Abstract

he experiment was conducted to determine biogas quality produced from mixture of dairy manure and bagasse with different C/N ratio level, and to know the optimal mixture ratio. There were three group treatments based on C/Nratio level : C/N ratio 22, 30, and 35 with three replications each. Nine units of 20 litre batch-feeding digesters were used during 30 days examination. The main data were BOD, COD, pH and VFA values, total biogas volume, andmethane value. C/N ratio of dairy manure and bagasse was 22, 12, and 198. Dry matter mixture was 7%. BOD value was decreased 54.33%, 27.89%, and 42.86%, COD value was decreased 35.85%, 8.29%, and 27.88% and biogas wasproduced as much as 29.42 litre, 68.51 litre, and 114.73 litre by each treatment with methane values of 24.16%, 27.5%, and 23.38%. VFA value were 27.19 ml mol, 27.86 ml mol, and 30.73 ml mol respectively during operation. BOD andCOD value decrease, pH value, biogas temperature, and total biogas volume was different among treatment. The optimal mixture was C/N ratio 30 treatment. The results indicated that bagasse could be used as material mixture in biogas production.(Key words : Biogas, Dairy manure, Bagasse, C/N ratio, Methane)

Abstract

This study was aimed to formulate strategies for increasing adoption innovation in the smallholder dairy farms. The study was conducted in May to July 2015. The research was conducted using survey method. The selection of respondents was conducted using multi-stage sampling to 270 dairy smallholder farmers in Yogyakarta Region, Central, and East Java Provinces. The method used was descriptive qualitative by using SWOT analysis (strength, weakness, opportunity and threat). The results of the research which is to optimize resources; provide a clear and continuous information about innovation; facilitate access to information by extending the dissemination of information; provide mentoring programs, counseling, and training with a demonstration of the innovation; increase the participation of dairy farmers; strengthen institutional dairy farmers; seek a tool to support innovation together; improve the quality of extension, media, and the delivery of information; and optimize government and NGO aid.

Abstract

 ABSTRACT Integrated farm is a model for the management of local resources’ potential by integrating livestock and crops into eco-friendly farming (zero waste). It can be the answer for the country’s basic needs of food that is still dependent on imports. Most of the habitants of Argorejo and Argosari village work as farmers and ranchers. Although the formal education of rural people is still low due to low income, the average member of activities to get information, knowledge and management skills in agribusiness sectors to understand the practical, economical, and effective ways in business development. It facilitates adoption of innovation and the process of information absorption. Adoption of innovation is a process of mental or behavioral changes in the form of knowledge (cognitive), attitudes (affective), and skills (psychomotor) since one recognizes the innovation until one decides to adopt it after receiving it. This is qualitative research, with research material in the form of members of agribusiness activities in the intervention program called Mandiri Bersama Mandiri. The method used is descriptive analysis. The data is collected by interview, observation, and records related to the activities of the group. The result of Classification Analysis with Adopter type shows that 7 of the 8 groups in the facilitation of innovation adoption of integrated farms have: 6.7% to 14.3% innovator; 8.3% to 18.2% early adopter; and the rest is 71.4% to 77.8% early majority. One group has no innovator and up to 50% early adopter. The innovators of Mandiri Bersama Mandiri program include the most influential people that have important positions, such as group leaders, village officials, or persons who have knowledge. It indicates that the potential success of the intervention program is located on the adopter type that exists in each member, absorption of new innovations adoption, and succeed in the community empowerment program.

Abstract

This research aims to understand the effect of metal ions bioabsorption which belong on different elemental groups to the methanol dehydrogenase (MDH) enzymatic activity in nitrogen-fixing bacteria Bradyhizobium japonicum USDA 110. Ten metal ions with each have 30μM concentration were added to grow Bradyhizobium japonicum USDA 110 in 10-1 diluted nutrient medium. The MDH activity test showed a similar result between the bacteria grown in medium without metal ions addition (control) and the bacteria were grown in a calcium ion/Ca2+ added media. The highest MDH enzymatic activity was shown on the bacteria grown in a magnesium/Mg2+ added medium, which showed 0.08 (U/mg) enzymatic activities. The addition of magnesium/Mg2+ metal ion accelerates the bacterial growth by 2.6 times and MDH activity by 1.28 times compared to control. The MDH enzyme is essential, especially for bacteria which exist in the soil environment, to adapt to high methanol concentration and to support bacterial anaerobic growth capacity along with plant symbiotic process. Moreover, the MDH activity staining method could also act as pollutant indicators like metal ions and hydrocarbon derivates. This research concluded that metal ions biosorption (calcium/Ca2+ and magnesium/Mg2+) are required for bacterial cells reproduction and oxidation of single carbon chain compounds like methanol. The nitrogen-fixing symbiotic bacteria, Bradyhizobium japonicum USDA 110 showed high MDH activity after the two metal ions absorption. However, contrary results were shown on vanadium/V3+, manganese/Mn2+, iron/Fe3+, copper/Cu2+, zinc/Zn2+, and aluminum/Al3+ absorption, which showed low MDH activity and cells biomass.

Abstract

Methanol dehydrogenase (MDH) enzyme was purified from Bradyrhizobium japonicum USDA110 cell-free extract. The bacteria were grown in a culture medium with direct 0.5% methanol addition aimed to stimulates the MDH catalytic enzyme activation. Bradyrhizobium japonicum USDA110 MDH enzyme was purified by using 25 mM 2-(N-morpholine) ethanesulfonic acid/MES pH 5.5 buffer and 1 M sodium chloride/NaCl which separated into three columns, the first column was PD-10 for buffer exchange; the second column was HiTrap Sepharose HP to obtain unbonded fraction in the column; and the third column was Mono S 5/50 GL integrated with two pumps HPLC (high-performance liquid chromatography) to obtain pure MDH enzyme for serial changing of 1 M NaCl-25mM MES pH 5.5 with the flow rate at 1 ml/min. The protein concentration and MDH catalytic enzyme activity were observed on each purification process starting from the cell-free extract to pure MDH enzyme. The pure MDH enzyme was obtained by Mono S 5/50 GL-HPLC purification which showed a single band on SDS PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis). The MDH enzyme purification from Bradyrhizobium japonicum USDA110 showed 90-fold purification, a sub-molecular weight of 63 kDa, specific activity at 2.69 U/mg, and optimum activity at a 35oC temperature and pH 9.

Abstract

This study aims to isolate and extract the chicken bone gelatin through hydrolysis process and was analyzed chemically the extracted gelatin product. Research activities included several stages: (1) chicken bone waste preparation (2) ossein extraction (3) gelatin production (4) gelatin chemical analysis. The extraction time was 60 minutes with 5 treatment levels of HCl concentration ie K1 = 2%, K2 = 3%, K3 = 4%, K4 = 5% and K5 = 6%. Each treatment was repeated three times. Gelatin analysis consists of water content, ash content, protein content, pH value, viscosity, and gel strength. The highest water content was 15,76% at 2% HCl concentration, the highest ash content was 44,40% at 2% HCl concentration, and the highest protein content was 23,23% at 3% HCl concentration. The highest pH value was 4.14 produced by gelatin at 2% HCl concentration, viscosity was 5,13 cP and gel strength of 252,43, the best bloom was produced by gelatin at 6% concentration level. The conclusion of this research is the best quality of gelatin is obtained by using chicken bones and the use of chloride acid for 24 hours and 6% HCl concentration level.
