Abstract

Millet is a cereal with high nutritional content and has the potential to be a substitute for wheat. This research aimed to determine the characteristics of millet flour fermented by Bimo CF at different fermentation times.  This research was conducted using a completely randomized design with one factor (fermentation time). It consisted of P1 (0 hours), P2 (12 hours), P3 (24 hours), P4 (36 hours), and P5 (48 hours). The resulting product of each treatment's water content, ash content, crude protein content, lightness, and yield were analyzed as tested parameters. Observation data were analyzed using ANOVA and further tested using Honest Significant Difference Test at a significance level of 5%. The result showed that fermentation period treatment using Bimo CF starter had a significantly different effect on water, ash, crude protein content, lightness, and yield. The 12-hour fermentation treatment is the recommended treatment to produce fermented millet flour, with a water content of 2.78%, ash content of 2.41%, crude protein content of 7.41%, lightness of 73.11, and yield of 45.5%.

Abstract

Histamine poisoning is a significant food safety concern associated with fish and fishery products. Enterobacter aerogenes is one of the histamine-producing bacteria capable of converting L-histidine into histamine through the histidine decarboxylase (HDC) enzyme. The accumulation of histamine in fish and fishery products can lead to allergic reactions and foodborne illnesses, highlighting the need for rapid detection methods. This study aimed to design and optimize primers targeting the HDC gene in E. aerogenes and validate their specificity using in silico PCR. The HDC gene sequence was retrieved from the NCBI GenBank database, and primers were designed using Primer3Plus software. The in silico PCR analysis was performed to assess the specificity of the primers against the genome of E. aerogenes and other histamine-producing bacteria species. The optimized Primer Pair 1 successfully amplified a 230 bp target region, showing high specificity for E. aerogenes in product size and annealing position with no cross-reactivity to other histamine-producing bacteria species. These findings demonstrate the potential of in silico PCR as a rapid and cost-effective screening tool for detecting histamine-producing bacteria in fish and fishery products. However, further in vitro validation is required to confirm the applicability of these primers in real-world food safety and allergy risk assessments.

Abstract

Fermented shrimp paste (terasi) is a traditional food product produced by micro, small, and medium enterprises (MSMEs) in Lombok Island, Indonesia. Due to its fermentation and handling processes, terasi is susceptible to contamination by foodborne pathogens. The Indonesian Food and Drug Authority has identified specific bacterial pathogens that pose a risk of contamination in shrimp paste, necessitating accurate and efficient detection methods. Molecular detection using polymerase chain reaction (PCR) is known for its high specificity and sensitivity, as well as its rapid processing time and cost-effectiveness. However, conventional PCR can only detect a single target pathogen in one reaction, limiting its efficiency in screening multiple contaminants simultaneously. This study aims to design and optimize Multiplex PCR primers for detecting multiple foodborne pathogens in terasi through an in-silico approach, followed by In Silico PCR verification to assess primer specificity and efficiency. Target pathogens include Salmonella spp., Staphylococcus aureus and Vibrio cholerae. DNA target sequences were retrieved from the National Center for Biotechnology Information (NCBI) database, and primer design was conducted using Primer3Plus software. The designed primer sets were validated using Primer-BLAST (NCBI), and in-silico PCR was performed to verify amplification efficiency and specificity. The designed primer set has been successfully obtained and evaluated using In Silico PCR and Primer-BLAST (NCBI), confirming its specificity to the target genes. These results demonstrate that the primers can be effectively used as a molecular detection tool for foodborne pathogens. The optimized conditions and validated primer set are expected to contribute to molecular-based food safety detection methods, particularly for Indonesian fermented food products.

Abstract

Roti tawar secara umum berbahan dasar tepung terigu yang kaya akan gluten. Gluten dapat menjadi penyebab penyakit celiac disease dan autoimun, sehingga diperlukan substitusi tepung terigu dalam pembuatan roti tawar. Substitusi terigu dapat dibantu dengan penggunaan bread improver seperti karagenan. Penelitian ini bertujuan untuk mengetahui pengaruh proporsi tepung komposit (terigu, sorgum dan maizena) dan konsentrasi karagenan terhadap mutu kimia, fisik dan organoleptik roti tawar rendah gluten. Metode penelitian menggunakan Rancangan Acak Lengkap dua faktor, yaitu proporsi tepung komposit terigu:sorgum:maizena (50:25:25, 40:30:30, 30:35:35) dan konsentrasi karagenan (0,2%, 0,4%). Hasil penelitian menunjukkan bahwa perlakuan proporsi tepung terigu:sorgum:maizena berpengaruh signifikan terhadap kadar abu, kadar serat kasar, daya kembang, elastisitas, aroma, rasa, warna crust, dan tekstur roti, sedangkan konsentrasi karagenan berpengaruh signifikan terhadap kadar serat kasar, warna crust, warna crumb, dan tektstur roti. Interaksi kedua faktor juga berpengaruh signfikan terhadap warna crumb dan tekstur roti tawar yang dihasilkan. Perlakuan proporsi tepung 30:35:35 dengan konsentrasi karagenan 0,2% adalah perlakuan yang paling disarankan untuk mengurangi penggunaan tepung terigu karena menghasilkan mutu organoleptik yang paling disukai panelis, dengan kadar air 25,21%, kadar abu 2,12%, kadar serat 7,14%, daya kembang 45,16%, elastisitas 64,95%, pori-pori yang kecil dan seragam 10,70-14,19 µm, dan waktu staling 48 jam, dengan karakteristik agak beraroma asam, tidak berasa asam, warna crust coklat muda, warna crumb putih kekuningan, dan tekstur agak lembut.

