Abstract

Lactococcus garvieae is a gram-positive ovoid cocci bacterium formerly classified as a member of the Lactococcus genus. This study aims to isolate L. garvieae from catfish rearing pond and characterize it as a potential probiotic candidate. L. garvieae was identified and characterized through phenotypic and genotypic observation, genomic % G~C content analysis, cell surface hydrophobicity assays, acidification test, in vitro antagonism, and a profile of antimicrobial activities. The MT597595.1 accession number corresponds to L. garvieae, as determined by a molecular identification test. Biochemical characterization was performed using API 50 CH kit. The genomics %G~C content of L. garvieae was 51.8. Findings from acidification ability tests, in vitro antagonism tests, and the ability of bacteria to grow in broth medium at pH 4 reveal that L. garvieae can inhibit the growth of Aeromonas hydrophila, Streptococcus agalactiae, Streptococcus iniae, and Edwardsiella ictaluri. However, it does not suppress the growth of L. garvieae  Edwardsiella tarda. Remarkably, L. garvieae has the ability to reduce the pH of neutral broth medium turning it acidic. Furthermore, L. garvieae’s hydrophobic cell surface exhibited an adhesive, hydrophobic, and protein surface cell content with a compact growth pattern consistent with postive SAT and MATH assay. Antimicrobial activity tests, encompassing 11 antibiotics, disclosed resistance to  Nalidixic acid while displaying intermediate sensitivity to Streptomycin and Trimethoprim. In conclusion, L. garvieae demonstrates an inhibitory effect on the growth of pathogenic bacteria, underlining its potential as a probiotic candidate.

Abstract

Tinfoil barb, Barbonymus schwanenfeldii (Bleeker, 1854), is a potential freshwater fish for Indonesian aquaculture. Before widely used, the candidate needs to be evaluated. This research aimed to evaluate the reproductive performance of the first generation (G1) and larval performance of the second generation (G2) of domesticated tinfoil barb. A total of 68 female and 24 male broodstocks were selected for the artificial breeding program. The gonadal maturity test was carried out by canulating the eggs every month. Before spawning, GnRHa hormone was injected into the dorsal area (0.5 mL/kg for female and 0.2 mL/kg for male). Fecundity, fertility rate, hatching rate, embryogenesis, and larvae ontogeny were recorded. The results showed that the first matured G1 males of tinfoil barb were at the standard length of 16.01 ± 1.18 cm, while females at 15.79 ± 1.23 cm. The mature broodstock indicated by the gonad maturity stage III and IV confirming higher estradiol concentration (above 400 pg mL-1). The fecundity of two mature broodstock-sized of 217.2 g and 197.3 g were 12,495 and 15,782 eggs, respectively. The spawning season of G1 tinfoil barb was in October and November (rainy seasons). The fertilized eggs latency time was 10 hours 44 minutes at 25°C and hatched after 23 hours 7 minutes. The fertility rate was 96.96 % and the hatching rate was 95.16 %. The survival rate of G2 normal larvae was 100 % at three days of the rearing period. The G2 larvae production in this experiment provides an excellent opportunity for fish diversification both for aquaculture and restocking.

Abstract

Catfish Clarias sp. is one of the leading commodities in demand and has economic value. Low survival due to cannibalism and disease is a serious problem. To overcome these problems is application of bioflocs and probiotics Bacillus NP5. This study aimed to analyze the appropriate dose of Bacillus NP5 probiotic addition to the biofloc system to improve the health status, growth performance, water quality, and resistance to A. hydrophila. The catfish fry (Clarias sp.) with an average weight of 1.79±0.05 g was used in the experiment for 40 days, reared in tanks with the dimension of 60×30×35 cm3. The experiment applied a completely randomized design (CRD) of five treatments with three replicates, namely KN (negative control), KP (positive control), BFT (biofloc application without probiotic bacteria), BFT4 (biofloc application with Bacillus NP5 104 CFU mL-1), BFT6 (biofloc application with Bacillus NP5 106 CFU mL-1). Treatments tested were challenged with A. hydrophila density 104 CFU mL-1 by immersion, except the negative control. After 40 days of rearing, biofloc and Bacillus NP5 treatments had a significant effect (P < 0.05) on growth performance, immune response, water quality, total bacteria in water and the gut compared to the control treatment (P < 0.05). In addition, total A. hydrophila in liver, kidney and water were lower (P < 0.05) in BFT4 and BFT6 treatments than the control. The conclusion of this study is that the bioflocs supplemented with Bacillus NP5 improved the growth performance, immune response and resistance of catfish to A. hydrophila infection.

