Abstract

Breast cancer, with its aggresive subtypes and resistance, is a major cause of cancer-related mortality among women. Advancements of breast cancer drug discovery is great and survival rate is better than in the past, but still advanced breast cancer remain difficult to treat, emphasizing the need for searching of alternative therapeutic agents with proven efficacy and safety. Curcuma mangga contains diterpenoid compounds that have demonstrated cytotoxic potential against breast cancer cells. This study aimed to elucidate the molecular mechanisms and pharmacokinetic properties of diterpenoids from the n-hexane extract of C. mangga against ER-positive breast cancer using a network pharmacology approach. Four diterpenoids—(E)-Labda-8(17),12-diene-15,16-dial, (E)-15,16-Dinorlabda-8(17),11-dien-13-one, m-camphorene, and p-camphorene—were evaluated. Breast cancer-related genes were retrieved from the NCBI database, while compound targets were predicted using SwissTargetPrediction and SEA Search Server. Nineteen overlapping genes were identified as Potential Therapeutic Target Genes (PTTGs) and further analyzed through protein–protein interaction, Gene Ontology, and KEGG pathway enrichment analyses. Key targets included EGFR, AKT1, CYP19A1, and PGR, which are involved in cell proliferation, survival, and hormone-regulated signaling pathways. ADME and toxicity predictions indicated favorable oral bioavailability for labdane diterpenoids, although potential hepatotoxicity and mutagenicity were observed in certain compounds. Overall, (E)-Labda-8(17),12-diene-15,16-dial emerged as the most promising candidate, supporting its further investigation as a potential therapeutic agent for ER-positive breast cancer.

Abstract

The principal issue frequently encountered in society is confusion regarding drug stability, especially beyond use date. Many people are unfamiliar with beyond use date and expiration date. This condition often leads to improper medication storage. The aim of this study was to analyze the relationship between a patient's knowledge about beyond use date and their behavior in storing and using medication. A cross-sectional study was conducted on 80 respondents at Sewon 1 Primary Health Center, Yogyakarta, from January to February 2025. Using a validated questionnaire and purposive sampling, data was analyzed with the chi-square test. The results of the univariate analysis showed that patients' knowledge about beyond use date was mostly in the high category, which included 42 respondents (52.5%). Accordingly, patients's medication storage behavior was also mostly classified as good, accounting for 48 respondents (60.0%). Based on descriptive statistics, the mean value for the knowledge was 21.0 ± 1.701, while the mean value for the behavior variable was 41.38 ± 3.605. The conclusion of this study showed that there was a significant relationship between patients' knowledge about beyond use date of non-sterile compounded drugs and their medication storage behavior, with a significance value of 0.015 (p<0.05).

Abstract

World Health Organization (WHO) menyatakan bahwa tuberkulosis (TB) menempati peringkat kedua penyebab kematian akibat penyakit menular di dunia. Obat Anti Tuberkulosis (OAT) merupakan terapi utama, namun efek sampingnya dapat memengaruhi kepatuhan pasien. Penelitian ini bertujuan untuk menganalisis efek samping OAT pada pasien TB paru rawat jalan serta hubungannya dengan tingkat kepatuhan minum obat pada fase intensif dan fase lanjutan. Metode penelitian yang digunakan adalah observasional deskriptif dengan rancangan time series di Puskesmas Payung Sekaki Kota Pekanbaru. Data diperoleh melalui kuesioner efek samping dan lembar evaluasi kepatuhan. Hasil penelitian menunjukkan pada fase intensif, efek samping tersering adalah mual/muntah (rifampisin 64,29%, isoniazid 57,14%, pirazinamid 50%, etambutol 57,14%). Pada fase lanjutan, efek samping dominan adalah urine seperti teh (rifampisin 52,63%). Tingkat kepatuhan pasien fase intensif sebesar 92,86% dan meningkat menjadi 100% pada fase lanjutan. Kesimpulannya, efek samping OAT bervariasi pada setiap fase, dengan tingkat kepatuhan tinggi menjadi faktor penting keberhasilan terapi.

