Tilapia productivity is influenced by many factors, one of which is Tilapia Lake Virus (TiLV) disease, which can cause a mortality rate due to infection reaching 90%. The development of nested PCR or semi-nested PCR methods can increase the sensitivity of detection of this virus. This study aims to detect the spread of TiLV attacking tilapia early using the semi-nested PCR method with primers designed in this study. Tilapia with TiLV symptoms were taken from a tilapia farmer in Sleman Regency, Yogyakarta Special Region Province. The fish is dissected, and the kidney and lymph target organs are for RNA isolation. The RNA isolation results were converted into cDNA and then amplified in two stages through the semi-nested RT-PCR method. The detection target is the third segment of the RNA of the TiLV. The first and second amplified amplicons were then electrophorized using agarose gel and observed under UV light. The detection results showed no visible band at the first amplification. In the second amplification, each sample showed positive results with the appearance of bands in the detection target gene. The semi-nested RT-PCR method is proven to increase detection sensitivity. Based on these results, it can be concluded that the semi-nested RT-PCR method in this study is effectively used to detect TiLV. It is necessary to develop research on a field scale to test the effectiveness of this method. This research is expected to provide an alternative detection of aquaculture diseases that are more effective and efficient.

Adolescence is an important period because it is a time when individuals seek self-identity and experience many problems. Depression is one of the biggest psychological disorders in adolescents, especially adolescents who by their keyatiman condition have to live in orphanages. This study aims to determine the relationship of social support with the incidence of depression in adolescents living in the Putri Aisyiyah Orphanage Yogyakarta, with a correlation design and cross-sectional time approach. Data was collected with questionnaires and analyzed with Spearman Rank. The results of this study showed that most adolescents in the Putri Aisyiyah Orphanage Yogyakarta received moderate social support as much as 23 (47.9%) and had a depression level in the category of not mild depression as many as 23 children (47.9%). The results of the Spearman Rank analysis found that the value of significance p = 0.000 means a significant relationship exists between social support and depression in adolescents living in the Putri Aisyiyah Orphanage Yogyakarta. It is therefore advisable for caregivers and orphaned adolescents to be able to increase social support, especially instrumental support both by caregivers and fellow adolescents in meeting material needs (finances and clothing) and support in solving personal problems and schoolwork.

The development of the digital world is accelerating with the presence of the COVID-19 pandemic around the world. The need for digitization is also needed in the health sector, such as EMR (Electronic Medical Records) to facilitate services in recording patient medical data. The Regulation of Physiotherapy Service Standards aims to provide a reference for the implementation of quality and accountable physiotherapy services to improve the quality of physiotherapy services to patients which is one of the technological advances in the industrial revolution 4.0 by providing EMR. With this EMR, it is hoped that it can facilitate physiotherapists' access to medical record data from patients that is neatly structured. EMRs with structured data can also help clinic owners make new decisions based on stored data. This research was conducted by observation, interview, and discussion methods for analysis and design. The results of the discussion and analysis of this study are structured, neat data according to the concept of relational data. 12 Entities generated from structured data and relationships from each entity that connects one to the other consist of 10 relationships. This study resulted in a pilot system for ERM specifically for Physiotherapy Clinics because it has structured and clearly documented data such as database design using ERD (Entity Relational Diagram) and DOD (Data Object Description), as well as system design and display.

Chronic energy deficiency (CED) is a condition of imbalance of energy and protein intake that can lead to pregnancy disorders such as anemia, bleeding, abnormal weight gain, and infection. This study aims to determine the characteristics of pregnant women with CEDs at the Galur II Kulon Progo Health Center. The research design used quantitative descriptive. The sample of this study was all pregnant women with CEDs as many as 49 respondents. The results showed that pregnant women with CEDs had age characteristics of 20-35 years (91.8%); have never given birth (51%); high school education level (61.2%); work as a housewife (67.3%); and did not have anemia (63.3%). This means that the risk of CED in pregnant women can be predicted based on the characteristics of young age, primipara, secondary education level, housewife's occupation, and although anemia does not occur.

Surgery is one of the medical procedures that will give an emotional reaction to the patient. Stressors that appear can be in the form of physiological stress, psychological stress (anxiety and fear) and social stress. The anxiety felt in pre-operative patients is mostly caused by excessive worry about what the patient will face during the surgical procedure. The implementation of pre-operative orientation can provide patients with an understanding of their condition and avoid negative things that arise in patients due to ignorance about the treatment they will receive. Orientation about the Central Surgical Installation room can be one of the interventions to increase patient knowledge so that anxiety can decrease. This research aims to develop educational media in reducing anxiety through audiovisual videos of the orientation of the Central Surgical Installation (IBS) room. The type of research planned is a quasy experiment research with a Pretest-Posttest Control Group Design approach. Sampling using accidental sampling. Data analysis used the Wilcoxon Signed Rank Test and Mann-Whitney Test. The results showed that there was a significant difference in anxiety between the intervention group and the control group after being given an educational intervention with a virtual video tour with a P value of
