Abstract

Bacillus is a bacterium that has the ability to survive in unfavorable environmental conditions, one of which is in nutrient-poor soils such as the Liwa Botanical Gardens. Bacillus isolates have been obtained from the soil of the Liwa Botanical Gardens which have lipolytic abilities. This study aimed to determine the activity of lipolytic bacteria isolates from Bacillus from Liwa Botanical Gardens. In this study, Bacillus was isolated from the soil of the Liwa Botanical Gardens using a graded dilution method, using Nutrient Agar media. The Bacillus isolates obtained were then purified in Nutrient agar media. Bacillus isolates were inoculated in lipase test media with a composition of 0.01% methyl red, 10 g peptone, and Nutrient Agar, 5% sterile olive oil, 2.5% Tween-80, 5 g NaCl, 0.1 g CaCl2.2H2O. The results of this study obtained 5 isolates of Bacillus which had fairly good growth, namely isolates BP 13, BP 14, BP16, TMA 13, TSR 5. With the results obtained the largest lipolytic activity was isolate BP 16 with an average index value of 6.34 cm while the lowest lipolytic activity was TMA13 isolate with an average index value of 2,345 cm.

Abstract

Bacillus sp is a bacterium that is commonly found in soil, water, air and plant debris, has the ability to produce enzymes and degrade natural substrates so that it contributes to the nutrient cycle. The Kebun Raya Liwa area is located at an altitude between 890 to 948 meters above sea level where at this altitude is included in a hilly area. With varying topographic conditions so that it has different soil conditions. This can trigger the diversity of Bacillus sp. The purpose of this study was to determine the diversity index of Bacillus sp in the Liwa Botanical Gardens soil. This research was conducted at the microbiology laboratory of the Mathematics and Natural Sciences faculty, Lampung University. The method used is sampling using five different types of soil, namely litter soil, biopori soil, arase sloping soil, ordinary soil, and ordinary arase soil. The isolation technique uses the pour plate method with multilevel dilutions. Calculation of diversity using a calculation method that refers to the calculation of Shanon Wiener. The results of this study obtained the highest diversity of Bacillus sp in litter soil with a total index of 2.488464 indicating a moderate level of diversity while the lowest total value was obtained in biopore soil with a value of 1.351604 indicating a moderate level of diversity. The high and low diversity of Bacillus sp bacteria can be caused by environmental conditions. In this study, the dominant morphology of Bacillus sp is circular colony shape.

Abstract

Fusarium sp is a type of fungus that is pathogenic especially on plants. This fungus attacks the pith tissue of the stem through the injured root. Yellow leaf color is an early indication of this fungal infection. An alternative in controlling this fungus could be to use Bacillus sp because these bacteria have growth inhibitory compounds. This study aims to determine the effect of Bacillus sp from Liwa Botanical Garden on Fusarium sp. Antagonist testing using dual culture method on media Mueller Hinton Agar (MHA) with 3 bacterial isolates was tested. The results of the antagonist test showed that the isolate Bacillus sp.TSR 6 had an inhibitory capacity of 19.7 mm with a strong inhibitory category against the fungus Fusarium sp.

Abstract

Kebun Raya Liwa’s soil has the potential to allow for a population of microorganisms such as the Bacillus sp. Bacillus sp. Kebun Raya Liwa are known to produce phytohormones that have the potential to help plant growth and development, one of which is the hormone Indole Acetic Acid (IAA). This study aims to determine the potential of Bacillus sp. from the soil of Kebun Rawa Liwa in producing IAA hormones. The research method included taking soil samples, preparing Nutrient Agar (NA) medium, measuring soil pH samples, isolating soil bacteria, purifying and characterizing Bacillus sp., preparing IAA standard curves, and testing the potency of Bacillus sp. IAA production was carried out in two ways, namely measuring the content of IAA with the addition of tryptophan and without the addition of tryptophan. IAA concentration was measured using a spectrophotometer with a wavelength of 520 nm. The data obtained is presented in tabular form. The isolation results obtained 10 isolates that were able to produce IAA hormone with and without tryptophan. The highest IAA production without tryptophan was isolate BP 5 with a concentration of 1.18 ppm, and the isolate that was able to produce the highest IAA with tryptophan was isolate BP 13 with a concentration of 3.90 ppm. This indicates that the isolate Bacillus sp. from the soil rhizosphere Kebun Raya Liwa can produce IAA hormones.

