Abstract

Identification of species Raja Ampat required in order to obtain marine biodiversity in Raja Ampat. Synaptula is one of the organism that inhabit in Raja Ampat and belonging to sea cucumber. Sea cucumber is an organism that difficult to distinguish through morphological characteristic.. This study aimed to identify Synaptula from Raja Ampat based on mitochondrial gene, cytochrome c oxidase subunit 1 (COI). The results of the study that Synaptula of Raja Ampat (UNP 101) is Plakobranchus ocellatus, the result is different with identification through morphological. Synaptula of Raja Ampat (UNP 101) has a high similarity with Plakobranchus ocellatus species from Sulawesi and the Philippines. Genetic distance Synaptula of Raja Ampat (UNP 101) with Synaptula reciprocans is 76.8%, while similarity values is 23.6%. The difference in the results of species  identification may be caused by contamination DNA Plakobranchus UNP 67A to DNA Synaptula UNP 101, contamination is likely to occur during the identification process which simultaneity. Abstract

Hipertiroidisme merupakan salah satu penyakit yang disebabkan oleh jumlah hormon tiroid yang disekresikan oleh kelenjar tiroid terlalu tinggi. Hipertiroidisme banyak terjadi pada wanita sebab selama proses kehamilan, janin yang sedang berkembang membutuhkan asupan yodium (iodin) yang lebih. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh induksi Natrium iodin sebagai penyebab Hipertiroidisme pada siklus estrus tikus betina (Rattus norvegicus) beserta gambaran histologi ovarium tikus betina (Rattus norvegicus). Metode yang dilakukan yaitu dengan melakukan induksi secara berlebih Natrium Iodin (NaI) yang menyebabkan Hipertirodisme kemudian dilakukan pengamatan siklus estrus dan pengamatan secara histologis melalui gambaran histologi ovarium. Hasil penelitian menunjukkan bahwa pengaruh Hipertiroidisme pada tikus betina menyebabkan siklus estrus tikus betina menjadi tidak normal dan berpengaruh terhadap fase estrus sedangkan gambaran histologis ovarium pewarnaan Hematoxylin-eosin menunjukkan tidak ada perbedaan antara tikus kontrol dengan tikus hipertiroid.   Kata kunci :  Hipertiroidisme, siklus estrus, ovarium.

Abstract

This research aims to analyze the effect of unripe sapodilla fruit extract on endogenous antioxidant expression in T1DM BALB/c mice and its free radical scavenging activity. Manilkara zapota extract (MzE) is an aqueous extract of unripe sapodilla fruit and was obtained by maceration and freeze-drying process. This study used 25 male BALB/c mice with 7-weeks-old of age. They were divided randomly into five groups (n=5) before treatment. A single high dose (145 mg/kg BW) of streptozotocin (STZ) was intraperitoneally injected to induce type 1 diabetes mellitus (T1DM). MzE was given orally once each day for 14 days. Liver cells were isolated and immunoassay with anti-superoxide dismutase (SOD) and anti-nuclear factor erythroid 2-related Factor 2 (Nrf2), and then the results were analyzed by flow cytometry. Diphenylpicrylhydrazyl (DPPH) assay was performed to analyze free radical scavenging. Data were analyzed statistically with one-way ANOVA (p<0.05). The result showed that the glucose levels in diabetic mice after MzE administration were significantly lower than in the DM group. MzE treatment increased the expression of Nrf2 and SOD in diabetic mice. MzE could scavenge DPPH with the IC50 value obtained at 48.35 μg/mL, while ascorbic acid as a control could scavenge DPPH with the IC50 value at 22.24 μg/mL. The increase in the scavenging activity is in line with the increase in extract concentration. In conclusion, this study revealed that MzE can be an endogenous antioxidant enhancer by improving the expression of Nrf-2, SOD and can inhibit free radicals as an exogenous antioxidant in T1DM.

