Abstract

Abstract

Three-dimensional scaffolds play a crucial role in supporting cell growth and tissue regeneration, especially in bone regenerative applications. Their utilization in dentistry demands effective cytotoxicity assessment, particularly for sponge-shaped scaffolds with a unique porous structure fostering a complex environment for cell growth. Therefore, the imperative lies in selecting cytotoxicity testing methods suitable for these characteristics. Relevant methods for such scaffolds include MTS, Alamar Blue, ATP, CCK-8, and MTT assays. MTS assay efficiently measures cell viability in a three-dimensional setting, while Alamar Blue and CCK-8 assays, with their water-soluble nature and high reproducibility, prove advantageous. ATP assay provides direct insights into cellular metabolic activity, and MTT assay assesses cell viability based on enzymatic capabilities. Due to their water-soluble characteristics, minimizing additional extraction steps, and ensuring high reproducibility, Alamar Blue and CCK-8 emerge as the most fitting options for cytotoxicity assessment in dentistry's sponge-shaped scaffolds.

Abstract

Background: An inadequate root canal treatment allows bacteria to thrive and survive, which is a major cause of endodontic failure. An appropriate treatment plan can be determined after a precise and accurate diagnosis has been made. Objective: The purpose of this case report is to demonstrate the root canal retreatment on a posterior mandibular tooth with periapical lesions. Case report: A 37-year-old female patient came with complaints of discomfort when chewing food with her lower right back tooth. The tooth had been treated ten years ago with a metal filling, which was replaced with a tooth-colored filling a year ago. Intra-oral examination of tooth 46 showed composite resin restoration on the distobuccal surface. Radiographic findings showed that there was restoration on the distal surface that reaches the floor of the pulp chamber and radiolucency in the furcation area. Root canal retreatment was performed based on the diagnosis of acute apical periodontitis et causa necrotic pulp. The treatment plan involved root canal retreatment with direct resin composite restoration. Root canal retreatment starts with the application of a rubber dam, followed by the removal of any existing restoration and caries, opening the cavity, root canal exploration, and root canal shaping. Biomechanical preparation was carried out with Protaper Hand Use with crown down technique, in conjunction with 2,5% NaOCl irrigation. The initial visit concludes with the disinfection of the root canal with calcium hydroxide. The following treatment involved a subjective and objective assessment, removal of the temporary filling, cleaning of calcium hydroxide, and final irrigation with 2,5% NaOCl, 17% EDTA, and 2% CHX. Obturation was accomplished utilizing the vertical condensation technique. The restoration was completed with composite resin. Conclusion: Root canal retreatment on teeth with acute apical periodontitis et causa necrotic pulp was successfully carried out.

Abstract

Periodontitis is a chronic, multifactorial, inflammatory disease, associated with plaque biofilms and characterized by the progressive destruction of the tooth-supporting structures. Periodontal diseases can increase the inflammatory response in patients, which might exacerbate the systemic symptoms and clinical course of coronavirus (COVID-19). The potential association of periodontitis and COVID-19 severity can be explained by the alteration in the expression of cellular receptors enhancing the virulence of SARS-CoV-2 and by periodontal pockets acting as viral reservoirs. These associations between periodontitis and COVID could be due to the fact that both diseases share common immune pathways such as the NF-κB pathway, NLRP3/IL-1β pathway, and IL-6 signaling pathway as well as common risk factors such as gender, lifestyle, and comorbidities. This suggests that the two diseases may be linked through the same genetic pathway.

Abstract

Introduction: Tooth loss affects a person's appearance and health, impacting their quality of life. Teeth play an essential role in chewing and aesthetics, particularly the anterior teeth. Dental trauma such as fractures can affect appearance and psychology, reducing self-confidence. Currently, aesthetics is a top priority in dental care. Tooth loss leads to complex oral problems, thus missing teeth need to be replaced with dentures. Methods: This study explores the use of various gingival retraction techniques before impression taking in prosthodontic treatment. The methods analyzed include mechanical, chemical, and surgical techniques, with a focus on retraction using retraction cords soaked in hemostatic agents to ensure accurate and blood-free gingival margins before impression taking. Discussion: Accurate impressions depend on proper tooth preparation and healthy soft tissue conditions. Effective gingival retraction is crucial for obtaining good impressions, especially at the cervical and gingival margins. Mechanical methods using retraction cords are commonly used due to their effectiveness, speed, simplicity, and affordability. However, chemical methods may pose risks to the gingiva, while surgical methods are more complex. Conclusion: Accurate impressions for fixed dentures require healthy soft tissues without periodontal disease. Gingival retraction before impression taking is essential for achieving accurate impressions, ensuring that the cervical margins are well defined. Retraction methods include mechanical, chemical, and surgical approaches, with mechanical methods being the common choice due to their effectiveness and simplicity.
