Abstract

Acne is a chronic skin disease involving inflammation of the pilosebaceous units with the main bacteria Staphylococcus epidermidis and Propionibacterium acnes has prompted the creation of cosmetic masks containing antibacterial agents. This study aims to evaluate the antibacterial activity of Lemongrass and Clove essential oils and to determine the synergistic effects of combining these essential oils in a peel-off gel mask formulation. The study was conducted using the liquid dilution method to determine the minimum inhibitory concentration and the disk diffusion method to test the inhibition zone diameter. Additionally, tests are conducted to determine the antibacterial activity against Propionibacterium acnes and Staphylococcus epidermidis, as well as to analyse the stability of the product both chemically and physically during storage. The results of the study showed that (1) the formulation containing a combination of 25% lemongrass and 18% cloves showed high inhibitory power and good stability, but caused moderate irritation; (2) lemongrass and clove essential oils were effective against Propionibacterium acnes and Staphylococcus epidermidis, but it was recommended to reduce the active ingredients to prevent potential irritation.

Abstract

Jerawat merupakan peradangan kulit umum yang sering kambuh dan dapat menurunkan mutu hidup. Infeksi bakteri seperti Cutibacterium acnes dan Staphylococcus aureus merupakan faktor penting dalam patogenesisnya. Tumbuhan berpotensi sebagai sumber antibakteri alamiah karena mengandung metabolit sekunder flavonoid, tanin, saponin, dan alkaloid yang mampu menghambat pertumbuhan bakteri, menurunkan risiko resistensi, dan menjadikannya kandidat terapi antimikroba prospektif. Penelitian ini bertujuan mengevaluasi aktivitas antibakteri kombinasi ekstrak daun jambu biji varietas kristal dan daun sambang darah terhadap bakteri penyebab jerawat (Cutibacterium acnes, Staphylococcus aureus, dan Staphylococcus epidermidis) menggunakan metode sumuran. Proses penelitian meliputi determinasi dan standarisasi simplisia, pembuatan ekstrak, serta pengujian aktivitas antibakteri. Hasil uji mutu menunjukkan simplisia dan ekstrak memenuhi semua standar parameter kimia (senyawa larut, kadar air, kadar abu) dan cemaran mikroba. Uji antibakteri menunjukkan bahwa kombinasi ekstrak pada rasio 1:2 (daun jambu biji : daun sambang darah) menghasilkan zona hambat yang lebih besar dibandingkan ekstrak tunggal pada bakteri Cutibacterium acnes (14,16 mm), Staphylococcus aureus (14,02 mm), dan Staphylococcus epidermidis (12,27 mm). Analisis one-way ANOVA dan post hoc menunjukkan hasil signifikan (p< 0,05), mengindikasikan adanya efek sinergis antar senyawa bioaktif kedua tanaman.

Abstract

Excessive free radicals accelerate skin degradation by causing oxidative damage to keratinocytes and fibroblasts, leading to structural protein breakdown (collagen, elastin, keratin), lipid peroxidation, and DNA damage. This process compromises skin barrier integrity, reduces elasticity, and promotes premature aging. Antioxidants play a crucial role in protecting the skin by neutralizing reactive oxygen species (ROS) and preventing lipid peroxidation. This study aimed to develop a topical cream containing jujube (Ziziphus mauritiana L.) and perilla (Perilla frutescens L. Britton) leaf extracts and evaluate its antioxidant activity and dermal safety. Extracts were obtained through kinetic maceration using 70% ethanol and formulated into oil-in-water creams at two concentration levels. The creams were assessed for physical characteristics (organoleptic properties, cream type, pH, homogeneity, spreadability, rheology, viscosity), stability under room and accelerated conditions (40°C for 4 weeks), antioxidant activity via DPPH assay, and irritation potential using albino rabbits. All formulations exhibited good physical properties, stability, and homogeneity. Irritation testing showed no erythema or edema, confirming the cream’s safety for topical application. Antioxidant testing revealed strong free radical scavenging activity, with IC50 values of 119.705 ppm (F1) and 66.938 ppm (F2). Accelerated storage slightly reduced antioxidant activity, while higher extract concentrations enhanced antioxidant effects compared to the base formulation. These findings indicate that jujube (Ziziphus mauritiana L.) and perilla (Perilla frutescens L. Britton) leaf extracts synergistically enhance antioxidant activity in a safe topical cream formulation, demonstrating potential as a natural preparation for skin protection against oxidative damage.

