Abstract

Limbah pada sedimen tambak bandeng berupa bahan organik, bersumber dari sisa pakan dan hasil metabolisme. Limbah ini mengandung protein yang tinggi, apabila tidak diproses dengan baik dapat mencemari lingkungan perairan. Aktinomisetes merupakan organisme tanah yang memiliki peran penting dalam proses dekomposisi bahan organik dengan bantuan enzim. Penelitian ini bertujuan untuk mengisolasi aktinomisetes yang mampu menghasilkan enzim protease. Jenis penelitian yang dilakukan yaitu deskriptif eksploratif. Sampel sedimen diambil dari dasar tambak ikan bandeng Kecamatan Tugu, Semarang pada 2 titik outlet tambak dengan metode coring modifikasi. Proses isolasi dilakukan menggunakan media IM5, IM6, IM7 dan IM8 dengan metode pour plate, sedangkan purifikasi dengan media IM6 dan IM8 menggunakan metode streak plate. Isolat diskrining dengan media spesifik Skim Milk Agar untuk melihat zona hambat di sekitar koloni sehingga diketahui Indeks Proteolitik (IP). Sebanyak 11 dari 17 isolat aktinomisetes mampu memproduksi enzim protease, tiga isolat berasal dari media IM6 dan 8 isolat dari media IM8. Diperoleh tiga isolat dengan kemampuan paling baik dalam menghasilkan enzim protease dengan nilai IP diatas 1,5, yaitu isolat VTB 1.1, VTB 1.2 dan YTB 1.2. Penelitian lanjutan perlu dilakukan untuk membuktikan bahwa ketiga isolat tersebut mampu menjadi agen pendekomposisi kandungan protein berlebih pada sedimen tambak.

Abstract

The Awur Bay and Kartini Beach in Jepara, Central Java, are fishing areas with blue swimming crab as a main catch. This study aims to examine the species of blue swimming crabs captured in the Northern Waters of Jepara from different locations, analyze the molecular characteristics based on gender differences, obtain GenBank accession numbers as genetic conservation, and construct their phylogenetic tree. The research used a quantitative descriptive method. Samples were collected using purposive sampling from fishermen's catches and selected based on the difference in crab claws and carapace color, with a total of 4 samples. The samples were extracted using a DNA extraction kit and tested using Polymerase Chain Reaction (PCR) to visualize mitochondrial DNA (COI gene) and processed using MEGA 11. BLAST results showed that the similarity of the samples to Portunus pelagicus ranged from 97.02% to 99.42%. The different colors of claws and carapace did not affect the DNA sequence composition, hence indicating sexual dimorphisms. The DNA sequences from this study were deposited to GenBank with accession number LC836055.1-LC836058.1. Based on the phylogenetic tree construction, the clade of all samples shows a close relationship with the species Portunus pelagicus.

Abstract

Ekosistem mangrove memiliki berbagai peranan ekologis, salah satunya adalah sebagai habitat organisme gastropoda. Gastropoda ekosistem mangrove berperan sebagai bioindikator dan dekomposer. Identifikasi spesies gastropoda secara molekuler di kawasan mangrove penting dilakukan untuk mengetahui spesies-spesies gastropoda yang ditemukan secara akurat. Penelitian ini bertujuan untuk mengidentifikasi spesies gastropoda dan tingkat kekerabatannya di Kawasan Mangrove Desa Tapak Kecamatan Tugu Kota Semarang yang dilaksanakan pada bulan September - November 2023. Metode yang digunakan dalam penelitian ini yaitu deskriptif-kuantitatif. Pengambilan sampel penelitian dilakukan dengan metode purposive sampling. Penanda genetik yang digunakan dalam penelitian ini adalah gen Cytochrome Oxydase I (COI) dengan penyejajaran sekuens DNA menggunakan metode MUSCLE. Analisis filogenetik berbasis algoritma neighbor joining (NJ) 1000 bootstrap pada software MEGA X. Hasil identifikasi molekuler diperoleh bahwa sampel GT1 memiliki kemiripan sebesar 96,12% (Percent Identity) dengan Pirenella alata dan sampel GT2 sebesar 99,69% (Percent Identity) dengan Telescopium telescopium. Hasil analisis filogenetik menunjukkan kladogram kedua spesies ini terdapat pada klade yang berbeda. Klade I terdiri dari sampel GT1 beserta 4 spesies pembanding lainnya dalam 1 genus, yaitu Pirenella alata, Cerithidea microptera, Pirenella nanhaiensis, dan Pirenella incisa. Klade II terdiri dari sampel GT2 beserta 4 spesies pembanding lainnya yaitu Telescopium telescopium, Varicinassa variciferus, Laevilitorina caliginosa, dan Monoplex krebsii.  Mangrove ecosystem has many ecological roles, one is as gastropods habitat. Mangrove ecosystems gastropods act as bioindicators and decomposers. Gastropods mangroves areas molecular identification is important to accurately the types of gastropod species. This research aims to identify gastropods species and the relationship levels in the Mangrove Area of Tapak Village, Tugu District, Semarang conducted from September − November 2023. This research using descriptive-quantitative method. Research sampling using purposive sampling method. The genetic marker used in this research is the Cytochrome Oxydase I (COI) gene with DNA sequence alignment MUSCLE method. Phylogenetic analysis based on the neighbor joining (NJ) 1000 bootstrap algorithm in MEGA X. The results of molecular identification showed that GT1 sample had a similarity of 96.12% (Percent Identity) to Pirenella alata and GT2 sample was 99.69% (Percent Identity) similar to Telescopium telescopium. The results of the phylogenetic of two species analysis show that the cladograms are in different clades. Clade I consist of sample GT1 along with 4 other comparison species in 1 genus, namely Pirenella alata, Cerithidea microptera, Pirenella nanhaiensis, and Pirenella incisa. Clade II consists of sample GT2 along with 4 other comparison species, namely Telescopium telescopium, Varicinassa variciferus, Laevilitorina caliginosa, and Monoplex krebsii.

