Abstract

Abstract. Rahayu SYS, Prihatini W. 2020. Characterization of metallothionein protein from hepatopancreas organ of Pilsbryoconcha exilis collected from Cikaniki River, Western Java, Indonesia. Nusantara Bioscience 12: 1-5. Freshwater environment, undergoing various changes due to the presence of dangerous toxic anthropogenic waste. It causes pressure on the freshwater biota that lives in it, such as Pilsbryoconcha exilis mussel at the bottom of freshwater. This pressure is controlled by the body through the synthesis of a set of stress proteins. Endogenous proteins, metallothionein (MT), in the body of freshwater biota absorb heavy metals in the body of biota, in the form of stress control. This research identified MT protein on P. exilis from contaminated waters such as the Cikaniki river with the average of mercury levels in water, sediment, and hepatopancreas of mussels using AAS method were 0.001 mg/L, 0.120 mg/L, and 1.318 mg/L respectively. Hepatopancreas of P. exilis was extracted using a Tissue Extraction Reagent I kit (Invitrogen), with procedures following the factory manual. The extract was purified by filtration using Sephadec 50; then, the filtration results were migrated together with the PageRuler TM Unstained Low Range Protein Ladder (Fermentas) in Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS PAGE) gel medium on Biorad Protein II electrophoresis. After completion of electrophoresis, the gel was stained using Page Blue Protein Staining Solution (Fermentas), following the factory manual procedure. Characterization at this research has succeeded in obtaining the MT-I isoform protein measuring 5, 10, and 25 kDa from the hepatopancreas organ of P. exilis.

Abstract

Abstract. Rahayu SYS, Fadila A, Fahmi MR. 2023. Identification of metallothionein protein in Anodonta woodiana as a biomarker of mercury (Hg) contamination. Nusantara Bioscience 15: 90-94. Heavy metal contamination can affect the survival of aquatic biota and will accumulate in the bodiesorganisms. Moreover, contamination identification at the molecular level can be analyzed using biomarker analysis. Biomarkers are responses measured individually, ranging from enzymes and xenobiotic measurements to organ and overall conditions. Biomarker analysis can be done by checking the metallothionein protein, this expression can be induced by Reactive Oxygen Species (ROS). Metallothionein (MT) has a thiol group with nucleophilic properties. As a result, this group can make Metallothionein able to find metals and free radicals. Therefore, prevention that can be done to reduce contamination at a higher trophic level requires monitoring the molecular level by observing the metallothionein protein. For example, Anodonta woodiana (Rea, 1834) or kijing taiwan induced by HgCl2 aims to characterize their absorption ability in the environment through metallothionein protein. That was conducted by the SDS-PAGE (Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis) method. SDS-PAGE electrophoresis results showed that the size of the metallothionein protein was 7 kDa, 15 kDa, and >30 kDa. Size >30 kDa is a group of proteins related to stress control or Heat shock protein (Hsp). The presence of Hsp is due to the body increasing stress protein synthesis and metallothionein to reduce normal protein synthesis. Based on the results obtained, this study revealed that A. woodiana could absorb HgCl2, as evidenced by the metallothionein protein characterization results.

Abstract

Rapid population growth has generated significant environmental challenges, particularly in waste management, which remains a national strategic issue in Indonesia. Waste management is regulated under Law No. 18 of 2008 on Waste Management and strengthened through Presidential Regulation No. 97 of 2017, which outlines the National Policy and Strategy for Household Waste and Similar Waste Management. One approach to reducing household waste is the TPS-3R (Reduce, Reuse, Recycle) program, which emphasizes minimizing waste volumes and improving waste characteristics prior to disposal at final landfill sites (TPA). This study aims to evaluate the performance of the TPS-3R program in Bogor City in terms of its effectiveness in reducing household waste. The evaluation applied the CIPP model (Context, Input, Process, and Product). The findings show that the Context aspect is strong, as the program aligns with national regulations, has a clear legal foundation, and meets the objective of reducing waste at its source through community participation. The Input aspect is adequate but requires improvement: TPS-3R facilities generally meet basic standards; however, human resources are still limited, with managerial and technical roles often carried out concurrently, and maintenance funding remains insufficient. The Process aspect is also rated adequate: available facilities are only utilized at 50–70%, while training and capacity building for TPS-3R managers are conducted more than three times annually. Funding primarily relies on community contributions, government allocations, and revenues from recyclable products. The Product aspect shows partial success: TPS-3R facilities contribute to waste reduction and generate by-products that provide supplementary income, yet overall waste reduction targets outlined in the Regional Waste Management Strategy (Jakstrada) have not been fully met. In conclusion, optimizing both input and process components is crucial to enhance TPS-3R performance and achieve the national target of reducing waste by 30% by 2025.

Abstract

Small urban lakes are increasingly threatened by domestic and industrial wastewater inputs that exceed their natural assimilation capacity. This study aimed to evaluate the water quality status and develop a dynamic pollution control model for Situ Tlajung Hilir, Bogor Regency, Indonesia. Water quality was assessed using the STORET method based on Government Regulation No. 22/2021 standards, while pollution dynamics were simulated using a system dynamics model consisting of population growth, domestic load, industrial load, and assimilation capacity submodels. Historical monitoring data (2019–2023) were used for calibration and behavioral validation through trend comparison. The lake was classified as heavily polluted, with BOD, Total Nitrogen, and Total Phosphate exceeding permissible limits and surpassing assimilation capacity. Simulation of management scenarios (2020–2035) showed that business-as-usual conditions lead to continuous degradation, whereas integrated domestic and industrial wastewater treatment can reduce pollutant loads by 60–80% and gradually approach ecological balance. The proposed integrated STORET–system dynamics framework provides a practical decision-support tool for sustainable lake restoration and urban water resource management.