Abstract

This study aimed to investigate the suitability of black soldier fly larvae (BSFL) combined with a commercial diet (CMD) on growth and feed performances of hybrid catfish (Clarias gariepinus x C. macrocephalus). A completedly randomized design was used to consist of four treatments and three replications with 100:0 (control), 75:25, 50:50, and 25:75 of CMD and fresh BSFL blended feeding. Fish were reared in 2 m2 inland canvas cages for 60 days. The results indicated that fish were fed 100% CMD had better average weight gain (AWG), daily weight gain (DWG), specific growth rate (SGR), food conversion ratio (FCR), feed efficiency (FE), and survival rate (SR) than the other treatments, but there was no statistically significant difference (p>0.05) when compared to 75:25 and 50:50 CMD:BSFL feeding. At the end of the experiment, the SR of fish was 81-91% and showed no significant difference (p>0.05) when compared between treatments. Therefore, hybrid catfish rearing can compensate CMD by fresh BSFL up to 50% of the feed amount per day. However, feeding with fresh BSFL can have side effects concerning steatosis

Abstract

Conventional microalgae culture is challenged by issues of light limitation and cell self-shading. This study aims to evaluate the impact of different cultivation modes on the growth and lipid content of Thalassiosira sp. The diatom, Thalassiosira sp., was grown in autotrophic, mixotrophic-suspended, and mixotrophic-biofilm conditions until the stationary phase was reached. After four (4) days of culture, analysis of the cell densities revealed a significant difference between groups, with cell densities of 7.3×105 cells mL-1 for control, 1.1×106 cells mL-1 for mixotrophic-suspended, and 1.9×106 cells mL-1 for mixotrophic-biofilm cultures. Both treatments are significantly higher than the control. However, mixotrophic-biofilm culture achieved the highest cell density among all cultivation modes, 161.81% higher than the control. The specific growth rate (SGR) of Thalassiosira sp. in mixotrophic-biofilm culture was highest among treatments, while the doubling time was significantly highest in the control. Moreover, mixotrophic-biofilm culture attained the highest biomass at 56 mg 100 mL-1. Thalassiosira sp. cultured under mixotrophic-biofilm also recorded the highest lipid content at 9.89%. It is both significantly higher than the control (3.06%) and the mixotrophic-suspended culture (6.15%). The cell density, algal biomass, and lipid content of Thalassiosira sp. under mixotrophic-biofilm culture highlight this culture strategy’s promising potential in improving microalgae growth and lipid content, ridding of light as an indispensable growth factor.

Abstract

Gram-positive spore bacteria are widely used as probiotics in general sectors. However, there are still limited bacterial isolates as probiotic candidates available from indigenous isolates, especially in aquaculture. This study aimed to obtain potential spore-forming isolates as probiotic candidate for tilapia. Tilapia fish samples were collected from Sukabumi, Ciamis, Serang, and Papua. Bacterial isolates were isolated from the digestive tract of tilapia. Bacteria were identified based on their morphological, molecular characteristics, complete genome composition, and cell surface identification based on hydrophobic properties. In this study, six bacteria were isolated and identified by molecular characteristics using 16S rRNA sequences. Based on the phylogenetic analysis, the 9 PP isolate was Priestia megaterium basonym: Bacillus megaterium, CMS 16N isolate was Brevibacillus halotolerans, PPN 10 isolate was Bacillus sp., 3.1 SKBM isolate was Bacillus mycoides, CMS 22 N and SRG32 isolate were Bacillus subtilis. Six bacteria had different phenotypicals, ATGC sequence compositions, and a higher proportion of total G~C sequence composition above 50%. The coherent cell surface hydrophobicity test was positive on the SAT, SA, AA, and compact growth patterns in soft-agar media for 9 PP, CMS 22 N, and SRG32 isolates. From our study, the indigenous spore-forming bacteria isolated from tilapia stomachs are enzymatic bacteria, which have a strong attachment to host tissue and high potential as a probiotic candidate for fish. Various hydrophobicity test results from each isolate indicate that the protein composition in the cell surface is different.

Abstract

Cultivation waste poses a severe threat to reducing environmental quality. However, the problem of low salinity cultivation waste can be overcome by converting it into plants. This research was conducted to assess the impact of salinity on the growth performance of Vannamei shrimp (Litopenaeus vannamei) and water spinach (Ipomoea aquatic Forsk) in a low-salinity aquaponic system. Floating raft system for planting water spinach. Furthermore, the role of water spinach in using N and P nutrients from the culture media of vannamei shrimp was analyzed. The experiment was carried out for 35 days, with two treatments and three repetitions, namely 5 and 10 ppt. Salinity affected the growth performance of shrimp and water spinach. Shrimp reared at 10 ppt exhibited higher growth rate, harvest size, and shrimp yield, along with lower feed conversion ratio (FCR) and water use compared to those reared at 5 ppt. Meanwhile, the performance of water spinach at 5 ppt salinity resulted in higher survival, plant height gain, root length gain, number of leaves, and yield of water spinach compared to 10 ppt salinity. The growth performance of water spinach decreased with increasing salinity, and the efficiency in reducing N and P from the 5 ppt culture media was 1.3 times higher than that of 10 ppt. Water spinach showed better results in the 5 ppt salinity media than 10 ppt, while vannamei shrimp grew at 5 ppt. Therefore, 5 ppt salinity was recommended as a suitable condition for integrated cultivation of vannamei shrimp and water spinach in low-salinity aquaponics. Both species were compatible and complemented each other’s role in developing low-salinity aquaponics.