Abstract

Kepiting merupakan komoditas perikanan bernilai ekonomi tinggi dan menjadi produk ekspor dari perairan utara Jawa Tengah. Berdasarkan kajian Komisi Nasional Pengkajian Stok Ikan (2022), potensi lestari kepiting di Laut Jawa diperkirakan 7.360 ton per tahun dengan tangkapan diizinkan 5.152 ton per tahun, dan tingkat eksploitasi telah mencapai 0,9. Penelitian ini bertujuan menganalisis pola pertumbuhan, ukuran tangkap pertama (〖CW〗_c), ukuran matang gonad pertama (〖CW〗_m), dan tingkat eksploitasi kepiting bakau (Scylla sp.) di perairan Pekalongan hingga Kendal sebagai upaya mendukung pengelolaan berkelanjutan. Abstract

Tambak budidaya udang banyak memproduksi limbah organik yang berasal dari sisa metabolisme dan sisa pakan dari kultivan budidaya. Keberadaan limbah organik yang melimpah dan tidak didampingi dengan manajemen kualitas air, akan mengakibatkan dampak buruk pada kultivan seperti kematian. Proses bioremediasi termasuk salah satu cara untuk membersihkan bahan pencemar pada lingkungan tambak yang dibantu oleh agen biologis yaitu bakteri. Bakteri amilolitik dapat digunakan sebagai agen bioremediator pada lingkungan karena menghasilkan enzim amilase. Amilase merupakan enzim yang mampu menghidrolisis pati menjadi senyawa yang lebih sederhana. Penelitian ini bertujuan untuk mengisolasi bakteri dari sedimen tambak udang vanname dan melakukan skrining enzim amilase menggunakan media Starch Casein Nitrat (SCN). Sedimen diambil pada 5 lokasi tambak pasca panen di Kabupaten Rembang. Sampel diisolasi menggunakan teknik pour plate dan purifikasi dengan teknik streak plate, sedangkan skrining enzim dilakukan dengan teknik dotting. Hasil penelitian yang didapatkan menunjukkan bahwa terdapat 20 isolat bakteri yang diisolasi dari sedimen tambak udang vanname dan 9 isolat bakteri menunjukkan aktivitas enzim amilase, diketahui dari zona bening yang dihasilkan pada isolat setelah ditambahkan iodine.

