Abstract

Background. The main etiology of endodontic treatment failure  is caused by bacteries that stay in the root canal. E.faecalis is a bactery that is found as an etiology of endodontic treatment failure. Cell wall of this bacteria is containing Lipoteichoic acid (LTA). LTA can penetrate into the periradicular tissue, act as endotoxin in host and cause periradicular inflammation and destruction. It occurs due to the capability of TGF-ß to enhance the proliferation collagen and MMP-1 to stop the collagen formation. The ability of enterococcus faecalis in enhancing inflamation process cause host can not reach the homeostasis phase and performing an even bigger tissue damage. Purpose. The aim of this study is to know about the expression of of TGF-ß and MMP-1 during the periapical tissue damage due to induction of E.faecalis. Method. This study used laboratory experimental with the post test only control group design. A total of 27 male rats were randomly divided into 3 main groups. Group A (negative control) : every tooth was’nt induced by anything. Group B ( positive control): every tooth was induced only by sterile BHIb and closed by GIC Fuji II as the final restoration. Group C (: every tooth was induced by 10 μl BHI-b E.faecalis ATCC212(106 CFU), and closed by GIC Fuji II as the final restoration. The animals were sacrificed after 21 days and prepared for histological examination of tissue damage, then we did the immunohistochemistry  followed by calculation on the light microscope. Result. The analysis revealed that the expression of MMP-1 increased significantly in group C when E.faecalis was induced. When expression of TGF-ß decreaced significantly in group C rather than group B.  Conclusion. From this study we know that the expression of TGF-ß and MMP-1 are make opposite pathway due to chronic apical periodontitis that induced by E.faecalis.

Abstract

Back Ground: Adherence of  Stretococcus mutans biofilm (S. mutans) to the surface of dental restorative materials is considered an important step in the development of secondary caries and periodontal disease. There are two type of nanofil composite: universal restorative and flowable restorative. That have different characteristic to induce S.mutans biofilm adherent in it surface.  Purpose: The aim of this study was to compare the adherence of S. mutans biofilm to two types of nanofil restorative materials, flowable restorative and universal restorative.. Materials and Methode: 32 disc-shaped specimens (∅ = 5.0 mm / thickness = 2.0 mm) of two types  composite were divided  to 4 groups (n = 8): group 1; universal restorative were immersed in pH cycling solution for 14 days ,group 2; Universal restorative  were  immersed in water for 14 day,group 3: flowable restorative were immersed in pH cycling solution,group 4: flowable restorative were immersed  for 14 days in water .  in day 15 th,  All speciments(n=32) were immersed for 24 hours in artificial saliva.. Streptococcus mutans cells were brought in contact with and grown on the speciments for 48 hours in BHI-B. Bacterial suspension was deposited onto each material and the adhesion of biofilm was evaluated trough optic density (OD) . Optic density   biofilm of S.mutans analyzed using Elissa reader’ Spectrophotometry. Statistical analysis was performed by Kruskall -wallis and Tukey HSD test  (α = 0.05). Result: Adherence of  S.mutans biofilm on flowable restorative (mean OD:1,933, SD: 0,633)  were significantly higher than universal restorative materials (mean OD: 1,240,SD:0,317). (P<0,05) Conclusion: The adherence of S.mutans biofilm on the surface of composites resin nanofil flowable restorative higher than universal restorative.

Abstract

Background: Enterococcus faecalis is one of the most common bacteria in root canal after endodontic treatment. This species is found in about 77% of the cases that resistant to treatment. It is necessary to develop an alternative for intracanal dressing, one of the potential substance is natural ingredients. The stem of ambonese banana (Musa paradisiaca var. sapientum) extract contains active substances such as tannin, flavonoid, alkaloid, saponin, and antraquinone, which have been known for their antibacterial potency. Purpose: The  aim of this study was to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of ambonese banana stem (Musa paradisiaca var. sapientum) extract against Enterococcus faecalis. Method: This research was a laboratory experimental study. Ambonese banana stem extract was made by maceration method with ethanol 96% and certain dilution was performed to obtain various concentration. Value of MIC and MBC of ambonese banana stem extract against Enterococcus faecalis were known by counting the growth of bacteria colonies on blood agar media in CFU/ml. Result: The percentages of bacteria colonies at concentration 85%; 82,5%; 80%; 77,5%; 75%; 72,5%; 70%; and 67,5% of ambonese banana stem extract in sequence were 0%; 0%; 0%; 0%; 0%; 0%; 7,44%; and 14,53% from the positive control. Conclusion: The stem of ambonese banana (Musa paradisiaca var. sapientum) extract had minimum inhibitory concentration (MIC) at 70% concentration and minimum bactericidal concentration (MBC) at 72,5% concentration against Enterococcus faecalis.

