Abstract

Pseudomonas fluorescens is plant growth promoting rhizobacteria (PGPR) often inoculated on plants as natural biocontrol agent capable of protecting the plants from soil-borne pathogens. Chemotactic motility allows populations of P. fluorescens to rapidly search for nutrients and is an important factor determining their competitive success to colonize plant root. Therefore, we investigated various glucose concentrations from 0% to 1% (w/v) to enhance chemotactic motility and growth of this rhizobia. Chemotactic motility was evaluated using swim plate assay and bacterial growth was measured using UV-Vis Spectrophotometer in LB and M9 medium. Glucose with low concentration (0.05%) showed to have optimum response in P. fluorescens chemotactic motility with colony diameter 38.3 mm in LB medium and 12.8 mm in M9 medium. Highest growth of P. fluorescens was seen in control condition of LB medium reaching a peak at 0.0246 OD600 (~±1,44x107 CFU ml-1 ) while growth in M9 medium supplemented with 1% glucose was just slightly lower with 0.0227 OD600 (~±1,32x107 CFU ml-1). Glucose in high concentration showed to repress chemotactic motility and first growth phase of P. fluorescens in LB medium due to catabolite repression.

Abstract

The organic content of palm liquid sewages is an indication of the abundance of microorganisms. This study aims to obtain anoxygenic photosynthetic bacteria (BFA) that produce carotenoid pigments from palm liquid sewages. Therefore, We isolated and screened of BFA from three palm liquid sewage disposal sites in Jambi Province. This research was conducted by an experimental method using a modified mineral medium. BFA isolates that growth and produced carotenoid pigments are visible because of the distribution of reddish-yellow pigments on the culture media. Screening results obtained 11 isolates of BFA sequentially Bg1K201, Bg1K202, Bg2k201, Bg3k201, Mr1k201, Mr1k202, Sl1k201, Sl1k202, Sl2k201, Sl2k202 and Sl3k202. The results of morphological and physiological characterization based on the Bergeys Manual of Determinative Bacteriology show that there are two types of BFA genera that have carotenoid pigments, respectively, the genus Rhodobacter (Bg1k201, Bg1k202, Bg3k201, Sl1k201, Sl1k202, and Sl1k202, and Sl2k202, and Sl1k202, and Sl2k202, and Sl1k202 and Sl2k202, and Sl2k202, and Sl2k202. Abstract

Mangrove ecosystem is a coastal wetland has biodiversities such as flora, fauna, and microorganisms. Microorganisms are an importants component in the mangrove ecosystem and have many roles, including source the nutrition, decomposer, and a source of biotechnology products. Cellulolytic bacteria are a group of cellulase-producing bacteria capable of breaking down cellulose into glucose monomers. This study aims to screen and determine the diversity of cellulolytic bacteria from the mangrove ecosystem. The research method has been experimental by exploring the cellulolytic bacteria in soil and sediment from the mangrove ecosystem. Three sampling locations were selected such as the litter soil, soil in the root, and soil in the tidal area. The screened of potential bacteria-producing cellulase was obtained by bacteria growth on Carboxy Methyl Cellulose (CMC) agar medium. The isolates were cultured for 48 hours at 37°C. Then, the bacteria that produced the cellulolytic activity were characterized by morphological and physiological characteristics. The results were adjusted according to Bergey's Manual of Determinative Bacteriology. The determination showed that six isolates had the same characteristics as Bacillus, Cellulomonas, Lactobacillus, and Micrococcus. The cellulolytic activity index showed that three isolates were sequentially MS06 (9,73), MS03 (6,30), and MS08 (5,41).

Abstract

Pro coffee is a fermented beverage using Bacillus cereus bacterial consortium with single-cell protein. This study aims to determine the best composition of the B. cereus consortium bacteria mixture in organoleptic aspects that panelists most prefer and the effect of adding B. cereus consortium bacteria on proximate analysis and pro coffee flavor. Organoleptic testing of probiotic coffee was conducted on 20 panelists, then continued with proximate analysis of pro coffee. The composition with the best organoleptic flavor is obtained in code D, with the treatment of adding 5% bacterial consortium B. cereus for 15 days. The analysis of taste obtained has a value of 0.000 <0.05, and the study results show that the significance value is smaller than the significance level. Hence, the treatment results give the effect of adding bacterial consortium to pro-coffee. The addition of bacterial consortium B. cereus to probiotic coffee, in the form of moisture, ash, protein, fat, and carbohydrate content, is 0.000 <0.05. The analysis results show that the significance value is greater than the significance level, so the treatment results do not give the effect of adding bacterial consortium to pro coffee

Abstract

Vannamei shrimp (Litopenaeus vannamei) is a fishery commodity widely cultivated in various parts of Indonesia. The main problem often found in vannamei shrimp production is poor water quality during the rearing period. This study aims to isolate and identify E. coli contained in water, sediment, and meat of vannamei shrimp cultivated in shrimp ponds and examine its sensitivity to antibiotics. The research was conducted from March to May 2023 at the Technical Implementation Unit (UPT) Vannamei Shrimp Ponds of the Maritime Affairs and Fisheries Service, Tanjung Punak Village, Rupat Utara District, Bengkalis Regency, Riau. The survey method was used in this study. The Most Probable Number (MPN) method was employed for E. coli isolation to calculate bacterial density, and the Kirby-Bauer method was used to test bacterial sensitivity to antibiotics. Based on the identification of the six isolates carried out, it was found that only two isolates were positively identified as E. coli bacteria, namely SD C 3.1 and IP B 1.1, which were isolated from sediment and the Wastewater Treatment Plant (WWTP) pond. The diameter of the inhibition zone for sediment isolate RF01 against the antibiotic chloramphenicol was 18.75 mm, indicating intermediate sensitivity. On the other hand, WWTP isolates RF02 exhibited an inhibition zone of 17 mm for chloramphenicol, indicating medium sensitivity as well. However, it showed resistance to penicillin with a diameter of 6.5 mm and resistance to ampicillin with a diameter of 2 mm.
