Abstract

Abstract

Porphyromonas gingivalis is an oppoturnistic microorganism in periodontal disease that is formed at the stage maturation of biofilm formation. One of the natural materials biofilm reduction is the Phaleria macrocarpa. Phaleria macrocarpa’s leaf contains chemical compounds such as flavonoids, tannins, alkaloids, saponins that function as antibacterials. The aim of this research is to determine the effectiveness of the nanoemulsion of the gel Phaleria macrocarpa’s leaf  on the thickness of the P. gingivalis bacterial biofilm. The research design was carried out with a post test only with a total of 28 samples, then divided into 4 groups of treatment of Phaleria macrocarpa’s leaf extract nanoemulsion gel preparations with variations in concentrations of 30%, 40%, and 50%, as well as a control group of 25% metronidazole gel. The reading of the anti-biofilm thickness test results was measured by OD (Optical density) using an ELISA-reader. One way Anova parametric test among obtained p<0.000 results showing the effect of Phaleria macrocarpa’ leaf gel nanoemulsion extract on the decrease in the thickness of the Porphyromonas gingivalis bacterial biofilm. It can be concluded that the nanoemulsion gel formulation of Phaleria macrocarpa’s leaf extract concentrations of 30%, 40%, and 50% is effective, with a concentration 50% was most effective against reducing the thickness of the bacterial biofilm Porphyromonas gingivalis.

Abstract

Background: In Indonesia, periodontitis is among the most prevalent periodontal disease. The dentist may administer extra adjunctive therapy as a gel to treat inflammation and the bacterium that causes periodontitis. The antibacterial and anti-inflammatory xanthones included in mangosteen peel extract gel speed up the inflammatory phase. Objective: This study aimed to determine the effectiveness of mangosteen peel extract gel in increasing the number of fibroblast cells in the healing process of periodontitis in Wistar rats (Rattus norvegicus). Material and Method: This research was an experimental laboratory design with a post-test-only design divided into four groups, 50% mangosteen peel extract gel group, 75% mangosteen peel extract gel group, metronidazole gel group, and metronidazole plus gel group. Result: The research data were analyzed using the One Way ANOVA test and showed a significant value difference with a p=0.000 (p<0.05). However, in the inter-group test with Bonferroni post hoc, it was found that there was no significant difference between the 75% mangosteen peel extract gel group and the metronidazole gel group with a value of p = 0.709 (p>0.05). Conclusion: This study concludes that mangosteen peel extract gel effectively increases fibroblast cells' number in the periodontitis healing process.

Abstract

Background: Phaleria macrocarpa’s leaf contains alkaloids, saponins, phenols, tannins, and flavonoids which have antioxidant, anti-inflammatory, and antibacterial activities. The nanoemulgel preparation was chosen because it can penetrate well and quickly into the mucosa thereby increasing absorption. Physical stability tests need to be carried out to ensure the quality, safety and benefits of the gel meet the expected specifications and stable during storage. Objective: This study aims to determine which concentration has the most stable physical stability of pH and viscosity which were observed for 28 days. Materials and Methods: This was an experimental laboratory design, with 30 preparation divided into 3 groups, group of 30%, 40%, and 50% concentration preparation of nanoemulgel leaves of Phaleria macrocarpa. which were tested for pH tests and viscosity tests for 28 days which was observed every 7 days in climatic chamber. The pH and viscosity tests among groups were compared and analyzed by the Repeated Anova and Pos Hoc test. Results: Average pH test and viscosity test 30% concentration with the average baseline and h+28 pH values were 5,02 and 4,97 and the baseline and h+28 viscosity values were 1610 cPas and 1520 cPas. The results of the average concentration of 50% pH value at baseline = 4,65 and h+28 = 4,60. The mean baseline viscosity value=1290 cPas and h+28=1170 cPas. The results of the post-Wilcoxon test showed p>0.05 in the 30% concentration group for pH and viscosity tests showed that there was no significant difference in pH and viscosity values at a concentration of 30%, It means the pH value and viscosity of the 30% concentration preparation of nanoemulgel were relatively constant from baseline to h+28. Conclusion: It can be concluded that the pH and viscosity test of the preparation of nanoemulgel leaves of Phaleria macrocarpa at a concentration of 30% was the most stable pH and viscosity compared to concentrations of 40% and 50%.
