Abstract

AbstractThe use of vaginal sponge in estrous synchronization has been a popular method to uniform estrous cycle of small ruminants. In order to increase the litter size, the use of the sponge is combined with PMSG. Therefore the objective of this study was to evaluate the effect of PMSG in combination with vaginal sponge on estrous response and litter size of Javanese local sheep. The study was conducted in Faculty of Animal Husbandry, Padjadjaran University, Indonesia with 32 non-pregnant local sheep. All these sheep was divided into four groups of treatment, i.e: 1) group of sheep was subjected to vaginal sponge (VS) treatment containing 30 mg Medroxyprogesterone acetat (MAP) inserted in the vagina for 14 days; 2) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection -1 d before VS removal; 3) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection at the day of VS removal; 4) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection +1 d after VS removal. The result showed that all treatment groups had 100%  of estrous response. However, the occurrence of estrous time varied among group after VS removal. Group 1 (G1) and G4 tend to have estrous occurrence at day 3 after VS removal by 55% and 75% response, respectively.  Whilst, G2 and G3 tend to have estrous occurrence respectively at day 1 (62,5%) and day 2 (50%) after VS removal. A 100% of pregnancy rate occurs in G2 and G4, while G1 and G3 have a 88,9% and 87,5%, respectively. No significant effects of the treatments on the ewes litter size. In conclusion, the use of PMSG administration 24 h prior, after, or at sponge withdrawal increase a tighter synchrony of oestrous and pregnancy rate, but has no effect on the increasing of litter size in Javanese local sheep.Keywords: Vaginal sponge, PMSG, estrous response, pregnancy rate, litter size, sheep

Abstract

Untuk mengetahui potensi antibakteri dan konsentrasi optimal jeruju (Acanthus ilicifolius), dilakukan uji daya hambat ekstrak dan fraksi daun terhadap pertumbuhan Vibrio harveyi, bakteri penyebab penyakit pada udang. Daun yang sudah dikeringanginkan diekstraksi dengan metanol, sehingga didapatkan ekstrak kasar (crude). Sebagian dari ekstrak kasar dilakukan fraksinasi dengan metode kolom silika gel dengan pelarut n-heksan, etil asetat, n-butanol, etanol, dan metanol. Uji daya hambat dilakukan dengan agar disc diffusion method. Perlakuan yang diberikan adalah ekstrak kasar, fraksi n-heksan, etil asetat, n-butanol, etanol, dan metanol yang masing-masing konsentrasinya 50-1000 ppm, yang diberikan pada kultur V. harveyi pada media Triptic Soy Agar (TSA). Fraksi etil asetat menunjukkan daya hambat terbaik (12 mm), diikuti ekstrak (11,33 mm), dan fraksi n-butanol (11 mm).

Abstract

Vibriosis was a crucial problem and a primer infection causing mortality of humpback grouper.Virulence factor of vibrio, such as vili, is an adhesion molecule protein based on haemaglutination test toblood serum of humpback grouper. This research was aimed to characterize an adhesion molecule of viliprotein of V. alginolyticus on epithelial cells of Humpback grouper as haemaglutinin protein. Themethods are laboratory exploration by isolate epithelial cells of Humpback grouper, characterize a receptorand test vili protein to adhesion molecule that is epithelial cell of Humpback grouper. The result of proteinhaemaglutinin test to vili V. alginolyticus show that an adhesion molecule with a molecule weight of38.98 kDa is an adhesin vili V. alginolyticus and a haemaglutinin. The adhesion pattern of V.alginolyticus on epithelial cells of Humpback grouper exposed to protein haemaglutinin of vili show bothdiffuse and localize pattern. Research conclusion was the haemaglutinin protein of vili V. alginolyticuswas an adhesin having a role to both virulence process of bacteria and attachment process to receptor ofepithelial cells of Humpback grouper.Keywords: adhesion molecule, epithelial cell, vili, V. alginolyticus, C. altivelis

