Abstract

PT. Kereta Api Indonesia is the largest transportation company in Indonesia. To maintain company productivity, employees need good physical fitness as the components of physical fitness are cardiopulmonary endurance, good muscle strength, and flexibility. Technicians, conductors, and machinists very much need this component of good physical fitness due to the high workload. A descriptive study with a cross-sectional design was conducted on secondary data, with 163 employees from PT. KAI DAOP 1 Jakarta in May-August 2017 using the Total Sampling method. The variables measured were cardiopulmonary endurance with the Young Men's Christian Association (YMCA) bench test, muscle strength with a dynamometer, and flexibility with a pleximeter. Prevalence of the very poor category was found in cardiopulmonary endurance (35.58%), right-hand grip strength (69.33%), left-hand muscle strength (71.78%), back muscle strength (53.37%), and leg muscle strength (55.21%). However, there are also some variables dominated by the moderate category, namely push strength (39.88%), pull strength (36.2%), and interestingly flexibility showed a very good category (76.69%). It is necessary to carry out a physical fitness program with principles that are following the physical activity itself, namely the FITT principle. This is because there are components of fitness that are dominated by the very poor category, namely cardiopulmonary endurance, right-hand and left-hand muscle strength, and back and leg muscle strength. The types of exercise that can be done in the physical fitness program are moderate-intensity aerobics, walking, cycling, lifting weights, and stretching.

Abstract

Personality is most commonly divided into five big traits1,2,3,4 namely Agreeableness, Conscientiousness, Extraversion, Neuroticism, and Openness to Experience. Agreeableness refers to the tendency to get along well with others and is associated with altruism or modesty. Conscientiousness refers to the extent to which an individual is careful, reliable, and persevering. Extraversion refers to the tendency to experience positive emotions and to have a positive outlook on life. In general, extroverts are talkative, sociable, outgoing, enthusiastic, and energetic. Neuroticism refers to the tendency to experience negative emotional states including anxiety, depression, or anger. Openness to experience refers to the tendency to be open-minded, imaginative, and curious. This study was cross-sectionall research with big five inventory (BFI) questionnaire. We used bahasa to validate translation of the BFI. The Participant N= 50 persons from health sciences and N= 116 persons from social science. All participant were lecturers and professors with aged interval 41-90 years. This questionnaire has 5 scale with 5 Likert scale mark with disagree strongly and agree strongly answer. The result showed all participant were included for three domain personality traits dimensions, which are agreeableness with 35, openness with 52, and conscientiousness with 29. Taken all together we concluded all lecturer and professors had tendency of agreeableness, openness, and conscientiousness for their personality traits dimensions.

Abstract

Background: One of the main problems of the Citarum River is the contamination of E. coli due to livestock activities, washing toilets, and industry. In addition, irrational use of antibiotics in the community and livestock can increase E. coli resistant strains to antibiotics. This study aimed to identify the presence of multidrug resistance (MDR) and extended-spectrum β-lactamase (ESBL) E. coli strains in Citarum river clusters, namely industrial, livestock, and residential clusters. Methods: This was a descriptive study. A sample of 100 mL surface water from each Citarum cluster. Culture, antibiotic sensitivity test, and PCR to identify blaCTX-M-15 gene carriers of ESBL E. coli were carried out in the sample. Results: There were 37 isolates of E. coli, with 24% of these isolates showing MDR properties, which can be found in industrial, livestock, and residential clusters at 13%, 8%, and 3% respectively. The most E. coli-resistant antibiotics found in these samples were ampicillin (45%), followed by tetracycline (37%), and azithromycin (29%). The PCR examination did not find the blaCTX-M-15 gene carrying ESBL properties in all three Citarum river clusters. Conclusion: The presence of E. coli isolates in each Citarum river cluster suggests the occurrence of river pollution due to animal, human or industrial waste.  Therefore, it is necessary to make better government regulations regarding sanitation and education for the surrounding community regarding the importance of keeping the river clean.

Abstract

Cardiac fibrosis is a pathological feature as a key factor in cardiac remodelling, thus progressing to heart failure. MicroRNAs (miRNAs) are crucial intracellular mediators of various biological processes, involved in the regulation of physiological and pathological processes. Recently, it has been shown that miRNAs were found to play a role in exercise-induced cardioprotection. Studies in human and animal subject with variety of acute and chronic exercise were observed. This review highlights impacts of exercise on the miRNA levels associated with cardiac fibrosis.

