Abstract

Diabetes Mellitus type 2 can be caused by the resistance of tissue towards insulin accompanied by relative deficiency in insulin secretion. Insulin resistance factor can result from obesity. This research aims to investigate anti- diabetic activity of the compound of FEA of curcuma (Curcuma domestica Val) and bitter melon (Momordica charantia L.). Subjects of this research were 40 albino Wistar rats (Rattus norvegicus) aged 5-8 weeks. The rats were randomly grouped into 8 experimental groups in which each group consisted of'5 rats. The tested animals were divided into 6 groups, KG) metformin 45 mg/Kg BB, P1: FEA curcuma 10 mg/ 200g BB, P2: FEA bitter melon 04 mg/ 200g BB, P3: Compound of FEA curcuma : FEA bitter melon 5 : 0,8 mg/200g BB, P4: Compound of FEA curcuma : FEA bitter melon 10 : 04 mg/200g BB, and P5: Compound of FEA curcuma : FEA bitter melon (20 : 02mg9/200g BB). The animals were inducted with insulin resistance with the giving of HFD-fructose. Abstract

The CYP2A6 gene, encodes the CYP2A6 enzyme, has a highly polymorphism. These ezyme have played a role in nicotine metabolism, one of the active compounds in cigarettes that causes of cigarette dependence. The active form of the CYP2A6 gene allele is * 1 allele and some inactive forms of the CYP2A6 gene allele are * 4, * 7 and * 9 alleles. The inactive alleles would reduce the activity of the CYP2A6 enzyme. The genotypes of CY2A6 gene (CYP2A6* 1, * 4, * 7 and * 9) among Chinese Indonesian smokers were determined by allele-specific polymerase chain reaction (PCR) followed by electrophoresis method. The cigarette dependence was analyzed using the Fagerstrom Test for Nicotine Dependence (FTND) method.This study showed that there are CYP2A6 polymorphism genes among 30 smokers who participate in these study. The frequencies of the genotypes of CYP2A6 alleles were CYP2A6 *1/*1 (46.7%), CYP2A6 *1/*4 (16.7%), CYP2A6 *1/*7 (3.3%), CYP2A6 *1/*9 ( 10%), CYP2A6 *1/*4/*7 (6.7%), CYP2A6 *1/*4/*9 (10.0%), CYP2A6 *1/*7/*9 (3.3% ) and CYP2A6 *4/*7/* 9 (3.3%). Abstract

The Novel compound of analog UK-3A defined as HF-1 (6-hydroxy-N-phenylnicotinamide) and HF-2 (6-hydroxy-N-phenylpicolinamide)was successfully synthesized by amidation reaction of aromatic carboxilyc acids and primary amine by adding the activator of DCC (dicyclohexylcarbodiimide), catalyst DMAP (4-dimethyl aminopyridine, and DMSO (dimethyl sulfoxide) as the solvent. Reaction run for 24 hours in 55oC. Result shows yield of HF-1 as much as 52%, and for HF-2 is 16%. The analogue compounds structure of UK-3A were characterized by spectrophotometer FT-IR, LCMS, and NMR. Citotoxicity assay against Murine Leukemia cells of HF-1 and HF-2 by MTT (3-(4,5-dimethylltiazo-2-yl-) 2,5-diphenyltetrazolium bromide) assay. The result of bioassay showed IC50(inhibitory concentration)value 71 µg/mL and 63 µg/mL for HF-1 and HF-2 respectively. It informed that the analogue compounds have lower activity than UK-3A compound which has IC50 value 38 µg/mL.

