Abstract

Vaccine research entered a new era when several useful molecular research tools were established. Instead of attenuated virulent microorganisms or killed virulent microorganisms, effective subunit vaccines were developed using recombinant DNA technology. By using the technology, selected genes of the virulent microorganisms can be amplified, cloned, expressed, and evaluated as vaccine components in challenge studies. However, a major bottleneck with the amplification of functional genes from Mycobacterium tubeculosis containing guanine plus cytosin-rich templates is often hampered by the formation of secondary structures like hairpins and higher melting temperatures. To solve this problem in this research, the amplification reaction was modified by addition of dimethyl sulfoxide (DMSO) into amplification reaction mixtures. It was found that 10% (v/v) of DMSO in the reaction mixture improved the amplification of GC-rich template of M. tuberculosis gene. This result indicating that amplification of unbalanced content of G and C deoxyribonucleotides genome could be improved using low-cost organic molecule, DMSO. Therefore, the DMSO should be widely useful as an enhancer to improve the amplification of GC rich construct from other genome. Abstract

ABSTRAKTujuan penelitian ini adalah untuk mengetahui cemaran baku mutu air limbah Rumah Potong Hewan (RPH) Sapi dan Ayam di Kota Mataram. Semua hasil melebihi baku mutu kecuali nilai pH. Kandungan ALT rerata air limbah RPH sapi 284004 x 104 koloni/ml dan mengandung mikroba jenis E.coli dan Salmonella. Sedangkan air limbah RPH ayam ALT rerata 101596 x 105 koloni/ml dan mengandung kuman jenis E. coli, Pseudomonas aeruginosa dan Klebsiella pneumoniae. Tingginya kandungan BOD, COD, TSS, ammonia, pH, minyak dan lemak maupun cemaran mikroba diduga disebabkan tidak difungsikannya Instalasi Pengolahan Limbah (IPAL) pada RPH sapi dan tidak optimalnya fungsi IPAL di RPH ayam.Kata kunci: RPH sapi, RPH ayam, Baku mutu limbah RPH, Cemaran mikroba, kota Mataram yABSTRACTThe purpose of this study was to determine the contamination of waste water quality standard and microbiologic contamination ofcattle and chicken slaughterhousein Mataram city. TPC average content of waste water of cattle slaughterhouse was 284004 x 104 colonies/ ml, and contained microbial species of E. coli and Salmonella. However, the TPC avarange of wastewater of chicken slaughterhouse accounted for 101596 x 105 colonies/ml and contained microbial species of E. coli, Pseudomonas aeruginosa and Klebsiella pneumoniae. Abstract

Latar Belakang: Biji pepaya telah digunakan sebagai antelmintik pada ternak. Teh biji pepaya memiliki aktivitas antiparamphistomum secara in vitro melalui metode adult mortality assay. Karakterisasi teh biji pepaya perlu dilakukan untuk menjamin mutu produk teh yang dihasilkan. Penelitian ini bertujuan untuk menentukan karakteristik fisik dan metabolit sekunder teh biji pepaya (Carica papaya L.). Metode: Kandungan senyawa teh biji pepaya dikarakteristisasi melalui skrining fitokimia. Evaluasi fisik teh biji pepaya meliputi organoleptik, pengukuran pH, penentuan bobot jenis, variasi bobot, dan kadar air. Hasil Penelitian: Teh biji pepaya memiliki rasa pahit, aroma khas, dan berwarna kuning bening, dengan pH 5.61, kadar air 6,876%, bobot jenis 1,000431887 g/ml, dan pada variasi bobo tidak ada bobot yang melebihi 12% limit variasi. Teh biji pepaya mengandung flavonoid dan steroid/triterpenoid. Kesimpulan: Teh biji pepaya memenuhi standar mutu teh. Teh biji pepaya mengandung flavonoid dan steroid/triterpenoid.

Abstract

This introduction to aquaponic planting techniques aims to provide examples of the application of advanced and sustainable agricultural technology to the people in Kayangan village, Kayangan sub-district, North Lombok district (KLU). This activity was carried out from December 2022 to February 2023 at the Bagek Kembar hamlet, Kayangan. The fish used were 60 days old fish seeds with a size of 10 cm x 5 cm which were reared in a 4x3 m tarpaulin container. The plant growth media used is in the form of used foam taken from one of the residents' houses with a size of 5 cm. Placement of the planting media uses used plastic cups and used bottles as containers for placing plastic cups. The vegetables used in making this aquaponics are mustard greens, lettuce and pakcoy. The nutrients used to support plant growth are in the form of fish manure and some additional organic nutrients which are made independently using organic waste from the kitchen. The results of using aquaponic technology as a cultivation technology show that the plants grow but the leaves of the plants turn yellow and the fish in the pond can grow large.

Abstract

Herbal drinks have become a popular alternative in maintaining people's health and well-being, especially in the modern era filled with the need for a healthy lifestyle. In Gondang village, Gangga sub-district, North Lombok district, the abundant potential of family medicinal plants (TOGA) has not been fully utilized. To address this challenge, an innovative herbal drink for Gondang village (MIDANG) aimed at increasing the added value of TOGA and improving public health was introduced to the Gondang village community. With a holistic approach, this activity covers the technical aspects of MIDANG production and the socio-economic implications of this product development. It is hoped that the MIDANG Herbal Drink will be a positive step towards sustainable prosperity for the people of Gondang Village and its surroundings.