Abstract

Acne is a common skin issue among teenagers, primarily caused by bacterial infections from Propionibacterium acnes and Staphylococcus epidermidis. Pineapple peel has been found to contain antibacterial properties that can help treat acne. This study aims to evaluate the antibacterial effectiveness of an anti-acne patch made from pineapple peel extract (Ananas comosus (L.) Merr.) against these two bacteria. The pineapple peel extract was obtained through a reflux extraction method, followed by phytochemical screening, which identified the presence of flavonoids, saponins, tannins, and triterpenoids. The extract was then formulated into patches with concentrations of 5%, 10%, and 15% and tested for antibacterial activity. Physical evaluations showed that the patches had an appropriate pH (4.5-5.5), uniform weight (CV < 5%), acceptable thickness (0.1-0.3 mm), moisture content below 10%, and did not cause irritation in volunteers. Antibacterial testing using the agar diffusion method showed the highest inhibition zones for P. acnes at 21.1 mm and S. epidermidis at 18.2 mm at a 15% concentration. Meanwhile, the well-diffusion method showed inhibition zones ranging from 12 to 21.8 mm for P. acnes and 12.7 to 24.7 mm for S. epidermidis across various concentrations. These results suggest that pineapple peel extract and its patch formulation have strong antibacterial activity, indicating potential as an acne treatment. Abstract

Green beans and potatoes are plants that have very high starch purity which can be used as excipients, especially disintegrants in tablet formulations. Diclofenac sodium is one of the most frequently prescribed anti-inflammatories and is consumed by many elderly patients. Meanwhile, elderly patients often have difficulty swallowing conventional tablets. Orally disintegrating tablet (ODT) is a solid dosage form that quickly disintegrates when placed on the tongue and is expected to be a solution for patients who have difficulty swallowing conventional tablets. This research aims to formulate Orally disintegrating Diclofenac Natrium tablets with a combination of disintegrating ingredients, namely potato starch and green bean starch and to obtain the disintegrating ingredient composition in the optimum formula. Orally disintegrating tablets are made using the direct compression method with a tablet weight of 200 mg. Tablet preformulation tests were carried out including flow time tests, angle of repose tests, bulk density tests and compressibility index tests. And the tablet physical quality evaluation tests include weight uniformity test, hardness test, firmness test, disintegration time test, wetting time test, and disintegration time test in the mouth. The research results showed that the preparation of Orally disintegrating Diclofenac Natrium tablets met the requirements for good physical properties. The fastest tablet disintegration time test was produced by formula 6 and the longest disintegration time test was produced by formula 1. Mung bean starch and potato starch can be used as disintegrants in the formulation of Orally disintegrating Diclofenac Sodium tablets. Abstract

Systemic Lupus Erythematosus (SLE) is a chronic autoimmune disease that requires long-term therapy to control symptoms and prevent organ damage. This study aims to determine the relationship between therapy factors and the level of compliance in SLE patients using the chi-square test with a sample of 64 respondents selected by purposive sampling. The research instrument, the MGLS questionnaire, was used to assess patient compliance. The results showed that the duration of treatment and drug side effects had a significant relationship (P<0.05) with the level of compliance, while the complexity of treatment did not have a significant relationship with the level of compliance (P>0.05). This study indicates that drug side effects are one of the important factors influencing the compliance of SLE patients with therapy

Abstract

The use of sunscreen is an essential measure to protect the skin from the harmful effects of ultraviolet (UV) radiation. Along with the increasing interest in natural ingredients, melinjo fruit (Gnetum gnemon L.) has the potential to be developed as an active ingredient source in skincare products. Melinjo is known to contain bioactive compounds such as phenols, flavonoids, and stilbenoids, which exhibit photoprotective activity and high antioxidant properties, enabling the absorption of UV radiation and the prevention of free radical formation that causes dull skin and premature aging. This study aims to explore the potential of melinjo fruit extract as a natural-based active ingredient for sunscreen, brightening, and anti-aging applications. The research methods included extraction of melinjo fruit using the maceration technique, formulation of the optimal cream containing 59.42% (w/v) olive oil and 6% (w/v) melinjo extract, and evaluation through Sun Protection Factor (SPF), tyrosinase inhibition, antioxidant activity, pH, and skin irritation tests. The results showed that the formulated sunscreen cream exhibited an SPF value of 56.12, classified as ultra protection. In addition, melinjo extract demonstrated tyrosinase inhibition activity and anti-aging potential with an IC50 value of 39.87 ppm. The irritation test indicated that the product was safe and did not cause skin irritation