Abstract

Ekosistem mangrove berada pada wilayah intertidal yang memiliki interaksi dinamis antara perairan laut, payau, sungai dan terestrial. Fungsi ekologis ekosistem mangrove sebagai pelindung pantai dari erosi, feeding ground, nursery ground, dan spawning ground bagi biota perairan. Biota yang beragam termasuk moluska kelas bivalvia banyak ditemui di daerah mangrove. Identifikasi spesies bivalvia secara molekuler di kawasan mangrove penting dilakukan untuk mengetahui spesies-spesies bivalvia yang ditemukan secara akurat. Tujuan penelitian ini adalah mengetahui spesies bivalvia di lokasi penelitian dan mengetahui hubungan filogenetik spesies yang telah ditemukan melalui analisis pohon filogenetik. Metode yang digunakan dalam penelitian ini adalah deskriptif kuantitatif dengan metode pengambilan sampel purposive sampling. Penelitian ini berlangsung pada bulan September – Desember 2023. Penanda genetik yang digunakan dalam penelitian ini adalah gen Cytochrome Oxydase I (COI) dengan penyejajaran sekuens DNA menggunakan metode maximum likelihood. Analisis filogenetik berbasis algoritma neighbor joining (NJ) 1000 bootstrap pada software MEGA XI. Hasil data sequencing kemudian dianalisis menggunakan BLAST dan menunjukkan sampel BM1 memiliki kemiripan sebanyak 99% dengan spesies Donax incarnatus sedangkan 99,85% pasa sampel BM2 dengan dengan spesies Perna viridis. Hasil analisis filogenetik menunjukkan kladogram kedua spesies ini terdapat pada klade yang berbeda. Klade I terdiri dari sampel BM1 beserta 4 spesies pembanding lainnya yaitu Donax incarnatus, Donax obesulus, Donax deltoides, dan Donax faba. Klade II terdiri dari sampel BM2 beserta 4 spesies pembanding lainnya yaitu Perna viridis, Perna canaliculus, Perna perna, dan Perna Picta.

Abstract

Cengklik Reservoir is one of the water and waterfront tourism destinations in the Boyolali Regency which has an area of 327,44 ha. The condition of Cengklik Reservoir is currently undergoing revitalization and tourism development. The purpose of this study was to analyse the suitability of reservoir waters as a tourist attraction based on the Tourism Suitability Index, to determine the perceptions of tourists potential in the reservoir, and community perceptions of tourism development in the reservoir. The research was conducted in March - April 2023. The research methods used a case study with quantitative analysis to calculate the value of the Tourism Suitability Index and qualitative analysis to determine the perception of tourism potential from 30 tourist respondents and tourism development from 30 community respondents. Results of the TSI calculation of the Cengklik Reservoir for boat tours amounted to 2.40 (suitable) and fishing amounted to 3.00 (very suitable). The tourists perception of the Cengklik Reservoir as well as the communitys perception of tourism development were rated positively. Abstract

White shrimp (P. merguiensis) is the dominant catch caught in Demak waters and has high economic value. The fishing gear used by fishermen is wangkong fishing gear. This research aims to determine the composition and size structure of the catch, as well as the relationship between length and weight and sex ratio. The research was carried out in April and May 2024. The research method used was a survey method. The shrimp sampling method used is the census method. The target catch of fishermen from Menco Village, namely Penaeus merguiensis, was 87.10% and fishermen from Babalan Village, namely Penaeus merguiensis, amounted to 66.63%. The non-target catches were Mugil spp, Plotosuscanius, Rastrelliger spp, Scylla serrata, Oratosquilla oratoria, and Anguilla bicolor. The length class mode for white shrimp in Menco Village is 74.4 – 79.2 mm and Babalan Village is 96.6 – 106.5 mm. The relationship between length and weight of male white shrimp from Menco Village was obtained as W= 0.000353293L2.724 and female white shrimp from Menco Village was obtained as W= 0.000244543L2.837. The relationship between length and weight of male white shrimp from Babalan Village was obtained as W= 0.001095644L2.402 and female white shrimp from Babalan Village was obtained as W= 0.000167463L2.960. The sex ratio for white shrimp showed that the ratio between males and females in Menco Village was 1:1.10 and Babalan Village was 1:1.36. The growth pattern of P. merguiensis is negative allometric and more female shrimp are caught than male shrimp. Keywords: Catch; Demak Waters; Wangkong; WhiteShrimp (P. merguiensis)

Abstract

The Wulan Delta, located in Wedung, Demak, was formed due to the sedimentation process of the Wulan and Serang rivers. Sediment transport starts from rivers to the sea; the process carries nutrients and various chemicals derived from agricultural, industrial, or household activities. This study aims to analyze the characteristics of hydrodynamics, suspended sediment, and bottom sediments in the Wulan Delta waters, Demak. Field data were collected in transitional season two (October) 2022. The obtained data were then analyzed in the laboratory at the Faculty of Fisheries and Marine Science. Results showed that the speed of the surface current in the study area ranged from 0 to 0.1 m/s in spring tide conditions with a surface current pattern coming from the direction of Semarang toward Jepara and 0–0.18 m/s in neap tide conditions with a current pattern coming from the direction of Jepara toward Semarang. The Wulan Delta has a sedimentary characteristic dominated by fine silt, which is as much as 82%–98%, with a deposition characteristic dominated by wave energy that causes successful sediment sorting with a sorting index of −2.25 to 2 and strongly asymmetrical to a small size with a skewness curve value of two.
