Abstract

Hypertension is a serious health problem and requires a good handling. An alternative treatment for hypertension is required due to the treatment of hypertension took a long period of time causing unwanted side effects. Melinjo seed hydrolyzed-proteins have an ACE-inhibitor activity, an antihypertensive effect. This study aimed to determine the antihypertensive effects of melinjo seed hydrolyzed-proteins in hypertensive rats. Wistar rats induced with prednisone 1,5 mg/kg body weight and 2% NaCl for 7 days and then treated. There were 7 groups: a normal group K, a group K(-) which is a hypertensive rats without treatment, a group K(+) which is hypertensive rats with captopril 2,5 mg/kg body weight on day 8th, and four treatment groups P1, P2, P3, and P4 which are hypertensive rats with melinjo seed hydrolyzed-proteins with a dose of 5, 10, 20, and 30 mg/kg body weight on day 8th, respectively. Blood pressure was measured before and after treatment every 1 hour for 5 hours. The results showed that melinjo seed (Gnetum gnemon) hydrolyzed-proteins can decrease blood pressure in hypertensive rats.   Keywords: Antihypertensive, Gnetum gnemon, Hydrolized-Protein, Blood Pressure

Abstract

Hydrolyzed protein isolates of melinjo seed using free alcalase has been known to have antioxidant potential. However, free alcalase can only be used once. Enzymes immobilization technique with entrapment method would overcome this problem. The immobilized enzyme characteristic was observed by SEM and hydrolysis degree value. The success of hydrolysis process was observed by SDS-PAGE. Antioxidant activity test was determined by the ability to inhibit hydroxyl radical and protection against DNA damage in 1% agarose gel. The result showed that protein isolates from melinjo seed had been successfully hydrolyzed by immobilized alcalase. Following hydrolysis showed that immobilized alcalase was effective to hydrolyze protein up to four times. Based on hydroxyl radical reduction activity test, antioxidant activity of hydrolyzed protein using immobilized alcalase had the same as that of glutathione as positive control antioxidant activity. In addition, hydrolyzed protein using immobilized alcalase had the ability toprotect DNA damage which showed by reversible change of supercoil band. Abstract

Hypertension is ranked third on the cause of death in Indonesia for all ages (6.8 %). Protein isolate of melinjo seeds (Gnetum gnemon L.) hydrolyzed with free alcalase has been known to have antihypertensive activity. However, the use of free alcalase is uneconomical since it can only be used once, thus we studied the immobilized alcalase effectivity to hydrolise the melinjo seeds protein isolate to be used as angiotensin converting enzyme (ACE) inhibitor. The immobilization method used entrapment method with DMDMOS/TMOS as matrix. Characterization of the immobilized enzyme was observed by FTIR and effectiveness of repeated hydrolysis was observed by the value of the degree of hydrolysis. The success of the hydrolysis process was determined by SDS-PAGE electrophoresis. Antihypertensive activity test was determined by the ability to inhibit ACE. The results showed that the alcalase has been immobilized in the matrix of silane and SDS-PAGE protein profile showed melinjo seed proteins had been successfully hydrolyzed. Hydrolysis process repeatedly demonstrated that immobilized alcalase was effective to hydrolyze protein isolates of melinjo seeds twice. Based on ACE inhibitory test, there were no significant differences between the protein isolate hydrolyzed by immobilized alcalase (Gg-PH), protein isolates hydrolyzed with free alcalase (Gg-PHB), and captopril. These findings suggest that the antihypertensive activity in Gg-PH is the same with that of captopril. Keywords: hypertension, antihypertensive, melinjo, enzymes immobilization, ACE inhibitor

Abstract

Hipertensi merupakan suatu keadaan seseorang ketika terjadi peningkatan tekanan darah sistolik ≥ 140 mmHg dan/atau tekanan darah diastolik ≥ 90 mmHg. Pengobatan jangka panjang hipertensi menimbulkan beberapa efek samping sehingga diperlukan suatu alternatif pengobatan untuk hipertensi, misalnya dengan protein isolat biji melinjo. Penelitian ini bertujuan untuk mengetahui efek antihipertensi dari protein isolat biji melinjo pada tikus hipertensi. Tikus wistar diinduksi dengan prednison 1,5 mg/kgBB dan NaCl 2% selama 7 hari kemudian diberi perlakuan. Terdapat 7 kelompok perlakuan, kelompok K merupakan tikus normal, kelompok K(-) adalah tikus hipertensi tapi tidak diberi perlakuan, kelompok K(+) adalah tikus hipertensi dan diberi kaptopril 2,5 mg/kgBB pada hari ke-8, kelompok P1, P2, P3, dan P4 merupakan tikus hipertensi dan masing-masing diberikan protein isolat biji melinjo dengan dosis 5 mg/kgBB, 10 mg/kgBB, 20 mg/kgBB, dan 30 mg/kgBB pada hari ke-8. Tekanan darah diukur sebelum dan setelah diberi perlakuan tiap 1 jam selama 5 jam. Data dianalisis secara statistik menggunakan uji Kruskal Wallis dan Mann. Dari hasil penelitian menunjukkan bahwa protein isolat biji melinjo tidak dapat menurunkan tekanan darah tikus model hipertensi.   Kata kunci: Antihipertensi, Gnetum gnemon, Protein Isolat, Tekanan Darah
