Abstract

The community service program held in Jendi Village, Girimarto District, Wonogiri Regency, was titled plastic waste processing training. The community service activity was carried out with the aim of implementing plastic waste management through educational activities and mentoring on managing plastic waste into items of economic value. This activity also aims to mitigate the impact of environmental damage caused by plastic waste. The method used in the community service was a participatory approach. This approach was carried out by involving the service team and the community in managing plastic waste. The location used was the Vimalakirti Temple complex, Jendi Village, Girimarto District. Meanwhile, the subjects assisted in the implementation of the service were Buddhists and residents of Jendi Village. This community service activity produced three results. First, the establishment of an ecosystem that cares about waste, especially plastic waste. Second, the realization of community understanding regarding how to sort plastic waste. Third, the community was educated about plastic waste management to generate economic benefits and have an impact on efforts to prevent environmental damage caused by plastic waste.

Abstract

The application of pectinase in industries will continue to be developed. To broaden the applicability of enzyme manufacturing in new industries, more research to investigate pectinolytic microorganisms with high activity and stability is still required. Therefore, this study aimed to obtain pectinolytic fungi that have the potential as pectinase producers. Pectinolytic fungi were isolated from rotten apple peels and selected based on clear zone formation on pectic agar media after cetyl trimethyl ammonium bromide (CTAB) staining. Pectinolytic fungi were identified based on 18S rRNA partial gene sequences and morphological characteristics. Pectinase production used Mandels and Weber medium with citrus pectin 20 g l-1 addition. Pectinase activity was determined based on the measurement of reducing sugars by a colorimetric method. A total of 5 fungal isolates were successfully isolated. All of the isolates had pectinolytic activities with clear zone diameters ranging between 0.99 to 7.32 mm. The isolate A3 showing the highest pectinolytic activity was identified as Rhizopus stolonifer. Microscopically, R. stolonifer A3 showed typical characteristics for Rhizopus, characterized by the presence of rhizoids, stolons and sporangiophores bearing a single spherical sporangium. The pectinase production of R. stolonifer A3 was optimum at initial pH 5.0, temperature 35 °C and incubation period of 3 days with pectinase activity of 14.75 U ml-1. R. stolonifer A3 produced acidic pectinase having optimum activity at pH 5.0 and temperature 50 °C. Thus R. stolonifer A3 has the potential to be used as a producer of acidic pectinase appropriate for use in the processing of fruit products.

Abstract

Keju merupakan salah satu produk olahan susu dengan nilai gizi yang lengkap serta memiliki cita rasa yang khas, sehingga digemari oleh masyarakat. Pada proses pembuatannya, keju menghasilkan limbah cair (whey) saat protein keju diendapkan dan dipadatkan.  Whey yang dibuang begitu saja tanpa diolah akan menimbulkan dampak bagi lingkungan dan dapat menimbulkan protes masyarakat sekitar. Di sisi lain, limbah cair keju masih memiliki kandungan nutrisi yang cukup untuk menjadi bahan baku pembuatan produk fermentasi seperti minuman atau produk lainnya yang memiliki manfaat bagi kesehatan. Minuman sinbiotik berisi probiotik, bakteri yang baik, serta prebiotik, yang merupakan nutrisi selektif untuk bakteri baik.  Pada jangka panjang, pengolahan lebih lanjut limbah cair keju ini dapat memberikan tambahan penghasilan bagi pemilik usaha melalui diversifikasi produk, serta berkontribusi untuk memenuhi kebutuhan masyarakat terhadap bahan pangan fungsional, juga meningkatkan perekonomian di lingkungan usaha mitra. Tujuan kegiatan ini adalah memberikan solusi pengelolaan limbah produksi keju di PT. Mazaraat Lokanatura Indonesia menjadi produk berupa minuman sinbiotik dengan menerapkan hasil riset di Grup Riset Biomateri Mikroba.  Selain itu, kegiatan ini juga akan ditautkan dengan program Merdeka Belajar Kampus Merdeka (MBKM) di prodi Biologi FMIPA UNS.   Luaran kegiatan ini berupa ide pemanfaatan whey serta penerapan teknologi pembuatan produk dari whey yang bisa dipasarkan untuk  mengurangi dampak limbah cair, serta rancangan kegiatan MBKM berupa magang industri di institusi mitra yang dapat direkognisi sampai 21 SKS untuk mahasiswa. Dari kegiatan ini dihasilkan produk minuman sinbiotik menggunakan bakteri probiotik Lactobacillus acidophilus serta prebiotik biji nangka, dengan penambahan ekstrak kunyit dan ekstrak kulit naga sebagai pewarna dan antioksidan.  Produk sinbiotik ini juga dijadikan sebagai bahan pembuatan produk perawatan diri berupa sabun serta lulur.