Abstract

One effort to reduce the transmission of malaria is the provision of antimalarial drugs. However, the use of drugs that are not according to standards causes resistance to Plasmodium. This condition triggers the exploration of various natural compounds to prevent malaria. Secondary metabolites derived from Streptomyces sp. are known to have antimalarial activity. However, information related to secondary metabolites from Streptomyces sp. strains InaCC A497 and AB8 as antimalarials are not yet known. The purpose of this study was to determine the metabolite compounds secondary contained in Streptomyces sp. strains InaCC A497 and AB8 as antimalarial candidates. This research method is descriptive by testing secondary metabolites which are carried out through chemical compound tests, thin layer chromatography (TLC), fourier transform infra-red (FT-IR), and gas chromatography-mass spectroscopy (GC-MS). Test results of chemical compounds and FT-IR analysis of Streptomyces sp. InaCC A497 contains alkaloids, flavonoids, and triterpenoids, while Streptomyces sp. AB8 contains alkaloids and tannins. In the TLC test, Streptomyces sp. InaCC A497 produced an Rf value of 0.257 and Streptomyces sp. AB8 of 0.314. Based on the GC-MS test, Streptomyces sp. InaCC A497 produces acetic acid compounds, ethyl ester including the ester group, and 1,2-benzene dicarboxylic acid, dioctyl ester including the alkaloid group. Streptomyces sp. AB8 produces the compound 2-pentadecyn-1-ol which belongs to the aromatic alcohol group and cochlioquinone A belongs to the quinone group. Both compounds have antimalarial activity.

Abstract

Kopi organik dapat dikatakan sebagai tanaman organik jika pertanian kopi tersebut setidaknya menanam kopi mereka selama tiga tahun tanpa menggunakan bahan kimia sama sekali. Standar tiga tahun diperlukan agar kondisi kebun kopi benar-benar teruji bersih dari sentuhan kimiawi. Legalitas sertifikat organik juga mewajibkan durasi penanaman tiga tahun ini. Sehingga Teh Daun Kopi merupakan olahan yang bermanfaat baik dari segi ekonomi maupun segi Kesehatan. Teh daun kopi adalah sebuah teh herbal yang terbuat dari dedaunan tanaman kopi (Coffea robusta atau             Coffea arabica). Setelah dibakar, dedaunan tersebut dapat dikumpulkan, kemudian diseduh atau direndam ke air panas dalam bentuk mirip teh. Tujuan kegiatan ini dilakukan adalah untuk mengenalkan pada masyarakat adanya manfaat dan nilai jual dari daun kopi, sehingga tidak hanya menjadi tumpukan sampah organik saja. Setelah kegiatan ini berlangsung dapat dilihat antusias warga pekon Suka jaya untuk melakukan pengembangan pengolahan produk teh yang berasal dari daun kopi.

Abstract

Learning media is one of the methods used in the learning process and is highly needed in facilitating material delivery. In presenting biology materials, such as biodiversity, attractive media is crucial in helping students understand without excessive memorization. Preserved specimen is an example of learning media in this situation. Wet and dry herbariums are one of the preserved specimens commonly used by teachers. Meanwhile, bioplastic creation using resin has not been a common practice. Some advantages of bioplastic creation include more beautiful and durable results while also can be created as keychains. The aim of this community service is to train teachers in SMA 15 Bandar Lampung in making animal and plant preserved specimens through various procedures. After the training, there was an increase in knowledge and skill in making preserved specimens. The average pre-test score was 56.25, while the average post-test was 90,63. The average score escalated by 34,38 points.