Abstract

We aimed to analyze the effect of CSN1S2 protein in Etawah crossbred goat milk and yogurt on the histopathology of the jejunum and the amount of cell damage in Complete Freund's adjuvant (CFA)-induced rheumatoid arthritis (RA) rats. The rats were divided into six groups: the untreated control rats (C), control rats were given CSN1S2 protein from Etawah crossbred goat milk (CM) or yogurt (CY), RA rats, RA rats given CSN1S2 protein from Etawah crossbred goat milk (RAM) or yogurt (RAY). Hematoxylin-eosin staining was conducted for the histopathological analysis of jejunum. Statistical analysis was done using one-way ANOVA (a significance value of p ≤ 0.05) followed by the Tukey test. Our study observed that the control, CM, and CY group have a normal histological structure of jejunum. The damage to the jejunum structure was reported in the RA group. The milk CSN1S2 protein was able to improve the structure of jejunum villi and increase the normal cell number in the jejunum of the RA group, similar to control. The RAY group showed an impaired jejunum structure and a high number of necrotic cells as in the RA group.

Abstract

The current study aimed to analyze the protective effect of the aqueous extract of temulawak (Curcuma xanthorriza) rhizome on the sperm quality of monosodium glutamate (MSG)-induced mice. This study used 30 male mice (aged 2.5-3 months old and weighing 25-30 g), which were then randomly divided into five groups: K- (healthy male mice received only aquadest), K+ (male mice exposed to 4 mg/kg BW MSG), MT1, MT2 and MT3 (MSG-induced male mice orally treated with 0.2, 0.4 and 0.6 mg/g BW temulawak (C. xanthorriza) rhizome extract for 14 days, respectively). At the end of treatment, all mice were sacrificed, and cauda epididymis was isolated. The obtained semen was analyzed for its quality, including motility, viability, concentration and spermatozoa abnormalities. MT2 group exhibited the highest sperm motility of MSG-induced mice (79.16±4.45%). The highest sperm viability was also observed in the MT2 (77.83±2.75%) followed by MT1 and MT3 groups (70.19±5.93 and 72.41±5.53%, respectively). MT2 and MT3 groups could increase (p<0.05) sperm concentration in MSG-induced mice by 14.03x106 and 14.46x106 cells/ml, respectively. While, sperm abnormalities of MSG-induced mice tend to decrease in all groups treatment by 38.10% (MT1), 36.32% (MT2) and 36.04% (MT3). In conclusion, the administration of 0.4 mg/g BW aqueous extract of C. xanthorriza rhizome could improve sperm quality by increasing the motility, viability and concentration of sperm and also altered the sperm abnormality of MSG-induced mice.

Abstract

Cancer death cases have increased yearly, and there are estimated to be 21.6 million cancer cases in 2030. Studies of herbal compounds for cancer treatment alternatives are essential because cancer treatment is relatively expensive and has adverse effects. Phyllanthus niruri (Pn) and Catharanthus roseus (Cr) are plants that are known as herbal medicines. Combining the two plants is expected to prevent and enhance the immune system in breast cancer cases. This study aims to analyze the anti-cancer and immunomodulatory effects of P. niruri and C. roseus extract (PCE) in modulating macrophage polarization in breast cancer mice. Experimental animals are divided into six groups and there is healthy control (normal mice), cancer (DMBA-induced mice), cancer mice with cisplatin administration, cancer mice with PCE administration with three different doses, including dose 1 (500 mg/kg Pn + 15 mg/kg Cr), dose 2 (1000 mg/kg Pn + 75 mg/kg Cr), and dose 3 (2000 mg/kg Pn + 375 mg/kg Cr). The mice were injected with DMBA once a week for six weeks to induce cancer in mice. The breast cancer mice model was administered with PCE orally for 14 days. The expression of CD11b+IL-10+ and CD11b+IFN-γ+ demonstrated macrophage polarization. The results showed that breast cancer induction using DMBA increased the level of IL-10 and decreased the level of IFN-γ significantly compared to the normal group (p < 0.05). In specific doses, administration of PCE could reduce IL-10 levels and increase the level of IFN-γ significantly (p < 0.05). PCE can modulate the polarization of macrophages by suppressing the M2-like macrophage and increasing the M1-like macrophage. The ability of PCE to modulate macrophage polarization indicates that the combination of P. niruri and C. roseus has activity as an anti-cancer.

