Abstract

Latar Belakang: Diabetes melitus adalah gangguan metabolisme yang ditandai dengan peningkatan kadar glukosa darah akibat gangguan sekresi insulin atau kerja insulin. Daun sukun ( Artocarpus altilis ) mengandung metabolit sekunder dengan potensi sifat antidiabetes. Tujuan: Penelitian ini bertujuan untuk mengevaluasi aktivitas antidiabetik ekstrak etanol daun sukun dalam menurunkan kadar glukosa darah pada tikus Wistar jantan yang diinduksi aloksan. Metode: Studi eksperimental dilakukan menggunakan 30 tikus Wistar jantan yang dibagi menjadi enam kelompok. Kelompok normal tidak diinduksi dengan aloksan, sedangkan kelompok lain diinduksi dengan aloksan. Ekstrak etanol daun sukun diberikan pada dosis 150, 300, dan 600 mg/kgBW. Kadar glukosa darah diukur pada hari ke-0, 5, 10, dan 15. Data dianalisis menggunakan ANOVA Satu Arah diikuti dengan uji Post Hoc Tukey HSD. Hasil: Pada hari ke-15, kadar glukosa darah rata-rata pada kelompok yang diberi ekstrak dengan dosis 150, 300, dan 600 mg/kgBW masing-masing adalah 251,0 ± 30,1 mg/dL, 251,6 ± 35,6 mg/dL, dan 192,4 ± 31,0 mg/dL. Dosis 600 mg/kgBW menunjukkan penurunan kadar glukosa darah terbesar dan menunjukkan kemanjuran yang mendekati glimepiride (174,4 ± 27,6 mg/dL). Kesimpulan: Ekstrak etanol daun sukun ( Artocarpus altilis ) menunjukkan aktivitas antidiabetes pada tikus yang diinduksi aloksan, dengan dosis 600 mg/kgBW menunjukkan efektivitas tertinggi. Kata kunci: Artocarpus altilis; aloksan; aktivitas antidiabetes; kadar glukosa darah.

Abstract

Gliocladium sp. is a fungus that functions as a biological control agent against plant pathogens. Research on the activity of Gliocladium sp. not yet against human pathogens. The aim of this study was to determine the antibacterial profile of ethyl acetate extracts of Gliocladium sp. T.N.C73 (LBKURCC3) using the TLC-bioautography method against E. coli ATCC 25922 and S. aureus ATCC 25923. The test results showed that the ethyl acetate extract of the fungus Gliocladium sp. exhibited antibacterial activity against S. aureus dan tidak menghambat E. coli whereas against, it showed antibacterial activity characterised by the formation of a clear area and verified with resazurin (blackish blue colour). Resazurin is used as a redox indicator in bacterial cell viability and proliferation tests. The conclusion of this study is that the ethyl acetate extract of Gliocladium sp. T.N.C73 (LBKURCC3) fungus can inhibit S. aureus bacteria.

Abstract

Chalcones are secondary metabolites belonging to the flavonoid class that exhibit sunscreen activity. The chalcone analog (E)-1-(2-hydroxyphenyl)-3-(4-methoxyphenyl)prop-2-en-1-one has the potential to be developed as an active ingredient in sunscreen formulations. In this study, the compound was formulated into an oil-in-water (O/W) lotion at three different active ingredient concentrations, namely F1 (0.1%), F2 (0.3%), and F3 (0.5%). The formulations were subsequently evaluated for their physical properties and acute dermal irritation in vivo using experimental animals. The physical evaluation demonstrated that all lotion formulations were homogeneous, had a pH of 6.90, exhibited an oil-in-water emulsion type, possessed a viscosity of 8,866 cps, showed pseudoplastic thixotropic flow behavior, and had a spreadability of 6.25 cm. Furthermore, no significant changes in the physical properties were observed after 8 weeks of storage. The acute dermal irritation test revealed that all formulations had an irritation index of 0, indicating a very slight irritation response. These findings demonstrate that all lotion formulations met the required quality criteria for lotion preparations and were well tolerated, supporting their potential as safe topical sunscreen formulations.

