Abstract

Aim of this research is to determine physical characteristic of ibuprofen-stearic acid due to compression force. Binary mixture of ibuprofen-stearic acid with weight ratio of 4:6, 5:5, and 6:4 was compressed under various compression force using hydraulic press with 13mm diameter flat punch.Identification of solid state interaction between these two components was performed by Hot Stage Microscopy (HSM). Physical characterization has been studied byDifferential Thermal Analysis (DTA), X-ray Powder Diffraction (XRPD), Scanning Electron Microscopy (SEM), and (Fourier Transform Infrared (FT-IR) spectroscopy. According to hardness measurement, 4:6 weight ratio has the highest tensile strength on 170.6N/cm2 under 19.93kN compression force. Interaction identification by HSM showed single blank line that indicates eutectic formation. Thermal analysis of DTA also revealed eutectic formation upon compression whichthe endotermic peak of ratio of 4:6has the lowest melting temperature of 53.2°C. X-ray diffraction of three peaksgenerally showed that peak intensity decreases as compression force increases, but at particular point it begins to increase again. Sintering phenomenon at the surface of compressed tablet was observed from SEM analysis. FT-IR study confirms the formation of simple eutectic. Key words: ibuprofen, stearic acid, binary mixture, compression force, physical characterization

Abstract

Ketoprofen is a non-steroidal anti-inflammatory drug with poor water solubility, so the absorption is less than optimal. One method to improve the solubility of ketoprofen is through the formation of multicomponent solid forms. The success of the formation of the multicomponent solid forms is strongly influenced by interactions between components in their solids. In this study, the analysis of the interactions in solid form of ketoprofen-coformers was carried out using the differential scanning calorimetry (DSC) and hot stage microscopy (HSM) with adipic acid and isonicotinamide as coformers. From the experimental results, the mixtures of ketoprofen-adipic acid show a solid-liquid phase diagram that indicates a simple eutectic system with eutectic points on the molar fraction of ketoprofen 0.9 and temperature at 92.9 °C. The ketoprofen-isonicotinamide mixtures have a eutectic system with the peritectic point. The solid-liquid phase diagram has indicated that the ketoprofen-adipic acid in eutectic composition forms a miscible liquid phase without interaction in its solid form, whereas the ketoprofen-isonicotinamide forms a miscible liquid phase accompanied by interaction with the excess component. The results of the HSM analysis showed the same phenomenon as the result of the DSC experiment and have confirmed with the FTIR analysis

Abstract

The aim of this research was to get optimal formula of levofloxacin tablet prepared with variation of PVP K-30 as binder and vivasol as disintegrant. The making of levofloxacin tablets was done by wet granulation. Tablet was prepared with various levels of PVP K-30 and disintegrant vivasol, compressed using a hydraulic press with 12 mm punch diameter, for 3 seconds. Physical quality (hardness, friability, and disintegration time) and dissolution rate of tablet was evaluated. The optimization of the formula was done by factorial design of 22 factorial experiments with 2 factors (PVP K-30 and vivasol) and 2 levels (2% and 4%). Optimization results showed that elevated levels of PVP K-30 increased tablet hardness, reduced friability of tablet, decreased disintegrating time, and increased dissolution rate of levofloxacin tablets. Meanwhile, elevated levels of vivasol increased the hardness of tablets, decreased the disintegrating time of tablets, decreased the dissolution rate of levofloxacin tablets, but did not affect the friability of tablets. In conclusion, the optimal tablet that meet the specifications of physical quality (hardness, friability, and disintegrating time) and dissolution rate was made by 2.4 to 3.7% of PVP K-30 and 2.0 to 3.2% vivasol as shown in the feasible area of design space.

Abstract

This research has been carried out to study inhibition of eutectic interaction which occurs on compressed binary mixture of ibuprofen-stearic acid by addition of corn starch. Corn starch was mixed into binary mixture of 40:60 (% b/b) ibuprofen-stearic acid with composition 10, 20, and 30% (w/w) and then compressed into tablet form. The physical characterization was conducted using tensile strength test, differential thermal analysis (DTA), powder X-ray diffraction (PXRD), Fourier transform infrared (FT-IR), and scanning electron microscope (SEM). According to tensile strength test, the value was reduced upon addition of 10 and 20% corn starch compared to the compressed binary mixture. Addition 20% corn starch to compressed binary mixture shifted the eutectic diffraction peaks with a decline in intensity during PXRD study. Thermal analysis of compressed binary mixture-20% corn starch exhibited an increase in eutectic point, and FT-IR spectrum was observed no difference compared to the compressed binary mixture. SEM analysis indicated that 20% corn starch addition was able to inhibit sintering phenomena by dispersed on particle’s surfaces. Physical characterizations result in the conclusion that corn starch can inhibit the eutectic interaction between ibuprofen and stearic acid upon compression

Abstract

Karakterisasi fisik tablet campuran biner meliputi uji kekuatan mekanis, difraksi sinar-X serbuk (DSXS), scanning electron microscope (SEM), differential thermal analysis (DTA), spektrofotometri inframerah, dan terakhir dilakukan uji disolusi.

Abstract

Background: In old ages there is a change in the immune system along with the aging process called the term immunosenescence. Neutrophil cells play an important role in natural immunity because they are the first immune cells to be deployed in the body's defenses. Objective: To prove that there is a difference in the function of neutrophil cell oxidative burst in older age group compared with the younger age group as well as the tendency of decreased oxidative burst function of neutrophil cells with increasing age.Method: This is a cross-sectional observational analytic study involving 48 healthy subjects. The subjects were divided into 3 age groups: young age (18-40 years old), middle age (41-59 years old), and old age ≥60 years old). Each of them were examined for the function of neutrophil cells oxidative burst. The data were then analyzed using one-way ANOVA test. The result was considered significant if p<0.05.Results: The obtained mean age 59.26±8.03 years old. The mean age for young age group was 28.75±6.66 years old. The mean of middle age group was 50.19±5.46 years old. The mean age of old group was 66.38±3.83 years old. The mean of netrofil oxidative burst cell function was 96.83±2.7% with mean of young age being 98.57±0.98%, middle age 97.71±1.64%, and old age 94.20±3.56%. One way ANOVA comparison analysis showed a significant difference with p = 0.000 (significant when p <0.05). The result of Rank Spearman test showed significant result with r=-0.590 (p=0.000).Conclusion: There are differences in the function of neutrophil cells oxidative burst between young and old age groups. There is a negative trend between the age group and the function of neutrophil cell oxidative burst. The increase of age causes decrease in the function of neutrophil cells oxidative burst.
