Abstract

ABSTRACTThe objective of this research was to evaluate the quality of chilled canine semen in Tris-egg yolk (TEY) extenders containing different concentrations of sericin. Semen were collected from four dogs by massage method. Canine semen was collected using sterile urine pots and evaluated. Sperm-rich fractions were pooled and divided into four equal aliquots, which were then diluted with TEY extenders supplemented with different concentrations of sericin (0%, 0.1%, 0.25%, 0.5%). The diluted semen aliquots were preserved at 4 ℃ in sterile centrifuge tubes and were then evaluated for spermatozoa motility, viability, plasma membrane integrity and acrosome integrity every 12 hours up to 72 h. The TEY extenders supplemented with 0.25% and 0.5% sericin resulted in higher spermatozoa motility and viability at 72 h compared to other TEY extenders (P0.05). The integrity of plasma membrane and acrosome of spermatozoa showed no significant differences among the groups extenders at 72 h. In conclusion, sericin in concentration of 0.25% and 0.5% were able to prevent the motility and viability of canine spermatozoa after storage for 72 h.

Abstract

The present study examines anatomy of Gayo mare reproductive organs. This study used three sample of Gayo mare reproductive organs (n= 3) for observation of morphology and morphometric of the mare reproductive organs. The ovarium was fixed in 4% paraformaldehyde solution then followed by histological method and stained using hematoxylin and eosin (HE) and Masson’s trichome (MT). Three mares were observed for diameter and changes overview of uterus during estrous cycle in real time using ultrasound. The results showed that, in general, the anatomy of Gayo mare’s reproductive organs was similar to other mares, but smaller in morphometry. The total length of the Gayo mare’s reproductive tract from labia to apex cornua was 48.00±1.00 cm. Weight of Gayo mare’s left ovary was 19.07±7.70 g and the right was 24.43±0.83 g. Histologically, there was no difference between Gayo mare’s structure and other mares. In cortex uteri there were some follicles surrounded by capillary, various development stages of follicles, healthy follicles, atretic follicle, and corpus albican; while in medulla there were a lot of connective tissues. Ultrasound of the uterus showed the change in diameter during estrous cycle with the largest diameter of corpus uteri was 4.43±0.10 cm in horses with estrous cycle of 21 days and 6.30±0.93 cm in horses with 24 days estrous cycle. In conclusion, the morphometry of Gayo mare reproductive organs are smaller than the other horses and there are differences in diameter of the uterus during the estrous cycle due to the changes of endometrium thickness.

Abstract

The Calophyllum genus has been reported to be rich in bioactive compounds that have medicinal properties. The author will also report the results of the isolation of compounds obtained from the ethyl acetate fraction of the bark of the plant C. macrophyllum Scheff, a type of Calophyllum found in Mount Kerinci, Jambi province. This paper will analyze the spectroscopic data of these compounds which include UV, IR, GC-MS, 1HNMR and 13CNMR spectra which are amplified by HMQC and HMBC spectra. The first step in bioactivity testing is the toxicity test using the BSLT (Brine Shrimp Lethality Test) method using Artemia Salina Leach shrimp larvae. The results of the spectroscopic analysis obtained the chemical formula as a derivative of fatty acid methyl ester (methyl-octadec-14 enoic), in the form of a yellow liquid with a boiling point of 173-175°C. The results of the initial screening for bioactivity through the toxicity test of the BSLT method on Artemia salina Leach shrimp larvae obtained an LC50 value of 141.2 g/mL. This compound still has the potential to have certain bioactivity

Abstract

An experiment was carried out to evaluate the effect of graded levels of cassava leaf meal (CLM) with varying cyanide content on growth performance of African catfish (Clariasgariepinus) fry. There were four groups of fish, each comprising 12 fish per group. One groupwas fed on a control diet (0 mg HCN/kg diet) with fish meal (FM) and soybean meal (SBM) asprotein source (30% CP). Groups two, three and four were fed graded levels of CLM to replace10, 20, and 30% of the total diet at the expense of SBM which contains 4.19, 7.47 and 11.96 mgHCN/ kg diet respectively. After 12 weeks, fish fed with increasing level of CLM showed asignificant growth depression and poor feed utilization compared to those fed the control diet.Apparent dry matter digestibility (ADMD) and apparent protein digestibility (APD) were alsosignificantly (P0.05) affected by cassava leaf meal inclusion, with the best values were found infish fed the control diet (52.26+0.24% and 83.11+1.17% for ADMD and APD respectively). Bodycomposition of fish fed higher CLM tended to have less protein and fat but more ash. The poormethionine availability as a result of low protein digestibility and the presence cyanide contentlimits the use of CLM in African catfish fry diet. Keywords: African catfish, cassava, growth, cyanide