Abstract

The widespread use of nonsteroidal anti-inflammatory drugs is often associated with gastrointestinal adverse effects, highlighting the need for safer alternatives. This study aimed to evaluate the analgesic and gastroprotective potential of a combination of honey pineapple and dragon fruit juices in mice. Analgesic activity was assessed using the acetic acid-induced writhing test with aspirin as the positive control, while gastroprotective effects were evaluated using an aspirin and hydrochloric acid-induced gastric ulcer model with sucralfate as the reference drug. The test samples consisted of three ratios of juice combinations (2:1, 1:1, and 1:2). Observed parameters included cumulative pain inhibition, number of gastric ulcers, gastric pH, and organ indices of the stomach and liver. The results demonstrated that all treatment groups exhibited analgesic activity, with cumulative inhibition values higher than the negative control (28.0±19.8%), namely 47.3±18.2%, 53.6±11.4%, and 45.0±13.7%, although still lower than the positive control (75.0±16.2%). In the gastroprotective assessment, treatment groups reduced gastric ulcer formation without significant changes in gastric pH and organ indices. Overall, the 1:1 ratio showed the most balanced profile, combining effective analgesic activity, gastroprotection, and safety, indicating its potential as a natural dual-function therapeutic agent.

Abstract

Cognitive impairment is associated with decreased cholinergic system activity and can be acutely modeled using atropine sulfate as a muscarinic receptor antagonist. This study aimed to evaluate the effectiveness of a combination of nutmeg essential oil (Myristica fragrans) and green tea leaf infusion (Camellia sinensis) on memory function in atropine-induced male mice. A quasi-experimental pretest–posttest control group design was used involving 30 mice divided into six groups. The observed parameter was escape latency in the Morris Water Maze test. The results showed that all groups had relatively homogeneous baseline learning ability, indicated by a progressive decrease in escape latency during the training phase. Atropine sulfate induction significantly increased escape latency in the negative control group (p = 0.043), indicating cognitive impairment. The combination of nutmeg essential oil and green tea leaf infusion significantly reduced escape latency compared to the post-induction condition (p = 0.043). However, no significant differences were observed between the combination and single treatments (p = 0.083) or compared to the positive control donepezil (p = 0.175). It can be concluded that the combination has the potential to improve cognitive function with effectiveness comparable to standard therapy.

Abstract

Breast cancer remains one of the leading causes of cancer-related mortality worldwide, necessitating the development of targeted therapies with improved efficacy and lower toxicity. This study aimed to evaluate the potential of alkaloid isolates from Voacanga foetida, namely voacangine and vobtusine, as tyrosine kinase inhibitors against MCF-7 breast cancer cells through an in silico approach. Molecular docking was performed using Molecular Operating Environment (MOE) 2024 against tyrosine kinase protein with PDB ID: 1T46, followed by ADME and drug-likeness prediction using SwissADME and ADMETlab. Doxorubicin was used as a positive control. The docking results demonstrated that voacangine and vobtusine exhibited competitive binding affinities of -7.80 and -7.70 kcal/mol, respectively, with RMSD values below 2 Å, indicating stable ligand–protein interactions. Both compounds formed hydrogen bond interactions with key active residues HIS790 and ARG791. ADME analysis revealed that voacangine possessed the best drug-likeness profile, while vobtusine showed favorable conformational stability and low probability as a P-glycoprotein substrate. In conclusion, voacangine and vobtusine exhibit promising potential as natural tyrosine kinase inhibitor candidates for targeted breast cancer therapy and warrant further in vitro and in vivo investigations.

Abstract

Non-compliance with antibiotic use is a key factor triggering therapeutic failure and increasing risk of antibiotic resistance. Various factors are suspected to influence patient compliance, including sociodemographics, knowledge, understanding of usage instructions, and patient perceptions toward antibiotics. This study aims to analyze factors influencing compliance with antibiotic use among dental clinic patients at Bakunase Public Health Center, Kupang City. This is an analytical observational study with a cross-sectional design. Sampling technique used consecutive sampling with a total of 50 respondents. Data were obtained through questionnaires and medical records, then analyzed using univariate analysis, chi-square test for bivariate analysis, and logistic regression for multivariate analysis. Results showed that respondent characteristics were dominated by 18-40 age group (48%), females (60%), secondary education level (50%), and those unemployed/having no income (58%). A total of 52% of respondents were classified as non-compliant. Analysis indicated that sociodemographic factors and perceptions had no significant effect on compliance (p > 0.05). Conversely, level of knowledge and understanding of instructions were most influential factors on antibiotic compliance (p = 0.017; OR = 5.069; 95% CI = 1.341-19.160), with understanding of instructions being the most dominant factor. Therefore, improving quality of education and counseling by healthcare professionals regarding proper antibiotic use is highly necessary to enhance patient compliance.

