Abstract

Senduduk bulu wild plant contains secondary metabolite compounds which used natural reducing agent on synthesis of silver nanoparticles (AgNPs). This research determine the optimum extract concentration and temperature in the process synthesis of silver nanoparticle, characteristics of silver nanoparticles and antibacterial activity of silver nanoparticles against S.aureus and E.coli. Synthesis of silver nanoparticles senduduk bulu leaf using variations extract concentration and temperature variations. The characteristics of silver nanoparticles senduduk bulu leaf were carried out using a UV-Vis Spectrophotometry, Particle Size Analyzer (PSA) and Scanning Electron Microscopy – Energy Dispersive X-Ray Spectroscopy (SEM-EDS). Antibacterial activity testing used the disc diffusion method. The results of research showed the silver nanoparticles of senduduk bulu leaf were successfully synthesized under optimal conditions an extract concentration of 1% and temperature of 60ºC indicated the colloid color changing to brown and maximum wavelength value of 413 nm with absorbance value of 2.9987. The size of colloidal AgNPs of senduduk bulu leaf was 59.2 ± 1.03 nm; zeta potential value -39.3±0.509 mV. The morphology of the AgNPs of senduduk bulu non uniform shape, the crystal size distribution is 50 – 230 nm with an average of 136 nm. The chemical composition of AgNPs crystals shows that silver (Ag) dominates with a weight concentration of 37.688% and atomic concentration of 7.605% at the peak of the 3 KeV spectrum. Feathered AgNPs had antibacterial activity against S.aureus of 14.1 ± 0.3 mm and E. coli of 13.5 ± 0.2 mm. Abstract

Cosmetic care products are designed to enhance human appearance. Centella asiatica (L). Urban, commonly known as gotu grain, is a natural ingredient that can be effectively used in anti-acne creams. The objective of this study is to investigate and compare the antimicrobial efficacy of cold cream and vanishing cream bases against Staphylococcus aureus, with a focus on determining their potential inhibitory effects. The preparation of cold cream involved a melt-emulsification process utilizing surfactants. In contrast, the vanishing cream was prepared using a melting method. Physical quality assessment was conducted through visual observation, evaluating organoleptic properties and homogeneity. The physical and antibacterial properties of the creams were evaluated using various standardized methods. Viscosity was measured using a Brookfield Viscometer, pH levels were assessed with a pH meter, spreadability and adhesion were evaluated using respective testers, and antibacterial activity was determined via the disc diffusion method. Statistical analysis of the physical quality test results was performed using the Independent Samples T-test, while antibacterial activity data were analyzed using One-Way ANOVA. The results of the physical quality evaluation revealed significant differences in the organoleptic properties among the gotu-gotu extract cream, cold cream base variation, and vanishing cream. However, no significant differences were observed in terms of homogeneity among the three formulations. The vanishing cream exhibited a significantly higher spreadability, with a mean value of 5.98 cm (n=3), compared to the cold cream base variation. Conversely, the vanishing cream displayed lower viscosity (mean: 3,560 cP, n=3) and adhesion (mean: 1.61 seconds, n=3) values compared to the cold cream base variation. The gotu-gotu extract cream formulated with a vanishing cream base exhibited potent antibacterial activity against Staphylococcus aureus, as evidenced by a significant inhibition zone diameter of 17.6 mm. This result meets the criteria for strong inhibition strength and is comparable to the positive control, demonstrating superior antibacterial efficacy relative to the cold cream base variation.

Abstract

Acne is experienced by 90% of teenagers due to infection with the bacteria Staphylococcus epidermidis. Bacterial infections are treated with antibiotics, but excessive use causes resistance, so medicinal plants are used that do not have side effects. (Aloe vera L.) have antibacterial activity because contain anthraquinone compounds. The aim of research is update the optimization of aloe vera leaf gel to increase the effectiveness of use. The research method uses Simplex Laticce Design with HPMC, Karbopol and TEA. The optimum formula concentration of 1.3379% HPMC, 2.540% Carbopol and 0.481% TEA produced predicted and experimental values for adhesion power (7.977 and 7.5 seconds), spreadability (7 and 6.87 cm), pH (6.128 and 6 ), viscosity (246.775 and 265.3 dPa.S) and inhibition zone (23.772 and 22.66mm). The results of the Paired Sample Test for verification data show a p value of 0.430 > 0.05, is not significant difference between the predicted and the experimental.

Abstract

Staphylococcus aureus is one of the bacteria that cause acne and can cause skin and organ damage because bacteria can defeat the body's defences. Carrot leaf extract has antibacterial power or inhibitory activity against S. aureus. This study aims to determine the inhibition value and antibacterial activity of ethanol extract and n-hexane fraction of carrot leaves. The research method consists of extraction using 96% ethanol and fractionation using water, ethyl acetate, and n-hexane. The antibacterial activity of carrot leaf extracts and fractions was carried out using the pitting method as well as the use of gentamicin positive control and DMSO negative control. The results showed that 96% ethanol extract of carrot leaves with a concentration of 30% had inhibition against S. aureus bacteria with a value of 7.8 mm or weak inhibitory activity, and the ethyl acetate fraction of carrot leaves with a concentration of 20% had inhibition against S. aureus bacteria with a value of 11.27 mm or strong inhibitory activity. Therefore, ethyl acetate fraction of carrot leaves have inhibition against S. aureus bacteria with strong inhibitory activity compared to other test solutions and is equivalent to gentamicin positive control

Biofilm degradation testing at a concentration of 50 mg/ml resulted in an average value of 60.43 ± 1.11, a concentration of 25 mg/ml with an average value of 50.05 ± 0.87 and a concentration of 12.5 mg/ml with an average value -mean 35.86 ± 1.05. The average EC50 value was 26.731 ± 1.1874. Tests using AAS on K+ metal ions showed ion leakage or cell contents coming out was 1,805 mg/L, Ca2+ metal ions resulting from this leak were 3,884. Morphological observations using SEM showed that the samples experienced shrinkage and reduced size from being perfectly round to oval to S.aureus bacteria in samples with water fraction

Abstract

Antibiotic resistance has become a critical issue in global health, one of which is MRSA. The dynamic problem related to the increasing MRSA infections is clear evidence that the treatment and management of this disease have not been optimally resolved, requiring the development of effective antimicrobial agents against MRSA. An exploratory method was used to assess the antibacterial activity, interaction patterns, and stability of sidaguri compounds. A total of 15 potential compounds from sidaguri were docked onto four molecular targets: PBP2a (4JCN), MecR1 (609S), FtsZ (8HTB), and SCCmec (4FAK), using the molecular docking methods AutodockTools and PyMol, followed by analysis of the amino acid residue similarity of each test ligand. The molecular docking results predicted that the compounds with the best binding affinity and interaction patterns similar to the natural ligand for their respective molecular targets were quercetin, ecdysone, and 24-methylenecholesterol. The validation parameters are calculated from RMSD and RMSF based on the In-Silico predictions. MD simulations were performed using YASARA Dynamics. The MD simulation results showed that, from the RMSD and RMSF graphs of the MRSA target molecules, quercetin, ecdysone, and 24-methylenecholesterol exhibited binding stability close to that of the natural ligand. The predicted pharmacokinetic profiles show that all three compounds have a good ADMET profile, with optimal absorption potential, adequate distribution, appropriate metabolism, and relatively low toxicity, making them promising candidates for development as antibacterial drugs against MRSA
