blood, increased serum NO levels, reduced MDA levels, increased serum SOD activity, and reduced serum TNF-α levels in L-NAME-induced hypertensive rats. It is concluded that ciplukan ethanol leaf extract exerts protective effects on L-NAME-induced hypertensive rats. These study results can strengthen the scientific basis of using ciplukan leaf ethanol extract to treat hypertension.

Abstract

Prevalence of tooth extraction or dental surgery was 48.5% of all dental care in Indonesia. Tooth extraction carries potential health risks and side effects such as pain, swelling, trismus and dysfunction of the oral cavity during recovery. Secretory leukocyte protease inhibitor (SLPI) is one of innate immunity proteins that can inhibit the activation of macrophages. We are expecting the provision of SLPI can decrease excessive inflammatory response in healing after tooth extraction. This study was to investigate the administration of SLPI on cellular expression of NF-Kβ and IL-1β on wound macrophages of Rattus novergicus post tooth extraction. The research design is in vivo experimental study. In total, 20 rats were randomly divided into four groups (each group n=5) and underwent tooth extraction on left incisor teeth of mandible. One of the groups did not receive SLPI administration (control group) and the socket was stitched after tooth extraction. Meanwhile, the remaining three groups (experimental groups) were given SLPI administration after tooth extraction with three different doses (0.1 µM, 0.5 µM and 2.5 µM, respectively). After SLPI administration, the socket of experimental groups was stitched. The effects of SLPI administration were evaluated by counting at the percentage of NF-Kβ translocation and the expression of expression of IL-1 in macrophages cells of the rat socket using immunohistochemistry analysis. The cellular expression of NF-Kβ and IL-1β were significantly decreased (p < 0.05) on groups with SLPI may decrease cellular expression of NF-Kβ and IL-1β on wound macrophages cells of rats post tooth extraction in a dosedependent manner.

Abstract

Pendahuluan: Kesehatan Fisik dipengaruhi dari kesehatan jiwa. Mayoritas masyarakat memiliki prioritas kesehatan secara fisikly seperti makan sehat, olah raga dan pola hidup sehat. Kekeliruan dari aspek perkembangan mental seseorang karena kehidupan lingkungan kurang sehat seperti tidak memiliki jiwa sehat, emosi labil, pemikiran negative, tidak nyaman dan tekanan mental lainnya menyebabkan jiwa kurang sehat. Kondisi demikian mempengaruhi peredaran darah yang tidak lancar. Tujuan: Tujuan penelitian ini untuk memberikan pemahaman secara umum kepada masyarakat untuk mampu memberikan kontribusi keilmuan dalam menjalani pola hidup sehat secara fisik dan psikis. Metode: Metodologi penelitian dengan menggunakan pendekatan kualitatif desain library research. Data diperoleh dari beberapa literature rivewe dan diberikan dukungan informan melalui buku digital maupun elektronik. Hasil: Hasil penelitian menggambarkan bahwa kehidupan sehat layaknya individu yang mampu menciptakan lingkuangan bersih secara fisik dan psikis. Mampu meminimalisir perilaku, pikiran dan sikap negative sebagai usaha sadar memberikan kontribusi terhadap kesehatan mental secara penuh menciptakan suasana bahagia dan meningkatkan kesehatan fisik. Kesimpulan: Upaya demikian salah satu bagian dari tindakan meminimalisir resiko serangan struk. Analisis studi literature memberikan deskripsi umum bahwa kesehatan mental memiliki peran penting dalam meningkatkan aspek kesehatan fisik. Siklus peredaran darah, situasi rileks dan bebas dari tekanan mental dapat membantu individu dalam menjaga kesehatan. Kekuatan mental yang sehat dapat mempengaruhi sugesti positif sehingga membentuk pola kehidupan sehat

Abstract

The purpose of this study is to determine the effect of histatin-1 protein increasing the concentration of fibroblast growth factor (FGF2) and fibroblast cell migration in human gingival fibroblast (HGF) culture. The HGF cell culture used was obtained from odontectomy patients and was the result of the 4th passage. Cell culture was divided into 4 groups: HGF control group (without treatment) and treatment groups with 5µM, 10µM, and 20µM concentration of histatin-1 respectively. The amount of fibroblast growth factor 2 (FGF2) was quantified by using ELISA method. In vitro fibroblast cells migration was measured using scratch/wound healing assay method. FGF2 concentration and HGF cell migration were measured at hour 0, 3, 11, 24, and 48. The results of the One Way Anova statistical test difference between FGF 2 (ΔFGF2) at hour 24 showed a value of P = 0.042 (P≤0.05) and HGF cell migration at hour 11 showed a P value = 0.003 (P≤0.05). The conclusion of this study is that Histatin-1 Protein is able to increase the concentration of Fibroblast Growth Factor 2 (FGF2) and the migration of Human Gingival Fibroblast cells.
