Abstract

Cayenne pepper has a high production demand. The high demand does not match the amount of production produced. One of the factors for decreased production is the presence of bacterial wilt disease caused by Ralstonia solanacearum. Bacterial wilt control can be done with the application of antagonistic bacteria, namely Bacillus sp. Bacillus sp. application requires an optimal formulation to maintain bacterial activity in the soil, one of which is Bacillus sp. encapsulation. This study aims to determine the effectiveness of Bacillus sp. encapsulation as a formulation of biological agent bacteria for the control of bacterial wilt disease and cayenne pepper plant growth. This study used the encapsulating material concentration and dosage factors for application to plants. Encapsulation was made using the extrusion method. The results showed that the concentration of 2% sodium alginate and 1.5% gelatin gave a significant difference in disease intensity of 18.89% and in the growth parameters of cayenne pepper plants were able to reach a height of 30.58 cm and a root length of 17.18 cm. The right concentration and high dose provide the ability to suppress intensity and encourage maximum growth

Abstract

Meloidogyne spp. is a root-knot nematode that can cause damage to plants up to 68%. The common control is the use of pesticides, but in the long term, it can damage the soil, so it is necessary to use biological agents that can control Meloidogyne spp. without causing environmental damage. The purpose of this study was to determine the ability of endophytic bacteria to control root-knot nematodes Meloidogyne spp.. The method used in this study was the exploration of endophytic bacteria from the Wonorejo mangrove ecotourism area, Surabaya using purposive sampling method. The five best endophytic bacteria were tested further with antagonistic tests. This test was carried out in vitro using 3 concentrations, namely 106, 108, and 1010 CFU/ml with 3 repetitions. The obtained results were tested using probit analysis to obtain the LC50 values. It was found that LC50 values for the most effective concentration to kill 50% of the Meloidogyne sp population is 109 and 1010 CFU/ml. It was also revealed that the isolate that is most capable of controlling Meloidogyne spp. nematodes is isolate A43. Keywords: Antagonist test, Endhopic bacteria, LC50, Mangrove, Meloidogyne spp.

Abstract

Padi (Oryza sativa L.) merupakan komoditas pangan utama di Indonesia, namun produktivitasnya sering terganggu oleh penyakit blas (Pyricularia oryzae) dan bercak cokelat (Helminthosporium oryzae). Kedua patogen ini dapat menurunkan hasil panen secara signifikan, terutama pada kondisi lingkungan yang mendukung. Penelitian ini bertujuan untuk menganalisis insidensi dan intensitas penyakit pada sistem pertanian organik dan pengendalian hama terpadu (PHT) di Mojokerto dan Jombang. Pengambilan sampel dilakukan di Desa Claket, Kecamatan Pacet, Kabupaten Mojokerto (950 m dpl) dan Desa Puton, Kecamatan Diwek, Kabupaten Jombang (50 m dpl) pada Juli-September 2024. Identifikasi jamur dilakukan melalui isolasi, uji postulat Koch, serta pengamatan makroskopis dan mikroskopis. Hasil penelitian menunjukkan bahwa insidensi H. oryzae mencapai 88% di kedua lokasi dengan intensitas 65% (Jombang) dan 63% (Mojokerto). Sementara itu, insidensi P. oryzae sebesar 86% di Jombang dan 82% di Mojokerto dengan intensitas masing-masing 66% dan 67%. Nilai insidensi yang tinggi menunjukkan kategori serangan sangat berat, meskipun intensitas masih tergolong sedang. Analisis uji T menunjukkan adanya hubungan signifikan antara faktor lingkungan (suhu dan kelembapan) dengan perkembangan penyakit (P<0.05) dan korelasi dengan r = 0.6-0.8. Hasil ini menegaskan bahwa kondisi agroekosistem dan varietas padi berperan penting terhadap tingkat serangan patogen, serta sistem budidaya organik maupun PHT mempengaruhi dinamika penyakit.

Abstract

Mangroves are unique ecosystems that support biodiversity, including endophytic microorganisms. Endophytic bacteria group is a group of microbes that have a mutualistic symbiosis to the host plant without harming the host, and can act as biocontrol agents and plant stimulants. Endophytic bacteria have a very wide host range. Each plant has an abundance and diversity of endophytic bacteria, depending on the internal and external factors that affect it. This study aims to determine the characteristics of endophytic bacteria and identify the diversity of endophytic bacteria found in Avicennia sp. Method Sampling from mangrove area of Gunung Anyar, Surabaya. Characterization of endophytic bacteria was done by Gram test, spore staining test, catalase test, oxidative-fermentative test, and selective media test (King's B and Yeast Dextrose Carbonate). The results showed the existence of several isolates of endophytic bacteria that have morphological diversity and different characteristics, such as shape, color, colony edge, and colony surface. Conclusions In this study has been successfully isolated some endophytic bacteria such as Lactobacillus sp., Corynebacterium sp., Bacillus sp., Xanthomonas sp., and there are also endophytic bacterial isolates that have not been identified. This study contributes to an early understanding of the diversity of microorganisms in mangrove ecosystems and opens up opportunities for further exploration of their potential applications, one of which is in agriculture

Screening of quorum quenching activity of rhizobacteria against Pectobacterium carotovorum subsp. carotovorum. Biodiversitas 23: 4336-4342. Quorum sensing (QS) is a mechanism for determining the virulence of the phytopathogenic bacterium Pectobacterium carotovorum. P. carotovorum carries out the QS mechanism with the chemical compound acyl-homoserine lactone (AHL) to signal and carry out its pathogenicity and virulence. The pathogenicity of P. carotovorum can be prevented through quorum quencher (QQ) activity derived from bacteria that degrade AHL compounds by producing AHL-lactonase. In the present investigation, AHL-lactonase-producing bacteria were isolated from the rhizosphere of carrot cultivation in Bumiaji Sub-District, City of Batu, East Java, Indonesia. The isolated bacteria were screened for quorum quenching activity. Out of thirty-seven isolates, 7 rhizospheric bacteria showed QQ activity against the Chromobacterium violaceum bioindicator. The hypersensitivity test on tobacco plants showed that, five strains SRL07, SRL09, SRL15, SRL21, and SRL35 did not cause a hypersensitivity response. The five isolates were able to suppress the virulence of P. carotovorum in carrot tubers. The 16S rRNA gene sequences of the five isolates had the highest similarity to Bacillus thuringiensis, B. subtilis, and B. cereus. The five isolates were detected to have the gene encoding AHL-lactonase (aiiA). The present study provides new information about the QQ activity of rhizosphere and its potential as a quorum quencher against P. carotovorum.