Abstract

Penelitian ini bertujuan mengetahui kejadian penyakit secara patologis pada ayam pedaging di Kabupaten Sukabumi dan Bogor pada bulan Februari (musim penghujan) dan Juni (musim kemarau) tahun 2012. Dari sejumlah peternakan yang dikunjungi total diperoleh 40 kasus penyakit. Diagnosis penyakit ditetapkan berdasarkan sejarah penyakit yang terjadi di peternakan, umur ayam yang sakit, gejala klinis, perubahan patologi anatomis, dan histopatologis. Berbagai faktor seperti kondisi iklim, tata laksana peternakan, pengobatan, dan sejarah vaksinasi digunakan sebagai data penunjang. Hasil penelitian menunjukkan bahwa penyakit yang teridentifikasi adalah colibacillosis (22,2%), asites (12,5%), gumboro (12,5%), Newcastle disease (ND) (10%), Salmonella pullorum (10%), dan necrotic enteritis (7,5%). Penyakit terutama terjadi pada ayam umur 11-21 hari (57,5%) dan terjadi pada musim penghujan (60%).

Abstract

To examine whether Javanese ginseng root has an antihyperglycaemic effect, twenty-one male Rattus norvegicus rats were randomly divided into four groups: negative control (n= 3), positive control (n= 4), treatment (n= 7), and placebo (n= 7). Alloxan was injected intraperitoneally at a single dose of 80 mg/kg BW to rats to induce hyperglycaemia. Infusion of Javanese ginseng root or distilled water was given at a dose of 2% w/v solution with a volume of 1.8 mL/200 g BW per day for 14 days through an enteral feeding tube to either the treatment or placebo group, respectively. Blood glucose levels were measured using the colorimetric method. One-way analysis of variance (ANOVA) was used to examine differences in the mean of delta blood glucose (post intervention minus post alloxan blood glucose levels) among groups, followed by Tukey’s post hoc analysis. A P value 0.05 was considered statistically significant. There was a significant difference (P= 0.0001) in delta blood glucose among groups. Post hoc analysis revealed that delta blood glucose in the treatment group (-102.99±2.26 mg/dL) was significantly (P= 0.0001) greater than in the placebo (2.45±0.29 mg/dL), positive control (3.05±0.70 mg/dL) and negative control (1.60±0.17 mg/dL) grou p. In conclusion, Javanese ginseng root has potential as a hypoglycemic agent in alloxan-induced diabetic animal models

Abstract

The objective of this research was to analyze the composition of saturated fatty acid, unsaturated fatty acids, and cholesterol in meat of Bali bull fattened using forage crops in small holder farming. The experiment was conducted for 3 months using nine Bali bull aged 2.5-3.5 years old with the initial body weight of 227-290 kg in the feedlots of Bero Sembada Farmers Group, Laen Manen Sub District, Malaka Regency, East Nusa Tenggara. The experiment was conducted based on the fattening system of local farmers in terms of feeding, housing, and health management. Feed given during the experiment was Centrosema pubences, Clitoria ternatea, fresh corn straw, king grass, Leucaena leucocepala, natural grass, Pennisteum purpureum, and Sesbania grandiflora. At the end of this study, cattle were slaughtered and the composition of saturated fatty acid, unsaturated fatty acids and cholesterol were measured. Data were analyzed using a descriptive statistic. Results revealed that the total of saturated fatty acid and unsaturated fatty acids in meat were 36.628% and 26.739%, respectively, while the cholesterol reached 79.18 mg/100 g of meat. In conclusion, meat of Bali bull fattened using forage crops in small holder farming has higher saturated fatty acid than unsaturated fatty acids.

Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas  oosit,  maka  oosit  yang  berkualitas  A  dimaturasi  secara  in  vitro. Oosit   hasil  maturasi  dipaparkan  ke  dalam  larutan  vitrifikasi  yang mengandung 30%  EG  + 0,5 M  sukrosa  dengan  lama  waktu paparan  3 menit,  oosit  dimasukkan  ke dalam  ministraw  0,25 cc  (French  straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan  sukrosa 0,5 M.  Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

The purpose of this research was to analyse the quality and storability of native chicken semen at room temperature after diluted with physiological saline solution (NaCl) supplemented with noni fruit extract. Research methodology used was nested randomized design which variables observed covering the quality of chicken spermatozoa macroscopically and microscopically with the doses of noni fruit extract of 0 % (P0), 10 % (P1), 20 % (P2), and 30 % (P3). Rate of spermatozoa motility at 0, 1, 2, and 3 hours on P0 were 83.70±4.74, 63.30±9.63, 32.90±8.52, and 15.00±6.20; P1 were 85.30±5.44, 72.70±10.06, 51.90±11.75, and 28.50±5.68; P2 were 84.60±6.40, 78.50±8.59, 57.90±10.73, and 33.70±9.06; and P3 were 81.80±7.30, 64.20±7.93, 40.30±11.66, and 19.80±7.47, respectively. Rate of spermatozoa viability at 0, 1, 2, and 3 hours on P0 were 85.30±9.09, 65.10±6.15, 32.10±11.86, and 10.30±9.09; P1 wer 87.40±6.22, 72.70±5.33, 50.80±13.59, and 26.50±10.99; P2 were 68.58±5.30, 77.70±4.79, 56.30±13.76, and 32.70±13.79; P3 were 81.20±8.04, 68.70±10.40, 36.20±16.61, and 16.90±11.93, respectively. Rate of spermatozoa abnormality at 0, 1, 2, and 3 hours on P0 were 8.70±3.40, 12.20±4.42, 17.80±5.67, and 20.30±6.38; P1 were 7.80±2.04, 9.80±2.69, 13.60±4.45, and 16.50±5.19; P2 wer 8.40±4.33, 10.00±2.45, 12.50±5.21, and 15.80±3.71; and P3 were 9.60±3.41, 10.90±2.64, 17.10±5.61, and 21.20±8.16, respectively. It can be concluded that the addition of noni fruit extract at the dose of 10 % and 20 % in physiological saline solutin maintain the native chicken spermatozoa quality up to 2 hours post storage at room temperature.

Abstract

Penelitian ini bertujuan mengetahui efek minyak jintan hitam (Nigella sativa L.) terhadap spermatogenesis tikus Wistar yang terpapar asap rokok. Jenis penelitian adalah eksperimental murni dengan subyek 24 ekor tikus yang terpapar asap rokok selama 21 hari dan dirandomisasi dalam 4 kelompok yaitu kelompok kontrol dan kelompok perlakuan dengan pemberian minyak jintan masing-masing 0,1; 0,2; dan 0,3 ml. Penegakan hipotesis dilakukan dengan menggunakan Krusskal-Wallis dan Mann Whitney dengan batas derajat kemaknaan P≤0,05, interval kepercayaan 95%. Pemberian minyak jintan hitam dapat memperbaiki derajat spermatogenesis.

Abstract

This research aims to evaluate the safety of ethanolic extract of Acalypha hispida (A. hispida) leaves with acute toxicity test using 15 female rats strain Sprague-Dawley. A single dose of different doses of extract (2, 4, 8, and 16 g/kg body weight) was administrated orally, and theobservation was conducted for 14 days. The results revealed that the ethanolic extract of A. hispida leaves was relatively harmless (LD50 16 g/kg BW), did not affect body weight, and did not show clinical signs of toxicity during the observation periods. The parameters of blood serumbiochemistry of all extract-treated groups (alanine aminotransferase, aspartate aminotransferase, creatinine, and urea) did not change significantly  compared to the control group. The histological observation of the liver showed a significant increase in eosinophilic cytoplasm and basophilic nuclei at all doses. However, the ethanolic extract of A. hispida leaves did not significantly affect glomerulus/Bowman’s capsule ratio, glomerular cell density, and the proportion of normal cell tubule. In conclusion, the ethanolic extract of A. hispida leaves was relatively harmless with LD5016 g/kg BW and seems to be safe in low doses (2 g/kg BW).