Abstract

Pengembangan ternak kambing lokal menghadapi tantangan dalam penyediaan pakan hijauan berkualitas secara berkelanjutan, terutama pada musim kemarau. Salah satu potensi lokal yang belum dimanfaatkan secara optimal adalah daun lamtoro (Leucaena leucocephala), yang diketahui memiliki kandungan protein dan TDN (Total Digestible Nutrients) yang tinggi. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memberikan pelatihan dan pendampingan kepada peternak kambing lokal di Desa Sambik Elen, Kecamatan Bayan, Kabupaten Lombok Utara, Provinsi Nusa Tenggara Barat mengenai teknologi fermentasi hijauan pakan ternak berbasis lamtoro sebagai alternatif pakan ternak yang bernutrisi dan ekonomis. Hasil pengamatan menunjukkan bahwa peternak dapat memahami dan mempraktikkan proses fermentasi hijauan pakan ternak, yang mencakup persiapan bahan, penambahan dedak dan starter, pencampuran, dan penyimpanan secara anaerob. Kesimpulannya, kegiatan pengabdian ini telah berhasil meningkatkan pengetahuan dan keterampilan peternak dalam pengolahan hijauan pakan ternak dengan teknologi fermentasi. Diharapkan kegiatan ini dapat berkontribusi pada keberlanjutan usaha peternakan kambing dan peningkatan perekonomian masyarakat peternak di daerah pedesaan.

Abstract

Pemasaran produk-produk yang dihasilkan masyarakat wirausaha Dusun Rangsot Timur, Kabupaten Lombok Utara belum mampu menembus pasar yang lebih luas karena kemasan dan label yang belum sesuai standar serta belum dimilikinya SPPIRT. Program pengabdian kepada masyarakat ini bertujuan untuk melakukan edukasi standar kemasan, label, dan PIRT kepada masyarakat wirausaha produk pangan di Dusun Rangsot Timur. Program dijalankan menjadi 3 (tiga) tahapan, meliputi studi pendahuluan, kegiatan edukasi, dan evaluasi. Metode pendekatan yang ditempuh adalah metode partisipatif. Instrumen evaluasi yang digunakan adalah angket pre-test dan post-test menggunakan Skala Likert (skor 1-4). Ketercapaian program pengabdian kepada masyarakat ditandai dengan terlaksananya keseluruhan tahapan program dan adanya peningkatan pengetahuan peserta program terhadap materi edukasi. Terdapat peningkatan pengetahuan peserta program tentang standar kemasan, label, dan PIRT dari kriteria sedang (68,95%) menjadi tinggi (80,08%) melalui kegiatan edukasi yang telah dilaksanakan. Terlaksananya program ini perlu diikuti kegiatan tindak lanjut oleh peserta bersama dengan pihak terkait untuk memperbaiki kemasan dan mengusahakan kepemilikan SPPIRT

Abstract

Stunting remains an unresolved issue in North Lombok, NTB, Indonesia. One of the government's efforts to reduce stunting rates involves ensuring nutritional fulfillment for mothers and expectant mothers, which can be obtained from local food sources. Nira (palm sap) and Trigona honey are local commodities available in Tanjung, North Lombok. Unfortunately, these commodities have not been optimally utilized for nutritional fulfillment or as income sources. Typically, Nira is only sold as fresh sap, fermented beverage (tuak), or palm sugar, which have relatively low market values. Combining Nira and Trigona honey to create high-value candy products represents an opportunity. This community service project aimed to improve welfare and nutrition in Sigar Penjalin Village by assisting in the production of hard candy from Trigona honey and Nira Aren as a potential local delicacy to combat stunting and enhance local income. The implementation involved 16 community members organized by UMKM Rangsot Kreatif. The training used ingredients like brown sugar, granulated sugar, glucose syrup, Trigona honey, and water. The training stages included preparation, material delivery, hands-on practice, and evaluation through pretests and posttests to measure participants' knowledge, skills, and interests. The pretest results showed a total score of 627 out of 960 (65.31%), indicating a moderate level of understanding. After the training, the posttest showed a significant improvement, with a total score of 829 out of 960 (86.35%), indicating a very high level of understanding. This activity highlights the importance of technological support and the utilization of local resources in developing nutritious and economical snack products, supporting improvements in community nutrition and economy, and providing alternative solutions in local product processing

