Abstract

Soybean Fusarium wilt and root rot disease caused by a necrotrophic ascomycete pathogen, F. oxysporum, triggered severe damage to the plant tissues and organs and impacted heavy losses. Biocontrol agents, Bacillus subtilis, were commonly used to produce a broad spectrum of antifungal substances and were gradually used in biocontrol studies for plant disease management. Investigation and determination of the inhibiting mechanism of antifungal substance produced by B. subtilis on F. oxysporum should be done to protect the soybean plant. This study revealed that basal nutrient broth (NB) gives the best antifungal activity. The stationary phase of the bacterial growth curve was obtained on two days of cultivation and showed the maximum antifungal activity against F. oxysporum. Ethyl acetate (EA) extraction of bacterial supernatant generated crude EA extract, which showed half inhibition (IC50) at 306.42 µg/ml obtained from the dose-response regression curve. Post-treatment mycelia of F. oxysporum with bacterial extract were demonstrated as hyphal deformation followed by malondialdehyde (MDA) accumulation. Furthermore, cellular leakage on fungal cells that may be triggered by antifungal compounds from strain W3.15 occurred. Last, the related antifungal compounds were predicted to be epicatechin and benzophenone from the LC-MS/MS analysis of crude EA extract. Accordingly, the biocontrol agent B. subtilis strain W3.15 promises a strong potency for biofungicide development.

Abstract

The trend of contaminant analysis methods is beginning to shift towards methods that are considered greener in an effort to support environmental sustainability. QuEChERS is known as a green extraction method because it requires less organic solvent and shorter pretreatment time than other conventional extraction methods. Deep Eutectic Solvent (DES) as a more environmentally friendly solvent has been developed to support the realization of green extraction. However, the use of DES specifically for the QuEChERS method has not been extensively investigated. Thus, the feasibility of using DES in the QuEChERS method has not been well elucidated. To fill this gap, this paper was prepared to provide an overview of the effectiveness of DES in the QuEChERS method. DES is known to act as an extractant and sorbent modifier. Recent advances in the use of DES based on the QuEChERS method, particularly in the extraction of pesticides, are discussed and future research needs related to its use on more complex matrices - such as biological matrices - are identified. In general, it is concluded that the use of DES based on the QuEChERS method is superior to extraction using synthetic organic solvents and fulfills the criteria of green chemistry.

Abstract

Pycnarrhena cauliflora Diels. known as Sengkubak, is a natural-based potential antioxidant and antibacterial from the Menispermaceae family. Streptococcus sanguinis is a primary colonizer bacteria that is consistent in biofilm formation. This study attempted to identify S. sanguinis inhibition from antibacterial and antioxidant bioactivity of sengkubak leaf extract. Sengkubak leaves samples were extracted with 70% ethanol through 72 hours (3x24) maceration. Phytochemicals were identified qualitatively and quantitatively through analysis of total quercetin and phenolic content. Antioxidants were tested using the DPPH method. S. sanguinis sensitivity was tested through liquid microdilution method. The extract of sengkubak leaves has a yield of 16.30% (w/w). Qualitative screening showed positive results for flavonoids, phenols, alkaloids, saponins, terpenoids, tannins, and steroids. TFC (quercetin) in extract was 5.22±0.969 QE/g and TPC was 4.08±1.005 mg GAE/g. The IC50 antioxidant value of extract was 40.136 µg/mL. The minimum inhibitory concentration value is shown at 5×10⁴ µg/mL. This research study proves that ethanol extract of sengkubak leaves holds various phytochemical compounds which are likely utilized as potent antioxidants and have inhibitory effect against S.sanguinis.

Abstract

The use of e-cigarettes has surged as an alternative to conventional smoking, with e-liquids containing various flavoring chemicals that enhance user experience. However, these flavoring compounds may pose significant health risks when inhaled. This study systematically reviews the chemical composition of e-liquid flavorings identified using Gas Chromatography-Mass Spectrometry (GC-MS). A comprehensive literature review was conducted, sourcing articles from PubMed, Google Scholar, and ScienceDirect published within the last decade. The review focuses on identifying chemical compounds in various e-liquid flavors such as vanilla, bubble gum, coffee, tobacco, strawberry, lemon, and cannabis. Key compounds identified include vanillin, ethyl maltol, cinnamaldehyde, benzaldehyde, and limonene, which contribute to flavor profiles but may also have adverse respiratory effects. Thermal degradation of these compounds can result in the formation of harmful carbonyl compounds like formaldehyde and acetaldehyde, both classified as carcinogens. Furthermore, nicotine, a major component in e-liquids, increases the risk of addiction and psychological disorders. This review underscores the need for strict regulations on e-liquid flavoring agents and further research on the long-term effects of inhaling these substances. It also highlights the importance of public awareness regarding the potential respiratory toxicity associated with e-liquids and vaping.
