Potential antioxidant activity of Lactobacillus fermentum KF7 from virgin coconut oil products in Padang, West Sumatra, Indonesia. Biodiversitas 23: 1628-1634. The isolation and characterization of five commercial Virgin Coconut Oil (VCO) products circulating in the local market of Padang city were conducted. In this study, eighteen isolates of lactic acid bacteria (LAB) were isolated and characterized microscopically, and biochemically. VCO-LAB isolates were Gram-positive bacilli and cocci, catalase-negative, and homofermentative (except VD was heterofermentative). There were three isolates of VCO-LAB (VB.3, VD and VE.4) that could produce gamma-aminobutyric acid (GABA). The highest concentration of GABA (19.5 mg/mL) was produced by isolate VB.3 after 72 hours of incubation and the addition of 7% MSG. VB.3 isolate shows the highest antioxidant activity with the addition of Monosodium glutamate (MSG), 68.13% for 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) ABTS and 88.02%. In addition, GABA-producing VCO-LAB could be grown in media containing 0.2-1.0 mM hydrogen peroxide, with the lowest optical density value of 0.82 nm. The antioxidant activity of VCO-LAB was affected by the GABA produced and by secondary metabolites such as peptides and antioxidant enzymes (NADH-oxidase, superoxide dismutase, NADH peroxidase and non-heme catalase). The highest antioxidant production was obtained in isolate VB.3 or Lactobacillus fermentum KF7.

The abundance of genetic diversity, especially local upland rice cultivars, makes an excellent opportunity to obtain local cultivars with the desirable character. The study aimed to assess the genetic variability and heritability of agronomic traits among local upland rice cultivars from West Sumatra. In this study, 28 local upland cultivars from West Sumatra were subjected to the analysis of variance. The research was conducted from November 2020 to April 2021 at the Research Station of the Faculty of Agriculture, Andalas University, Padang. The evaluation was conducted using a Randomized Complete Block Design with three replicates. Results showed variation in each agronomic trait, ranging from intermediate to tall in plant height, short to moderate in panicle length, low to moderate in the number of tillers, days to flowering and days to maturity, and light to moderate in grain yield. Plant height, number of tillers, days to flowering, and days to maturity were traits with high to moderately high GCV values and high heritability values. The number of tillers and days to maturity could be chosen as selection criteria for assembling superior varieties. Both traits performed high heritability estimates and significant genotypic correlation with grain yield.

Abstract. Rahma H, Nurbailis, Busniah M, Kristina N, Larasati Y. 2021. The potential of endophytic bacteria to suppress bacterial leaf blight in rice plants. Biodiversitas 23: 775-782. Endophytic bacteria are potential as biocontrol agents against bacterial leaf blight (BLB) disease caused by Xanthomonas oryzae pv. oryzae in rice to support sustainable agriculture. This study aimed to select and characterize 22 endophytic bacteria isolated from healthy rice, determine their ability to promote plant growth and suppress bacterial leaf blight disease in rice and also identify potential endophytic bacterial isolates. The study was arranged in a Completely Randomized Design with 24 treatments and three repetitions. The treatments used in the current study consisted of Xanthomonas oryzae infected plants and treated with endophytic bacterial isolates; infected plants without endophytic bacteria treatment (positive control), non-infected plants (negative control). Identification of potential endophytic bacteria was performed based on 16S rRNA sequences. Three out of 22 bacterial isolates, i.e., LmB1, LmA6, and LmB2 were able to suppress bacterial leaf blight disease with severity levels of 35.82%, 23.78%, and 23.78%, respectively. Based on the rice plant growth parameters, three bacterial isolates (LmA6, LmB1, and LmB35) were able to increase the growth of rice plants with an average value of 69.56%, 56.51%, and 47.82%, respectively. Two bacterial isolates, i.e., LmB 1 and LmA6 suppress the development of bacterial leaf blight disease and increase the growth of rice plants. Based on DNA sequence comparisons of DNA fragments amplified by 16S rRNA related marker of the selected bacterial isolates and database, then LmA6, LmB2, LmB1, and LmB35 had similarities with Bacillus cereus MD152 (96.87%), Bacillus thuringiensis ATCC 10792 (98.20%), Ochrobactrum intermedium strain OI1 (97.52%), and Stenotrophomonas maltophilia strain A1w2 (97.92%), respectively. Our study revealed that the indigenous endophytic bacteria from rice plants could be potential biological agents for controlling bacterial leaf blight disease and increasing plant growth.

