Abstract

Seagrass ecosystems services in the coastal waters are included as primary producers, nutrient recycler, bottom stabilizers, sediment traps, and erosion barriers. Gleaning fisheries in seagrass bed in Mokupa waters could cause damage on seagrass, that can be marked by changes in seagrass cover. The purposes of this study were to identify the types of seagrasses and to determine the health condition of seagrass bed ecosystem in Mokupa waters. The research method used in this study is quadrat transect method. Data collection was carried out by laid three transects (100 m) with distance between each transect was 50 m. A square frame (50×50 cm2) which is divided into 4 squares is placed on the right side of the transect, with 10 m distance between frame. This study found four types of seagrasses, namely Enhalus acoroides, Thalassia hemprichii, Halodule pinifolia and Halophila ovalis. E. acoroides have the highest percentage cover (16.19%) followed byT. hemprichii (6.91%), H. pinifolia (4.50%) and H. ovalis (1.56%) respectively. Seagrass cover in the study area is considered medium (26-50%) while the health condition was poor (29.25%).

Abstract

One type of enzyme whose application is very widespread is protease enzymes. Microorganisms that have not been widely explored as a source of protease enzymes are bacteria that are in conjunction with Nudibranch. The study aims to identify the morphological characteristics of bacteria, then determine the hydrolysis ability of protease enzymes from bacteria symbiont Nudibranch. Identification of morphological characteristics is done macroscopic, and microscopic with gram staining. Proteolytic activity tests are performed qualitatively by measuring the proteolytic index of bacteria grown on Skim Milk Agar medium. The results of macroscopic identification of symbiotic bacteria with Nudibranch are dominated by irregular colony shapes, undulated edges, growth of the entire colony has a flat height, milky white colony color, buttery texture and opaque bacterial colonies. Based on microscopic identification, two gram-negative isolates are obtained, namely RL.S14, S1.1.1, and gram positive isolates, namely AK.S00, AK.S01, AK.S02, AK.S03, S.PP1, RL.S6, S2.1.5, S2.2.1, S3.3.1, S3.3.4, RL.S10. Nudibranch symbiont Bacterial has colony cells were dominated by the diplobacillus form. The results of the protease activity test of Nudibranch symbiont bacteria showed that 12 positive isolates had proteolytic activity with the highest proteolytic index produced by isolate AK.S00, which was 2.5 mm. Keywords: Nudibranch, symbiotic bacteria, characteristics, protease

Abstract

Padina australis is a species of marine algae from the Phaeophyta division (brown algae) which is generally distributed in marine waters. This algae is able to live in waters with high levels of heavy metals such as in the Kima Bajo waters. This study aims to isolate arsenic (As)-resistant bacteria in P. australis, observe cell shape and bacterial characteristics, and determine bacterial resistance to antibiotics. The method used is conventional bacterial isolation which has been tested on As2O3 compounds. The results obtained were that P. australis contained arsenic resistant bacteria at concentrations of 250 ppm, 500 ppm, and 1000 ppm. The bacteria obtained were Gram positive, in the form of bacilli. These arsenic-resistant bacteria are also resistant to antibiotics such as amoxicillin, cefixime, and doxycycline. Keywords: Padina australis, Arsen (As), Kima Bajo

Abstract

The long-term goal of this research is the management of vaname shrimp culture that is certified "Good Fish Cultivation" (GFC) based on environmentally friendly and sustainable technology so that its products can be accepted in domestic and foreign markets. The specific target is the implementation of phytoremediation technology in shrimp aquaculture at the Marine Field Station owned by FPIK UNSRAT which will be developed into the North Sulawesi Marine Education Center (NSMEC) area. The long-term goals and specific targets above will support the leading areas of maritime and maritime affairs in the Unsrat Research Master Plan. Analysis of water quality parameters and shrimp growth response using controlled-scale phytoremediation technology was carried out to see the remediation ability of Eucheuma denticulatum seaweed. Research method using phytoremediation technology application on white shrimp cultivation in the form of demonstration plots. The average growth in the plots provided with seaweed ranged from 8.3 g to 8.4 g. While the control that was not stocked with seaweed, the average weight growth was 6.6 g. This figure shows quite different values for different treatments. Water quality parameters such as DO, pH showed better water quality conditions in the plots spread with seaweed. This shows that Eucheuma denticulatum seaweed is capable of being a phytoremediator for the environment where the vannamei shrimp (Litopenaeus vannamei) is present. Seaweed Eucheuma denticulatum has the ability to absorb waste such as the faces of white shrimp (Litopenaeus vannamei). Keywords: phytoremediation, seaweed, vaname shrimp, East Likupang.

Abstract

Indonesia has abundant biological resources that produce many potential bioactive compounds that are useful for pharmaceuticals and other materials, one of which is antioxidants. Antioxidants are substances that can prevent or slow down cell damage caused by free radicals. This study aims to analyze how strong the antioxidant activity is in the body of asteroidea (Linckia laevigata) taken in Tongkaina waters, Bunaken District, Manado City. And this study found that the inhibition value of the ethanolic extract of Linckia laevigata had strong antioxidant activity where the extract increased and the concentration was 10 mg/L with an average value of 67,3 %, a concentration of 15 mg/L  of 67,8 %, a concentration of 20 mg/L of 68,8 % and the highest concentration of 25 mg/L getting an average value of 69 %. The increace in the percentage of inhibition and the ethanolic extract of Linckia laevigata, showed that the concentration of the extract contributed greatly to the ability of the extract to reduce free radicals. In this study, the DPPH test was carried out by observing the decrease in absorbance wavelength of 517 nm using UV-Vis spectrophotometer, where the decrease in absorbance value as a result and a decrease in color intensity. While vitamin C as a comparison has a greater yield and sample extract of Linckia laevigata which proves that the antioxidant content and the sample has a fairly stable result. Keywords: Bioaktif compounds, Linckia laevigata, Antioxidant, Vitamin C

Abstract

The presence of coral reef fishes, especially family Chaetodontidae is one of bioindicators to environment health condition of coral reef. Fish indicator research in Bunaken Island waters aims to find out the community structure, such as the composition, abundance, density, equality, diversity and dominance. Data collection was conducted at 5 (five) locations in the Bunaken Island Traditional Zone where there are 5 (five) observation points respectively, namely TP-1 Mandolin, TP-2 Alung Banua, TP-3 Sachiko, TP-4 Muka Kampung, and TP-5 Bunaken Timur. Data collection by underwater visual census (UVC) techniques using SCUBA. The results showed there were 23 species of indicator fish from family Chaetodontidae, total individuals number of each species in 5 research sites as many as 97 individuals. The highest fish density is 0.06 at TP-5 while the lowest value in TP-2 with density value 0.02. Through indicator fish community structure indicates that the coral reef quality conditions is stable indicated by diversity values (H') in all locations were moderate category and inversely proportional by low dominance values, high leveling values (E). The similarity rate (IS) of indicator fish species in all locations differs from each other indicating the condition of coral reefs in each location grows independently without being influenced by other locations. Keywords: Reef Fish, Coral Reef, Indicator Fish.