Abstract

Lip cream is widely used as a lip color because lip cream can provide an even and long-lasting color. Red spinach contains anthocyanins which function as antioxidants. The lifestyle patterns of people who are starting to return to the back to nature lifestyle are one of the backgrounds for research to formulate lip cream preparations from natural dyes from red spinach (Amaranthus tricolor L.). This study aims to provide information on the use of red spinach as a natural dye. The study was conducted experimentally, including the manufacture of lip cream formulations with red spinach ethanol extract at concentrations of 15% and 17%. Examination of the physical quality of the preparations included organoleptic tests, stability, homogeneity, pH, adhesion, spreadability, irritation, and hedonic tests calculated using SPSS. The results showed that all preparations had good organoleptics, good stability, met the requirements of the homogeneity test, and the appropriate pH. In addition, the preparations also met the adhesion and spreadability tests, and did not cause irritation. The most popular formula is F2 formula with 17% concentration of red spinach ethanol extract, which shows the best performance in these aspects. This study shows the potential of red spinach as a safe and effective natural colorant in lip cream formulation.

Abstract

Hypertension requires long term therapy that often involves drug combinations, thereby increasing risk of drug interactions that may affect effectiveness and safety of treatment. This study aimed to evaluate potential for antihypertensive drug interactions among outpatients at Bhayangkara Hospital Kupang from January to June 2025, based on severity levels and interaction mechanisms. This was a descriptive retrospective study utilizing patient prescription data. A total sampling technique was applied, obtaining 300 prescriptions that met inclusion and exclusion criteria. Potential drug interactions were screened using Medscape Drug Interaction Checker and Drugs.com, followed by descriptive analysis. Results showed that out of 300 prescriptions, 205 (68.33%) had potential drug interactions. Amlodipine was most frequently prescribed drug (52.56%). A total of 412 cases of potential drug interactions were identified, which were dominated by moderate severity with 322 cases (78.15%), followed by major severity with 47 cases (11.41%), and minor severity with 43 cases (10.44%). Based on mechanism, pharmacodynamic interactions were most dominant with 336 cases (81.55%), while pharmacokinetic interactions accounted for 76 cases (18.45%). In conclusion, the potential for antihypertensive drug interactions at Bhayangkara Hospital Kupang remains high, predominantly characterized by moderate severity and pharmacodynamic mechanisms. Optimal therapeutic monitoring is essential to enhance patient safety.

Abstract

Kappaphycus alvarezii is the primary source of carrageenan widely utilized in pharmaceutical, food, and cosmetic industries. Carrageenan quality is significantly influenced by post-harvest drying methods. This study aimed to analyze effect of drying methods on moisture content and carrageenan content of red variety K. alvarezii from Letbaun Village, Semau Island, East Nusa Tenggara. A quantitative descriptive design was applied using two treatments: natural sun drying for ±3 days and oven drying at 60°C for 48 hours. Moisture content was determined by gravimetric method, while carrageenan content was quantified using High Performance Liquid Chromatography (HPLC) after alkaline extraction with 8% KOH. Statistical analyses were performed using Shapiro–Wilk test, Levene's test, and independent sample t-test (α = 0.05). Oven drying yielded a moisture content of 27.32% and a mean carrageenan content of 25.98%, whereas sun drying yielded 28.85% moisture and 24.13% mean carrageenan. Both methods complied with SNI 2690:2015 standards (≤ 30%). The independent t-test indicated no statistically significant difference between two methods (p = 0.471; p > 0.05), although oven drying numerically produced higher carrageenan content. Oven drying is recommended as a more controlled and consistent post-harvest alternative for producing high-quality carrageenan raw materials for pharmaceutical applications.
