Abstract

Dexamethasone (Dex) is synthetic corticosteroid, known as anti-inflamation drug to ameliorate autoimmune diseases. It worked by inhibiting production of proinflamatory citokines. The aim of this experiment was to confirm the effect of administration of Dex on erythroid progenitor cells, TER-119+VLA-4+ and erythrocytes, TER-119+VLA-4- expression from bone marrow compartments. Two weeks old BALB/c mice were used and grouped into 3 injection treatments of Dex with six replications i.e. no injection (control), 0.5 mg/kg BW (dose 1), 10 mg/kg BW (dose 2). To investigate the therapeutic effect of dexamethasone, the mice were sacrified on day-7. The bone marrow cells were isolated and analyzed by flow cytometry. Data analysis was confirmed with the Kruskal-Wallis test followed by Mann-Whitney test with significance level (α) of 0.05. The result showed that administration of Dex on BALB/c mice increase the expression of erythroid progenitor cells, TER-119+ VLA-4+ and erythrocytes, TER-119+VLA-4- from bone marrow.Key words: Dex, erythrocytes, erythroid progenitor cells

Abstract

ABSTRAK Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian formalin dan benzo(α)pyrene terhadap struktur jaringan hati dan paru mencit (Mus musculus). Mencit jantan berumur dua bulan terpapar formalin dosis 2 mg/kg berat badan dengan perlakuan oral selama 60 hari dan terpapar benzo(α)pyrene dosis 250 mg/kg berat badan dengan perlakuan injeksi intraperitoneal sebanyak empat kali setelah 30 hari dengan selang waktu satu hari. Stuktur histologi hati dan paru mencit dianalisis secara deskriptif. Jumlah hepatosit yang mengalami nekrosis dianalisis dengan menggunakan uji Mann-Whitney pada software SPSS 16.0 for Windows. Hasil dari penelitian ini menunjukkan bahwa pemberian formalin, benzo(α)pyrene dan kombinasi antara formalin dan benzo(α)pyrene meningkatkan jumlah hepatosit yang mengalami nekrosis. Nekrosis terjadi di sekitar area portal triad dan di sekitar area vena sentralis pada hati mencit, sedangkan nekrosis terjadi di sekitar area vena pada paru mencit. Abstract

ABSTRAK Studi mengenai hubungan kekerabatan, evolusi dan identifikasi spesies secara molekuler tidak terlepas dari isolasi DNA dan amplifikasi gen COI dan 16s rRNA. Keberhasilan isolasi DNA dan amplifikasi gen COI dan 16S rRNA merupakan langkah awal yang penting untuk studi mengenai hubungan kekerabatan, evolusi dan identifikasi spesies. Penilitian ini bertujuan untuk mengamplifikasi gen COI dan 16S rRNA dari invertebrata laut yang diambil dari Raja Ampat, Plakobranchus ocellatus. Sepasang primer COI universal LCO-1490 dan HCO-2198 digunakan untuk mengamplifikasi fragmen gen COI dari P.ocellatus, sedangkan amplifikasi gen 16s rRNA menggunakan sepasang primer 16Sar-L dan 16Sbr-L. Fragmen gen COI dan 16S rRNA berhasil teramplifikasi dengan panjang amplikon masing-masing sekitar 680 bp dan 450 bp.   Kata kunci: COI, gen, identifikasi, Plakobranchus ocellatus dan 16S rRNA ABSTRACT The first important step to study about organism relationship, evolution and species identification are DNA isolation and amplification of 16S rRNA and COI genes. This research aimed to amplify COI and 16S rRNA genes of marine invertebrates taken from Raja Ampat, Plakobranchus ocellatus. A pair of COI universal primer LCO-1490 and HCO-2198 was used for ampification a fragment COI gene P.ocellatus, while a fragment of the 16s rRNA gene was amplified using 16Sbr-L and 16Sar-L primer. Either  a fragment COI or 16S rRNA genes was successfully amplified, with a length amplicon about 680 bp and 450 bp, respectively. Abstract

Tujuan penelitian ini adalah untuk mengetahui efek lama pembekuan semen terhadap kadar MDA spermatozoa dan persentase viabilitas spermatozoa. Sampel yang digunakan berupa  semen beku sapi Limousin dengan lama pembekuan 2 bulan dan 3 bulan yang dikoleksi dari BBIB Singosari. Semen beku yang telah dithawing diencerkan dengan Phosphate Buffer Saline (PBS) kemudian disentrifugasi untuk memisahkan spermatozoa dan seminal plasma. Kadar MDA spermatozoa diukur dengan menggunakan uji TBA dan spektrofotometer. Pengamatan viabilitas spermatozoa dilakukan dengan menggunakan pewarnaan eosin-negrosin. Data berupa kadar MDA spermatozoa dan persentase viabilitas dianalisis dengan uji-t berpasangan dan korelasi pearson menggunakan spss 16.0. Hasil analisis menunjukkan bahwa kadar MDA spermatozoa pada lama pembekuan 2 bulan (0,0777±0,008 nM) dan 3 bulan (0,0919±0,016 nM) tidak berbeda nyata (p<0,05). Terdapat korelasi negatif antara persentase viabilitas dan kadar MDA pada spermatozoa. Kata kunci: MDA, semen beku, spermatozoa