Abstract

L-Arginine is a semi essential amino acid and has a function in arterial vascular relaxation and improves blood flow. Single-bulb black garlic was produced from ordinary single-bulb garlic clove by process from maillard browning at temperature at 700C and 95Y of humidity controlled room for a month. It is known that single-bulb black garlic has benefits as antidiabetes, uric acid, cholesterol, hypertension, endurance and others. This study aims to determine the content of L-Arginine compound on single-bulb garlic and single-bulb black garlic by using HPLC (High Performance Liguid Chromatography)method.The mobile phase used consists of buffer A and buffer B. Buffer A consists of Na-Acetate (pH 6.5), Na-EDTA, Methanol, and Tetrahidrofuran. While B buffer consists of Methanol and Water Pro Injection. Thermo Scientific Column ODS-2 Hyersil C18, flow rate 1 ml / min, using Flouresensi detector with 350 nm wavelength. Based on the results obtained L-Arginine compound contained in a single-bulb garlic of 2.504 while for single-bulb black garlic that is egual to 06704. The content of L-Arginine compounds in single garlic is greater than that of a single black garlic. Diabetes mellitus(DM) and dyslipidemia are high prevalence diseases which also as one of risk factors for coronary heart disease. Knowledge and adherence are important factors in the treatment of DM-dyslipidemia either to control blood glucose and cholesterol or to prevent from complications. Education is one of the ways to increase knowledge and adherence. The aim of this research is to determine the impact of intervention (Patient Information Leaflet (PIL) and motivation messages by SMS) to knowledge and adherence of DM-dyslipidemia patients in PKU Muhammadiyah Yogyakarta Hospital. This research is a quasi experimental study with analytical quantitative. The data taken from February 2016 to April 2017 at PKU Muhammadiyah Yogyakarta Hospital. Inclusion criterias in this research are patient with age more than 18 years old, with diagnosed by DM-dyslipidemia, using 1 or more of DM and dyslipidemia medicine, not deaf, and cooperative. Interventions that given to the patients are Patient Information Leaflet (PIL) and motivation messages by SMS which routinely given in a week after first we met. Knowledge is assessed using questionnaire and adherence is assesed using pill count method. The data that need analyzed are characteristic patients and the impact of intervention using Wilcoxon and paired t-test. The result of intervention in 56 patients showed the increasing number of patient that have good knowledge after intervention, from 58,9% to 71,4%, otherwise it not differ significantly (p 0.071). Other than that, the adherence also increased that showed from the patients who have adhere category after intervention, from 58,93% to 91.07% and it differ significantly with p 0.001 (p<0.05). Abstract

Researched about the influence of ethanol consentration to entrapment of nifedipin in ethosome formula. This research suppose to know about the influence of etanol consentration tonifedipine entrapment, form and size of ethosome, to decide the optimal consentration of etanol forming ethosome with high PDE (Percent Drug Entrapment) in nifedipin, and to know the ethosome characteristic. Ethosome formula  made in by hot method, using variation consentration of ethanol, phosphatidilcholin 1.5% and nefidepin 0,05%. Nifedipin concentration that entrapment in ethosome measured byUV-VIS Spectrophotometer at wavelength 336 nm, form and ethosome size fixed by using microscope.  Ethosome with  high  percent drug entrapment have taken from formula by using ethanol 50% with entrapment 71.440%. The formed ethosome is Multy Lameral Vesicle (MLV) and Multy Vesicular Vesicle (MVV) with the size about 1 μm until 15μm.

Abstract

To prevent hyperglycemia, people need alternative source of carbohydrate with low glycemic index. Porang bulbs from Amorphophallus muelleri Blume plant is rich on glucomannan, that could prevent elevation of blood glucose level. The aim ofthis study is to determine aglycemic index and intestinal transit index of porang bulbs flour. Glycemic index determined by glucose tolerance test method and intestinal transit index determined by measuring the intestinal transit distance of mice administered 1,5 kg BW intragastically the porang suspense. Forty-five minute later mice was given carbo absorben as a marker. At 15 minute later mice was sacrificed by cervical dislocation and the distance of charcoal marker was measure from pillory. The results shows that porang has glycemic index 85 compare with glucose 100 and treatment with porang flour caused the intestinal motility was faster (intestinal transit index -0,42) rather than wheat four (0,32). Its mean that, porang flour shorten the retention time of chymus in intestinum s0 it will inhibit the absorbtion of'glucose. Abstract