Abstract

Penggunaan bahan-bahan alami sebagai agen antibakteri dan antioksidan topikal semakin mendapat perhatian, terutama dalam pengembangan formulasi untuk kulit berjerawat. Penelitian ini bertujuan untuk memformulasi dan mengevaluasi gel yang mengandung kombinasi ekstrak buah asam gelugur dan ekstrak daun perilla, serta untuk menilai aktivitas antibakteri, antioksidan, dan keamanan topikalnya. Tiga formulasi gel dibuat, gel dasar tanpa ekstrak (F0), gel dengan konsentrasi ekstrak optimal (F1), dan gel dengan konsentrasi ekstrak dua kali lipat (F2). Formulasi gel dievaluasi untuk sifat organoleptik, homogenitas, pH, daya sebar, viskositas, dan stabilitas fisik menggunakan cycling test. Aktivitas antibakteri diuji terhadap Cutibacterium acnes dan Staphylococcus epidermidis menggunakan metode difusi cakram, aktivitas antioksidan dinilai melalui uji penangkal radikal bebas DPPH. Uji iritasi kulit primer dilakukan pada kelinci albino untuk menentukan keamanan topikal gel. Hasil menunjukkan bahwa semua formulasi memenuhi karakteristik fisik dan stabilitas gel topikal yang dipersyaratkan. Sebelum dan sesudah cycling test stabil pada bakteri C.acnes F0 (0,00 mm), F1 (14,35 ± 0,32 mm), dan F2 (26,42 ± 0,08 mm). Sedangkan pada bakteri S.epidermidis F0 (0,00 mm), F1 (11,20 ± 0,24 mm), dan F2 (20,15 ± 0,05 mm). Nilai IC50 sebelum dan sesudah cycling test F0 (664,857 ppm), F1 (106,518 ppm menjadi 113,25 ppm), dan F2 (63,01 ppm menjadi 71,50 ppm). Uji iritasi menunjukkan skor 0,0 ± 0,0, yang menunjukkan bahwa gel aman untuk aplikasi topikal.

Abstract

Radiasi ultraviolet (UV) berpotensi menyebabkan kerusakan kulit mulai dari eritema hingga kanker kulit. Penggunaan tabir surya sintetis seperti oksibenzon berisiko menimbulkan iritasi, sehingga pengembangan tabir surya alami menjadi alternatif yang diperlukan. Mengembangkan formulasi tabir surya alami dari kombinasi minyak biji bunga matahari (Helianthus annuus L.) dan ekstrak daun kecombrang (Etlingera elatior (Jack.) R.M. Sm.) serta mengevaluasi stabilitas dan aktivitas fotoprotektifnya. Minyak diekstraksi dengan cold pressing, sedangkan ekstrak diperoleh melalui maserasi etanol 70%. Kedua bahan distandardisasi berdasarkan parameter mutu. Aktivitas SPF diukur secara in-vitro menggunakan spektrofotometri UV-Vis pada panjang gelombang 290-320 nm dengan persamaan Mansur. Stabilitas formulasi diuji melalui cycling test. Formulasi kombinasi (2:1) menunjukkan stabilitas fisik yang baik selama pengujian dan aktivitas SPF yang signifikan. Hasil pengukuran SPF in-vitro mengonfirmasi potensi fotoprotektif dari kombinasi kedua bahan alam tersebut. Kombinasi minyak biji bunga matahari dan ekstrak daun kecombrang berpotensi sebagai tabir surya alami yang efektif dengan stabilitas memadai, menawarkan alternatif fotoproteksi yang aman dari bahan alam.