Abstract

Penelitian ini bertujuan untuk menganalisa efek pemberian alpha-S2 kasein  susu kambing PE terhadap perubahan morfologi dan histologi membran sinovial tikus Rheumatoid Arthritis (RA). Tahapan penelitian meliputi pembuatan RA menggunakan multidosis CFA (Complete Freud Adjuvant. Kelompok perlakuan dibagi menjadi (C: Kontrol; CM: Kontrol + alpha-S2 kasein; RA: Rheumatois Arthritis; RAM: Rheumatois Arthritis + alpha-S2 kasein). Treatment RA dilakukan  selama 2 bulan dengan dosis 2mg/kg. Analisa morfologi dilakukan dengan skoring perubahan bentuk kaki dan histologi menggunakan metode HE yang divisualisasi menggunakan mikroskop BX53. Hasil penelitian menunjukkan adanya perbaikan morfologi yang ditandai dengan penurunan hasil skor klinis (C = 0; RA = 4; CM = 0; RAM = 1.3). Perbaikan histologi terletak pada penurunan jumlah hiperplasia pada kelompok RA + alpha-S2 kasein menuju kondisi normal dimana penurunan level tersebut mengidikasikan alpha-S2 kasein dapat digunakan sebagai alternatif penyembuhan RA.

Abstract

Porang (Amorphophalus oncophyllus Hook.), a tuber plant, is intensively explored for export commodities from East Java and as a raw food stuff material for domestic industry. The prospects of Porang tubers as the healthy food is interesting, however some studies are needed for their allergy effects. It was thought that the allergy effects of Porang are caused by the composition of Calcium Oxalate crystal that leads the itching and irritation in the mouth. On the other hands, the food allergy is usually caused by unchanged protein during food digesting.  Some proteins have been known that play a role in Calcium Oxalate crystal (Matrix proteins) formation, they are: Asp-rich and Ser-rich acidic proteins that are tightly bound to Calcium. So, substantially the allergen of Porang tuber is from the matrix protein, not from its crystal. The allergen in the food is usually a protein that is not change during the food digestion. In this study, it has been done the allergic test of some Porang tuber variants, namely: Sumber Baru-Jember; Klangon, Saradan-Madiun and Sumber Bendo, Saradan-Madiun, by ELISA-IgE from rat’s blood serum that have been orally treated by raw Porang tubers powder before.  The SDS-PAGE analysis shows that the proteins bands profiles are similar between raw and steamed Porang tubers (appearance Molecular Weight: 18 and 15 kDa). But, those raw Porang tubers bands are thicker than the steamed ones. The thickness of those raw Porang tubers decrease from 25 to 50 %, compare to those steamed ones. It means that the steaming of Porang is effective to decrease the potential allergy of Porang for consumptions. The allergic test analysis by ELISA-IgE revealed that the potential allergy of Porang are: Sumber Baru (Jember) > Porang var. Klangon - Saradan (Madiun) > Porang var. Sumber Bendo - Saradan (Madiun), respectively.

Abstract

Hematopoiesis play important roles on maintaining body homeostasis. Dexamethasone (Dex) known as synthetic glucocorticoids and widely affect many regulations i.e. immune system, inflammation, reproduction and all aspects of metabolism. Generally Dex were used as therapeutic agentsto ameliorate many kind of diseases including cancer and auto-immune diseases. The aims of this research were to investigate the effect of Dex administration toward expression of hematopoietic stem cells and blood progenitor cells. Two weeks old BALB/c mice were used and treated with 3 groups of Dex administration i.e. untreated-mice, 0.5 mg/kg BW, and 10 mg/kg BW. Mice were sacrificed on day-7 after administration of Dex. The bone marrow stromal cells were isolated, then followed by flow cytometry analysis. The result showed that, Dex administration with dose 0.5 mg/kg BW significantly increase the expression of hematopoietic stem cells, CD34+, chemokines, SDF-1+, erythroid progenitor cells, Gr-1-TER-119+, granulocytes, Gr-1+, and B progenitor cells, B220+.

Abstract

Tolerance between donor and recipient is a key for successful allogeneic transplantation. To determinethe involvement of CD8+ T cells in regulating T cell homeostasis in allogeneic transplantation, spleen cellswere analyzed post bone marrow transplantation. In this study, C57BL/6 mice were used as donor andBALB/c mice were used as recipients. BALB/c mice were irradiated with a lethal dose of 850 rad prior tothe transplantation. Twenty-four hours post a radiation BABL/c mice were transplanted with bone marrowcells (10x106) derived from C57BL/6 mice that free from mature T cells. Analysis of hematopoietic cells inthe spleen was done two months post-transplantation. The results showed that CD8+CD25+FOXP3+ T cellin collaborasion with CD4+ regulatory T cell contribute to the prevention of graft-versus-host diseases(GVHD). In this study at was demonstrated that naïve CD4+CD62L+ T cells dominated the sub set of Tcells, whereas the development of CD4+CD44+ memory T cells are very limited. In conclusion, that regulatoryT cells maintain normal homeostasis by maintaining T cells remained at naïve status.

