Abstract

Abstract. Diltiazem Hydrochloride (HCl) is commonly used in hypertension therapy for its beneficial effects, such as reducing blood vessel pressure resistance and relaxing the heart muscles. In an in-vivo study, Diltiazem HCl is known to be decrease by 60% of its bioavailability due to first-pass metabolism. Thus, it is necessary to develop a new administration route, some of which is buccal drug delivery route in mucoadhesive buccal film. Moringa gum is a polyuronide polymer with a hydroxyl group in its side chains, giving the potential use as a mucoadhesive polymer. However, it has poor folding endurance and its release kinetics study has not been investigated. Polyvinyl alcohol (PVA) is a water-soluble polymer with hydroxyl side chain. PVA is known for its high tensile strength and flexibility, though it can be affected by humidity. The aim of this research is to determine the right amount of moringa gum and PVA to obtain a film with good physical characteristics and mucoadhesive residence time. In this study, the optimization of moringa gum and PVA was carried out using factorial design. Mucoadhesive buccal film was prepared by solvent casting method, the films were later evaluated and the obtained responses were analyzed by Software Design Expert 11.0.0. The optimization result shows the film containing the amount of moringa gum 2% and PVA 5% can produce the optimum formula. The mucoadhesive buccal film formula has surface pH of 6.083, mucoadhesive residence time of 445.055 min, swelling index of 4.067, and 87.9913% drug release in in-vitro drug release study after 420 min following the Higuchi model release kinetics. Therefore, it can be concluded that the obtained optimum formula of mucoadhesive buccal film can be used in the preparation of mucoadhesive buccal film Diltiazem HCl.. Abstract

Abstract. Mud crab shells contain chitin which can be converted into chitosan. Co-processed excipient or combining two or more excipient ingredients can increase the compressibility, compactibility of tablets. This research aims to formulate the active ingredient chitosan which is synthesized from mangrove crab (Scylla serrata) shells and then make chitosan tablets with a co-processed excipient consisting of two variations of primogel. The difference in formula, namely F1 (primogel 4%) and F2 (primogel 8%) was then seen as to its effect on the physical quality of the tablet. The tablet printing method is direct compression. The physical quality of the tablets was evaluated and the results showed that the formula met the test requirements for tablet weight uniformity, friability, hardness, disintegration time but did not meet the requirements for tablet size uniformity. Then the evaluation results of the co-processed excipient met the specifications for the flow speed test, angle of repose, compressibility index, Hausner ratio, water content test and particle size distribution. Thus, it can be concluded that chitosan from mangrove crab shells (Scylla serrata) can be formulated into tablet preparations from co-processed excipients using the direct compression method but there is no significant difference between Primogel 4% and 8% as a crushing agent on the characteristics of chitosan tablets. mud crab (Scylla serrata) shell. Abstract

Abstract. Osteoarthritis (OA) is a degenerative joint disease characterized by damage to joint cartilage, subchondral bone, and osteophyte formation. The prevalence of OA is still large with major therapy is non steroid anti inflammatori (NSAID) drug. However, prolonged use of NSAIDs leads to side effects on gastrointestinal tract. Zingiber officinale var. amarum is beneficial herbal for anti-inflammatory therapy in OA. The damage of chondrocytes in OA triggers the activation of inflammatory process through the COX-2 pathway, resulting in the production of chemical mediators prostaglandin D2 (PGD2) and thromboxane A2 (TXA2), causing pain and functional decline. Thus, this research aimed to predict active compounds of Z. officinale var. amarum rhizomes that potential to inhibit PGD2 and TXA2 as anti-inflammatory agents for OA through molecular docking. Method This research use 20 active compounds of Z. officinale var. amarum using molecular docking method with proteins target including of PGD2 (ID:1S2A) and TXA2 (ID:6IIV). The docking process was conducted using the PyRX 0.8 application, and the results were visualized in the BIOVIA Discovery Studio 2020 application. Result the research on the 20 active compounds in the rhizome of Z. officinale var. amarum indicates that compounds such as [6]-Gingerdiol 3,5-diacetate, 10-gingerol, gingerdione, and gingerdiol can interact with the active site of the protein 1S2A through hydrogen bonding with Gln-222. Furthermore, compounds like [6]- dehydrogingerdione, paradol, gingerdiol, geraniol, 10-gingerol, gingerdione, [6]- Gingerdiol 3,5-diacetate, isoborneol, zingerone have been identified to interact with 6IIV through hydrogen bonding with Gln-301 and Thr-81. This indicates that these compounds exhibit activity as inhibitors of PGD2 and TXA2. The interactions occurring between the complex compounds of Z. officinale var. amarum and 1S2A and 6IIV can lead to inhibitory effects on PGD2 and TXA2.
