Abstract

Background: Poor blood pressure control of hypertensive patients is caused partially by bad lifestyle management, which increases the risk of a cardiovascular event by stimulating the defective endothelial function. Objective: This study aimed to investigate the effect of night sleep duration on systolic blood pressure and Nitric Oxide concentration in hypertensive patients. Methods: 60 hypertensive patients age 40 years based on the medical record participated in this study. The sleep duration and sleep quality were performed using the PSQI questionnaire. The 40 participants thus selected randomly for the NO concentration assay using NO Salivary Elisa Kit ab65238. Results: 53 participants experienced night sleep duration ≤ 6 hours, and 44 (73.7%) participants had poor sleep quality. Night sleep duration was associated significantly with hypertensive patients' systolic blood pressure (p = 0,000 0.05; Odds ratio=0.008). Conclusion: The longer duration of sleep at night among hypertension patients, the lower systolic pressure were found. There was no correlation between night sleep duration on NO concentration (p 0.05). These findings provide evidence that night sleep duration is associated strongly with the increased systolic blood pressure but not directly caused by the alteration of endothelial function. Keywords: sleep duration. systolic blood pressure, hypertension, endothelial function, Nitric Oxide

Abstract

Hyperglycemic crisis is a metabolic emergency that causes morbidity or death. Diabetic Ketoacidosis (DKA), and Hyperosmolar Hyperglycemic State (HHS), are 2 forms of hyperglycemic crisis that often occur in patients with uncontrolled diabetes mellitus. A 45-year-old female patient came to the hospital emergency department with decreased consciousness since 12 hours and shortness of breath since 2 days. The patient has a history of diabetes mellitus. On physical examination, E2V2M4 consciousness, blood pressure 140/90, oxygen saturation 40% with a reservoir bag oxygen mask 15 liters per minute, coarse rhonchi and chest retractions. From the results of investigations, there was an increase in blood glucose (523 mg/dL), a chest X-ray of pulmonary edema and bilateral bronchopneumonia. The patient was diagnosed with a hyperglycemic crisis with unstable hemodynamics. However, before being transferred to the intensive care unit, the patient was declared dead. Hyperglycemic crisis is a metabolic emergency whose management must be fast and appropriate in order to maintain hemodynamic stability and prevent death Keyword :Diabetes, Diabetic ketoacidosis, Hyperglycemic crisis, Hyperosmolar hyperglycemic state

Abstract

Cancer is a disease that appears characterized by abnormal cell growth which can then infect adjoining parts of the body and or spread to other organs. One type of cancer that often occurs is Oral Squamous Cell Carcinoma (OSCC). There are several target receptors involved in cell proliferation, which are COX-2 and CDK-4/6 receptors. One of the herbal treatments for OSCC can be done using Chrsanthemum cinerariifolium (Trev.) herb extract. This research aims to find active compounds in Chrysanthemum cinerariifolium (Trev.) using UPLC-QToF-MS/MS method, the analysis of physiochemical prediction, toxicity analysis and molecular docking results on the proteins of COX-2 and CDK-4/6. This research combines two types of research, which are descriptive experimental for metabolite analysis and preexperimental one shot case study type for in silico test. The analysis of Metabolite compounds is using UPLC-QToFMS/MS Instrumentation, and for in silico tests using some SwissADME for physiochemical tests, PkCSM and Protox Online Tools for toxicity tests and Biovia Discovery Studio Visualizer and PyRx 0.8 for molecular docking tests. The results of this study obtained 28 compounds from metabolite profiling. And the results of molecular docking showed 1 compound with the best affinity on the tethering of both receptors. The compound is Diosmetin. There are also amino acid with 15 residues obtained in this process. However, the bonding value of native ligand with target protein is higher than the bonding of test ligand with target protein

Abstract

Background:   Forensic examination of semen in cases of sexual intercourse (e.g., sexual assault) aims to detect the presence of spermatozoa or male-specific biomarkers in vaginal swab samples from victims. Conventional methods, including microscopic examination with staining and chemical tests (acid phosphatase and prostate-specific antigen/PSA), are routinely used but have limitations in sensitivity and specificity. In the past five years, advancements have emerged in semen detection technology, including Loop-mediated Isothermal Amplification (LAMP), Y-STR (Y chromosome short tandem repeat) analysis, sperm-specific aptamers, and Convolutional Neural Networks (CNN) for automated identification. Objective: This review article aims to summarize current literature (2019–2024) regarding spermatozoa detection methods in vaginal swab samples, covering both conventional and innovative techniques. Methods: Literature was searched through international and national databases using relevant keywords including “forensic sperm detection,” “vaginal swab semen,” “LAMP for sperm,” “Y-STR sexual assault,” “aptamer sperm detection,” and “CNN sperm microscope” within the publication range of 2019–2024. Results: Recent studies confirm that microscopy (histological staining) and chemical testing (acid phosphatase and PSA) remain the initial standard for semen examination. However, both are presumptive and susceptible to false-positive or false-negative results. LAMP and Y-screen methods demonstrated high sensitivity in detecting male-specific DNA, comparable to Y-STR profiling. Fluorescent DNA aptamers selectively bind to sperm cells, enhancing detection speed and accuracy. The application of CNN to colored microscopic images has enabled automated “sperm hunting” with accuracy exceeding 90%, significantly expediting laboratory analysis. Conclusion:  Conventional methods such as microscopy and chemical assays remain essential in forensic semen analysis, but show limited reliability in low-sperm samples or cases involving azoospermic perpetrators. Emerging techniques based on DNA, biochemical markers, and artificial intelligence offer improvements in sensitivity, specificity, and detection efficiency. Integration of multi-method approaches is recommended to optimize outcomes. Further standardization and research are needed to facilitate widespread implementation of these novel technologies in forensic laboratories.