Abstract

Cough is a natural defense mechanism that functions to clear secretions and foreign particles from the respiratory tract. Allergic cough is commonly treated with a combination of dextromethorphan hydrobromide, glyceryl guaiacolate, and chlorpheniramine maleate. This study aimed to develop and validate a High-Performance Liquid Chromatography (HPLC) method for the simultaneous determination of these compounds in tablet dosage forms. The HPLC method employed a reversed-phase system using a C18 column with a mobile phase consisting of methanol : acetonitrile : distilled water (20:5:75, v/v), a flow rate of 1.0 mL/min, and UV detection at 270 nm. Method validation was performed in accordance with analytical guidelines, including linearity, sensitivity (LOD and LOQ), selectivity, precision, and accuracy. The results demonstrated good linearity for all analytes with correlation coefficients (r) greater than 0.998. The LOD and LOQ values met the required criteria. Precision studies showed %RSD values ≤ 2%, while accuracy ranged from 99% to 101%. Assay results of two commercial tablet samples complied with pharmacopeial requirements. The validated HPLC method proved to be accurate, precise, selective, and suitable for routine analysis of dextromethorphan hydrobromide, glyceryl guaiacolate, and chlorpheniramine maleate in tablet formulations.

Abstract

Gastritis is an inflammation of the gastric mucosa with Proton Pump Inhibitors (PPIs) as the first line of treatment. This study aims to evaluate PPI regimen use and its relationship with treatment success in outpatients with gastritis. The study design is an observational analytical study with a retrospective approach. The sample in this study consisted of 107 patients, namely outpatients diagnosed with gastritis who used PPIs from January 2024 to December 2025. The data were analyzed univariately to describe the characteristics of the variables in frequency distribution and percentage. The results showed that most patients were in the 46-70 age range (46.7%) and were female (76.6%). The most common level of education was high school (37.4%), and the majority of patients were unemployed (63.6%). A total of 58.9% of patients had comorbidities, with hypertension (13.1%) and diabetes mellitus (11.2%) being the most common comorbidities. The pattern of therapy use shows that the combination of lansoprazole and sucralfate was the most commonly used regimen (48.6%). The most commonly prescribed drug was lansoprazole, with 78 uses (41.3%). The evaluation of treatment outcomes showed that 78.5% of patients were declared successful, with 55.1% of patients not returning for follow-up visits, 97.2% not requiring escalation of treatment, and no patients requiring hospitalization.

Abstract

The use of traditional medicine is in great demand because it is relatively safe and rarely causes side effects. Bioactive compounds in plants generally require large biomass to be extracted, so endophytic microbes, especially actinomycetes, are used as an alternative source because they are able to produce secondary metabolites without damaging the host plant. This study aims to isolate and test the antibacterial activity of endophytic actinomycetes from turmeric and galangal plants against Staphylococcus aureus and Escherichia coli. Actinomycete isolation was carried out using Humic Vitamin Agar (HVA) media, and purification using ISP 4 media and continued with antibacterial activity tests. The results of the research showed that of the 26 isolates of endophytic actinomycetes from turmeric and galangal plants that had been isolated, there were 15 isolates that had the ability to inhibit the growth of Staphylococcus aureus and Escherichia coli bacteria. The diameter of the inhibition zone against Staphylococcus aureus ranged from 7.14-14.16 mm, while against Escherichia coli it ranged from 8.11-18.17 mm, with isolate DK4 showing the highest antibacterial activity. These results indicate that endophytic actinomycetes from turmeric and galangal plants have the potential to produce secondary metabolites that are antibacterial.