Abstract

Sand crab (Emerita sp.) is a marine biodiversity, but it has not been used as a fish feed ingredient. This study aimed to evaluate the nutritional value of sand crabs and to understand its effect when used as feed ingredient on the performance of Koi carp. The study was conducted in two steps, which is evaluation of the nutritional value of sand crab and its effect on fish performance. The proximate composition, amino acids, and fatty acids were measured using AOAC methods, and then the carotenoid content was determined by spectrophotometry. In the second step, the sand crab was added to fish feed at doses of 0, 5, 10, and 15 percent, and fed to koi fish for 42 days. The parameters observed were length gain (LG), weight gain (WG), and feed efficiency (FE). Nutritional data were analyzed by description and compared with fish feed ingredients from previous studies. Fish performance were analyzed by one-way ANOVA. When significant, Tukey’s significant mean test was applied. The result showed that the nutritional value of sand crab was comparable to other feed ingredients with a protein content of 37.88%, while carotenoid content was superior. The best performance of Koi carp was obtained with a dose of 15% sand crab in the diet, with LG, WG, and FE values of 0.93 ± 0.05 cm, 0.48 ± 0.06 g, and 63.50 ± 7.05 %, respectively. Based on this result, it can be concluded that sand crab has a high nutritional value and can be used up to 15% in Koi carp diet.

Abstract

Vibrio parahaemolyticus is one of the pathogens in crustaceans that can cause mass death in vannamei shrimp farming. This study aimed to evaluate the effect of administering the commercial herbal supplement Phycurma Aquatic (PA) through feeding at different doses to prevent V. parahaemolyticus infection in vannamei shrimp. This study used a completely randomized design (CRD) consisting of five treatments and three replications, which are negative control group, positive control group, and PA at doses of 2.5, 5.0, and 7.5 mL kg-1 feed. Shrimps were reared for 30 days and fed five times a day. On the 31th day, shrimps were challenged with V. parahaemolyticus at a dose of 105 CFU mL-1, except for the negative control group. The results showed that the administration of 5.0 mL kg-1 of PA in the feed gave the best growth performance (P<0.05) compared to other treatments. The administration of 5.0 mL kg-1 PA in feed also enhanced shrimp health status and significantly increased the total hemocyte count, phagocytic activity, respiratory burst, and phenoloxidase. Furthermore, the administration of PA also increased antioxidant activity, reduced malondialdehyde levels, decreased V. parahaemolyticus population in the intestine, and reduced hepatopancreas tissue damage. Moreover, the survival rate of vannamei shrimp before and after the challenge test in the treatment group with a dose of 5.0 mL kg-1 of PA was significantly higher (P<0.05) compared to other treatments.

Abstract

Javaen barb is a native fish in Indonesian inland water with economic value and the potential to be developed as cultured fish resources and ornamental commodities. In the development of aquaculture, there are still problems, such as the low adaptability of the larvae, so that their survival is also low. This study was conducted to evaluate the effect of melatonin supplementation on the productivity of Javaen barb fish juveniles in culture containers. This study was conducted experimentally using a completely randomized design consisting of four treatments with different doses of melatonin supplementation, i.e., A) control (without melatonin supplementation), B) 0.2 mg/100 g of feed, C) 0.4 mg/100 g feed, and D) 0.6 mg/100 g feed, each treatment was repeated three times. Javaen barb juveniles were kept in an aquarium measuring 30×36×60 cm with a water level of 40 cm, consisting of 12 units. Each aquarium was stocked with 150 individuals and given an aeration system with the same air pressure intensity. Feeding was carried out ad-libitum with a frequency of three times daily for 180 days of rearing. The results showed that melatonin supplementation of as much as 0.6 mg/100 g of feed was able to increase the growth of Javaen barb fish juvenile with the highest survival rate (81.33 ± 0.54 %) and feed conversion ratio (2.61 ± 0.14). Melatonin supplementation of 0.6 mg/100 g of feed had higher total leukocyte (3.41±0.73×104 cells/mm3) and hemoglobin (5.07±0.12 g%) values and provided the best production performance in Javaen barb juveniles.