Worsen asthma symptoms is associated with low vitamin D levels that increases asthma attacks risk. Physical activity is one factor that affects vitamin D levels in the blood. This study aimed to identify relationship physical activities effects with vitamin d levels on asthma and non-asthma patients. The study was conducted in March-June 2018. The subjects were asthma patients and non-asthma adults and didn’t have other comorbidities. Data analysis used pearson test to determine physical activity effect with vitamin D levels. There were significant differences in vitamin D levels (P <0.000) and physical activity (P <0.000) in asthma and non asthma respondents. The results of the correlation test between vitamin D levels and physical activity in Approximate Significance value was 0.965, which means there were very strong relationships between vitamin levels and physical activity on the respondents of asthma and non asthma. So, the asthma patients with sufficient physical activity will have normal vitamin D levels to improve control of asthma symptoms. This study aimed to identify relationship physical activities effects with vitamin d levels on asthma and non-asthma patients. The study was conducted in March-June 2018. The subjects were asthma patients and non-asthma adults and didn’t have other comorbidities. Data analysis used pearson test to determine physical activity effect with vitamin D levels. There were significant differences in vitamin D levels (P <0.000) and physical activity (P <0.000) in asthma and non asthma respondents. The results of the correlation test between vitamin D levels and physical activity in Approximate Significance value was 0.965, which means there were very strong relationships between vitamin levels and physical activity on the respondents of asthma and non asthma. So, the asthma patients with sufficient physical activity will have normal vitamin D levels to improve control of asthma symptoms.

Abstract

Swamedikasi yang tidak rasional dipengaruhi oleh rendahnya tingkat pengetahuan, keterampilan, dan perilaku masyarakat dalam pemilihan obat. Tujuan dari penelitian ini adalah untuk mengetahui efek samping apa saja yang paling sering muncul, serta mengetahui hubungan antara kejadian efek samping obat dengan cara penggunaan obat, dan mengetahui faktor apa saja yang menimbulkan efek samping dalam penggunaan obat bebas, obat bebas terbatas, OWA pada masyarakat di Kecamatan Kotagede, Yogyakarta. penelitian ini didisain secara observasional dengan metode survei,pengambilan data menggunakan kuisionerpada 100 pasien. Data yang diperoleh dianalisis dengan metode Uji Fisher dengan taraf kepercayaan 95%. Hasil penelitian diperoleh bahwa efek samping yang umum dirasakan oleh pasien adalah mengantuk, nyeri pada perut, dan jantung berdebar. Berdasarkan hasil Uji Fisher menunjukan bahwa tidak ada hubungan yang bermakna antara cara penggunaan obat dengan kejadian efek samping obat, faktor yang mempengaruhi efek samping pada penggunaan obat-obat tanpa resep adalah kurangnya pengetahuan masyarakat tentang bagaimana cara penggunaan obat yang baik dan benar, ketidak patuhan pasien dalam aturan pemakaian obat yang sudah dijelaskan oleh apoteker, danadanya perbedaan respon tubuh terhadap obat yang dikonsumsi tiap pasien. Kesimpulan, hasil uji statistik tidak terdapat hubungan yang bermakna antara penggunaan obat yang benar atau tidak benar dengan muncul atau tidak muncul efek samping obat, namun pada kenyataannya penggunaan obat-obat yang dibeli tanpa resep dokter dapat menimbulkan berbagai efek samping dan adanya perbedaan respon tubuh terhadap obat yang dikonsumsi tiap pasien.

Abstract

Treatment with risperidone 1.5-6.0 mg/day provides good benefits for schizophrenia treatment, severe psychiatric disorder with long-term disability potential and limited availability of treatment options. According to the average percentage of patients with abnormal liver function tests, patients with clinically significant improvement (> 3 times of the normal upper limit for SGPT, aspartate aminotransferase.Abnormalities that are commonly without any symptom occur within 6 weeks of antipsychotic use (Correll et al., 2015). The purpose of this study is to analyze the liver function of schizophrenic in-patients who take risperidone and lorazepam in Amino Gondohutomo’s psychiatric hospital of Central of Java Province. The research subjects were 132 schizophrenic diagnosed patients who take risperidone and combination of risperidone and lorazepam. We used Chi Square Tests, Paried T Tests and One Sample Test.Based on the research results, it can be concluded that there is a correlation between readmisi and age group, in age group > 29 years more risk relapse than age group <29 years. Also, it can be concluded the combination of risperidone and lorazepam at risk 0,933 times will increase SGOT than get treatment with risperidone only and therapy the combination of risperidone and lorazepam at risk twice will increase SGPT rather than receive treatment with risperidone only.