Abstract

Background : Dentinal hypersensitivity (DH) is a painful clinical condition and is characterized by pain arising from exposed dentin in response to various stimuli. Various treatment modalities are available to treat dentinal hypersensitivity which include at-home and in-office treatment. At home treatment generally consists of a variety of dentrifices containing different constituents like strontium chloride and bioactive glass (Novamin). These agents cause occlusion of dentinal tubules thereby reducing hypersensitivity.Purpose: This study  was  to evaluate the effects of different desensitizing dentifrices on dentinal tubule occlusion by scanning electron microscopy (SEM) of strontium chloride and bioactive glass (Novamin) pastes in the treatment of dentinehypersensitivity (DH) Methods: sixteen extractedspecimens from bovine incisors teeth with randomized into 2 groups (n=8). The crowns were removed from the root and the crown  were sectioned longitudinally into two parts (in a mesiodistaldirection). The cervical toothwere resulting in two samples per tooth.Dentinal tubules were exposedand thesamples provided a 4 mm x 4 mm area of exposed dentinal tubules.Group 1 (treated with strontium chloride), Group 2 (treated with  bioactive glass (novamin))After each treatment for seven days, tubule occlusion on dentin were analyzed by scanning electron microscopy  (SEM).The data were analyzed using “mann-whitneytest” (p<0.05).Result: Groups bioactive glass (novamin) showed tubule occlusion highest when compared with groups strontium chloride.Conclusion: bioactive glass (novamin) paste showed tubule occlusion highest with strontium chloride

Abstract

Background. Chlorhexidine gluconate is one of endodontic irrigants that has excellence capability to penetrate into dentin tubules and kill the pathogenic bacteria there. On the other hand, chlorhexidine gluconate has side effects to cause allergic reactions of the tissue and discoloration of the teeth. Xanthone from mangosteen pericarp can be considered as a natural alternative irrigant that usually has a good tolerance to the body. Purpose. The aim of this study compared the cytotoxicity between 0,78% xanthone from mangosteen pericarp and 0,2% chlorhexidine gluconate toward BHK-21 fibroblast cells.  Methods. This study used experimental post-test only control group design. Xanthone from mangosteen pericarp preliminary cytotoxicity tested in various concentrations. Xanthone from mangosteen pericarp classified as a non-toxic concentration at 0,78%. Cytotoxicity of 0,78% xanthone from mangosteen pericarp compare with cytotoxicity of 0,2% chlorhexidine gluconate using MTT assay method. Cytotoxicity of material can be seen from % of cell viability. Viable cell measured by the result of optical density that read by ELISA reader 620 nm. Result. 0,78% xanthone from mangosteen pericarp showed lower cytotoxicity than 0,2% chlorhexidine gluconate toward BHK-21fibroblast cells. One-way ANOVA showed a significant difference between the study groups (P<0,05). Conclusion. 0,78% xanthone from mangosten pericarp showed lower cytotoxicity than 0,2% chlorhexidine gluconate toward BHK-21 fibroblast cells.

Abstract

Stem cells provide new hope to fasten the healing process of various oral diseases including periodontal tissue(periodontitis). At present time, there is not yet found a material that can fix the teeth perfectly. Adult stem cells frombone marrow widely used for treatment. Numbers of stem cell are limited so growth factor is necessary to promoteproliferation of stem cells. Growth factor that has been used is expensive and difficult to get. Because of theseconstraints, it is necessary to develop alternative uses of medicinal plants as growth factor that accelerates woundhealing process. This study is aimed to proven what extent the potential Pegagan (Centella asiatica L) as a naturalgrowth factors to stem cells. The ability of proliferation and osteogenic differentiation in mesenchymal stem cells(MSC)was analyzed by adding terpenoid fraction of Centella asiatica. MTT assay data showed that the addition ofPegagan on cultured MSCs can increase stem cell proliferation. Pegagan increases the ability of MSC proliferationand differentiate into osteoblasts It is concluded that pegagan increase the ability of MSC proliferation anddifferentiate into osteoblasts.