Abstract

Pengembangan ternak kambing lokal menghadapi tantangan dalam penyediaan pakan hijauan berkualitas secara berkelanjutan, terutama pada musim kemarau. Salah satu potensi lokal yang belum dimanfaatkan secara optimal adalah daun lamtoro (Leucaena leucocephala), yang diketahui memiliki kandungan protein dan TDN (Total Digestible Nutrients) yang tinggi. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memberikan pelatihan dan pendampingan kepada peternak kambing lokal di Desa Sambik Elen, Kecamatan Bayan, Kabupaten Lombok Utara, Provinsi Nusa Tenggara Barat mengenai teknologi fermentasi hijauan pakan ternak berbasis lamtoro sebagai alternatif pakan ternak yang bernutrisi dan ekonomis. Hasil pengamatan menunjukkan bahwa peternak dapat memahami dan mempraktikkan proses fermentasi hijauan pakan ternak, yang mencakup persiapan bahan, penambahan dedak dan starter, pencampuran, dan penyimpanan secara anaerob. Kesimpulannya, kegiatan pengabdian ini telah berhasil meningkatkan pengetahuan dan keterampilan peternak dalam pengolahan hijauan pakan ternak dengan teknologi fermentasi. Diharapkan kegiatan ini dapat berkontribusi pada keberlanjutan usaha peternakan kambing dan peningkatan perekonomian masyarakat peternak di daerah pedesaan.

Abstract

Jerowaru sub-district is one of 21 sub-districts in East Lombok district. Many people raise buffaloes to fulfill their need for food of animal origin. However, the business is still hampered by diseases caused by worm infestation, which is economically detrimental. This study aimed to obtain data on the prevalence of helminthiasis in buffaloes and identify the types of worms that infest the animals. Fecal samples were collected from buffaloes kept in smallholder farms in several villages in the Jerowaru sub-district. Examination and identification of samples were conducted at the Microbiology and Biotechnology laboratory of the Faculty of Animal Science, University of Mataram, and the Banyumulek Animal Health laboratory. The results show that the prevalence of helminthiasis in buffaloes kept in smallholder farms in the Jerowaru subdistrict was 14.60%. The types of parasites infecting buffaloes consisted of Eimeria (13.48%) and Toxocara (1.12%). Although parasite infection intensity is categorized as mild, it is necessary to pay attention to the cleanliness of cattle pens, especially during the rainy season, so they are not wet and humid.

Abstract

Foot and Mouth Disease (FMD) is a viral disease that affects livestock and can cause significant economic losses.  Vaccination has been recognized as the primary strategy for FMD prevention, but vaccine development, especially conventional vaccine production, has time, cost, and effectiveness limitations. Using software-based immunoinformatics methods has cost and time efficiency for simulation development and calculations in vaccine development research.  This study aimed to design FMD vaccine candidates using an epitope-based in-silico approach, focusing on identifying potential epitopes of the pathogen that causes FMD. The in silico approach was used to analyze FMD virus genome sequences from UniProtKB (https://www.ebi.ac.uk/Tools/msa/clustalo) and the National Library of Medicine application (https://www.ncbi.nlm.nih.gov).  Furthermore, the prediction of FMD virus epitopes was analyzed using the Immuno-Epitope database, and the prediction of peptide antigenicity levels using VaxiJen 2.0 software. The analysis identified eight potential epitope candidates, such as CSIQKR, TEFGFHPNA, EIRPMEKVRA, YEGVEL, SFARRGT, APGLPWALQGKRRGALIDFESGTV, MASLEDKGKPF, and TLPTSFNYGAI with antigen binding affinities of 2. 9424nM, 1.65nM, 1.4702nM, 1.2966nM, 1.2643nM, 1.1967nM, 1.1921nM and 1.0143nm respectively. These epitopes are expected to form the basis for developing more selective and safe peptide vaccines. By focusing on epitopes, the resulting vaccine can improve the effectiveness and safety of vaccination and accelerate the development of new vaccines for FMD prevention.

Abstract

The role of farmers is a determining factor in the success of efforts to control Foot and Mouth Disease (FMD), especially the level of knowledge, attitude, and skills of farmers. This study aims to determine the correlation and influence between knowledge, attitude, and skills of farmers with the location and characteristics of farmers. The study was conducted using a survey method on samples selected purposively in three sub-districts, namely Praya Tengah, Pujut and Praya Barat Daya with a total of 100 respondent farmers. This study uses correlation analysis and linear regression for data analysis. The results of the study indicate that there is a correlation value between the level of knowledge, attitude and skills of farmers towards controlling FMD with the location and characteristics of farmers, including: gender, age, education level, occupation, ownership status, experience/length of farming and the number of cattle owned by the farmer. As well as the influence of the location of the farmers residence and the characteristics of the farmers on the level of knowledge, attitude and skills of the farmers.