Abstract

Penelitian konstruksi dan kloning gen CelE, yang merupakan penyandi enzim selulase dari Ruminococcus plavefaciens, muncul sebagai respons terhadap kebutuhan mendesak akan sumber daya enzim yang efisien dalam industri pangan dan agroindustri. Enzim selulase, dengan fokus pada CelE, memiliki peran vital dalam proses degradasi selulosa, sebuah komponen utama dalam dinding sel tanaman. Keberadaan Ruminococcus plavefaciens sebagai sumber gen CelE menarik perhatian karena mikroorganisme ini terdapat dalam sistem pencernaan hewan ruminansia dan memiliki potensi besar untuk menghasilkan enzim selulase yang efisien. Tujuan dari penelitian ini adalah untuk membuat dan mengkloning plasmid pET15b dengan gen CelE. Konfirmasi gen CelE pada DNA rekombinan dilakukan dengan metode identifikasi resistensi bakteri host pada media dengan antibiotik dan koloni PCR. Penelitian ini dilaksanakan dengan menerapkan metode sebagai berikut: Pelaksanaan Optimasi Kodon dan Konstruksi Plasmid Rekombinan (CelE), Persiapan Sel Kompeten, Transformasi Sel, Uji PCR Koloni Transforman Hasil penelitian menunjukkan bahwa penelitian ini telah berhasil mendapatkan bakteri transforman yang diharapkan dengan hasil Uji PCR Koloni Transforman mendapatkan product size yang sesuai yaitu 205 bp, sehingga dapat disimpulkan plasmid pET15b dengan gen CelE yang telah dioptimasi berhasil dikonstruksi dan dikloning.

Abstract

UMKM pangan lokal memainkan peranan penting dalam peningkatan perekonomian masyarakat setempat yang tentu saja bermuara pada peningkatan perekonomian nasional. Salah satu wilayah di Lombok, Provinsi Nusa Tenggara Barat yang memiliki UMKM pangan lokal adalah Dusun Rangsot Timur, Desa Sigar Penjalin, Kabupaten Lombok Utara. Target pasar yang lebih luas untuk peredaran produk dapat dicapai apabila produk pangan lokal tersebut sudah terjamin mutu dan keamanan pangan. Hal ini menjadi tantangan bagi pelaku UMKM pangan lokal sebab produk yang tidak terjamin mutu dan keamanannya akan berdampak pada kesehatan konsumen dan keberlangsungan usaha itu sendiri. Penjaminan mutu dan keamanan pangan dapat dicapai dengan menerapkan sanitasi yang baik selama proses produksi. Oleh karena itu, sangat penting untuk membentuk kesadaran dan pengetahuan mengenai praktik sanitasi yang baik di kalangan pelaku UMKM pangan lokal. Kegiatan pengabdian ini dilaksanakan dalam bentuk edukasi aspek dan implementasi sanitasi dalam industri pangan. Tahapan program pengabdian masyarakat dibagi menjadi 3 (tiga), yaitu persiapan, pelaksanaan, dan evaluasi program. Ketercapaian program pengabdian diukur dengan menggunakan Skala Likert dengan skor kuesioner 1-4. Hasil kuesioner para peserta dianalisis menggunakan metode deskriptif. Kegiatan pengabdian berhasil meningkatan pengetahuan peserta tentang sanitasi industri pangan yang signifikan sebesar 8,71%. Kegiatan pengabdian ini diharapkan dapat mendorong UMKM pangan di Dusun Rangsot Timur untuk mengimplementasikan praktik sanitasi yang efektif, meningkatkan kualitas dan daya saing produk.

Abstract

Prevention of the transmission of dengue hemorrhagic fever (DHF) is carried out by breaking the chain of dengue transmission and administering vaccines, but to date, this has not achieved the expected target. Dengue virus tests using RT-PCR require skills and relatively expensive equipment. Serological test of IgM and IgG often shows false negatives or false positives, especially in dengue-endemic areas. The antibody test against NS1 using the ELISA method has weaknesses because anti-dengue IgM is often not detected in secondary infections. The development of serodiagnostic tests for rapid, affordable, sensitive, and specific detection of dengue virus infection is very necessary. Recombinant multivalent envelope proteins domain III (ED-III) dengue virus is a biomarker that has the potential to be developed to detect all dengue virus serotypes. One of the proteins that has high antigenicity is glycoprotein E which is found in the envelope of the dengue virus and is the most antigenic part of the virus. This research aims to combine several parts of the antigenic protein found in all dengue virus serotypes as immunoserodiagnostic material. This research is an analytical survey research, that compares the results of the atypical lymphocyte test, RT-PCR, and ELISA using the multivalent ED-III antigen. The number of samples used was 26 samples obtained from patients who were diagnosed with dengue fever using an accidental sampling technique. The results of the atypical lymphocyte examination showed 14 positive samples, while the results of the RT-PCR and ELISA examinations were 23 and 24 positive respectively. The average Optical density (OD) of examination using the ELISA method was 1.902 with sensitivity and specificity levels of 92% and 96%. There is no difference result of the RT-PCR compared with the ELISA test. Therefore, recombinant multivalent envelope protein domain III (ED-III) dengue virus can be used as a diagnostic tool to detect dengue fever infection.