Abstract

BackgroundPhysiological and immunological changes in the elderly may contribute to the composition and patterns of the gut microbiota, including Escherichia coli, as well as antimicrobial susceptibility patterns. The research was conducted to assess the effect of aging on Escherichia coli growth and susceptibility to several antibiotics. MethodsAn experimental laboratory study using 7 young (6-month-old) (YWR) and 7 old (24-month-old) Wistar rats (OWR) as subjects. Rat stool specimens were used as representing the subjects. Antibiotic susceptibility was determined using the Kirby-Bauer method. The independent t-test and Mann-Whitney test were used to analyse the data. ResultsAll fecal specimens collected from both groups were positive for E. coli growth. The YWR showed a low density of E. coli, while the OWR showed a statistically significant increase in the population of E. coli (p=0.001). Overall, the E. coli isolates showed a high proportion of resistance to erythromycin (100%), ampicillin (86%), and oxytetracycline (58%). The isolates collected from YWR were significantly more resistant to streptomycin (83% vs. 19%, p<0.001) and tetracycline (47% vs. 3%, p<0.001) than the isolates from OWR. Young Wistar rats also had a higher resistance rate than OWR to the most frequently used antibiotics, such as ampicillin (97% vs. 76%, p<0.001). ConclusionOur study demonstrated an increase in the fecal E. coli population with increasing age of the Wistar rats. In addition, the results of susceptibility tests for several antibiotics showed that age may not be associated with an increase in the population of a resistant strain of E. coli.

Abstract

Citarum is the longest river in West Java and one of the most polluted rivers in Indonesia and the world. Heavy metals from agriculture, industry, and household waste pollute the Citarum river. Heavy metals enter humans and animals by consuming contaminated food and beverages. Several studies have identified mitochondria as a primary target for heavy metal poisoning, resulting in impaired mitochondrial energy production, induction of oxidative stress, apoptosis, and mitophagy. This study aims to investigate the impact of heavy metals from the upstream and downstream areas of the Citarum river on mitochondrial gene expression of cox4i1, sod2, baxa, mfn1b, and ppargc1a in the skeletal muscle of zebrafish (Danio rerio) using in vivo models for biological monitoring of early detection of environmental heavy metal pollution. This experimental study was conducted from December 2019 to March 2020. The study involved four treatment groups and one control group of zebrafish, which were exposed to river water and sediments collected from two sites in the Citarum river area in December 2019. The biomarker levels were analyzed using multivariate analysis. Although all heavy metal levels except mercury were below the WHO threshold in all samples, this study's results showed that the RT-PCR results indicated that the levels of cox4i1, baxa, and ppargc1a in all samples were generally higher than those in the control. There were significant differences (p<0.05) using multivariate analysis in sod2 in Cibeureum water compared to Cibeureum sediment and ppgarc1a compared to control and Balekambang sediment. In conclusion, heavy metals from the upstream and downstream areas of the Citarum river had an impact on mitochondrial gene expression of cox4i1, sod2, baxa, mfn1b, and ppargc1a in the skeletal muscle of zebrafish. Biomarkers pparc1gc and sod2 could be further studied to identify the most valid and reliable parameters for biological monitoring, which may aid in the early detection of environmental damage to humans and animals.

Abstract

Endurance training, a form of physical activity that relies on continuous aerobic exercise and repetitive muscle contractions, is widely acknowledged for its positive effects on overall physical fitness. Aerobic exercise, an essential component of endurance training, has numerous benefits including improved cardiovascular and respiratory health, increased muscle endurance, and enhanced resistance against fatigue. It has also been found to contribute to skeletal muscles, potentially by stimulating the synthesis of proteins involved in muscle fiber formation. Although resistance exercise has been favored for promoting muscle growth, some suggests that aerobic exercise can also produce skeletal muscle hypertrophy comparable to that of resistance exercise if performed correctly. The duration, intensity, and specific type of aerobic exercise play important roles in determining skeletal muscle mass. The mammalian target of rapamycin (mTOR) known as a key regulator of muscle protein synthesis that associated with exercise activity. Several signaling pathways, such as Akt/mTOR and MAPK, are involved in controlling muscle protein synthesis during exercise. This review aimed to understand the impact of aerobic exercise intensity and other training parameters on skeletal muscle, to provide valuable insights for optimizing exercise programs and fostering muscle hypertrophy. In this review, we had systematically searched PubMed and Google Scholar from January 2013 to May 2023. Our result indicated that aerobic exercise can be expected to promote skeletal muscle hypertrophy and improve muscle mass and function. The regulation of skeletal muscle mass is complex, involving various signaling pathways such as mTOR, as well as the influence of hormones and growth factors.