Morpho-anatomical characterizationand DNA barcoding of Achillea millefolium L. Biodiversitas 23: 1958-1969. One of the important things to study the distribution of secondary metabolites in the plant body is to carry out the identification process. Morphological markers have several limitations to recognize plants, therefore supporting data is needed so that the information becomes more comprehensive. This study aims to identify Achillea millefolium L. based on morphological, anatomical, and DNA barcoding markers to obtain specific data and avoid confusion. Morphological studies were carried out descriptively using vegetative organs, while anatomical studies of rhizomes, stems, and leaves used the paraffin method. The DNA barcoding was performed by analyzinggenes from 3 different individuals. The research was carried out by amplifying and sequencing the partial gene in the ribulose-bisphosphate carboxylaselarge subunit (rbcL) regions and maturase-K (matK). The results showed that the plant had taproots, short stems due to root rosette and the leaves were double compound. The rhizome and stem tissues had almost the same structure, while the leaves had a tissue arrangement that was similar to other plants in general, except that the mesophyll tissue was undifferentiated. The results of DNA barcoding showed a percentage of identity above 98% for both the rbcLand matK genes.

Abstract. Hervani D, Sari SP. 2022. The abundance of arthropods and natural enemies on two growth phases of hybrid corn, Solok District, Indonesia. Biodiversitas 23: 2966-2972. Solok District, West Sumatra, Indonesia is one of Indonesia's hybrid corn production regions. As yet, the study of arthropods is still limited. This study aimed to study the abundance and diversity of arthropods in the hybrid maize agroecosystem in Solok District, West Sumatra. The research was conducted from May to November 2020 in Tanah Garam Sub-district, Solok District. The sampling was carried out four times in two growth phases of hybrid corn: vegetative (the age of 20 and 40 days after planting/DAP) and the generative phase (the age of 60 and 80 DAP). The arthropod collection was conducted using a yellow trap and a modified vacuum technique. The identification process was carried out at the Insect Bioecology Laboratory, Andalas University. The study found two classes of arthropods in a hybrid cornfield in Solok District, namely Arachnida and Insecta. The Shannon-Weiner diversity index value (H') was 1.383, and the evenness index of species (J') was 0.997. A total of  2,927 individual arthropods, 80 families, 8 orders,  and 3 classes, were recorded in the hybrid corn ecosystem in Solok District. Insect class (2,866 individuals) dominated the diversity of arthropods, where the majority of the arthropods found belong to groups of natural enemies such as predators and parasitoids. Conservation of natural enemies (predators and parasitoids) is one of the main factors in supporting the success of pests biologically, so the act of identifying natural enemies in an area is very important.

Less. is a member of the Asteraceae family and is popular as a medicinal plant. It is very important to authenticate plants using various markers to confirm the labeling of plants and to reveal the bioactive compounds for pharmaceutical research. This study aims to characterize the morpho-anatomical and DNA barcoding aspects of P. indica. Three samples obtained from Taman Husada Graha Famili, Surabaya, Indonesia were used in this study. Morphological studies were carried out descriptively, while the paraffin method was used for anatomical observations. DNA barcoding analysis was performed by amplifying and aligning the rbcLand matKgenes. Morphologically, P. indicahas organs like Asteraceae plants in general, with specific characteristics in leaves and flowers. P. indica has an anatomical organ similar to that of Asteraceae plants globally, only that there are specific histological features on its leaves, including anomocytic stomata, glandular cells, and multicellular glandular trichomes. Alignment and reconstruction of the phylogenetic tree showed that the sample plants were closely related to P. indica in the GenBank database. Based on morpho-anatomical characters and DNA barcodes, the plant under study was confirmed as P. indica L. (Less).

Arthropods can be found on the surface of the soil and plant canopy. West Pasaman District is one of the centers of corn production in Indonesia. This study aimed to determine the abundance and the diversity of arthropods in Luhak Nan Duo Sub-district and Kinali Sub-district, West Pasaman District, West Sumatra District, Indonesia. The research was conducted using a purposive sampling technique from October to December 2020. The arthropods sampling was done by two techniques: yellow trap and modified vacuum technique. The identification process was conducted at the Insect Bioecology Laboratory, Faculty of Agriculture, Universitas Andalas, Padang, Indonesia. A total of 5477 arthropods were found in maize plantations in West Pasaman. The arthropods found were classified into two classes, 8 orders, 85 families, and 269 morphospecies. The individual number was dominated by the Insect class, reaching 97.09% (5318), while the Arachnida class was only 2.99% (159). Data were analyzed for the number of species (S), the number of individuals (N), Shannon-Wiener diversity index (H'), and Evenness index of species (J'). Regarding the location, Luhak Nan Duo has a higher number of individuals, Shannon-Wiener diversity index, and Evenness index than Kinali. However, the pattern of arthropod abundance in the two maize cultivation locations in West Pasaman was similar, increasing with the age of the maize plant. However, in general, the diversity values of arthropods in maize cultivation in Luhak Nan Duo and Kinali were more than 3. It means that the West Pasaman is still quite balanced and safe from the risk of damage due to pests because of the highest abundance of arthropods on maize plantations.