Abstract

Acute lung injury tends to be induced by infection or sepsis that disrupt alveolar and vascular permeability, neutrophil influx, and edema. Those impairments are worsened by the increase of oxidative stress along with hyperinflammation response. Oxidative stress in lung tissue could be indicated by malondialdehyde (MDA) and nuclear factor erythroid 2-related factor 2 (Nrf2) expression. This research aimed to evaluate the efficacy of Baby Java citrus peel extract (BJE) in suppressing oxidative stress and preventing lung injury in lipopolysaccharides (LPS)-induced mice. Twenty-five male BALB/c mice were divided into five groups consisting of untreated (N), LPS (A), and LPS-induced followed by treatment using BJE at various doses: 75 mg/kg BW (BJE-1), 105 mg/kg BW (BJE-2), and 150 mg/kg BW (BJE-3). Lungs were isolated for histopathological analysis also detection of MDA and Nrf2 using flow cytometry. BJE at the dose of 105 mg/kg BW could inhibit the alteration of lung histology following LPS challenge including alveolar and interstitial neutrophil infiltration, proteinaceous debris, and septal thickening. The same dose also showed good potency in suppressing MDA and Nrf2 levels as oxidative stress indicators. Our findings demonstrated protective effects of Baby Java citrus peel in acute lung injury and oxidative stress prevention after LPS exposure.

Abstract

Maintaining immunoglobulin levels in the body is important to protect the body from exposure to pathogens. One effort can be made by consuming herbs containing immunomodulatory compounds, such as Cheral®, which includes a combination of herbs Phyllanthus niruri and Curcuma longa. This research aims to determine the expression of immunoglobulin M (IgM) and immunoglobulin G (IgG) following the administration of Cheral® to Wistar rats. The study was conducted in vivo, utilizing 24 healthy male Wistar rats for a 90-day treatment period. The research was divided into four treatment groups, including a control group and three dosage groups: Dose 1 (156.25 mg/kg BW), Dose 2 (312.5 mg/kg BW), and Dose 3 (468.75 mg/kg BW). IgM and IgG were isolated from the spleen and analyzed using flow cytometry. Flow cytometry data were analyzed using SPSS with a one-way ANOVA and post hoc test (p-value <0.05). The analysis showed that the relative number of IgM-producing cells in the control group was significantly higher than in the treatment groups, with a difference of 44.40%. In contrast, the relative number of IgG-producing cells in Dose 3 was significantly lower than all other treatment groups, showing a decrease of 29.21%. Overall, the expression of IgG and IgM did not differ substantially across all treatments. The lower IgG and IgM profiles compared to the control group indicate Cheral®'s ability to prevent infections and maintain the immune system of the rats throughout the treatment period.

Abstract

Background: Obesity is a global epidemic caused by excessive body fat, which is increasing free fatty acids in the liver, causing oxidative stress and liver cell damage. Green tea yogurt (GTY) with encapsulated Lacticaseibacillus paracasei E1 (GTY-LpE1) might have a beneficial effect in reducing liver cell damage. This study was conducted determine GTY-LpE1 effect on superoxide dismutase (SOD) expression, malondialdehyde (MDA) expression and liver histopathology in high-fat high-fructose diet (HFFD) mice. Material and Methods: A completely randomized design (CRD) with 7 groups, including normal diet (ND) group, HFFD group, 1.3 mg/kg BW simvastatin (SIM)-administered HFFD group, 5 g/kg BW probiotic yoghurt (PY)-administered HFFD (PY), 2.5 g/kg BW GTY-administered HFFD (2.5 GTY), 5 g/kg BW GTY-administered HFFD (5 GTY), and 10 g/kg BW GTY-administered HFFD (10 GTY). The diet was given for 16 weeks, followed by oral administration of sim/yoghurt during the last 4 weeks. Mice were sacrificed and the liver was collected. SOD and MDA expression were analyzed by flow cytometry. Histopathology analysis was done by evaluating hematoxylin-eosin (HE) staining of the liver.Result: The percentage of necrotic cells were 34.55, 34.31, and 21.95%, when treated with 2.5, 5, and 10 g/kg BW with GTY-administered HFFD, respectively, these were lower than the ones in the HFFD group (69.49%). The percentage of MDA expression were 15.55, 18.69, and 22.42%, respectively, these were lower than the ones in the HFFD group as well. The percentage of SOD expression were 9.49, 7.85, and 11.11%, respectively, these were higher than the ones in the HFFD group (3.44%). Conclusion: GTY-LpE1 could decrease the number of necrotic cells in the HFFD mice livers and improve the hepatocyte damage by reducing MDA expression and enhancing SOD expression. GTY-LpE1 can be used as an alternative food to control obesity.Keywords: alginate, chitosan, encapsulation, green tea, probiotic
