Abstract

Mangrove ecosystem is a coastal wetland has biodiversities such as flora, fauna, and microorganisms. Microorganisms are an importants component in the mangrove ecosystem and have many roles, including source the nutrition, decomposer, and a source of biotechnology products. Cellulolytic bacteria are a group of cellulase-producing bacteria capable of breaking down cellulose into glucose monomers. This study aims to screen and determine the diversity of cellulolytic bacteria from the mangrove ecosystem. The research method has been experimental by exploring the cellulolytic bacteria in soil and sediment from the mangrove ecosystem. Three sampling locations were selected such as the litter soil, soil in the root, and soil in the tidal area. The screened of potential bacteria-producing cellulase was obtained by bacteria growth on Carboxy Methyl Cellulose (CMC) agar medium. The isolates were cultured for 48 hours at 37°C. Then, the bacteria that produced the cellulolytic activity were characterized by morphological and physiological characteristics. The results were adjusted according to Bergey's Manual of Determinative Bacteriology. The determination showed that six isolates had the same characteristics as Bacillus, Cellulomonas, Lactobacillus, and Micrococcus. The cellulolytic activity index showed that three isolates were sequentially MS06 (9,73), MS03 (6,30), and MS08 (5,41).

Abstract

This research was conducted from February to June 2023. A sampling of Avicennia marina and Sonneratia alba mangroves was conducted in the mangrove ecosystems of Mengkapan Village, Sungai Apit District, Siak Regency. This research aims to identify the species and test the antimicrobial activity of endophytic fungi found in the leaves and stems of A. marina and S. alba mangroves in the mangrove ecosystem of Mengkapan Village using survey and experimental methods. This research began by determining the sampling location and measuring the physical and chemical parameters of the mangrove environment. Then, sampling and preparation of A. marina and S. alba mangrove samples were carried out to isolate the endophytic fungi. After isolation, macroscopic and microscopic characterization was done to identify endophytic fungi. The diameter increase of the endophytic fungi was measured and calculated, and the antagonism activity test of the endophytic fungi was carried out. The antimicrobial index of endophytic fungi was measured against the pathogenic bacteria Vibrio sp and E. coli. This research obtained that 12 endophytic fungi were isolated from the mangroves A. marina and S. alba: two genera Rosellinia, two genera Pythium, four genera Hansfordia, and four genera Aspergillus. Based on the test results, all genera found in this research inhibited the growth of E. coli with inhibitions of 75.99% (Rosellinia sp), 87.70% (Pythium sp), 73.42% (Hansfordia sp) and 86.88% (Aspergillus sp). However, only three genera inhibited the growth of Vibrio sp by more than 50%, with inhibitions of 85.23% (Rosellinia sp), 87.87% (Hansfordia sp), and 86.07% (Aspergillus sp).

Abstract

Cobia fish (Rachycentron canadum) is one of the marine carnivorous fish that is relatively new and has the potential to be cultuvited. This study aims to determine the effectiveness of larval rearing and nursery cobia fish found in the Center for Marine Aquaculture (BBPBL) Lampung. Held 20 January to 19 February 2021. The mature fish were mated in a 2x2x1 m concrete tank filled with 4,000 L of water. The eggs produced were harvested using an egg collector, counted and hatched in a 0.8x0.6x0.5 m aquarium. After hatching, the larvae were moved into nursery tanks with a density of 5-7 fish/L. Larvae were reared until they were 25 days old and fed naturally with Branchionus sp, Nannochloropsis sp, and artemia. The larvae were then counted, weighed, measured, graded and their growth analyzed. The results of this study showed that not all of these cobia eggs were fertilized by male cobia sperm at the time of spawning. Spawning was carried out on January 16, 2022. Total eggs was 648,000, total fertilized eggs was 480,000 and fertilization rate was 74.04 %. The results of the initial larval count were 75,000, 17,040 were harvested with a survival rate of 22.72%. Water quality (pH, DO, temperature, salinity, nitrite and ammonia) during the research was still normal.

Abstract

Pro coffee is a fermented beverage using Bacillus cereus bacterial consortium with single-cell protein. This study aims to determine the best composition of the B. cereus consortium bacteria mixture in organoleptic aspects that panelists most prefer and the effect of adding B. cereus consortium bacteria on proximate analysis and pro coffee flavor. Organoleptic testing of probiotic coffee was conducted on 20 panelists, then continued with proximate analysis of pro coffee. The composition with the best organoleptic flavor is obtained in code D, with the treatment of adding 5% bacterial consortium B. cereus for 15 days. The analysis of taste obtained has a value of 0.000 <0.05, and the study results show that the significance value is smaller than the significance level. Hence, the treatment results give the effect of adding bacterial consortium to pro-coffee. The addition of bacterial consortium B. cereus to probiotic coffee, in the form of moisture, ash, protein, fat, and carbohydrate content, is 0.000 <0.05. The analysis results show that the significance value is greater than the significance level, so the treatment results do not give the effect of adding bacterial consortium to pro coffee