Abstract. Permata DA, Susandarini R. 2021. Morphological diversity and phenetic relationship of wild and cultivated Begonia based on morphology and leaf venation. Biodiversitas 23: 928-937. Begonia is an ornamental plant with intense cultivation and hybridization practices because of the uniqueness and beauty of its leaves. Begonia with a very large number of species is interesting to study, especially because Indonesia is one of the resource centers of this genus diversity as indicated by the increasing number of new species found every year. This study aimed to explore the morphological diversity of wild and cultivated Begonia and reveal the phenetic relationships of the species and hybrids based on morphology and leaf venation patterns. In this study, we used 19 Begonia samples belonging to five species collected from their natural habitat and 14 species and hybrids obtained from the nurseries. Morphological observations of living specimens and examination on microscopic slides of cleared leaf resulted in 28 characters which were used to determine phenetic relationships using cluster analysis. The high variability of leaf venation patterns described in this study disclosed the micromorphological diversity of Begonia which has not been widely exposed. The results of morphological observations showed high variabilities in leaf color, leaf shape, and petiole color. The results of cluster analysis and principal component analysis indicated that the grouping of species was mainly determined by the existence of trichomes on leaf surfaces and petiole, leaf venation pattern, leaf variegation, and leaf shape. The grouping of species resulting from cluster analysis can be used in the selection of potential parents to produce new Begonia hybrids through crossing experiments.

The preservation of local rice is essential to preventing genetic drain. In Deli Serdang District, North Sumatra, Indonesia, local rice is in great abundance. A total of 23 Deli Serdang local rice were collected and studied from coastal to highland areas. This study aims to determine the morphological characteristics of 23 local rice genotypes of Deli Serdang and to obtain superior characters to be developed into superior local varieties. A completely randomized design with three replicates was used in this study. Morphological characteristics of 23 local rice genotypes obtained various characters. The culm length was short to very long; culm diameter was thin to thick; the leaf-blade length was long to very long; leaf-blade width was intermediate to broad; culm number was low to high. The auricle, ligule, and collar were almost completely whitish, except for Sijambi yellowish-green and green, 2-cleft acute ligule shape. The harvest age was medium to deep. The grains were white, straw, gold, brown, brown spots, brown lines, and black. The pericarps were white, light brown, red, and black. Most of the organs indicated broad genetic variability having the opportunity to be developed into superior local varieties. All local rice was scattered into five groups based on morphological markers with a similarity of 32.12%.

Abstract. Hayati PKD, Witari S, Rozen N, Sutoyo, Widiarsih S. 2022. Effect of gamma irradiation on the germination, pollen viability, and morpho-agronomic of Pachyrhizus erosus cv. Kota Padang. Biodiversitas 23: 1231-1238. Yam bean or Pachyrhizus erosus (L.) Urb., an unexploited plant that produces the storage root tuber, is used in a variety scale of food processing and cosmetic industries in Indonesia. Novel approaches to induce variability in yam bean cv. Kota Padang will accelerate the breeding efforts and improvement of the cultivar. The purpose of the study was to elucidate the effects of gamma irradiation on seed germinability, pollen viability, and morpho-agronomic performance of yam bean cv. Kota Padang. Seeds were irradiated with gamma-rays on 0, 100, 150, 200, 300, 400, and 500 Gy, then LD50 was determined based on the germination percentage. Observation of the irradiated yam bean growth in the field was carried out individually. Results showed that gamma irradiation decreased seed germinability, pollen viability, and agronomic traits. Still, it increased the variability of the traits among the mutant populations, except for the number of branches. Correlation analysis confirmed the results, indicating that selection in the next M2 generation must consider semi-dwarf and pod yield performances. The LD50 was 150 and 176 Gy; hence, the irradiation doses of 150 and 200 Gy were appropriate in producing high genetic diversity with low physical damage in yam bean cv. Kota Padang.