E-mail: ririndwi68@yahoo.com ABSTRAK Sapi Friesian Holstein (FH) merupakan bangsa sapi yang memiliki produksi susu tertinggi dibandingkan bangsa-bangsa sapi perah lainnya. Banyak peternak yang menginginkan peningkatan produktifitas sapinya dengan asumsi setiap 1 ekor sapi setidaknya melahirkan 1 ekor anak sapi setiap tahun. Namun banyak masalah yang ditimbulkan pada sapi postpartum, salah satunya akibat peningkatan radikal bebas pada tubuh sapi postpartum, sehingga asumsi tersebut tidak terpenuhi. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian selenium dan vitamin E secara intramuskular terhadap kadar Luteinizing Hormone (LH) serum sapi FH postpartum. Hewan coba yang digunakan adalah 6 ekor sapi bunting 7 bulan yang dibagi menjadi 2 kelompok, masing-masing 3 ekor untuk kelompok kontrol (P0/ tidak diinjeksi selenium dan vitamin E) dan perlakuan (P1/di injeksi sodium selenite 1,5 mg/ml + vitamin E 50 mg/ml). Injeksi dilakukan 5 kali yaitu saat sapi bunting 7 bulan, 8 bulan, saat 2 minggu sebelum melahirkan, 7 hari dan 14 hari setelah melahirkan. Pengambilan serum untuk pengujian kadar LH dilakukan dua kali dari masing-masing hewan coba, yaitu pada 25 hari dan 45 hari postpartum. Pemeriksaan kadar LH dilakukan dengan teknik ELISA (Enzyme-Linked Immunosorbent Assay). Hasil penelitian menunjukkan bahwa pemberian sodium selenite 1,5 mg/ml + vitamin E 50 mg/ml cenderung menyebabkan peningkatan LH serum sapi FH pada 25 dan 45 hari postpartum.   Kata kunci: LH, sapi FH, selenium, vitamin E ABSTRACT Holstein Friesian dairy cows (FH) is a group of cows that have the highest milk production compared to other dairy cattle groups. Many farmers want to increase the productivity of the cow with the assumption that every one cow give birth to at least 1 cow each year. But many problems caused in postpartum cows, one of which is due to the increase of free radicals in the postpartum cows body, so the assumptions are not met. This study was conducted to analyze the effect of selenium and vitamin E intramuscularly in LH levels serum of dairy cow FH postpartum. There are 6 cows divided into two groups, 3 cows for each group. First group is control (P0) which no administration of sodium selenite and vitamin E, and second group is treatment (P1) that intramuscularly injected of sodium selenite 1.5 mg/ml + vitamin E 50 mg/ml). Injection of selenium and vitamin E conducted 5 times when 7 months, 8 months cows gestation, two weeks before calving, seven days after calving and 14 days after calving. Serum sampling for testing levels of LH take in 25 days and 45 days postpartum on each cows. The level of LH was conducted by ELISA (Enzyme-Linked Immunosorbent Assay). The results showed that administration of sodium selenite 1.5 mg/ml + vitamin E 50 mg/ml causes increased LH levels serum that taken 25 days, 45 days postpartum.   Keywords: dairy cow FH, LH, selenium, vitamin E

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak ethanol kemangi (Ocimum canum Sims.) terhadap struktur histologi testis mencit jantan (Mus musculus). Penelitian ini menggunakan 9 ekor hewan coba mencit strain Balb/c jantan berumur 3 bulan, berat badan 20 - 30 g. Hewan coba dibagi menjadi tiga (3) kelompok kontrol tanpa pemberian ekstrak kemangi (P0), kelompok yang diberi ekstrak kemangi dengan dosis 100 mg/kgBB (P1) dan 200 mg/kgBB (P2). Ekstrak kemangi diberikan setiap hari secara oral selama 35 hari. Parameter yang diamati adalah diameter tubulus seminiferus, tebal epitel germinal, jumlah lapisan germinal dan jumlah sel spermatogenik meliputi spermatogonia, spermatosit dan spermatid. Data yang diperoleh dianalisis dengan One-way ANOVA (p ≤ 0,05). Jika menunjukkan perbedaan yang bermakna, maka dilanjutkan dengan uji Least Significant Difference (LSD). Hasil penelitian menunjukkan bahwa pemberian ekstrak kemangi dosis 200 mg/kgBB meningkatkan diameter tubulus seminiferus, tebal epitel germinal, dan jumlah sel spermatid