Abstract

Background: Regenerative endodontic treatments are classified based on approach in the procedures and principles. One is cell-free approach that involves a revitalization procedure to achieve continued root growth while the other is cell-based approach which involves pulp/dentin regeneration through isolation and ex vivo expansion of stem cells seeded in the scaffold and then transplanted into the canal space. Technically, the cell-free approach is simpler than cell-based approach because the former does not concern about external stem cell isolation and culture. Currently, the gold standard and the one that has been clinically performed is cell-free approach. Cell-based approach is still not a standard because currently the cell-based approach is still in the clinical trial stage. Purpose: to evaluate the biological basis and clinical protocols recently used in regenerative endodontic treatment and discuss potentially future treatment approaches. Review: The literature review was searched in PUBMED with the keywords: immature permanent teeth, pulp revascularization, stem cells and regenerative endodontics. The regenerative endodontic treatments with various procedures and techniques result in a significant increase in root length and dentinal wall thickness. Stimulation of stem cells in apical root canal system is required to induce tissue formation and continued root maturation. Conclusion: The success of regenerative endodontic therapy relies on the development of a technique that enables clinicians to create functional pulp tissue within cleaned and shaped root canal systems. It is hoped that further research studies are required to define advantages and limitations of cell-free and cell-based approach.

Abstract

Background: Pulp capping is one of the treatments for reversible pulpitis and aims to maintain pulp vitality. This treatment requires a material that can protect the pulp with good biocompatibility. The physical and mechanical properties, bio interactivity and bioactivity of pulp capping materials are very important for the formation of reparative dentin. Calcium hydroxide (Ca(OH)2) as the gold standard material in pulp capping treatment also has some disadvantages. Another alternative for pulp capping material is blood clam shell because it contains 98% calcium carbonate (CaCO3), which is a compound with a bone-like structure and can induce pulp cell differentiation. Objective: To investigate and explain the difference in compressive strength between CaCO3 from blood clam shells and Ca(OH)2 as a candidate pulp capping material. Methods: This research is a laboratory experimental study with post test only control group design method. Ca(OH)2 and CaCO3 samples were formed with a mixture of powder and aquadest with 4x6 mm sample size. The samples were dried at room temperature and the compressive strength was measured using a universal testing machine (UTM). Result: There is a significant difference in compressive strength between Ca(OH)2 and CaCO3 blood clam shells in the Mann-Whitney test results (p<0.05). Conclusion: The results of the compressive strength test between the mixture of Ca(OH)2 with aquadest in a ratio of 1:1 are greater than the mixture of CaCO3 blood clam shells with aquadest in a ratio of 3:1 so that pure CaCO3 blood clam shells with distilled water without other additives cannot be used as a candidate for capping pulp material.

Abstract

Introduction: Diabetes mellitus is a chronic metabolic disorder characterized by high blood sugar levels. It is a global health problem that can increase the risk of tooth decay and requires conservative dental treatment. Several factors impact the severity of tooth decay, sugary drink consumption is one of them. This study aimed to investigate the association between tooth decay in patients with diabetes mellitus who regularly consume sugary drink versus those who do not. These patients visited Conservative Dentistry Section, Universitas Airlangga Dental Hospital Surabaya, Indonesia, during Jan-Mar 2023. Methods: This study involved 37 patients with diabetes mellitus and posterior tooth decay. Demographic data were gathered from personal interview and dental records. Statistical analysis was done using chi-square analysis. Results: sugary drink consumption has a significant difference (p<0.05) among the patients with posterior tooth decay and diabetes mellitus. Conclusion: sugary drink consumption influences the severity of tooth decay in diabetes mellitus patients.

Abstract

Education is the human’s right as the main fondation in the development of the nations. The school zoning system is implemented in the process of accepting new student in order to acess the education evenly. The problem of this research are related to the rules of accepting new students through the zoning system and the authority in determining their zones. This research is aimed to be a critic related to school zoning process which is still becoming an issue nowdays. This research was normative analyzed by using three methods such as statue approach, fact approach and analytical conceptual aproach. The results in this research showed that there is vagueness norm in determining the school zone which has caused some problems for the new students. Therefore, the government’s authority is needed to interpretation this vagueness, and give freedom in determining the zoning area for new student who cannot enter the school because of the long distance. It should be done in order to implement the educational justice and enforcement of human rights in Indonesia.
