Abstract

Leaves agarwood (Aquilaria microcarpa Bail) is one of the natural materials that can be used as an antibacterial. This study aims to determine the content type of secondary metabolites and antibacterial activity against Staphylococcus aureus and Proteus mirabilis by the method of Kirby-Bauer disc diffusion. Simplicia leaves agarwood macerated with ethanol 96%. Results of phytochemical screening of ethanol extract of leaves agarwood contain flavonoids, phenols, tannins, saponins and steroids. Furthermore, the TLC for the assertion of their chemical compounds in extracts of leaves agarwood using a mobile phase of butanol: acetic acid: water (4: 1: 5) with the stationary phase silica gel GF254 with spotting FeCl3 5% for phenolic and AlCl3 5% for flavonoids later in UV detection at 254nm and 366nm. Control positive used is siprofloksasin 50 μg while control negative used is DMSO 20 % . Based on the results of the average diameter zone obstruent extract ethanol leaves agarwood in staphylococcus aureus by concentration of the 300 mg / ml, 400 mg / ml, 500 mg / ml is 12.50 mm, 13,51 mm, 15,80 mm. while in proteus mirabilis by concentration of the 300 mg / ml, 400 mg / ml, 500 mg / ml is 12,10 mm, 13,26 mm, 15.19 mm. This shows that extracts ethanol leaves agarwood having activity on Gram positive and Gram negative.

Abstract

Quantitative analysis of organic acid in acidifier product is needed to maintain product effectiveness in repressing microbial growth and lowering feed and gastrointestinal tract pH. The aim of this research is determining the level of formic and lactic acid in two acidifiers from the market. Analysis were performed using reversed phase High Performance Liquid Chromatography (HPLC) with LiChrospher® 100 RP-18 column (250 x 4.0 mm, 5μm, Merck), buffer potassium dihydrogenphosphate 50 mM-TEA 0,5% pH 4,00 as mobile phase and flow rate 0,6 mL/min. Wavelength used in the analysis was 214 nm. Validation methods provide results of recovery by 98.02%-101.97% for formic acid and 97.09%-102.78% for lactic acid, RSD <0.59% for formic acid and 1.28% for lactic acid, LOD 63.05 µg/mL for formic acid 4.55 µg/mL for lactic acid, LOQ 210.16 µg/mL for formic acid and LOD 15.18 µg/mL for lactic acid. The analysis method gives linearity in the range of 439.2 -1764.61 µg/mL for formic acid with correlation coefficient (r) 0.9992 while lactic acid linearity range was 47.88 µg/mL-193.44 g/mL with a correlation coefficient (r) 0.9994. Conformity product A to its label provides the results of 108.19%-109.82% formic acid and 156.88%-167.90% lactic acid whereas the result for product B were 101.65%-109.95% formic acid and 151.10%-172.82% lactic acid.

Abstract

Selection and usage of rational drugs determine the success of expected therapeutic effect, especially for pediatrics. Acute bronchitis in pediatrics is one of the highest incidents of Acute Respiratory Infection (ARI) in Army Hospital Kartika Husada Kubu Raya. This study aimed to evaluate rationality of drug doses conformity in outpatient pediatric acute bronchitis in Army Hospital Kartika Husada Kubu Raya on 2015. This research was a descriptive observational research with cross-sectional design. The sample was taken with purposive sampling amounted to 36 prescriptions. Then the evaluation of drug doses conformity was counted according to the literature. The results showed that drugs which were used for acute bronchitis were mucolitics and expectorants, antibiotics, bronchodilators, multivitamins, decongestans, analgesics, and antihistamins. The result of drug dosage conformity evaluation showed that the under-dose drug item (R/) was 51%, over-dose was 15% and appropriate dose was 34%.

Abstract

The development of anticancer drugs from ethyl p-methoxycinnamate (EPMC) derivatives has been done to compounds high activity in inducing cancer cells apoptosis and minimal side effects. p-Methoxycinnamoyl hydrazide derivates, modified from EPMC structure, were docked into the ligand-binding pocket of Check point kinase 1 enzymes (2YWP) and the aromatase enzyme (3S7S) using software Molegro Virtual Docker (MVD) Ver.5.5. We compared the Rerank score of native ligand with p-Methoxycinnamoyl hydrazide derivates. Rerank scores of compounds 4b and 4c (-99.98 Kcal/mol and -99.80 Kcal/mol) were lower than the native ligand A42 in inhibiting the enzyme checkpoint kinase 1. Rerank values of p-Methoxycinnamoyl hydrazide derivate compounds were greater than the native ligand EXM in inhibiting the enzyme aromatase. p-Methoxycinnamoyl hydrazide derivate compounds, especially compounds 4b and 4c, had anticancer mechanism by inhibiting the checkpoint kinase 1 enzyme pathway and showed no activity in inhibiting the aromatase enzyme.

Abstract

Acute respiratory infections (ARI) is one of the major health problems in Indonesia. There are three classifications of cases of ARI, consist of pneumonia, severe pneumonia and non pneumonia. This study aimed to find an overview of the use of antibiotics and the evaluation of suitability prescriptions performed in cases of non pneumonia Acute Respiratory Infections (ARI) in IMCI Polyclinic, Cengkareng District Community Health Centre (Puskesmas Cengkareng), West Jakarta. Evaluation for the prescription suitability was useful to find the percentage of the suitability of prescribing on the existing management. This activity was done using retrospective method with data of 100 children patients suffered from non pneumonia cough obtained from registration book on IMCI Polyclinic during February 2016. The existing data was then matched to the patient status and prescription books that went into the dispensary unit at second floor. Furthermore, we evaluated the suitability of the reference book titled Integrated Management Scheme for Toddler Patients/Bagan Manajemen Terpadu Balita Sakit (MTBS), issued by the Ministry of Health of the Republic of Indonesia in 2011. The results showed that the percentage of antibiotic used in the sample was 59.6%, which the most frequent antibiotic used was amoxicillin. Therefore we concluded that the prescription compliance with IMCI guidelines in Puskesmas Cengkareng at February 2016 was inadequate yet.

Abstract

The snakehead (Channa striata) extract contains albumin (in its water phase) and omega-3 and omega-6 (in its oil phase) which have been proven to be effective in wound healing process. This research was aimed to determine wound healing effect of snakehead extract-containing ointment on 6 groups of male Wistar rats. The rat’s skin was wounded to produce acute stage II opened wound, the ointment was applied for sixteen days, and wound healing process was observed. The width of the wound was measured by using Macbiophotonic Image J program. The AUC results of each groups from smallest to largest were normal group (702.84% x day), negative group (749.56% x day), positive group (765.146% x day), water phase-containing ointment 10% group (795.146% x day), oil phase-containing ointment 10% (837.282% x day), and water-oil-phase combination ointment 10% (874.901% x day). The data were analyzed using One-Way ANOVA and Post-Hoc LSD test. The result of this research showed significant differences (p<0.05) between groups of water-oil phase combination of snakehead extract ointment with a negative control group. This research result showed that ointment containing water-oil phase combination of snakehead extract had wound healing effect on acute stage II opened wound.

Abstract

Type 2 diabetes mellitus is a degenerative disease with long-term treatment that requires knowledge and self-management to control blood sugar level. The purpose of this study was to evaluate the provision of booklet and pharmacist education in improving knowledge and behavior of self-management of type 2 diabetes mellitus patient at Mayapada Hospital Tangerang. This study respondents were 90 people who were divided into 3 groups. Group 1 got booklet and education, group 2 got booklet only and control group did not get any treatment. Fasting Blood Glucose (FBG) levels before the intervention were in poorly controlled category and after intervention there was no significant change. There was an increase in the level of knowledge and self-management in group 1 and group 2. There was no significant change in FBG. Provision of booklet with education by pharmacist showed a higher increase in knowledge (p=0.02) and self management behavior (p<0.001) than provision of booklet only. There was no relationship between knowledge and self-management behavior with blood sugar levels. It could be concluded that booklet and education by pharmacist could improve knowledge and behavior of self-management of DM type 2 patients in each intervention group. However, there was no relationship between knowledge and self-management behavior with FBG.

Abstract

Mucoadhesive dosage form is a pharmaceutical dosage form with prolonged gastric residence time which can increase bioavailability of drugs. An excipient with suitable swelling and bioadhesive characteristics plays important role to obtain good mucoadhesive dosage form. Our pre-eliminary study showed that chitosan-xanthan gum polyelectrolyte complex (CXPC) in ratio 1:1 exhibit suitable swelling index to be developed as mucoadhesive dosage forms. This research was performed to study CXPC characteristics as matrix for mucoadhesive granules dosage form matrix. In this study CXPC 1:1 was utilized as the matrix in the mucoadhesive granules with drug-CXPC ratio of 1:1, 1:2, and 1:3, using diltiazem HCl as a drug model. The granules were prepared by wet granulation method, and the in vitro bioadhesive properties of the granules, as well as the drug release profile, were evaluated. The results showed that formula containing diltiazem HCl and CXPC in ratio of 1:2 possessed a good bioadhesive ability up to 8-12 hours and showed controlled drug release profile up to 8 hours. The results can be concluded that the CXPC is a suitable excipient as a mucoadhesive matrix.

Abstract

Chitinase is an enzyme that degrades chitin on β-1,4-acetamido-2-deoxy-D-glycoside bound to produce Chitin oligosaccharide or its soluble monomer, N-acetylglucosamine (NAG). Chitinase are usually produced naturally by chitinolytic microorganism that are found in soil or water environment. Chitinase have been widely used for biocontrol agents of plant pests. The product from chitinase hydrolysis can be used as antiinflammatory agent and anti hyperpigmentation. This research was aimed to screen and evaluate the chitinolytic ability of 9 isolate. The best isolate was the one which produced high chitinolytic activity and hydrolyzed chitin into NAG. 9 isolate were examined and BPPTCC-2 was the best isolate which produced highest level of chitinolytic activity 0.1780 U/ml. The chitinase was able to hydrolyzed chitin resulting high yields of NAG 11.5% as the only product.

Abstract

Dengue fever is a major health issue in the world. Preventive measure with temephos/abate has long been used to combat this problem. Istiana et al reported that there were cases of resistance of Aedes aegypti larvae to temephos in various parts of the world, including Indonesia. Indonesian Cinnamon (Cinnamomum burmannii) has been proven to possess larvacide effect and can be used as an alternative insecticide. This research was meant to compare the larvacide effect of Indonesian cinnamon (Cinnamomum burmannii) essential oil to the larvacide effect of temephos against Aedes aegypti larvae. This research used laboratory experimental design analysis. The total number of samples were 20 larvae of Aedes aegypti at 3rd and 4th instar which were tested four times on each four experimental groups. The collected data were the total number of Aedes aegypti larva deaths observed 24 hours after the variables were applied. The analysis of the results was done by using statistic analysis program. There were significant differences on larva deaths due to the essential oil and abate variables with p value < 0.001. The results were as follows: negative control death (0%), emulsifier (2.5%), essential oil (22.5%), and abate (100%). The larvacide effectiveness of abate is significantly better than that of Indonesian cinnamon essential oil.
